Pulmonary neuroendocrine tumours – different biological entities?
Background: Lung cancer is the leading cause of cancer-related deaths worldwide, accounting for more than 40,000 deaths each year in Germany. Twenty-five percent of all lung tumours belong to the group of neuroendocrine tumours, encompassing typical (TC) and atypical carcinoids (AC), large-cell neuroendocrine carcinoma (LCNEC) and small-cell lung cancer (SCLC). Even though all four subgroups share some morphological and molecular properties, it is still unclear whether they are different biological entities or form one distinct tumour entity displaying different grades of differentiation. They were previously reported to arise from serotonin producing pulmonary neuroendocrine cells (PNEC). It is suggested that LCNEC and SCLC originate from the same or similar precursor cells. However, it remains unclear how carcinoids can be integrated into this scheme.
Material and Methods: In order to address this question, 74 representative tumour specimens were used for sequencing analysis performed on a MiSeq instrument covering the 221 most important mutation hotspots related to human neoplasias. Additionally, mRNA-expression profiles of 80 tumour samples were determined for 91 selected genes using NanoString nCounter technology. Furthermore, three unequivocal samples of each tumour entity were chosen to be tested for their miRNA signature via 384 well TaqMan low-density array real-time qPCR for the expression of 768 unique miRNAs. Finally, the pulmonary neuroendocrine tumour samples were analysed via TaqMan qPCR and immunohistochemistry for expression of the five most important subunits of the 26S proteasome.
Results: 44 miRNAs were identified which showed a significantly different miRNA expression between the subtypes of pulmonary neuroendocrine tumours. For 12 miRNAs, the difference was highly significant (p<0.01). Eight of these were negatively correlated with the grade of tumour biology, the other four were positively correlated. Six miRNAs are significantly associated with survival. The mRNA expression analysis showed 48 significant correlations with tumour type. Sixteen most significant (p≤0.001), 20 highly significant (p≤0.01) and 12 significant (p≤0.05) candidates were identified. Nine significant correlations with progression-free survival were found. Overall survival correlated significantly with nine genes. Of the 643 variants that passed the applied filter processing as described above, 122 variants were related to TC, 150 to AC, 164 to LCNEC and the final 207 variants were detected in the SCLC samples. We found a number of molecular features associated with pulmonary neuroendocrine tumours.
Conclusion: In summary, we hypothesise that there is adequate evidence that neuroendocrine tumours should be considered an overall biological family of neoplasms. The family of pulmonary neuroendocrine tumours can be biologically well distinguished from all other forms of lung cancers. Additionally, better biological understanding of carcinoids and neuroendocrine lung carcinomas will increase the chance of finding new drug targets, maybe resulting in a more personalized therapeutic concept for this group of malignancies
Biomechanical and motor performance differences between female and male soccer players - analysis of gender-specific game patterns, kicking performance and shoe development
Im Vergleich zum Männerfußball fand der Frauenfußball in der bisherigen Forschung deutlich weniger Beachtung. Das allgemein zunehmende Interesse und die steigende Anzahl fußballspielender Frauen verdeutlichen jedoch die Notwendigkeit, sich auch auf sportwissenschaftlicher Ebene mit dem Frauenfußball ausführlicher zu beschäftigen und so bestehende Forschungslücken zu schließen. Dazu leistet diese Arbeit einen Beitrag, indem sie durchgeführte Studien zum Spielverhalten und zur Ballbehandlung von weiblichen und männlichen Fußballspieler/innen sowie zur geschlechtsspezifischen Fußballschuhkonstruktion vorstellt.
Zunächst werden mithilfe einer systematischen Spielanalyse der Frauen-Fußballweltmeisterschaften 2003 und 2011 und der Männer-Fußballweltmeisterschaft 2002 geschlechtsspezifische Unterschiede in der Spielweise sowie Entwicklungen im Frauenfußball aufgezeigt. Auf der Grundlage dieser Ergebnisse, werden weiterhin die Schussgenauigkeit und die bei Genauigkeitsschüssen verwendeten Schusstechniken analysiert.
Im zweiten Teil der Arbeit liegt der Fokus auf die Fußballschuhkonstruktion. Hierfür wird der geschlechtsspezifische Einfluss des Fußballschuhs auf die Laufzeiten in einem Parcours und auf die plantare Druckverteilung erörtert. Abschließend wird mithilfe einer Fragebogenanalyse untersucht, ob sich weibliche und männliche Fußballspieler/innen hinsichtlich ihrer Anforderungen an die Fußballschuhkonstruktion unterscheiden.
Die Ergebnisse der Spielanalyse zeigen, dass das Spiel der Männer durch eine höhere Frequenz von Aktionen mit dem Ball gekennzeichnet ist. Die Entwicklung im Frauenfußball geht hin zu einem vermehrten und variableren Einsatz des Kurzpassspiels. Insgesamt spielen beide Geschlechter den Ball am häufigsten mit der Innenseite. Jedoch existieren geschlechterabhängige Unterschiede in der Verwendung der Techniken für dieselbe Aktion mit Ball: Fußballspielerinnen verwenden für lange Pässe häufiger den Innenspann als Fußballspieler.
Die Schussgenauigkeitsstudien zeigen, dass die weiblichen Probandinnen aus allen Entfernungen ungenauer und seltener mit einem stabilen Fußgelenk schießen als die männlichen. Weiterhin wechseln sie bereits bei geringeren Entfernungen ihre Schusstechnik von der Innenseite hin zum Innen- und Vollspann. Die Männer können dagegen – aufgrund ihrer größeren Kraftfähigkeiten – größere Entfernungen mit dem Innenseitstoß überbrücken, welche eine höhere Genauigkeit ermöglicht.
Eine Untersuchung des Einflusses des Fußballschuhs auf die Laufzeiten und auf die plantare Druckverteilung zeigt keinen statistisch signifikanten Interaktionseffekt von Geschlecht und Schuhbedingung.
Hinsichtlich der präferierten Fußballschuheigenschaften geben weibliche und männliche Fußballspieler/innen eine ähnliche Reihenfolge an: Bequemlichkeit, Ballgefühl, Halt im Schuh und Stabilität auf dem Untergrund sind die wichtigsten Eigenschaften. Jedoch unterscheiden sich die Geschlechter bezüglich der Gewichtung einzelner Eigenschaften: Für weibliche Spielerinnen sind die Bequemlichkeit und der Halt im Schuh wichtiger als für die männlichen Spieler. Der Schutz vor Verletzungen ist zwar für die Frauen ebenfalls wichtiger, er wird jedoch von beiden Geschlechtern als relativ unwichtig beurteilt. Bezüglich der Änderungswünsche aktueller Schuhmodelle wünschen sich mehr als ein Drittel der weiblichen Spielerinnen eine schmalere Passform und knapp ein Viertel längere Stollen.
Anhand der durchgeführten Studien kann die Trainingsempfehlung gegeben werden, dass weibliche Spielerinnen im Techniktraining den Fokus auf den Innenspannstoß legen sollten, da sie diese Technik vermehrt einsetzen. Außerdem wirkt sich vermutlich eine funktionale Kräftigung der fußgelenksstabilisierenden Muskulatur positiv auf die Schussgenauigkeit und auf die -geschwindigkeit aus. Hinsichtlich der Fußballschuhkonstruktion sollte dem Wunsch der Frauen nach längeren Stollen nicht unreflektiert nachgegangen werden, um das Risiko einer Kniegelenksverletzung nicht weiter zu erhöhen. Fußballschuhe für Frauen sollten jedoch auf die weibliche Fußmorphologie angepasst werden und insbesondere eine schmalere Passform besitzen.Introduction
Women’s soccer has become increasingly popular in recent years. The international soccer association FIFA reported a 37 % increase of female soccer players from 21,9 million in the year 2000 to 30 million in 2014 (FIFA, 2007; FIFA, 2015). Nevertheless, previous research mostly addressed men’s soccer, disregarding gender issues.
This paper aims to identify differences between female and male soccer players in view of their playing behaviour, kicking techniques and kicking performance. Furthermore, it focuses on the demands of gender-specific soccer shoe development.
Methods
Four different studies were conducted to gain a better understanding of female and male soccer:
1) Game analysis of the women’s World Cups 2003, 2011 and the men’s World Cup 2002
A video analysis was performed of the finals, semi-finals and quarterfinals of the women’s World Cup in 2003 (USA) and 2011 (Germany) and the men’s World Cup in 2002 (Korea/Japan). Every individual action involving the ball or close to the ball was categorised and located on the playing field. Furthermore, the employed kicking techniques for ball controls, passes and shots were recorded.
2) Kicking accuracy of female and male soccer players
Two kicking studies were carried out to analyse kicking accuracy, velocity-accuracy trade-off and kicking techniques of female and male recreational soccer players. First, subjects were asked to kick a ball at an electronic target from a distance of eight and eleven metres on artificial turf in a laboratory. Kicking accuracy, velocity and employed techniques were recorded. The second study took place on natural turf. Subjects performed kicks from six different distances ranging from eight to 23 metres. A soccer goal was divided into 15 areas to measure kicking accuracy; again the kicking techniques were documented.
3) Effect of gender and shoe condition on running time and plantar pressure distribution
Recreational female and male soccer players ran through a soccer-specific traction course. The course included straight accelerations and decelerations as well as cutting and turning movements. Subjects performed four runs, each with one of four different shoe conditions in a randomized order. Furthermore, pressure patterns under the foot were recorded while wearing the four shoe conditions during walking and running.
4) Gender-specific soccer shoe development
A questionnaire was distributed to non- and semi-professional female and male soccer players. They were requested to rank different shoe characteristics from most important to least important. Moreover, they were asked about their own soccer shoes, potential problems with these and whether they would change current shoe models.
Results
1) The number of actions with ball per minute of net playing time increased in the women’s game from 9,9 in year 2003 to 10,4 in 2011. Men (2002) performed 12,3 actions with ball per minute. These actions with ball consisted predominantly of short passes and ball controls (Women 2011: 82,1 %; Women 2003: 73,7 %; Men 2002: 81,5 %). Women and men most frequently controlled, passed and kicked the ball with the inside of the foot (Women 2011: 65 %; Women 2003: 52 %; Men 2002: 60 %). Regarding long passes, women more often used the instep of the foot (Women 2011: 77 %; Women 2003: 73 %; Men 2002: 66 %).
2) Compared to men, women kicked less precisely and had more ankle movement during the ball contact. No gender differences were found concerning absolute kicking velocity at a distance of eight metres. However, while men increased kicking velocity as the distance increased, women still kicked with the same velocity and also kicked at a much higher relative velocity (= based on individual maximum kicking velocity). Female and male soccer players most frequently used the inside of the foot to kick the ball from distances up to 14 metres. At 17 metres or more, women changed their kicking technique and most frequently used the instep, while men still very often used the inside of the foot.
3) The shoe condition did not influence running times of the functional traction course. In general, elapsed times of female subjects were about 10 % higher than those of male subjects. Plantar pressure distribution was affected by shoe condition but not by gender. Notably, pressure distribution did not mirror different stud configurations of the shoe models.
4) Comfort was ranked as most important shoe feature by both genders. Ball sensing (touch of the ball), stability (less movement between foot and shoe) and traction (less movement between shoe and ground) were also desirable shoe features. Comfort and stability were even more important for female players. Both features are closely related to shoe fit and last design. Injury protection was seen as quite unimportant by all soccer players, although it was more important for female than male players.
About 50 % of the female players would like to change shoe fit in the mid- and forefoot area; most of them would prefer a narrower shoe fit. Just as many male players required changes, but half of them favoured wider and the other half narrower shoes.
Conclusion
The game analysis as well as the kicking accuracy studies showed gender differences in kicking the ball. Probably due to less muscular strength, female players far more often used the instep of the foot whereas male players mostly kicked the ball with the inside. Therefore, female players should particularly practice this kind of kicking. Furthermore, they could improve kicking accuracy by strengthening muscles which cause ankle movements to reduce joint movement during the ball contact. A soccer-specific shoe construction should be based on the gender-specific foot morphology to improve shoe fit. Numerous female soccer players are not satisfied with current shoe models: They would prefer a narrower shoe width which better matches female feet.

Fortschritte in der automatisierten Probenvorbereitung für die Gaschromatographie: Festphasen-Mikroextraktion, Dampfraum-Analyse, Festphasenextraktion
Despite the widespread use of more and more effective detection technologies such as tandem-mass spectrometry, it is becoming increasingly clear that proper sample preparation
is still the key to a successful analysis. The most important demands are thereby complete automation, sensitivity, reliability, economic efficiency and also implementation of “green” technologies that conserve our environment, e.g., by avoiding the use of larger volumes of organic solvents. Modern microextraction techniques can fulfill these demands and are already capable of replacing many traditional, manual sample preparation approaches. This
thesis presents advances in the field of microextraction technologies. The primary scope is thereby the development and evaluation of a novel solid-phase microextraction device (SPME). PAL (prep and load) SPME Arrow retains all advantages of the classical SPME
fiber and augments this proven concept with improved mechanical reliability and sorption phase volume. Initial evaluation of the method shows significantly increased extraction yields
and a potential to further advance the field of SPME. Polycyclic aromatic hydrocarbons (PAHs) could be analyzed from the freely dissolved fraction in water with detection limits
that are at least one order of magnitude better as in case of the classical SPME fibers. In the field of headspace analysis (HS), a systematic comparison between available techniques has
been carried out using 41 volatile analytes. The results clearly show how enrichment techniques such as in-tube extraction (ITEX), SPME or PAL SPME Arrow deliver better sensitivity than static headspace. The instrumental mode of action is thereby of larger
influence to method sensitivity and reproducibility than its type of automation. This is demonstrated by correlating the results of a loop and trap sampler to those generated with the syringe-based static headspace option and various enrichment methods. Sorption phase material selection had the smallest influence in this context and the beneficial potential of special sorption phases seems to be limited to the analysis of compounds capable of specific
molecular interactions. As for the classical solid phase extraction (SPE) the novel ITSP (instrument-top sample preparation) option in terms of miniaturized cartridges (MSPE) has
been evaluated. By using these, the complete sample preparation in terms of extraction, elution and further refinement such as evaporation and derivatization can be carried out by
PAL-type samplers. First results indicate that this option has a lot of potential but still requires further improvement, e.g., in terms of time efficiency.Trotz der Nutzung immer leistungsfähigerer Detektionstechniken wie der Tandem-Massenspektrometrie, wird stetig klarer, dass gute Probenvorbereitung nach wie vor der Schlüssel zu einer erfolgreichen Analytik ist. Die wichtigsten Anforderungen sind dabei vollständige Automatisierbarkeit, Empfindlichkeit, Zuverlässigkeit, ökonomische Effizienz und der Einsatz „grüner“ Technologien welche die Umwelt schonen, z.B. durch den Verzicht
auf größere Mengen organischer Lösungsmittel. Moderne Mikroextraktionstechniken können
diese Anforderungen erfüllen und dabei bereits zahlreiche, klassische, manuelle Probenvorbereitungstechniken ersetzen. Diese Arbeit präsentiert neue Entwicklungen im Feld der Mikroextraktionstechniken. Der primäre Fokus lag dabei auf der Entwicklung und Evaluation einer neuartigen Festphasen-Mikroextraktionstechnik (SPME). PAL (prep and
load) SPME Arrow behält die Vorteile der klassischen SPME-Faser bei und wertet diese um bessere mechanische Zuverlässigkeit und größere Sorptionsphasenvolumina auf. Erste Evaluationen dieser neuen Technik zeigen erhebliche Zugewinne an Extraktionsausbeute, sowie Potential das generelle Feld der SPME weiter zu entwickeln. Dabei konnten polyzyklische, aromatische Kohlenwasserstoffe (PAKs) aus der frei im Wasser gelösten
Fraktion bestimmt werden, wobei Nachweisgrenzen erreicht wurden die mindestens eine Größenordnung besser waren als im Falle der klassischen SPME-Faser. Im Umfeld der
Headspace-Analytik (HS) wurde anhand von 41 volatilen Analyten ein systematischer Vergleich von sechs Probenvorbereitungsoptionen durchgeführt. Dabei wurde klar, dass Anreicherungsverfahren wie In-Tube Extraction (ITEX), SPME und PAL SPME Arrow im Hinblick auf die Empfindlichkeit den statischen Headspaceverfahren überlegen sind. Das
Operationsprinzip ist dabei klar wichtiger als die Art seiner Automatisierung, was durch Gegenüberstellung eines Loop- und Trap-Samplers mit einer spritzenbasierten, statischen
Headspace Option sowie verschiedenen Anreicherungsoptionen gezeigt werden konnte. Die Auswahl unterschiedlicher Materialien als Sorptionsphase hatte dabei klar den geringsten
Einfluss und scheint eher für Analyten wichtig zu sein, welche zu spezifischen, molekularen Interaktionen fähig sind. Für die klassische Festphasenextraktion (SPE) wurde die neuartige
ITSP (instrument-top sample preparation) Option Evaluiert. Mit diesen miniaturisierten Kartuschen lässt sich der komplette SPE-Ablauf samt einer Einengung und Derivatisierung
komplett auf einem PAL-Sampler abbilden. Erste Resultate zeigen neben dem großen Potential dieser Option, noch bestehenden Bedarf an Optimierung z.B. bzgl. der zeitlichen
Effizienz
Functional diversity with asymmetrically located comparison and its use for steering angle acquisition
Elektronik und Dutzende elektronische Steuereinheiten (ECUs) dominieren mittlerweile das Automobil und alle seine Funktionen. Ein Lenkwinkelsensormodul stellt beispielsweise verschiedensten Fahrzeugfunktionen die aktuelle Fahrtrichtung bereit. Fehlerbedingte Ausgabe falscher Winkel führt in einer verknüpften Assistenzfunktion mit eigenständiger Beeinflussung der Quer- und Längsdynamik des Fahrzeugs zu einem unvertretbaren Gefahrenrisiko. Zur Risikominderung werden sich bei Versagen gefährlich auswirkende Funktionalitäten gemäß der Norm ISO 26262 entwickelt. Dazu werden in dieser Norm unter anderem ein geeignetes Sicherheitskonzept und seine Anwendung gefordert. Um die höchste normgemäße Sicherheitsintegritätsstufe ASIL D zu erreichen, ist das altbewährte Sicherheitskonzept EGAS in aller Regel zu schwach, weil es nur ein nichtredundantes Rechnersystem (MC) vorsieht.
Unter der Bedingung, ebenfalls mit einem einzigen MC auszukommen, wird zur Lösung dieses Problems ein neuartiges Sicherheitskonzept entwickelt. Es sieht vor, von MC berechnete Ausgangsgrößen funktionell diversitär auf redundante Sensorgrößen umzurechnen. Die im zweiten Sensorbaustein integrierte und damit asymmetrisch angeordnete Vergleichseinrichtung (AAV) stellt unabhängig von MC und für jeden einzelnen von MC erarbeiteten Funktions- und Ausgabewert die Integrität sowohl der Daten als auch der Rechner- und Sensorhardware sicher. Weiterhin vereinfacht dieser Aufbau den Verifikationsaufwand entscheidend, weil weder Sensoren noch umfangreiche MC-Software, sondern allein die Funktion der weit weniger komplexen AAV verifiziert werden muss. Die Beschränkung auf neben MC nur zwei weitere integrierte Schaltungen (ICs) stellt ebenfalls eine für die funktionale Sicherheit vorteilhafte Vereinfachung dar, denn zwei gleiche, jedoch funktionell diversitär erfassende Sensor-ICs verringern die Komplexität des neuen Konzepts auf das Notwendigste. Im Gegensatz zum EGAS-Konzept ist allmähliche Leistungsabsenkung sowie Notlauf einzelner Funktionalitäten möglich. Dies wird durch von Ende zu Ende abgesicherte Freigabe- bzw. Abschaltbotschaften erreicht, die AAV nach Vergleichen unabhängig von MC an die Aktorik sendet. Im konkreten Einsatz zur Lenkwinkelerfassung wird demonstriert, wie bzw. dass die höchsten normativen Anforderungen an die Hardwaresicherheitsintegrität für eine ECU mit nur einem Rechnersystem erfüllt werden. Anschließend wird in einer tiefgreifenden und umfassenden Bewertung der Sicherheitsintegrität in Systemen mit dem vorgestellten Sicherheitskonzept verallgemeinernd seine Eignung für Fahrzeugfunktionalitäten mit Sicherheitszielen bis ASIL D gezeigt und nachgewiesen
Establishment and improvement of a high-resolution method to analyse the phosphorylation state of the mitogen activated protein kinases ERK1 and ERK2
Fehlregulationen der ERK1/2-Signalkaskade könnten nicht nur in der Onkogenese, sondern auch im Kontext der Alzheimer-Krankheit und der Amyothrophen Lateralsklerose involviert sein (Kim und Choi, 2010; Deschenes-Simard et al., 2014). Vollständig aktiviert werden die Mitogen-aktivierten Proteinkinasen ERK1 und ERK2 über eine doppelte Phosphorylierung an einem konservierten Threonin-Glutamat-Tyrosin (TEY)-Motiv. Monophosphorylierte Isoformen von ERK1/2 sind ebenfalls in lebenden Zellen nachzuweisen und besitzen in vitro bei physiologischen ATP-Konzentrationen eine messbare Kinaseaktivität. Eine hochauflösende Trennung und Detektion der unphosphorylierten, einfach- und zweifach-phosphorylierten Isoformen von ERK1 und ERK2 sowie ihre semi-quantitative Analyse sind durch den Einsatz der isoelektrischen Fokussierung in Mikrokapillaren mit nachgeschaltetem Immunnachweis (CIEF-Immunoassay) möglich.
Im ersten Teil der vorliegenden Arbeit wurde ein optimiertes Arbeitsprotokoll entwickelt, um die Phosphorylierung von ERK1/2 in Zelllysaten mit dem CIEF-Immunoassay zu analysieren. Es wurden verschiedene Lysesysteme getestet, wobei sich herausstellte, dass Reduktionsmittel die ERK1/2-Detektion erheblich beeinflussen können. Das kommerziell erhältliche M-PER-Reagenz eignete sich am besten, da es eine höhere Effizienz zur Lyse der Zellkerne zeigte als andere Lysepuffer, keine Reduktionsmittel enthielt und eine hohe Ausbeute an phosphorylierten ERK1/2-Isoformen ermöglichte. Die Reproduzierbarkeit der relativen Quantifizierungen im CIEF-Immunoassay erwies sich für solche Signale als akzeptabel (Variationskoeffizient < 15 %), deren relativer Flächeninhalt mindestens 1 % der aufsummierten Flächeninhalte aller ERK1- bzw. ERK2-Signale betrug. Insgesamt lagen die Abweichungen in einem Bereich, der in der Regel sowohl einen Vergleich von Proben innerhalb eines Laufes als auch zwischen mehreren Läufen erlaubte.
Als wichtige Weiterentwicklung der bisher publizierten analytischen Möglichkeiten des verwendeten CIEF-Immunoassays gelang im Rahmen der hier vorgestellten Arbeit die zusätzliche Unterscheidung und eindeutige Zuordnung von Threonin-monophosphoryliertem und Tyrosin-monophosphoryliertem ERK1 und ERK2. Dieser Nachweis wurde durch die Kombination synthetischer phospho-ERK1/2-Peptide mit verschiedenen anti-phospho-ERK1/2-Antikörpern in kompetitiven Blockierungsexperimenten, welche in dieser Form erstmalig im CIEF-Immunoassay eingesetzt wurden, erreicht.
Um die praktische Anwendbarkeit des optimierten Arbeits- und Analyseprotokolls zu überprüfen, wurde im zweiten Teil der Arbeit zunächst exemplarisch die Kinetik der ERK1/2-Phosphorylierung in den Zelllinien SH-SY5Y und THP-1 sowie in humanen peripheren mononukleären Blutzellen nach Stimulation der Zellen zeitabhängig untersucht. Die ERK1/2-phospho-Form-Verteilung schien dabei abhängig vom jeweiligen Zelltyp und eingesetzten Aktivator zu variieren.
Die erfolgreich etablierte Methode wurde schließlich in Form einer orientierenden Pilot-Studie mit einer klinischen Kohorte an humanen peripheren mononukleären Blutzellen von Patienten mit Alzheimer-Demenz, leichten kognititven Einschränkungen, Amyotropher Lateralsklerose und nicht-dementen Kontrollprobanden angewandt. Trotz der sehr kleinen Stichprobe konnten statistisch signifikante Gruppenunterschiede, nach erfolgter Aktivierung durch Gabe eines Stimulus, hinsichtlich der Inaktivierung (Dephosphorylierung) der doppelt-phosphorylierten und Threonin-monophosphorylierten Formen von ERK1 beobachtet werden. Dies könnte auf eine mögliche krankheitsassoziierte Veränderung in der Aktivität von Phosphatasen hinweisen. Die Ergebnisse sind allerdings aufgrund der kleinen Gruppengrößen als vorläufig zu betrachten. Für den Einsatz mit einer größeren Kohorte ist die Optimierung weiterer Parameter erforderlich. Insgesamt ermöglicht der CIEF-Immunoassay eine detaillierte Studie zur relativen Verteilung der phospho-ERK1/2-Isoformen in Zelllysaten.The dysregulation of the ERK1/2 signalling cascade appears to be involved not only in oncogenesis but also in the context of Alzheimer’s disease and amyotrophic lateral sclerosis. The mitogen activated protein kinases ERK1 and ERK2 are fully activated by dual phosphorylation at a conserved threonine-glutamate-tyrosine (TEY) motif. Monophosphorylated isoforms of ERK1/2 are also present in living cells and have appreciable kinase activities in vitro at physiological ATP concentrations. The highly sensitive detection and differentiation of the unphosphorylated, monophosphorylated and diphosphorylated isoforms of ERK1 and ERK2 as well as their semi-quantitative analysis can be achieved by capillary isoelectric focussing followed by immunodetection (CIEF-immunoassay).
In the first part of the present work an optimised working procedure was elaborated for the detailed analysis of phosphorylation of ERK1/2 in cell lysates by CIEF-immunoassay. When different lysing systems were tested, it turned out, that reducing agents can substantially influence the ERK1/2 detection. The commercially available M-PER reagent was most suitable as it possessed higher efficiency in lysing nuclei than other buffers, it contained no reducing agents and offered a high recovery of ERK1/2-phospho-forms in the resulting cell lysates. The reproducibility of the relative quantifications in the CIEF-immunoassay was acceptable (coefficient of variation < 15 %) for signals with a relative abundancy greater than 1 % of the added abundancies of all ERK1 and ERK2 signals, respectively. Generally, the observed variation allowed for reasonable comparison of samples within the same assay as well as in different, independent assays.
An important improvement of previously published analytical possibilities of the CIEF-Immunoassay is the additional differentiation and classification of the threonine- and tyrosine-monophosphorylated isoforms of ERK1 and ERK2, which is presented within this work. The differentiation and unequivocal peak identification were achieved with a combination of synthetic phospho-ERK1/2 peptides and several anti-phospho-ERK1/2 antibodies in competitive blocking experiments, which were performed for the first time in the CIEF-Immunoassay in this form.
As a proof of principle the optimised standard operating procedures were applied to exemplarily analyse the kinetics of ERK1/2 phosphorylation and the time dependent occurrence of phospho-ERK1/2 after stimulation in SH-SY5Y and THP-1 cell lines as well as in peripheral blood mononuclear cells in the second part of the work. The ERK1/2-phospho-form distribution seemed to depend on the particular cell type and the applied stimulus.
The optimised method was furthermore applied to an orienting pilot study on human peripheral blood mononuclear cells from a small clinical cohort of patients with Alzheimer’s disease, mild cognitive impairment, amyotrophic lateral sclerosis and non-demented controls. Despite of the very small sample size, statistically significant differences between the diagnostic groups could be observed, after activation by applying a stimulus, for the inactivation (dephosphorylation) of the diphosphorylated and threonine-monophosphorylated forms of ERK1. These changes could possibly be linked to a disease related dysregulation of phosphatase activity. Due to the small size of the groups within this study, the results have to be considered as preliminary. For additional confirmative studies with a greater sample size an optimisation of several parameters will be necessary. Altogether, the CIEF-immunoassay enables detailed studies concerning the relative ERK1/2-phospho-form distribution in cell lysates
Coalgebraic Behavior Analysis — From Qualitative To Quantitative Analyses
In order to specify and analyze the behavior of systems (computer programs, circuits etc.) it is important to have a suitable specification language. Although it is possible to define such a language separately for each type of system, it is desirable to have a standard toolbox that allows to do this in a generic way for various – possibly quite different – systems.
Coalgebra, a concept of category theory, has proven to be a suitable framework to model transition systems. This class of systems includes many well-known examples like deterministic automata, nondeterministic automata or probabilistic systems. All these systems are coalgebras and their behavior can be analyzed via the notion of final coalgebra or other category theoretic constructions.
This thesis investigates how to improve and build upon existing results to explore the expressive power of category theory and in particular coalgebra in behavioral analysis. The three main parts of the thesis all have a different focus but are strongly connected by the coalgebraic concepts used.
Part one discusses adjunctions in the context of coalgebras. Here, well-known automata constructions such as the powerset-construction are (re)discovered as liftings of simple and well-known basic adjunctions.
The second part deals with continuous generative probabilistic systems. It is shown that their trace semantics can be captured by a final coalgebra in a category of stochastic relations.
The final contribution is a shift from qualitative to quantitative reasoning. Via the development of methods to lift functors on the category of sets and functions to functors on pseudometric spaces and nonexpansive functions it is possible to define a canonical, coalgebraic framework for behavioral pseudometrics.Um das Verhalten von Systemen (Computerprogrammen, Schaltkreisen etc.) zu spezifizieren und zu analysieren, ist es wichtig eine geeignete Spezifizierungssprache zu finden. Obwohl es möglich ist, eine solche Sprache separat für jeden Typ von System zu definieren, ist es erstrebenswert einen standardisierten Ansatz zu haben, welcher es ermöglicht, dies in einer generischen Weise für diverse – möglicherweise stark unterschiedliche – Systeme zu tun.
Koalgebra, ein Konzept der Kategorientheorie, hat sich als geeignetes Modell zur Modellierung von Transitionssystemen herausgestellt. Diese Klasse von Systemen umfasst viele bekannte Beispiele wie deterministische, nichtdeterministische oder probabilistische Automaten. Alle diese Systeme sind Koalgebren und ihr Verhalten kann mittels finaler Koalgebren oder anderer kategorientheoretischer Konstruktionen analysiert werden.
Diese Dissertation beschäftigt sich mit der Frage, wie existierende Resultate verbessert und erweitert werden können, um die Ausdrucksmächtigkeit von Kategorientheorie und besonders Koalgebra in der Verhaltensanalyse zu untersuchen. Die drei Hauptteile dieser Arbeit haben alle eine unterschiedliche Ausrichtung, sind aber durch die verwendeten koalgebraischen Methoden stark miteinander verbunden.
Der erste Teil behandelt Adjunktionen im Kontext von Koalgebren. Hierbei werden übliche Automatenkonstruktionen wie die Potenzmengenkonstruktion als Lifting einfacher und wohlbekannter Basisadjunktionen (wieder-)entdeckt.
Im zweiten Teil werden kontinuierliche, generative probabilististische Systeme betrachtet. Es wird gezeigt, dass deren lineares Verhalten von einer finalen Koalgebra in einer Kategorie stochastischer Relationen erfasst werden kann.
Der letzte Beitrag ist ein Wechsel von qualitativer zu quantitativer Argumentation. Durch die Entwicklung von Methoden, die dazu dienen Funktoren von der Kategorie der Mengen und Funktionen zu Funktoren auf pseudometrischen Räumen und nicht-expansiven Funktionen zu erweitern, ist es möglich eine kanonische, koalgebraische Herangehensweise für Verhaltensmetriken zu entwerfen
Online power management with embedded optimization for a multi-source hybrid with real time applications
The focus of this thesis is to develop a suitable power management optimization strategy for a three-source hybrid vehicle powertrain. This strategy takes into account the integration of optimized parameters that limit the battery and fuel cell current by utilizing a third power source, namely supercapacitor. The goal is to develop a modular structure with decoupled online and offline parts such that implementation in case of real driving conditions is feasible. Based on the literature review it can be concluded that providing optimal solutions in terms of multiple objectives online is an issue. Adaption of optimized control strategy to real driving data is another concern. The developed strategy employs an online rule-based control with embedded offline-optimized parameters. The parameters are optimized with respect to multiple and conflicting objectives such as fuel consumption and state-of-charge deviation minimization. By a suitable selection of parameters, operation of all three sources within desired working ranges is possible, keeping in mind the load demand. By varying the weights between the objectives, one or more objectives can be given more priority than others. The application of this concept to fuel cell-battery-supercapacitor hybrid is discussed in this thesis. Detailed modeling of all components along with verification and plausibility assessment is done. For the purpose of experimental validation, the real powertrain components are replaced by controllable power sources and sinks that emulate the dynamics of real components. Finally, a brief concept is presented to integrate the developed power management optimization in real driving scenarios. For validation/verification purposes, a driving simulator environment is connected to the experimental hybrid electric vehicle set-up and with the help of an illustrative example, the desired predicted optimal values are calculated online and displayed to the human driver by a suitable interface. The absence of online tuning of controller parameters in this example is counteracted by developing a concept based on literature. With the help of this concept, the adaption of the power management control concept, developed in this thesis, can be realized
Verhalten von toxikologisch relevanten Silberverbindungen und Bariumsulfat-Partikeln in komplexen Medien: von der Synthese zu biologischen Untersuchungen
This work comprises the synthesis procedures of toxicologically relevant particles, and their behavior in complex biological media in terms of stability and dissolution, with specific biological responses to the synthesized materials. Two toxicologically relevant particulate systems were studied. As a model of a biologically active material, silver compounds, especially silver nanoparticles, were investigated. In contrast, barium sulfate was examined as a bioinert system.
The formation of silver nanoparticles during the reduction with glucose in the presence of poly(vinylpyrrolidone) as a capping agent was followed for more than 3000 min. It was shown that spherical silver nanoparticles were formed first, but in later stages, an increasing fraction of nano-triangles and also a few nanorods developed. By electron microscopy, the growth of spherical and trigonal nanoparticles with time was described. From X-ray powder diffractometry, it was concluded that the domain size in the spherical nanoparticles increased proportionally to the particle diameter and was always about ¼ of the diameter, indicating that twinned seeds were formed very early in the process and then simply grew by extending their size. The lattice constant of the nanoparticles was systematically increased in comparison to microcrystalline silver, but did not change as a function of particle diameter.
The behavior of silver ions was investigated in biologically relevant concentrations in different media, from physiological salt solution over phosphate-buffered saline solution to protein-containing cell culture media. The results showed that silver ions that were initially present were bound as silver chloride due to the presence of chloride. Only in the absence of chloride, glucose was able to reduce Ag+ to Ag0. The precipitation of silver phosphate was never observed in any case. It was concluded that the predominant silver species in biological media is dispersed nanoscopic silver chloride, surrounded by a protein corona which prevents the growth of crystals and leads to colloidal stabilization. Using bacteria (S. aureus) and cell culture experiments (human mesenchymal stem cells; T-cells; monocytes), it was possible to show that a toxic effect occurred at the same silver concentration of ionic silver and synthetically prepared silver chloride nanoparticles. Furthermore, the dissolution of silver nanoparticles and the solubility products of silver chloride and silver nitrate were quantified in protein-containing cell culture media.
Barium sulfate micro-, sub-micro-, and nanoparticles were synthesized for evaluation of inflammatory potential. The synthesis of fluorescent labeled BaSO4 particles with a narrow size distribution was achieved. The resultant powders were uniform in form, chemical composition and surface modification by the fluorescent dye. All of the particles were colloidally dispersed in cell culture medium with serum. From the cellular uptake studies on rat alveolar macrophages, it was concluded that a significant difference occurred in the case of barium sulfate microparticles for suspended and adherent cell fractions. This was explained by the sedimentation of synthesized particles over time, and confirmed by stability studies of dispersed particles in biological medium by dynamic light scattering. However, in a particle-induced cell migration assay for evaluation of the inflammation potential of the system, no dose- and size-dependency was found
The role of the sympathetic nervous system during Friend retrovirus infection
In der vorliegenden Arbeit wurde die Rolle des sympathischen Nervensystems (SNS) im Verlauf einer Friend Retrovirus (FV)-Infektion bei Mäusen untersucht. Es konnte gezeigt werden, dass es zum Höhepunkt der virus-induzierten Splenomegalie zu einem massiven Verlust des sympathischen Neurotransmitters Noradrenalin (NA) in der Milz kommt. Dieser Effekt war transient und am Ende der akuten Infektion nicht mehr zu beobachten. Die FV-induzierte NA-Depletion konnte durch die pharmakologische Blockade der Katecholamin-degradierenden Enzyme Monoaminoxidase A (MAO-A) bzw. Catechol-O-Methyltransferase (COMT) mithilfe der selektiven Inhibitoren Clorgylin bzw. 3,5-Dinitrocatechol verhindert werden. Diese Intervention führte in FV-infizierten Mäusen zudem zu einer signifikanten Reduktion in der Viruslast und zu einer verminderten virus-induzierten Splenomegalie. Eine periphere chemische Sympathektomie vor der FV-Infektion hatte hingegen eine verstärkte Pathologie zur Folge. Sympathektomierte FV-infizierte Mäuse wiesen im Vergleich zu intakten Tieren eine signifikant höhere Viruslast auf und zeigten eine signifikant stärkere Splenomegalie. Eine Behandlung mit selektiven adrenergen Agonisten bzw. Antagonisten hatte keinen Einfluss auf den Infektionsverlauf. In chronisch FV-infizierten Tieren führte die chemische Sympathektomie ebenfalls zu einer Vergrößerung der Milz. Direkte Effekte von NA auf die Virus-Replikation konnten in in vitro Experimenten ausgeschlossen werden. Die Expression von β-adrenergen Rezeptoren in Erythrozyten-Vorläuferzellen deuten auf eine mögliche Interaktion des Neurotransmitters mit den Zielzellen des Virus hin, was jedoch in weiterführenden Experimenten bestätigt werden muss. Zusammengefasst zeigen die Ergebnisse dieser Arbeit, dass es während der Pathogenese einer FV-Infektion zu Veränderungen im Neurotransmitterstoffwechsel in einem wichtigen Zielorgan der Infektion kommt. Pharmakologische Interventionen, die eine experimentelle Erhöhung bzw. Erniedrigung der NA-Konzentration in der Milz bewirken, haben eine abgeschwächte bzw. verstärkte Pathologie zur Folge. Des Weiteren demonstriert diese Arbeit, als eine von wenigen präklinischen Studien, dass sich das SNS, während einer retroviralen Infektion, für den Wirt als äußerst vorteilhaft erweisen kann. Somit wird deutlich, dass ein dringender Bedarf an klinischen Studien besteht, die das Risiko und das therapeutische Potential von adrenergen Agonisten/Anatgonisten oder von MAO-A/COMT-Inhibitoren in Patienten retroviraler Infektionserkrankungen, sorgfältig beurteilen.The present thesis investigated the role of the sympathetic nervous system (SNS) during Friend retrovirus (FV) infection in mice. Results show that the splenic concentration of the sympathetic neurotransmitter noradrenaline (NA) was massively reduced at the peak of the virus-induced splenomegaly. This effect was transient and could not be observed at the end of the acute infection. FV-induced NA depletion was prevented by pharmacological inhibition of the catecholamine degrading enzymes monoamine oxidase A (MAO-A) and catechol-O-methyltransferase (COMT) using the selective inhibitors clorgyline (CLG) and 3,5-dinitrocatechol (OR-486). Moreover, this intervention resulted in significantly reduced viral load and a less pronounced virus-induced splenomegaly in FV-infected mice. In contrast, chemical sympathectomy prior to FV-infection aggravated the pathology of the disease. Sympathectomized and simultaneously FV-infected mice exhibited both, a significant higher viral burden and a more pronounced increased splenomegaly compared to intact FV-infected mice. Noteworthy, treatment with selective adrenergic agonists or antagonists did not influence the course of FV infection. Chemical sympathectomy in chronically FV-infected mice also resulted in a significant enlargement of the spleen. Direct effects of NA on viral replication could be excluded by in vitro studies. The expression of β-adrenergic receptors in erythroid precursor cells suggests a possible interaction between the sympathetic neurotransmitter and the viral target cells. However, this has to be confirmed in future experiments. Together, the results of this thesis demonstrate that FV infection leads to substantial changes in neurotransmitter metabolism in the spleen, one of the main sites of viral replication. Pharmacological interventions, that increase or decrease splenic NA concentration, can have a major impact on viral pathology. Furthermore, this work belongs to the minority of pre-clinical studies that were able to demonstrate, that the SNS plays an important and beneficial role for the host in terms of retroviral infections. Thus, there is an urgent need for clinical studies that carefully evaluate the risks and therapeutic potential of adrenergic agonists/antagonists or MAO-A/COMT inhibitors in patients suffering from retroviral infections
Beschreibung der pflegerischen Versorgungsfelder "Chronische Wunde" und "Schmerz" - Eine Querschnittsuntersuchung im Universitätsklinikum Essen
Vor dem Hintergrund der Relevanz quantitativer Untersuchungen zu pflegerelevanten Gesundheitsstörungen für die Pflegeforschung und der mangelnden Ausgangslage an verlässlichen Daten sollten in dieser Arbeit die Versorgungsfelder „Chronische Wunden“ und „Schmerzen“ im Universitätsklinikum Essen auf Grundlage von Dokumentationssichtungen und Patientenbefragungen/-beobachtungen aus dem Jahr 2009 beschrieben werden. Die zentralen Fragen sind dabei einerseits, in welchen Häufigkeiten und bei welchen Patienten chronische Wunden und Schmerzen auftreten sowie andererseits, mit welchen Risikofaktoren und Einschränkungen chronische Wunden einhergehen und welche Intensitäten und Folgen bei den Schmerzpatienten ermittelt werden konnten. Weiterhin wird der Frage nachgegangen, wie sich auf Grundlage der vorliegenden Daten das Schmerzmanagement gestaltet. Die gewonnenen Ergebnisse werden mit denen vorliegender Studien und pflegerischer Expertenstandards in Beziehung gesetzt, um so eine realistische Einschätzung dieser zwei pflegerischen Versorgungsfelder im Universitätsklinikum Essen zu erhalten, welche einem weiteren Ausbau des Versorgungsmanagements als Grundlage dienen kann
