    A contribution to validating of simulation models in production and logistics

    In this dissertation a new way of thinking in the validation of simulation model methods in production and logistics systems is presented. The focus is on finding the most important parameters for the validation of simulation models, the relationships between these parameters in the form of correlations which influence the complexity of the models, the required availability of data for these parameters, and the uncertainty which is largely dependent on data availability. Further, a short assessment is made of sequential bifurcation as a method for model validation in the context of finding important parameters, reducing complexity, and exploring uncertainties

    Zugriff auf den Volltext ist gesperrt, neue Version unter DuEPublico-ID 40890 Architectural tensile surface structures – often made from coated woven fabrics – carry external loads only by activating tensile stress in the membrane plane, typically in the form of biaxial stress states. One important and challenging aspect of structural fabric analysis is the determination of stiffness parameters that sufficiently model the stress-strain behaviour for these stress states. The particular difficulty is that coated woven fabrics exhibit very complex stiffness behaviour. In general, it is nonlinear, nonelastic and considerably anisotropic. Nevertheless, current membrane structure design practice is based on a simplified orthotropic linear-elastic plane stress relationship, where the elastic constants are “tensile modulus” and “Poisson’s ratio”. The elastic constants must be determined for each material using biaxial tensile tests. The intention of the present work is to develop principles for determining elastic constants that closely approximate the actual fabric stress-strain response for any fabric structure and all common types of coated woven fabrics. The focus lies on the most commonly utilised materials: PVC coated polyester fabrics and PTFE coated glass fibre fabrics. The foundation of the present work is a comprehensive survey of the structural behaviour of all types of membrane structures – anticlastic, synclastic and plane – as well as of the stiffness properties of coated woven fabrics for architectural applications. Discussion of the mechanical background to the constitutive law for orthotropic linear-elastic plane stress provides the frame of application for elastic constants, particularly in relation to the boundaries of the Poisson’s ratios. An analysis of internationally established biaxial test and evaluation procedures identifies the strengths and weaknesses of current practice. Gaps in knowledge are closed with experimental investigation into the full range of commonly used architectural fabrics. Combining all insights, principles for refined biaxial test and evaluation procedures are stated with the objective of determining elastic constants for design purposes. As a basic principle, procedures for anticlastic structures and for synclastic or plane structures are developed separately. Their commonality lies in the fact that they are based on what is defined as the stable state of the fabric. Using stable state elastic constants makes it possible to calculate with the nominal prestress in the fabric structure analysis. Example application of the refined procedures illustrates that deviations between the measured and calculated strain on a specific evaluation stress level are low throughout. Die elastischen Konstanten müssen für jedes Material in biaxialen Zugversuchen bestimmt werden. Das Ziel der vorliegenden Arbeit ist die Entwicklung von Prinzipien zur bemessungsorientierten Bestimmung elastischer Konstanten derart, dass sie das tatsächliche Spannungs-Dehnungs-Verhalten von allen üblichen Architekturgeweben für alle Tragwerksformen gut approximieren. Der Fokus liegt dabei auf den gebräuchlichsten Produkten: PVC-beschichtete Polyestergewebe und PTFE-beschichtete Glasfasergewebe. Das Fundament der vorliegenden Arbeit ist sowohl eine umfassende Studie des Tragverhaltens aller typischen Membranbauformen – antiklastisch, synklastisch und eben – als auch der Steifigkeitseigenschaften der Architekturgewebe. Eine Erörterung des mechanischen Hintergrunds zum orthotropen, linear-elastischen Materialgesetz bei Anwendung auf den ebenen Spannungszustand liefert die Randbedingungen für die Anwendung der elastischen Konstanten, besonders im Bezug auf Grenzwerte für die Querkontraktionszahlen. Eine Analyse internationaler Biax-Versuchs- und Auswerteprozeduren identifiziert die Stärken und Schwächen der aktuellen Praxis. Vorhandene Wissenslücken werden durch experimentelle Untersuchungen an der ganzen Bandbreite der gebräuchlichen Gewebetypen für die textile Architektur geschlossen. Aus der Kombination aller Erkenntnisse werden Prinzipien für fundierte Biax-Versuchs- und Auswerteprozeduren abgeleitet. Das Ziel ist die Bestimmung von elastischen Konstanten, die als Eingangsparameter in der Bemessung dienen. Grundsätzlich wird zwischen Prozeduren für antiklastische und synklastische bzw. ebene Strukturen unterschieden. Beiden ist allerdings gemein, dass sie den “eingespielten Zustand eines Gewebes” nutzen. Erst die Nutzung von elastischen Konstanten im eingespielten Zustand ermöglicht es, die Strukturberechnungen auf den nominellen Vorspannungszustand zu gründen. Beispielhafte Anwendungen der weiterentwickelten Prozeduren verdeutlichen, dass die Abweichungen zwischen gemessenen und berechneten Dehnungen auf einem zuvor für die Auswertung definierten Spannungshorizont durchweg klein sind. Dies zeigt eindrucksvoll, dass das linear-elastische Materialgesetz durchaus sehr geeignet ist, auch das Spannungs-Dehnungs-Verhalten aller typischen PVC-beschichteten Polyester- und PTFE-beschichteten Glasfasergewebe zu beschreiben

    Effectiveness of leech therapy in osteoarthritis of the knee - a randomized, controlled trial

    Die Gonarthrose ist weltweit die häufigste Gelenkerkrankung des erwachsenen Menschen. Ihre Prävalenz nimmt mit steigendem Alter zu. Die Beschwerden der Patienten sind gekennzeichnet durch starke Schmerzen und Funktionsminderung. Therapieziel ist die Bekämpfung der klinischen Symptome. Das therapeutische Spektrum ist jedoch entweder in seiner Wirksamkeit limitiert oder mit erhöhten gesundheitlichen Risiken und Nebenwirkungen verbunden. Blutegel könnten hier eine sinnvolle und effektive Therapiealternative sein. Ihr therapeutischer Nutzen bei lokalen Schmerzsyndromen wurde in der Literatur wiederholt beschrieben. In der vorliegenden kontrollierten, monozentrischen, randomisierten Studie wurde über einen Zeitraum von 91 Tagen (April bis Juli 2002) die Wirksamkeit der Blutegeltherapie bei symptomatischer Gonarthrose klinisch evaluiert. 51 Patienten (m:f = 18:33) im Durchschnittsalter von 60-65 Jahren mit klinisch gesicherter Gonarthose (Stadium II-III) erhielten entweder eine einmalige Applikation von 4-6 Blutegeln periartikulär am schmerzhaften Kniegelenk (Prüfgruppe) oder eine zwei- bis dreimal tägliche topische Anwendung von Diclofenac-Gel über 28 Tage (Kontrollgruppe). Alle Auswertungen basieren primär auf der „intention-to-treat“ Population. Fehlende Werte wurden nach der „last-observation-carried-forward“ Methode ersetzt. Das sequentielle Studiendesign zur Analyse des Hauptzielkriteriums wurde im Verlauf durch einen F-Test ersetzt. Die absolute Veränderung des WOMAC-Schmerzscores zeigte sich sieben Tage nach Therapiebeginn mit p < 0,0001 hochsignifikant für die Blutegeltherapie (primäres Zielkriterium). Obwohl sich dieser Unterschied im nachfolgenden Beobachtungszeitraum nicht länger nachweisen ließ, zeigte die Untersuchung der sekundären Zielkriterien einen deutlichen, bis zum Ende der Studie anhaltenden, signifikanten Unterschied für die Blutegeltherapie im Funktions-, Steifigkeits- und Gesamtscore. Eine Beeinflussung der Ergebnisse durch Confounder wurde nicht nachgewiesen. In weiteren, größeren Fallzahlstudien sollte daher der klinische Nutzen der Blutegeltherapie einschließlich der Wirkungsweise weiter evaluiert werden.Background: Leech therapy was commonly used in traditional medicine for treating localized pain. Clinically significant pain relief after leech therapy for osteoarthritis of the knee has been demonstrated by preliminary data. Objective: To evaluate the effectiveness of leech therapy for symptomatic relief of osteoarthritis of the knee. Design: Randomized, controlled trial. Setting: Outpatient department for integrative medicine of an academic teaching hospital. Patients: 51 patients with osteoarthritis of the knee (leech therapy: 24 patients, mean age [±SD], 62.5 ± 10.2 years; topical diclofenac therapy: 27 patients, mean age [±SD], 65.5 ± 6.7 years). Intervention: A single treatment with 4 to 6 locally applied leeches (leech therapy group) or a 28-day topical diclofenac regimen (control group). Measurements: Mean of the pain, function, and stiffness subscores of the Western Ontario and McMaster Universities Osteoarthritis Index and physical sum score of the Medical Outcomes Study 36-Item Short-Form Health Survey with group comparisons at days 3, 7, 28 and 91. Results: The primary end point, pain at day 7, was reduced from a mean (±SD) of 53.5 ± 13.7 to 19.3 ± 12.2 after leech therapy compared with 51.5 ± 16.8 to 42.4 ± 19.7 with topical diclofenac (estimated group difference, -23.9 [95% CI, -32.8 to -15.1]; p < 0.001). Although the difference between group pain scores was no longer significant after day 7, differences for function, stiffness, and total symptoms remained significant in favor of leech therapy until the end of study and for quality of life until day 28. Results were not affected by outcome expectation. Conclusions: Leech therapy helps relieve symptoms in patients with osteoarthritis of the knee. The potential of leech therapy for treating osteoarthritis and the pharmacologic properties of leech saliva remain to be clarified. Ann Intern Med. 2003; 139: 724-730

    Experiments on light induced transport processes on Mars and in protoplanetary disks

    Diese Arbeit befasst sich mit den Effekten der Photophorese und des thermischen Kriechens im Hinblick auf den Transport von Teilchen auf dem Mars und in protoplanetaren Scheiben. In verschiedenen Experimenten und Messkampagnen werden einzelne Teilaspekte dieser Phänomene näher betrachtet. Die durch thermisches Kriechen induzierten Gasströme können einen signifikanten Einfluss auf Teilchen haben. Dies funktioniert allerdings nur dann effizient, wenn die Größe des Teilchens oder seiner Kapillaren beziehungsweise Poren ungefähr der mittleren freien Weglänge der Gasmoleküle entspricht. Diese Bedingung ist für den Marsboden gut erfüllt. Daher kann dort durch Temperaturgradienten Gas durch die Poren gepumpt werden, wodurch ein Überdruck unter der Oberfläche auftritt. Dieser Überdruck übt eine hebende Kraft auf das Staubbett aus, wodurch Staubtransport begünstigt werden kann. Die Transportgrenze beschreibt die Windgeschwindigkeit, ab der Transport stattfindet. Diese wird im Experiment unter Einfluss von Beleuchtung untersucht. Reskaliert auf den Mars kann die Transportgrenze um bis zu 12% durch Sonneneinstrahlung reduziert werden. Diese Reduktion kann einen Teil der Diskrepanzen zwischen beobachteten Windgeschwindigkeiten, die Transport nach bisherigen Erkenntnissen selten zulassen, und den häufig zu beobachteten Transportphänomenen zu erklären. Die Photophorese ist eher für die Bewegung einzelner freier Teilchen bedeutsam und es wird untersucht, in wie weit sie für verlängerte Verweildauern von Staub in der Marsatmosphäre verantwortlich sein kann. Für die untersuchten Basaltteilchen liegt die Kraft nur im Prozentbereich der Gravitation und spielt daher für diese Klasse von Teilchen nur eine untergeordnete Rolle. In einer protoplanetaren Scheibe hingegen erfahren die Teilchen nur wenig Gravitation, da sie sich in einer Umlaufbahn befinden; durch die Gasreibung entsteht eine Restgravitation, die die Teilchen nach innen und zur Mittelebene sedimentieren lässt. Hier können auch kleine Kräfte die Bahn der Teilchen signifikant beeinflussen. Metergroße Körper driften in einer protoplanetaren Scheibe mit einer vom Scheibenmodell abhängigen Geschwindigkeit (der Größenordnung 100 m/s) radial in Richtung Stern und sind damit schnell für das Planetenwachstum verloren. Die Photophorese kann diese Geschwindigkeiten soweit beeinflussen, dass diese Körper sogar eine signifikante Geschwindigkeit radial nach außen aufweisen. Dieser Effekt wird bei porösen Körpern noch durch thermisches Kriechen durch die Poren verstärkt. Allerdings funktioniert die Photophorese durch direkte Strahlung vom Stern nur dort, wo die Scheibe optisch dünn ist. Dies ist bei den meisten Scheibenmodellen nur an der Oberfläche der Scheibe der Fall. Die Grenze zwischen optisch dünn und optisch dicht wird im Zuge der Evolution der Scheibe durch verschiedene Prozesse kontinuierlich radial nach außen verschoben.This work is about the effects of photophoresis and thermal creep in terms of transport of particles on Mars and in protoplanetary disks. Individual aspects of these phenomena are considered in various experiments and measurement campaigns. Thermal creep induced gas streams may have a significant influence on the behaviour of the examined object. This will only work if the size of the object or of its capillaries or pores is approximately equal to the mean free path of the gas molecules. This condition is well satisfied for the Martian soil. Therefore it is possible on Mars that gas is pumped through the pores by temperature gradients, creating an overpressure below the surface. This pressure exerts a lifting force on the dust bed, whereby dust transport can be promoted. From the experimental results a reduction of the transport limit – which is the minimum wind velocity for transport – of up to 12 % can be deduced. The photophoresis rather moves individual particles and its influence concerning the prolonged residence time of dust in the atmosphere is examined. For the investigated basalt particles the force is only a few percent of gravity and therefore just plays a minor role for this class of particles. When considering a protoplanetary disk this effect is more important, since the particles in orbit around the star experience only small residual gravity; induced by the gas friction, the particles settle to the mid plane and drift inward. Especially at the meter-barrier where meter sized objects drift inward with a velocity depending on the disk model (in the order of 100 m/s), and hence would quickly be lost for planet growth, photophoresis can influence these speeds to such extent that the bodies even move outward with significant speed. This effect is enhanced in porous bodies due to thermal creep through the pores. However, this applies only for optically thin regions of the disk. For most disk models this is only the case at the disk surfaces. The border of the transition between optically thin and optically thick moves continually outward during the disks lifetime

    Fast Differential-Geometric Methods for Continuous Muscle Wrapping Over Multiple General Surfaces

    Musculoskeletal simulation has become an essential tool for understanding human locomotion and movement disorders. Muscle-actuated simulations require methods that continuously compute musculotendon paths, their lengths, and their rates of length change to determine muscle forces, moment arms, and the resulting body and joint loads. Musculotendon paths are often modeled as locally length-minimizing curves that wrap frictionlessly over moving obstacle surfaces representing bone and tissue. Biologically accurate wrapping surfaces are complex and a single muscle path may wrap around multiple obstacles. However, state-of-the-art muscle wrapping methods are either limited to analytical results for a pair of simple surfaces, or they are computationally expensive. This thesis describes a new method for the fast and accurate computation of a massless musculotendon’s shortest path that wraps frictionlessly across an arbitrary number of general smooth wrapping surfaces. Furthermore, an explicit formula for the path’s exact rate of length change is presented, as well as an algorithm for simulating path lift-off and touchdown. The total path is regarded as a concatenation of straight-line segments between local surface geodesics, where each geodesic is naturally parameterized by its start point, direction, and length. The shortest path is computed by finding the root of a vector-valued global path-error constraint equation that enforces that the geodesics connect collinearly with adjacent straight-line segments. High computational efficiency is achieved using Newton’s method to zero the path error with an explicit, banded Jacobian that maps natural variations of the geodesic parameters to path-error variations. Simulation benchmarks demonstrate that the proposed method computes high-precision solutions for path length and rate of length change, and that it allows for wrapping over biologically accurate surfaces that can be described either parametrically or implicitly. By using the explicit path-error Jacobian, the proposed method is very efficient and thus allows for simulating muscle paths over hundreds of surfaces in real time

    Approach for Improved Signal-Based Fault Diagnosis of Hot Rolling Mills

    Der hier vorgestellte Ansatz ist in der Lage, zwei spezifische schwere Fehler zu erkennen, sie zu identifizieren, zwischen vier verschiedenen Systemzuständen zu unterscheiden und eine Prognose bezüglich des Systemverhaltens zu geben. Die vorliegende Arbeit untersucht die Zustandsüberwachung des komplexen Herstellungsprozesses eines Warmbandwalzwerks. Eine signalbasierte Fehlerdiagnose und ein Fehlerprognoseansatz für den Bandlauf werden entwickelt. Eine Literaturübersicht gibt einen Überblick über die bisherige Forschung zu verwandten Themen. Es wird gezeigt, dass die große Anzahl vorheriger Arbeiten diese Thematik nicht gelöst hat und dass weitere Untersuchungen erforderlich sind, um eine zufriedenstellende Lösung der behandelten Probleme zu erhalten. Die Entwicklung einer neuen Signalverarbeitungskette und die Signalverarbeitungsschritte sind detailliert dargestellt. Die Klassifikationsaufgabe wird in Fehlerdiagnose, Fehleridentifikation und Fehlerprognose differenziert. Der vorgeschlagene Ansatz kombiniert fünf verschiedene Methoden zur Merkmalsextraktion, nämlich Short-Time Fourier Transformation, kontinuierliche Wavelet Transformation, diskrete Wavelet Transformation, Wigner-Ville Distribution und Empirical Mode Decomposition, mit zwei verschiedenen Klassifikationsalgorithmen, nämlich Support-Vektor Maschine und eine Variation der Kreuzkorrelation, wobei letztere in dieser Arbeit entwickelt wurde. Kombinationen dieser Merkmalsextraktion und Klassifikationsverfahren werden an Walzkraft-Daten aus einer Warmbreitbandstraße angewendet.The approach introduced here is able to detect two specific severe faults, to identify them, to distinguish between four different system states, and to give a prognosis on the system behavior. The presented work investigates the condition monitoring of the complex production process of a hot strip rolling mill. A signal-based fault diagnosis and fault prognosis approach for strip travel is developed. A literature review gives an overview about previous research on related topics. It is shown that the great amount of previous work does not cope with the problems treated in this work and that further investigation is necessary to provide a satisfactory solution. The design of a new signal processing chain is presented and the signal processing steps are detailed. The classification task is differentiated into fault detection, fault identification and fault prognosis. The proposed approach combines five different methods for feature extraction, namely short time Fourier transform, continuous wavelet transform, discrete wavelet transform, Wigner-Ville distribution, and empirical mode decomposition, with two different classification algorithms, namely support vector machine and a variation of cross-correlation, the latter developed in this work. Combinations of these feature extraction and classification methods are applied to rolling force data originating from a hot strip mill

    Detection and analysis of misuse in SIP-based networks

    Die Sprachkommunikation über „Voice over IP“-Netzwerke, basierend auf dem Session Initiation Protokoll (SIP), verbreitet sich auf Grund von Funktionalitäts- und Kostenvorteilen zunehmend und wird die klassischen Telefonnetze in den nächsten Jahren vollständig ablösen. Zusätzlich zu den Netzen der Telefonanbieter wird die Sprachkommunikation über das SIP-Protokoll auch im Unternehmens- und Privatanwenderumfeld unverzichtbar. So bietet VoIP die Möglichkeit, sich unabhängig von dem aktuellen Aufenthaltsort über das Internet bei dem jeweiligen Heimatnetzbetreiber oder der eigenen Firma anzumelden und über das dortige Nutzerkonto Gespräche zu führen. Da die Telefonie somit von einer geschlossenen und vergleichsweise sicheren Plattform auf eine viel offenere Plattform in das Internet migriert wird, ergeben sich neue Risiken und Missbrauchsmöglichkeiten im Bereich der Telefonie. In dieser Dissertation werden Angriffe untersucht, die mit der Einführung von SIP-basierten Sprachdiensten im Internet entstehen und nicht aus Bedrohungen der Netzwerkschicht oder aus rechtlichen Vertragsbestimmungen resultieren. Das Ziel dieser Angriffe ist das Erlangen eines finanziellen Vorteils, indem ein Angreifer kompromittierte Zugänge für Auslandstelefonate oder für Anrufe zu Premiumnummern auf Kosten der Anschlussinhaber nutzt („Toll Fraud“). Für die Realisierung der Bedrohungsanalyse und der Angriffserkennung wurden Konzepte, ein Versuchsnetzwerk sowie die notwendigen Softwarekomponenten ergebnisorientiert entwickelt. Im Vergleich zu anderen Forschungsarbeiten wurden Untersuchungen mit Ködersystemen (Honeypots) weiterentwickelt und es wurde ein System für eine verteilte, automatische Angriffserkennung entwickelt. Dafür wurden SIP-Verkehrsdaten über einen Zeitraum von sechs Jahren in zwei Class-C-Netzwerken aufgezeichnet und mit einem neuen Analyseansatz unabhängig von einzelnen SIP-Nachrichten automatisch ausgewertet. Die Ergebnisse des Feldversuches in dieser Dissertation zeigen, dass die Bedrohungen für die SIP-Infrastruktur ansteigen und dass bereits eine Weiterentwicklung und Optimierung der Angriffswerkzeuge nachzuweisen ist. Die zunehmende Anzahl der Toll Fraud-Versuche mit internationalen Anrufzielen (und auch zu Premium-Rufnummern) verdeutlicht, dass bei einem unzureichenden Schutz der SIP-Server für die Nutzer und Betreiber sehr schnell ein erheblicher finanzieller Schaden entstehen kann. Es ist daher unerlässlich, die vorgeschalteten, systematischen Angriffsstufen frühzeitig zu erkennen und Abwehrkomponenten zu benachrichtigen. Für die automatisierte, verteilte Angriffserkennung in Echtzeit und für die Maximierung des Beobachtungsgebietes wurde für diese Dissertation das „Security Sensor System“ entwickelt. Mit Hilfe von leichtgewichtigen Sensoren wurde eine weltweite signaturbasierte Angriffserkennung realisiert. Zusätzlich zu der standortbezogenen Angriffserkennung werden Angriffe durch einen zentralen Dienst korreliert. Dadurch können Angreifer netzwerkübergreifend bzw. länderübergreifend identifiziert und somit Gegenwehrkomponenten in Echtzeit benachrichtigt werden. Der Vergleich der verschiedenen Messstellen im Internet belegt, dass die analysierten Angriffsmuster nicht nur im Netzwerk der Universität Duisburg-Essen, sondern zeitlich zusammenhängend auch an anderen Standorten auftreten. Dadurch wird deutlich, dass die ermittelten Ergebnisse auch für andere Netzwerke gültig sind und dass die Toll Fraud-Problematik bereits für alle Betreiber von SIP-Servern relevant ist.Voice over IP networks based on the Session Initiation Protocol (SIP) are becoming more and more widespread in the Internet due to functionality and cost advantages and will soon replace the classic telephony networks. Therefore, support of open SIP-based interfaces is an increasingly important requirement for IP-based Public Branch eXchanges (PBXs) and provider systems. The VoIP service allows using the personal or company VoIP account from any location worldwide. The migration of the telephony service from a closed and comparatively secure environment to a network with open interfaces creates security issues and opens up new opportunities for misuse and fraud. In this thesis, attacks are analyzed which result from introducing SIP-based voice services and do not belong to the area of contract regulations or attacks on the network layer. The attacker’s goal is to gain immediate financial benefit by making toll calls (international, cellular, premium services) via cracked third party accounts (“Toll Fraud”). To realize the threat analysis and the attack detection concepts, a SIP-based testbed and required software components were developed. In comparison to the related work, analyses with Honeypots were enhanced and a mechanism for automatic, distributed attack detection was realized. Therefore, for gathering the required data, a Honeynet with two class-C networks captured the SIP traffic for a period of six years. The automatic analysis is based on attacks and operates independently of single SIP messages. The field test results of this thesis demonstrate that SIP-based threats increase over time and attack tools are optimized and enhanced. The increasing number of Toll Fraud attempts to international or premium numbers reveals that Toll Fraud attacks can cause the account owner substantial financial damage in a very short amount of time if there is insufficient attack detection and mitigation. Hence, it is necessary to implement an attack detection which is able to identify the different attack stages and sends a notification to mitigation components before a Toll Fraud call is established. In this thesis, the Security Sensor System was developed to maximize the monitoring scope and to realize the distributed, automatic attack detection in real-time. The light-weight sensor component provides worldwide signature-based attack detection. Additional to the location-based attack detection, all attack notifications are sent to a central service which correlates the incoming alarm messages and provides a comprehensive attacker identification to inform mitigation components in real-time. The comparison of different sensor nodes in the Internet shows that the analyzed attack patterns do not only occur in the University testbed, but also temporally coherent in other networks. Thus, the results are valid for different network environments and it is crucial to know that Toll Fraud attacks are already performed in reality

    The role of different GFI1-expressionlevel in the progression of MDS and AML

    The presented data of this doctoral thesis suggest that the transcription factor GFI1/Gfi1 plays an important role in the progression and manifestation of AML. AML patients with low GFI1 expression in their blasts showed an inferior prognosis and a shortend EFS and OS. Furthermore, the KD of GFI1 resulted in a block of differentiation in the bone marrow of mice. The KO and the KD of GFI1/Gfi1 lead to an enhanced AML progression in the MLL-AF9 mouse model. In addition, upregulation of Gfi1 resulted in an increase of differentiation of bone marrow cells and a decrease of monocytosis in a persisting AML. It could be shown that the KO of Gfi1 impeded AML progression whereas the KD of GFI1 accelerated AML progression in the NUP98/HOXD13 mouse model. In cooperation with the the IRCM in Canada it could also be shown that H3K9 acetylation at GFI1 target genes is increased in the bone marrow of GFI1-KD-NUP98/HOXD13 mice. Further analysis of gene expression (Gene Set Enrichement Analysis) showed an upregulation of those genes that belong to "cluster 5" which were defined by Valk et al. as a subgroup of AML patients with poor prognosis. These data suggest, that the GFI1-KD-NUP98/HOXD13 mice recapitulate the situation of AML patients with an inferior prognosis very well. Moreover, the essential role of GFI1/Gfi1 for AML progression and manifestation in AML1-ETO9a- and CBFB-MYH1- mouse models could be demonstrated. Furthermore the here depicted results open up new vistas in treating AML patients with epigenetic drugs. As well as in murine as in human leucemic bone marrow cells it could be shown that cells with low GFI1 expression responded better to the treatment with the HATi CTK7a. This could be also confirmed in first in vivo experiments. Taken together, the presented data suggest that GFI1/Gfi1 plays a level- and dose-dependent role in progression of AML in the here used mouse models and that different GFI1/Gfi1 expression levels could be exploited as new prognostic factor in order to establish a new epigenetic therapy for MDS/AML patients

    Bestehende Bemessungsvorschläge zur Berücksichtigung einer Schweißnahtnachbeandlung mittels höherfrequenten Hämmerns (HFH) decken lediglich Stahlsorten bis maximal S960 und Blechdicken größer gleich 5 mm ab. Das Ziel der vorliegenden Arbeit ist die Untersuchung des Einflusses einer HFH-Behandlung auf das Ermüdungsverhalten geschweißter, ultrahochfester Feinkornbaustähle mit Streckgrenzen von 960 N/mm2 und höher im oberen Zeitfestigkeitsbereich und die Entwicklung eines Vorschlags zur Bemessung von HFH-behandelten Schweißverbindungen für diese Stahlsorten. Innerhalb von Ermüdungsversuchen wird der Einfluss einer HFHBehandlung an vier kranbautypischen Kerbdetails aus ultrahochfesten Feinkornbaustählen S960, S1100 und S1300 erfasst. Mit Hilfe analytischer und numerischer Methoden wird der Einfluss der aus dem Schweißprozess resultierenden Imperfektionen Axialversatz, Winkelverzug sowie der lokalen Nahtgeometrie auf den Beanspruchungszustand an der Nahtübergangskerbe untersucht. Auf Grundlage der eigenen Versuchsergebnisse und aus der Literatur ausgewerteten Versuchsreihen wird abschließend ein Bemessungskonzept zur Erfassung des Einflusses einer HFHBehandlung auf die Ermüdungsfestigkeit geschweißter, ultrahochfester Feinkornbaustähle S960 und höher entwickelt.Existing design recommendations for the consideration of the post weld treatment method high frequency hammer peening (HFHP) are limited to steel grades of S960 and plate thicknesses of 5 mm and higher. The intention of the present work is to investigate the influence of the HFHP treatment on the fatigue behaviour of welded ultra high strength fine grained structural steels with yield strengths of 960 N/mm² and higher within the upper finite fatigue life region and to develop a design proposal for HFHP treated welded joints for these steel grades. Within fatigue tests the influence of HFHP treatment on four typical welded notch details of mobile crane structures made of S960, S1100 and S1300 is determined. The influence of weld imperfections like linear misalignment, angular misalignment and the local weld geometry on the stress at the weld toe is evaluated by analytic and numerical methods. Based on the results of the own fatigue tests and evaluated fatigue test results of the literature a design concept for the consideration of the influence of HFHP treatment on the fatigue strength of welded ultra high strength fine grained structural steels S960 and higher is proposed
