Generic Adaptation Support for Wireless Sensor Networks
Wireless Sensor Networks are used in various and expanding application scenarios and are also considered to be important elements of the Internet of Things. They monitor and deliver data, which is not only used for research but to an increasing degree also in business environments. With the increasing complexity of these scenarios and the increasing dependency on the availability of the sensor network data, the requirements to a Wireless Sensor Network increase at the same pace.
Since Wireless Sensor Networks are typically implemented using resource-constrained platforms, sensor network algorithms are typically optimised for specific operating conditions such as static or mobile networks, high or low traffic etc. However, due to scenario complexity and dynamic real-world conditions a static configuration of a Wireless Sensor Network software cannot always meet the requirements. Moreover, these requirements of the sensor network's user can change over time, for example concerning accuracy. Therefore, the sensor network software has to adapt itself to cope with dynamic system conditions and user requirements.
This thesis presents the TinyAdapt and TinySwitch frameworks to solve the aforementioned problems. TinyAdapt, our generic adaptation framework for Wireless Sensor Networks, allows for the autonomous adaptation of arbitrary sensor network algorithms based on explicit and intuitively defined user preferences and on automatically monitored network conditions. Due to a two-phase approach, run-time adaptation is executed completely and efficiently on standard sensor node hardware and does not need support from, e.g., the base station. The creation of adaptive applications is guided by a complete workflow, which is presented as well.
When changing parameters of an algorithm is not enough to achieve the desired adaptation results, the algorithm has to be exchanged completely. However, several limitations of TinyOS and the sensor node hardware limit the use of simple code exchange by node reprogramming for efficient adaptation. TinySwitch, our generic switching framework, allows to switch between alternative algorithms that are already installed in parallel. TinySwitch analyses these algorithms, determines their dependencies and creates all code to enable one of the algorithms while isolating all others. Due to its minimal overhead, TinySwitch is perfectly suited for run-time adaptation in TinyAdapt
Preparation of Epitactically Grown Bismuth Films on Si(111) and NaCl and Investigation of their Electrical Conductivity
Bisherige Arbeiten zur elektrischen Charakterisierung dünner und dünnster
Wismutfilme erfolgten üblicherweise entweder an epitaktischen Filmen auf einem halbleitenden Siliziumsubstrat oder in Form von nicht-epitaktischen Filmen auf verschiedenen Substraten wie Quarzglas oder Glimmer. Im Rahmen der vorliegenden Arbeit entstand eine Ultrahochvakuumanlage, die neben der Präparation der Filme unter reproduzierbaren Bedingungen auch die elektrischen Messungen unter Ultrahochvakuum (UHV) - Bedingungen ermöglichte. Damit konnte im Gegensatz zu vielen bisherigen Arbeiten der Einfluss der Oxidation des Filmes an Luft ausgeschlossen werden. Weiterhin wurden die Untersuchungsmöglichkeiten an dünnen Wismutfilmen deutlich erweitert, indem erstmalig epitaktische Filme auf einem isolierenden Substrat gewachsen wurden. Zu diesem Zweck wurden NaCl - Einkristalle in einer neu entwickelten Präparationstechnik durch kurzzeitigen Kontakt mit Wasser bis hin zu einer atomar glatten Oberfläche geätzt. Auf dieser Fläche konnte schließlich Wismut epitaktisch auf NaCl aufgewachsen werden. Um Vergleichbarkeit mit bisherigen Veröffentlichungen herzustellen, wurden die Systeme Bi auf Si und nicht - epitaktisches Bi auf verschiedenen Substraten in die elektrischen Leitfähigkeitsmessungen einbezogen.Former experimentals works on the electronical properties of thin bismuth
films have shown controversially discussed results about the existence or nonexistence of a semimetal to semiconductor transition in such films. Most of these experiments had been performed either on a semiconducting silicon substrate or on substrates where no epitaxial film growth could be achieved. For the experiments presented in this thesis, a newly built ultrahigh vacuum system was planned and erected for preparation and electrical characterisation within one single vacuum chamber assembly.
In this work, new experiments are presented starting with epitaxially grown,
nearly single-crystalline films on insulating substrates as well as on low conductivity silicon substrates. To obtain a comparability with former works,
non-epitaxial films have been characterized as well under UHV conditions to

Study of the effects of the small molecule RITA on the growth of medulloblastoma
Zusammenfassend lässt sich sagen, dass RITA (Restoration of p53 and Induction of Tumor Apoptosis) eine Wirkung gegen humane Medulloblastomzelllinien hat. Der Signalweg ist bisher nicht sicher geklärt und verstanden.
Im Rahmen dieser Arbeit konnte gezeigt werden, dass RITA eine antiproliferative und apoptoseauslösende Wirkung auf Medulloblastomzellen in vitro und in vivo unabhängig des p53-Status hat. In den in vitro- Versuchen wurde ein Anstieg von den Proteinen p21, p53 und MDM2 nach RITA Behandlung gezeigt. Zusätzlich konnte ein Zellzyklusarrest bestätigt werden. Es sollte aber weiterhin im Rahmen von (auch klinischen) Studien untersucht werden, ob sich RITA eignet, Patienten mit Medulloblastomen zu behandeln, die keine p53-Mutation aufweisen, und evt. sogar auch Patienten, deren Medulloblastome ein mutiertes p53 haben.
In einigen anderen in vitro Studien konnten bisher die synergistischen Effekte RITAs mit anderen Molekülen untersucht werden, welche die p53 Funktion wiederherstellen sollen. Daher scheint es sinnvoll diese Kombinationsstudien auch für das Medulloblastom durchzuführen.
Die in vivo- Versuche zeigten eine signifikante Tumorwachstumsreduktion durch die RITA- Behandlung im Vergleich zur Kontrollkohorte. Da RITA in bisherigen in vivo Studien keine toxischen Effekte zeigt und zusätzlich die bisher damit untersuchten/behandelten Tumoren keine Resistenzentwicklung zeigen, lohnt es, sich die Funktionsweise weiterhin genauer zu untersuchen und sich RITA zu Nutze zu machen. Hiermit eröffnen sich neue, vielversprechende Möglichkeiten für therapeutische Anwendungen in der Klinik.Background: Medulloblastoma accounts for about 20 % of the brain tumors in children. With an overall survival of 40-70% despite multimodal therapy approaches (surgery, radiation and chemotherapy), medulloblastoma still represents a major clinical challenge in pediatric oncology. Since dysfunction of p53, because of mutations in the TP53 tumor suppressor gene, is rare in neuroectodermal embryonal tumors, including medulloblastomas, other mechanisms inactivating p53 have to be identified. One of the mechanisms is an overexpression of MDM-2. Based on the existing data, functional reactivation of p53 and/or inhibition of the p53/MDM-2 axis in medulloblastoma is widely considered to be a promising therapeutic option for this most common brain tumor in children. RITA (Restoration of p53 and Induction of tumour apoptosis) has been identified to be able to disrupt the MDM-2/p53 axis, as well as it has been proven to induce apoptosis very effectively so far in colon carcinomas and neuroblastomas for example. This project aims at identifying the therapeutic potential to interrupt the MDM-2/p53 axis in medulloblastoma with the small-molecule RITA.
Results: Through treatment of medulloblastoma cell lines with RITA the expression of MDM-2, p53 and p21 increased in vitro. Treatment of the cells with RITA reduced cell viability, although this was done independent of the p53 status. Another effect of the RITA treatment was a cell cycle arrest in the S- phase. Even the transfection of medulloblastoma cell lines with a dominant negative p53 did not change RITAs effect on the cells, in that matter that it still induced apoptosis.
We could also show an effect of RITA against the medulloblastoma in vivo. RITA was able to significantly reduce the tumor growth in mice treated with RITA as well as it enabled a significantly longer overall survival for the mice.
Concluding, RITA seems to be a promising substance in the line of small molecules to treat medulloblastoma in vitro and in vivo. But further understanding on how it works exactly is necessary in order to be able to use it as a therapeutic approach in medulloblastoma patients
Die Rolle der AAA-ATPase p97 und ihrer Kofaktoren beim regulatorischen Abbau von Substratproteinen nach DNS Schäden oder Replikationstress
The cellular response to radiation is governed by a complex signaling network that is regulated by the ubiquitin-proteasome system in multiple ways. The AAA-type ATPase p97 (also known as VCP or Cdc48) has emerged as a central element of the ubiquitin-proteasome system as it facilitates the degradation of critical substrate proteins by the proteasome. As such, it is involved in replication stress and DNA damage responses, and is currently explored as a cancer drug target in clinical trials. However, its function is still poorly understood. p97 cooperates with protein cofactors, three of which have been linked to the radiation response: the heterodimeric Ufd1-Npl4 ubiquitin adapter, DVC1 and UBXD7 that acts as an adapter to cullin-RING ubiquitin ligases (CRLs).
This work provides insight into three different aspects of the role of p97 and its cofactors in the response to replication stress and DNA damage in human cells. First, we investigated the relevance of p97 for DNA damage checkpoints. Second, we examined the significance of p97 and its cofactors for survival of human cancer cells after replication stress. Third, we explored the role of a novel p97 cofactor, UBXD7 that was linked to DNA-related functions of p97. Analyses of the G1/S and intra S checkpoints showed that p97Ufd1-Npl4 may only play a minor role in these checkpoints. However, DVC1 is crucial for the intra S checkpoint. Importantly, work from this study contributed to demonstrating that the p97Ufd1-Npl4 complex ensures robustness of the G2/M checkpoint by facilitating the degradation of the phosphatase CDC25A (published in Riemer et al, 2014). This study confirmed the relevant target protein of p97, CDC25A, in the checkpoint and contributed to the identification of the cofactors involved in CDC25A degradation, including the novel p97 cofactor UBXD7. Moreover, we could demonstrate that in mammalian cells, unrepaired DNA damage from interphase manifests in mitotic segregation errors in cells depleted of p97Ufd1-Npl4 and that the lack of either of these components decreased cell survival after replication stress. Furthermore, p97 inhibition with the inhibitor NMS-873 showed a synergistic effect with the genotoxic chemotherapeutic agent doxorubicin. Additionally, NMS-873 was able to delay the degradation of CDC25A in the DNA damage response. Finally, by investigating the significance of UBXD7 for diverse p97 substrate proteins, we found that UBXD7 is not generally involved in CRL- and p97-dependent degradation both in the nucleus and the cytoplasm. Moreover, a detailed structure-function analysis revealed that binding to CRLs and p97 is essential for UBXD7 function.Die zelluläre Strahlenantwort wird von einem komplexen Signalnetzwerk gesteuert, das auf vielfältige Weise vom Ubiquitin-Proteasom-System reguliert wird. Ein zentrales Element des Ubiquitin-Proteasom-Systems stellt die AAA-ATPase p97 (auch bekannt als VCP oder Cdc48) dar, welche die Degradation von kritischen Substratproteinen durch das Proteasom unterstützt. p97 ist involviert in die zelluläre Reaktion auf Replikationsstress und DNA-Schäden und wird gegenwärtig als Wirkstoffziel für Krebsmedikamente in klinischen Studien erforscht. Dennoch ist seine Funktion noch wenig verstanden. p97 kooperiert mit Protein-Kofaktoren, von denen drei mit der zellulären Strahlenantwort in Verbindung gebracht wurden: der heterodimere Ufd1-Npl4-Ubiquitin-Adapter, DVC1 und UBXD7, welches als Adapter für Cullin-RING-Ubiquitin-Ligasen (CRLs) fungiert.
Diese Arbeit bietet Einblick in drei verschiedene Aspekte der Rolle von p97 und seinen Kofaktoren in der Reaktion auf Replikationsstress und DNA-Schäden in menschlichen Zellen. Zunächst wurde die Relevanz von p97 für die Zellzyklus-Kontrollpunkte untersucht. Danach wurde die Bedeutung von p97 und seinen Kofaktoren für das Überleben humaner Krebszellen nach Replikationsstress überprüft. Schließlich wurde der neue p97-Kofaktor UBXD7, der mit Funktionen in Bezug auf DNA in Verbindung gebracht wurde, untersucht. Analysen des G1/S- und des intra-S-Kontrollpunkts haben gezeigt, dass p97Ufd1-Npl4 möglicherweise eine untergeordnete Rolle in diesen Kontrollpunkten spielt. DVC1 jedoch ist entscheidend für den intra-S-Kontrollpunkt. Arbeiten dieser Studie haben zu dem Nachweis beigetragen, dass der p97Ufd1-Npl4-Komplex die Robustheit des G2/M-Kontrollpunkts gewährleistet, indem er die Degradation der Phosphatase CDC25A unterstützt (publiziert in Riemer et al, 2014). Diese Arbeit bestätigt CDC25A als das relevante Zielprotein von p97 im Kontrollpunkt und trug zur Identifikation der an der CDC25A-Degradation beteiligten Kofaktoren bei, einschließlich des neuen p97-Kofaktors UBXD7. Darüber hinaus konnte gezeigt werden, dass sich in Säugerzellen mit herunterreguliertem p97Ufd1-Npl4 unreparierte DNA-Schäden aus der Interphase in der Mitose in Segregationsdefekten manifestieren. Zudem minderte der Mangel an einer dieser Komponenten das Überleben der Zellen nach Replikationsstress. Darüber hinaus wies p97-Inhibition mit dem Inhibitor NMS-873 einen synergistischen Effekt mit dem genotoxischen Chemotherapeutikum Doxorubicin auf. Zusätzlich verzögerte NMS-873 die Degradation von CDC25A in der DNA-Schadenantwort. Durch die Erforschung der Relevanz von UBXD7 für diverse p97-Substratproteine konnte schließlich gezeigt werden, dass UBXD7 nicht generell in CRL- und p97-abhängigem Abbau im Nukleus und Zytoplasma involviert ist. Eine detaillierte Struktur-Funktionsanalyse hat ergeben, dass die Bindung an CRLs und p97 für die Funktion von UBXD7 essentiell ist
Online Profiling - analysis and intervention to protect privacy
Online-Profiling wird insbesondere im Online-Marketing intensiv genutzt, um eigentlich anonyme Webseitenbesucher_innen zu kategorisieren und ihnen auf Basis ihres Surfverhaltens bestimmte Eigenschaften zuzuschreiben. Erweiterte Einsatzzwecke, wie etwa das Kreditscoring, zeigen allerdings, dass die vermeintlich anonymen Profile Auswirkungen auf die informationelle Selbstbestimmung und letztlich die Autonomie des_der Betroffenen haben. Neben einer Analyse der Hintergründe von Profiling und den Auswirkungen auf Privatheit, stellt diese Arbeit Möglichkeiten für Nutzer_innen vor, die Transparenz von und Intervenierbarkeit in Profile herstellen können. Dazu wurde ein neues Verfahren zur user-zentrierten Online-Tracking-Analyse entwickelt und der Nachweis erbracht, dass die vorgestellten Methoden zur Obfuscation (Verschleierung) einen Effekt auf die Interessenprofile eines Anbieters haben.Online-Profiling is heavily used in online marketing to categorize website visitors that consider themselves to be anonymous. Profiles are assigned based on Online-Tracking techniques that try to reconstruct a user's web history. The use for these profiles is continuously expanded and currently also in use for credit scoring. This development highlights the influence profiling has on the informational self-determination and therefore the autonomy of those being profiled. Besides an analysis of Online-Profiling this dissertation contains new approaches to transparency and intervenability for web users. A new methodology is used for a user-centered Online-Tracking study and we provide evidence that the obfuscation method we developed is effective in influencing the interest profiles created by one large tracking provider
Studies about the thaumarchaeal prolyl-isomerase NmPin from Nitrosopumilus maritimus
Peptidyl-Prolyl-Bindungen (Xaa-Pro) nehmen eine Sonderstellung innerhalb von Peptiden und Proteinen ein, da sie im Gegensatz zu nicht-prolinhaltigen Peptidindungen stabile cis-Isomere ausbilden können. Cis-ständige Xaa-Pro-Bindungen nehmen je nach Position innerhalb eines Proteins großen Einfluss auf dessen dreidimensionale Struktur und Funktion. Enzyme, welche die Rotation zwischen cis- und trans-Isomeren katalysieren, werden Peptidyl-Prolyl-cis/trans-Isomerasen (PPIasen) genannt und sind ubiquitär in allen Domänen des Lebens zu finden. In der Zelle erfüllen PPIasen eine wichtige Rolle bei der Faltung und Regulation einer Vielzahl von Proteinen.
In dieser Arbeit konnte erstmals die endogene Expression und detaillierte Charakterisierung eines archaealen Parvulins gezeigt werden. NmPin aus dem marinen, chemolithoautotrophen Thaumarchaeon Nitrosopumilus maritimus ist eine kleine single domain PPIase vom Parvulintyp (sdPar), welche in einem Protease-gekoppelten Aktivitätsassay Isomeraseaktivität gegenüber einer Reihe von unphosphorylierten, peptidischen Substraten zeigte. Mit Hilfe von NMR-Titrationsexperimenten und Mutagenesestudien konnte das aktive Zentrum und einzelne für die Katalyse benötigte Reste (D42, S82) in der dreidimensionalen Struktur von NmPin lokalisiert werden. Eine detaillierte Untersuchung der Ligandenbindung zeigte zudem die Beteiligung eines Netzwerks von oberflächennahen, hydrophoben, verzweigten Aminosäureseitenketten bei der Fixierung des Liganden in der Prolinbindetasche.
Bei genauer Analyse der Proteinoberfläche von NmPin konnte eine positiv geladene, Lysin-reiche Fläche identifiziert werden, die laut in vitro-Sedimentationsstudien für die Bindung von NmPin an Lipidvesikel verantwortlich ist. Bereits der Austausch eines zentralen, innerhalb der Thaumarchaeota konservierten Lysins (K7) gegen ein Glutamat führte zu einem Verlust der Bindungsfähigkeit. In Kooperation mit der Universität Aberdeen konnten immunofluoreszenz- und transmissionselektronenmikroskopische Aufnahmen von NmPin in N. maritimus-Zellen gemacht werden, welche die Bindung des Proteins von außen an die Zellmembran in vivo bestätigen. Diese Ergebnisse lassen eine Lokalisation der PPIase in einem pseudo-periplasmatischen Raum zwischen der Zellmembran und dem S-Layer vermuten. Potentielle zelluläre Substrate für NmPin stellen in diesen Raum sekretierte Proteine oder Proteine des S-Layers dar.Peptidyl-prolyl-bonds (Xaa-Pro) are exceptional in peptides and proteins due to their ability to form stable cis-isomers in contrast to all other non-proline peptide bonds. Relative to their position in a protein cis-isomeric Xaa-Pro bonds have a strong impact on the three dimensional structure and function of the protein. Enzymes which catalyse the rotation between cis- and trans-isomers are called peptidyl-prolyl-cis/trans-isomerases (PPIases) and are ubiquitously found in all domains of life. In the cell PPIases play a crucial role in protein folding and protein regulating processes.
This study is the first description of an endogenously expressed and in detail characterized archaeal Parvulin. NmPin from the marine, chemolithoautotrophic thaumarchaeon Nitrosopumilus maritimus is a small parvulin-type single domain PPIase (sdPar) which showed isomerase activity against a subset of unphosphorylated peptidic substrates in a protease-coupled activity assay. By using NMR titration experiments and mutagenesis studies the active site as well as single residues essential for catalysis (D42, S82) were identified in the three dimensional structure of NmPin. In addition a more detailed investigation of the ligand binding mechanism showed the participation of a network of surface exposed, hydrophobic, branched residue sidechains in ligand fixation inside the proline binding pocket.
A thorough topological analysis revealed a positively charged, lysine-rich patch on the protein surface which targets NmPin to lipid vesicles in vitro. The exchange of a single central lysine (K7), which is conserved in thaumarchaeotes, against a glutamate already leads to a loss of the binding ability. In cooperation with the University of Aberdeen immunofluorescence- and transmission electron microscopy of NmPin in N. maritimus cells was done, that confirms that the protein is attached to the outer cell membrane in vivo. These results point towards a PPIase located in a pseudo-perisplamatic space between the cell membrane and the S-Layer. Hence potential cellular substrates for NmPin are proteins secreted into this space or proteins of the S-Layer
Silver-nanoparticles with defined morphology - Synthesis, properties and biological impact
Im Rahmen dieser Dissertation wurden verschiedene Methoden zur Darstellung von Silber-Nanopartikeln mit unterschiedlicher Morphologie, aber vergleichbarer Größe, Ladung und Funktionalisierung erarbeitet. Primäres Ziel war die Anwendung im biologischen System, woraus sich besondere Anforderungen an die Reinheit, Konzentration und Stabilität der Partikel ergaben. Es wurden im Wesentlichen fünf verschiedene Partikelsysteme synthetisiert und mit verschiedenen analytischen Methoden charakterisiert. Weiterhin wurde die Auflösungskinetik und die biologische Wirkung gegenüber eukaryotischen und prokaryotischen Zellen bestimmt.In this work, silver nanoparticles with different shapes but comparable size and identical surface functionalization were prepared. The primary objective was the application in biological media, resulting in specific requirements concerning the purity, concentration and stability of the particles. Mainly, five different particle systems were synthesized and characterized by different analytical methods. Furthermore, dissolution kinetics were determined and the biological effect towards eukaryotic and prokaryotic cells was examined
“Constructing a New Socialist Countryside” in Contemporary China: Strategic Groups and Institutional Change
The Chinese Communist Party (CCP) leadership’s recent approach to narrowing the widening urban-rural gap is set forth in the comprehensive development policy for rural China: “Constructing a New Socialist Countryside” (shehui zhuyi xinnongcun jianshe, CNSC). This policy was a core part of the 11th Five-Year Plan in 2006. Like all other central rural development programs, CNSC is ultimately implemented by county and township governments. Arising from the idea of the “developmental state”, this dissertation delineates how the state is involved in the political economy of institutional change at local levels. Further elaborating on Oi’s notion of “local state corporatism” and Heberer and Schubert’s “strategic group” approach that provide a set of concepts for analysis, this study applies and contributes to these theoretical and analytical frameworks by examining the roles of local officials, coal mine owners and villagers, as well as the agency problems and relationship between the local state and businesses.
Employing a qualitative inquiry strategy and using in-depth interviews, field observations and extensive archival research of the comparative case studies in Fugu County, this dissertation examines how the local state used managerial strategies - “Local State Entrepreneurial-Developmentalism” - to change the ownership structure from private dominated to mixed ownership in order to reinstate gradually the state ownership of the private enterprises and to expand the tax base to promote the county-wide economy. Consequently, the institutional change in ownership structure reduced the total information and transaction costs and the bargaining power of private capital, on the one hand, and on the other hand, it reinforced the state-business symbiotic relationship and intensified agency problems.
Furthermore, using the “strategic group” approach in the analysis, this study investigates how the county and town cadres established strategic cooperation to co-opt local coal mine owners/entrepreneurs into “constructing a new socialist countryside”. It argues that Corporate Social Responsibility (CSR) was used by the local officials as a political label to justify their efforts to establish new redistributive institutions. Formal and informal institutions closely interacted in the process of CNSC implementation, in other words, in the formation of strategic groups and in the process of extracting matching funds, redistribution and conflict mediation through pairing schemes. These empirical patterns of local entrepreneurial-developmental state and strategic groups contribute to clarifying how agencies structure within China as a developmental state
Charakterisierung zirkulierender Tumorzell-Subgruppen bei Nicht-kleinzelligem Lungenkarzinom (NSCLC) und ihre klinisch-prognostische Relevanz
Zirkulierende Tumorzellen (CTC) haben sich aufgrund ihrer klinisch-prognostischen Aussagekraft zu einem vielbeachteten Gegenstand der Tumorforschung entwickelt.
Im Rahmen dieser Arbeit wurden zirkulierende Tumorzellen (CTC) von Patienten, die an einem Nicht-kleinzelligen Lungenkarzinom (NSCLC) leiden, mithilfe immunomagnetischer Beads angereichert und isoliert. Diese Zellen wurden auf ihre Anwendbarkeit hinsichtlich eines möglichen prognostischen Vorhersagewertes mittels der Multi-Immunfluoreszenzfärbung und der real-time quantitativen Reverse-Transkriptase-Polymerasekettenreaktion untersucht.
Es wurden solche Marker ausgewählt, die eine Differenzierung zwischen epithelialen, mesenchymalen und stammzellartigen Eigenschaften ermöglichen. Ein etwaiger Zugewinn mesenchymaler oder stammzellartiger Eigenschaften der Tumorzellen im Kontext einer Epithelial-Mesenchymalen Transition wird in der Literatur mit einem aggressiveren Wachstum, höherem Metastasierungspotential und gesteigerter Resistenz gegenüber Chemotherapeutika in Verbindung gebracht. Das CTC-„Profil“ eines Patienten könnte so also in der Lage sein, wertvolle Informationen über den individuellen Krankheitsverlauf zu liefern.
In der vorliegenden Arbeit weisen vor allem die Anwesenheit N-Cadherin-positiv gefärbter Zellen bei Patienten sowie verhältnismäßig erhöhte Expressionen von Keratin18, EpCAM und der Aldehyddehydrogenase1 auf eine prognostische Relevanz hin.
Diese Arbeit zeigt Ansätze auf, sich der Anwendbarkeit von zirkulierenden Tumorzellen bei NSCLC-Patienten als prognostisches Werkzeug und als Möglichkeit zum individuellen Therapiemonitoring bzw. zur Therapiesteuerung einer Platin-basierten Chemotherapie zu nähern.
Ein entscheidendes Ergebnis dieser Arbeit ist der Nachweis individueller Variationen der Zusammensetzung zirkulierender Tumorzellen beim NSCLC, welche mit unterschiedlichen klinischen Parametern korrelieren.
Weiterhin konnte belegt werden dass diese Zellen sich als Ausgangspunkt für sogenannte „Downstream“-Analysen eignen (beispielsweise FICTION-Technik)
