    The making of minority language policies in Chinese schools. Street-level bureaucracy ad curriculum decisions in Xishuangbanna, Southwest China

    This dissertation explores the relationship between institutions and decision-making within policy implementation at schools. Based on the street-level bureaucracy framework, an analytical framework that views policy implementation as a process that depends on the decisions by personnel in delivery agencies against the background of institutions, this dissertation examines the following research question: How do institutional settings of school and curriculum management affect the implementation of education in minority languages at school level? This research question is being scrutinized with an explorative case study on education in Dai-language at a set of schools that are all located in Xishuangbanna, a multilingual prefecture in Southwest China, but that differ in their institutional settings. The study is based on data that derives from interviews that the author has conducted with school personnel, with officers from the educational administration, and with parents and students, as well as from classroom observations and the analysis of official statistics. The first chapters present the theoretical framework, the most relevant policies concerning education in minority languages in China, the institutional background for school-based decision-making, as well as the linguistic characteristics of Xishuangbanna. Against this background the main part analyses decision-making by school personnel. This analysis focuses on three aspects: considering roles and interests, interpreting policies and institutional spaces, and making decisions and justifying them. The existing institutional settings concerning education in Dai-language structure the implementation process by opening up spaces for decision-making, which have to be filled-up by school personnel through interpretation and consideration. School personnel thus gain the role of decision-makers, although they suffer from the dilemmas that vague policy goals provide. In a final part this thesis also discusses possible consequences of policy implementation through school personnel’s decision-making on both the representation of ethnic minorities and the adjustment of school education towards students’ needs. Concerning both questions, however, decision-making by school personnel is unable to meet the high expectations that have been formulated by researchers and politicians. Instead, this mode of policy implementation rather allows the state to present local diversity at schools as a promise to China’s ethnic minorities, without threatening the state’s authority in prescribing educational goals.Diese Arbeit untersucht das Verhältnis von Institutionen und Entscheidungsfindung bei der Politikimplementierung an Schulen. Basierend auf dem „street-level bureaucracy“-Ansatz, welcher Politikimplementierung als einen durch die Entscheidungen von Personal in Dienstleistungsagenturen vor dem Hintergrund von institutionellen Rahmenbedingungen geformten Prozess begreift, geht sie der Fragestellung nach, wie institutionelle Rahmenbedingungen von Schul- und Curriculumsverwaltung die Implementierung politischer Vorgaben zum Unterricht in Minderheitensprachen durch Schulpersonal beeinflussen. Diese Fragestellung wird mittels einer explorativen Fallstudie zu Dai-sprachlichem Unterricht an Schulen mit verschiedenen institutionellen Rahmenbedingungen in Xishuangbanna, einer multilingualen Präfektur im Südwesten Chinas, untersucht. Die Studie basiert auf Daten, die der Autor während zweier Feldforschungsphasen durch Interviews mit Schulpersonal, Beamten der Bildungsbehörden, Eltern und Schülern, durch Unterrichtsbeobachtungen sowie durch die Auswertung offizieller Statistiken gesammelt hat. Zunächst wird in der Arbeit der theoretische Rahmen von Politikimplementierung, dann die relevanten Politiken zu Minderheitensprachbildung in China, der institutionelle Hintergrund für schulbasierte Entscheidungsfindung sowie die linguistische Charakteristika der Fallregion Xishuangbanna vorgestellt. Darauf aufbauend wird dann im Hauptteil die Entscheidungsfindung von Schulpersonal unter drei Aspekten analysiert: die Berücksichtigung von institutionellen Rollen und Interessen, die Interpretation von Politiken und Entscheidungsspielräumen und das Fällen und Begründen von Entscheidungen. Die existierenden institutionellen Rahmenbedingungen zum Dai-sprachlichen Unterricht in Xishuangbanna strukturieren den Implementierungsprozess, indem sie Entscheidungsspielräume eröffnen, welche das Schulpersonal durch Prozesse der Interpretation und Abwägung füllen muss. Das Schulpersonal erhält dadurch die Rolle von Entscheidern, leidet jedoch auch unter dem Dilemma unklar definierter Politikziele. In einem Ausblick blickt die Arbeit auf mögliche Konsequenzen der Politikimplementierung durch Entscheidungen von Schulpersonal auf die Repräsentation ethnischer Minderheiten in der Gestaltung von Politik und der Ausrichtung von Schulbildung an den Bedürfnissen von Schülern in China. In beiden Bereichen verhindern die gegebenen Institutionen der Evaluierung von Schulen jedoch, dass sich die von Politik und Forschung in die Entscheidungsfindung durch Schulpersonal gesetzten Hoffnungen erfüllen können. Stattdessen erschafft diese Form der Politikimplementierung ein System, welches dem chinesischen Staat die Gelegenheit gibt, mit vordergründig lokaler Diversität in Schulen ein Versprechen an die ethnischen Minderheiten des Landes einzulösen, ohne seine Autorität über die Festschreibung von national einheitlichen Bildungszielen zu verlieren

    Short and long term radiation effects after ionizing radiation and its dependence on chromatin modification and repair inhibition in human lung cell lines

    The dynamic nature of chromatin and its modifications play a critical role in transcriptional gene regulation and thus can affect several cellular functions such as differentiation, replication, DNA damage response and activation of cell death. Alterations in cellular epigenetic and genetic structure after IR can lead to long term effects, such as survival and genomic instability. Additionally, DSB repair is thought to be differentially regulated in euchromatin and heterochromatin. Histone methylation is an important modification related to the compactness and transcriptional activity of chromatin. Thus histone methyltransferases (HMT) play a pivotal role in determining the functional status of chromatin. The human HMT SUV39h1 and SUV39h2 are altered in various types of human cancers. Here we show that the knockout Suv39h1/2 mice fibroblasts (Fbs) have significantly lower clonogenic survival, which is attributed to reduced DSB repair capacity of these cells. The downregulation of HMT SUV39H1 in human lung cancer cell lines leads to significantly reduced number of radiation-induced residual γH2AX. However, the effect of SUV39H1 downregulation on clonogenical survival was less marked as compared to the mouse knockout model. Additionally, the HMT inhibitor Chaetocin, significantly reduces the radiation dose necessary to control 50% of a plaque-monolayer culture after treatment with low concentrations, reduces the cell proliferation and significantly increased apoptosis in lung cancer cell lines. Moreover Chaetocin was found to remodel the nuclear chromatin structure by forming chaetocin-induced chromatin condensation/clustering which is mainly associated with heterochromatin domains in primary human Fbs, transformed mFbs and in the tumour cell line H1299

    Conformational Equilibria of Organic Adsorbates on Nanostructures in Aqueous Solution. MD Simulations

    Atomistic molecular dynamics (MD) simulations of bulk aqueous solution of NaCl and functionalized gold nanoparticles (GNPs) in aqueous solution were conducted at 300 K. Bulk NaCl solution systems of different concentration have been studied using various force field parameter sets to investigate aggregation of ions and to choose suitable force field parameters for ions. Gold nanoparticles functionalized with linear and branched hydrocarbon chains with different grafting density (d f =1, 2/3 and 1/3), chain length ((CH 2 ) l , l=5, 11, 17 and 23), position of the branching point (4th, 8th and 12th carbon) and terminal groups (CH 3 , COO − and NH +3 ) were modeled to investigate various phenomena at the microscopic level. The solution of a functionalized GNP with ionic terminations was neutralized by excess Na + and Cl − ions. Focus has been given to the penetration depth of water and ions into the diffuse shell of alkanethiols as a function of grafting density, functionalization and chain length. Special focus has been given to the relative accessibility of the gold core by water in dependence of grafting density, chain length and chain shape. The orientation of water molecules inside the hydrocarbon chain environment (from the surface of the gold core to the terminal groups) and near the terminal groups has been studied in detail. Hydrogen bonds between water and polar terminal groups have also been studied. Special attention has been given to the solvent accessible surface area (SASA) of functionalized GNPs, and the coating asymmetry and ways to prevent it. It is observed that the penetration of water and ions into the hydration shell of alkanethiols increases with decreasing grafting density, irrespective of the terminal group of the hydrocarbon chains. High grafting densities lead to more extended hydrocarbon chains which behave more rigidly. The solvent accessible surface area (SASA) is dependent on the chain length and the terminal group. SASA (per unit chain length) of COO − group terminated GNPs with chain length (C l ) 5 is the highest among all GNP coated with linear chains. Water molecules orient near the surface of the gold core pointing one hydrogen towards the center of the gold core irrespective of chain length, terminal group and grafting density. Water molecules cannot access the surface of the gold core of a branched chain GNP as easily as in the linear chain GNP. Branched chains protect the core better than the corresponding linear chains. Among all studied systems, nonpolar group terminated branched chains with chain length 17 and branched at the 4th carbon atom provide the best protection to the nanoparticle core. Coating asymmetry is generally more prominent for longer chains (>11), and the asymmetry can be prevented by introducing branching points

    Towards understanding bacterial induced sepsis: RNA recognition by murine TLR13 and human TLR8

    Immune defense against infection depends on recognition of invading pathogens by innate pattern recognition receptors (PRRs) localized in the plasma- or endosomal membrane and the cytoplasm. Toll-like receptors (TLR) are an important family of PRRs detecting bacterial pathogen associated molecular patterns (PAMPs). Blood borne bacteria tend to over-amplify inflammatory signals upon infections, which often causes septic shock. Blockade of TLR2 and -4 in combination with antibiotic therapy efficiently inhibits sepsis pathology upon Gram-negative bacterial infection in mice. Major host receptors of Gram-positive bacteria beyond TLR2 had not been identified previously. Here, I show that recognition and killing of Gram-positive bacteria by macrophages depends on an endosomal TLR beyond TLR3, -7, -8, -9, -11 and -12. Immune activation upon Streptococcus pneumoniae D39 infection or heat inactivated Staphylococcus aureus (hiSa) challenge was abrogated in murine MyD88/Trif-/- bone marrow derived macrophages (BMMs). In contrast, Tlr23479-/- BMMs recognized and killed these bacteria like wt cells, yet 3D/Tlr24-/- BMMs lacking endosomal TLR function were impaired in both. According to common view, the bactericidal activity of host cells involves production of reactive oxygen species (ROS) or nitric oxide (NO). However, p47/Phox-/- and iNOS-/- BMMs were unremarkably competent in S. pneumoniae D39 killing. Bacterial recognition and killing capacity was carried by cell culture supernatants since bacteria were killed when confronted with cell free supernatant from pre-stimulated wt BMMs, indicating extracellular killing independent of phagocytosis. My results suggest involvement of other effector molecules such as antimicrobial peptides in macrophage bactericidal activity. I also show that murine TLR13 recognizes a conserved bacterial 23S ribosomal (r)RNA segment of 10 nt (5'-CGGAAAGACC-3') which also binds macrolide, lincosamide and streptogramin (MLS) antibiotics (including erythromycin). RNase A treatment of hiSa abrogated Tlr23479-/- cell activation. Consequent analysis of bacterial ssRNA molecular subspecies indicated 23S rRNA as TLR13 ligand. Detection of S. aureus RNA was conferred by TLR13 upon its over expression in HEK293 cells, while its mRNA knockdown upon siRNA transfection into BMMs abrogated it. The TLR13 activating 23S rRNA segment contains adenosine (A)2085 in S. aureus. Its N6-adenosine methylation or mutation mediated by specific methyltransferases (ErmC and ErmB) is known to confer antibiotic resistance and at once abrogated activation of TLR13. Hence, 23S rRNA from erythromycin resistant clinical S. aureus isolates grown in the presence of erythromycin as well as synthetic oligoribonucleotides (ORN) methylated at the respective adenosine or mutated towards guanosine mimicking resistance conferring modifications failed to activate TLR13. Over expression of ErmB or ErmC in erythromycin susceptible bacteria resulted in N6-adenosine methylation as shown by HPLC and masking towards TLR13. Thus, my data identify both TLR13 and its highly conserved ligand, as well as a mechanisms of antibiotic resistance qualifying as bacterial immune escape strategy to avoid TLR13 driven recognition. Furthermore, I show that human TLR8 not only replaces TLR13 but also exceeds its specificity by promiscuously sensing bacterial and mitochondrial rRNA in a merely URR motif dependent manner. RNase A treated hiSa activated hPBMCs only via TLR2, implying ssRNA as major immune stimulant. PBMCs were MyD88 dependently activated by bacterial 23S (Sa19) or mitochondrial Sa19-like 16S rRNA (BtmtD3_4) derived ORNs also when methylated or mutated, as well as by all bacterial rRNAs. Comparative transcriptome profiling of Sa19 responsive differentiated human monocytoid THP-1 cells indicated correlation of TLR8 expression with Sa19 responsiveness. Accordingly, Unc93b1-/-- and Tlr8-/--THP-1 cells as well as siRNA driven knockdown and ectopic TLR8 expression confirmed this statement. Responsiveness of Tlr8-/-- and Unc93b1-/- THP-1 cells towards bacterial infection was abrogated upon TLR2 blockade (mAb T2.5). Additional endosomal inhibition (chloroquine) resulted in a substantial decrease of PBMC activation. Endosome inhibition by chloroquine and simultaneous antibiotic treatment efficiently inhibited bacterial infection driven inflammatory activity of human whole blood. Therapy at the early phase of bacterial sepsis might thus benefit from TLR8 and -2 blockade besides antibiotic therapy.Die Immunabwehr gegen Infektionen hängt von der Erkennung invasiver Pathogene durch Mustererkennungsrezeptoren (PRR) ab, die in der Plasma- oder endosomalen Membran bzw. im Zytoplasma lokalisiert sind. Toll-like Rezeptoren (TLR) bilden eine wichtige PRR Familie, die z. B. bakterielle Pathogen-assoziierte molekulare Muster (PAMP) erkennt. Das angeborene Immunsystem immunsupprimierter Individuen tendiert zu überschießenden inflammatorischen Reaktionen auf bakterielle Infektionen. TLR2 und -4 Blockade kombiniert mit Antibiotika-Gabe schützt mit Gram-negativen Bakterien infizierte Mäuse vor Sepsispathologie. Spezifische Wirtsrezeptoren für Gram-positive Bakterien von TLR2-ähnlicher Wichtigkeit waren zuvor unbekannt. Ich zeige hier, dass die Erkennung und Abtötung Gram-positiver Bakterien durch Makrophagen in Abhängigkeit eines endosomalen TLR, jenseits von TLR3, -7, -8, -9, -11 und -12 erfolgt. Immunaktivierung durch eine Streptococcus pneumoniae D39 Infektion oder eine Konfrontation mit Hitze-inaktiviertem Staphylococcus aureus (hiSa) war in murinen MyD88/Trif-/- Knochenmarksmakrophagen (BMM) abwesend. Tlr23479-/- und wt BMM erkannten und töteten die Bakterien gleichermaßen. 3D/Tlr24-/- BMM, deren endosomale TLR unfunktional sind, waren in beiderlei Hinsicht nahezu inaktiv. Die bakterizidale Aktivität hängt insofern lediglich in geringem Maße von der Produktion reaktiver Sauerstoffspezies oder Stickstoffmonoxid (NO) ab, als dass p47/Phox-/- und iNOS-/- BMMs normal reagierten. Die Erkennung und Eliminierung der Bakterien wurde anhand der Zahl lebender Bakterien sowie des Gehaltes an NO und Zytokinen in Kulturüberständen untersucht. Bakterien wurden bereits durch die Konfrontation mit zellfreien Überständen von vor-stimulierten wt BMM abgetötet. Dies spricht für eine Phagozytose-unabhängige und extrazelluläre Abtötung. Dieses Ergebnis impliziert eine Beteiligung anderer Effektormechanismen an der bakterizidalen Aktivität von Makrophagen, wie möglicherweise die Freisetzung antimikrobieller Peptide. Ich zeige hier auch, dass der murine TLR13 ein hochkonserviertes 23S rRNA Segment einer Länge von 10 nt (5'-CGGAAAGACC-3') erkennt, das ebenso eine Bindestelle von Makrolid, Lincosamid und Streptogramin (MLS) Antibiotika ist. RNase A Behandlung von hiSa hob seine Aktivierung in Tlr23479-/- BMM auf. Die Untersuchung bakterieller RNA Subspezies identifizierte 23S rRNA als TLR13 Ligand. TLR13 Überexpression in HEK293 Zellen und Knockdown mittels siRNA in BMM wies ebenfalls darauf hin. Mutation (A→G) tragen um Resistenz-vermittelnde RNA-Modifikationen zu immitieren, aktivierten TLR13 nicht. Überexpression von ErmB oder ErmC in zuvor Erythromycin sensitiven Bakterien hatte den gleichen Effekt. Die N6-Adenosin Methylierung war mittels HPLC von 23S rRNA darstellbar. Meine Daten identifizieren daher sowohl einen hochkonservierten TLR13 Liganden, als auch dass Antibiotikaresistenzmechanismen als bakterielle Immunevasions-Strategien wirksam sind. Desweiteren zeige ich, dass humaner TLR8 im Gegensatz zu murinem TLR13 jegliche bakterielle und auch mitochondrielle rRNA erkennt. Die TLR2 abhängige PBMC Aktivierung durch RNase A behandelten hiSa implizierte ssRNA als stimulatives PAMP. PBMC wurden in Abhängigkeit von MyD88 durch bakterielle 23S (Sa19) oder mitochondrielle 16S rRNA (BtmtD3_4, Sa19 abgeleitetes ORN) aktiviert, auch wenn diese methyliert oder mutiert waren, sowie durch alle bakteriellen rRNAs. Das ermittelte Konsensus Motiv ist UR/URR. Eine vergleichende Transkriptomanalyse von Sa19 responsiven differenzierten humanen monozytoiden THP-1 Zellen im Vergleich mit Kontrollen implizierte TLR8. Unresponsive Unc93b1-/-- und Tlr8-/--THP-1 Zellen, sowie siRNA vermittelter Knockdown und ektopische TLR8 Überexpression implizierten ebenfalls TLR8 als Rezeptor. Die Aktivierung von Tlr8-/-- and Unc93b1-/- THP-1 Zellen durch bakteriellen Infektionen wurde von einer TLR2 Blockade (mittels mAb T2.5) verhindert. Zusätzliche endosomale Hemmung (Chloroquin) führte in PBMC zu substantiell verminderter Aktivierung. Eine TLR8 Blockade mittels Verabreichung von Chloroquin und zeitgleicher Antibiotikabehandlung verhinderte die Bakterien getriebene endzündliche Immunaktivität einer humanen Vollblutkultur. Die kombinierte TLR8 und -2 Blockade mit Antibiotika Gabe könnte unseren Daten zufolge der Therapie der frühen Phase bakteriell induzierter Sepsis zuträglich sein

    Laserbasierte Modifikation von porösen TiO2 -, TiO2/Au- und Au- Materialien

    Der Fokus dieser Arbeit liegt auf der laserbasierten Modifikation von porösen Titandioxid- und Gold-Materialien. Poröse Materialien sind aufgrund ihrer großen Oberfläche und den damit verbundenen Eigenschaften z.B. für photovoltaische und -katalytische Anwendungen von Interesse. So kann der Lichteinfang und die Anzahl von katalytisch aktiven Zentren mit steigender Oberfläche erhöht werden. Große Oberflächen können durch Nanomaterialien, also Materialien mit einer Strukturgröße unter 100 nm, erreicht werden. Nanomaterialien können in verschiedenen Formen vorliegen. Die bekannteste Form sind Nanopartikel. Diese können großtechnisch mit verschiedenen Verfahren in ausreichender Menge hergestellt werden. In Form von Pulvern ist der Einsatz von Nanopartikeln allerdings nur begrenzt möglich. Erst durch die Verbindung der Nanopartikel untereinander und somit der Bildung eines festen, nanoporösen Netzwerks kann der elektrische Kontakt hergestellt und die elektrische Leitfähigkeit verbessert werden. Die Bildung eines nanoporösen Netzwerks kann durch Sintern von Nanopartikeln erreicht werden. Konventionell werden dabei Nanopartikelschichten im Ofen erhitzt, so dass die Partikel miteinander versintern. Dieses Verfahren hat den Nachteil, dass es nur eine homogene Bearbeitung der kompletten Schicht erlaubt. Häufig ist es jedoch von Vorteil nicht eine homogene, sondern eine für die Anwendung optimierte, strukturierte Schicht zu erhalten. Neben einer Anpassung der Struktur kann auch die chemische Modifikation nanoporöser Materialien für viele Anwendungen von Vorteil sein. Eine Möglichkeit, nanoporöse Materialien im Mikro-/Nanometer-Bereich zu bearbeiten und somit die porösen Strukturen und die chemische Zusammensetzung für verschiedene Anwendungen anzupassen, bietet die Laserbearbeitung. Bei der Laserbearbeitung wird ein Laserstrahl auf die Oberfläche fokussiert. Durch Absorption des Laserlichts erwärmen sich die Partikel und können lokal versintern. So können verschiedene Strukturen erzeugt oder die Materialien chemisch modifiziert werden. Durch die Verwendung von Lasern unterschiedlicher Wellenlänge kann die Laserenergie selektiv in verschiedenen Materialien absorbiert werden und dort verschiedene Prozesse induzieren. Die Laserbearbeitung zeichnet sich darüber hinaus durch einen geringen technischen Aufwand, eine hohe Bearbeitungsgeschwindigkeit sowie ein einfaches Upscaling im Hinblick auf eine Serienfertigung aus. Ziel dieser Arbeit ist es, die Laserbearbeitung von nanoporösen TiO2 - und TiO2/Au-Nano-partikelschichten sowie von nanoporösem Gold für photovoltaische und spektroskopische Anwendungen zu demonstrieren. Die Veränderung dieser Materialien durch die Laserbearbeitung wird in Abhängigkeit von den Laserparametern analysiert. Aus diesen Daten werden Rückschlüsse auf die zugrundeliegenden Prozesse und Mechanismen gezogen. Darüber hinaus wird der Einsatz solcher laserbearbeiteter Schichten in Farbstoffsolarzellen bzw. laserbearbeiteter, nanoporöser Goldsubstrate in spektroskopischen Anwendungen dargestellt. Hierdurch werden neue Wege zur Herstellung und Modifikation von nanoporösen Materialien aufgezeigt.The focus of this work is the laser-based modification of porous titanium dioxide and gold materials. Porous materials are of interest for photovoltaic applications and -catalysis due to their large surface area and associated properties. Thus, the light trapping and the number of catalytically active centres can be increased with increasing surface. Large surfaces can be achieved by nanomaterials, i.e. materials with a feature size of less than 100 nm.The most popular form of nanomaterials are nanoparticles. These can be prepared industrially by various methods in sufficient quantity. In the form of powders, the use of nanoparticles, however, is limited. Only by connection of the nanoparticles with each other and hence the formation of a solid, nanoporous network, the electrical contact is made and the electrical conductivity can be improved. The formation of a nanoporous network can be accomplished by sintering of nanoparticles. A conventional method is to heat nanoparticle layers in the oven, so that the particles sinter together. This method has the disadvantage that it only allows a homogeneous treatment of the entire layer. Often, however, it is advantageous to create patterned layer, that is optimised for a specific application. In addition to an adjustment of the structure, the chemical modification of nanoporous materials can be beneficial for many applications. Laser processing offers a way to process nanoporous materials in micro- and nanometer range and thus optimise the porous structures and chemical composition. In laser processing, a laser beam is focused onto the surface. Through absorption of the laser light, the particles heat up and sinter locally. Thus, various structures can be created or the materials can be chemically modified. By the use of lasers of different wavelength, the laser energy can be selectively absorbed in different materials and there induce various processes. The laser processing is further characterised by low technical complexity, a high processing speed and a simple upscaling with a view to mass production. The goal of this work is to demonstrate laser processing of nanoporous titania and hybride titania/gold nanoparticle layers and nanoporous gold for photovoltaic and spectroscopic applications. The modification of these materials by the laser processing is analysed as a function of the laser parameters. From these data, conclusions about the underlying processes and mechanisms are drawn. Moreover, the use of such laser-processed layers in dye sensitised solar cells or laser-processed nanoporous gold substrates is shown in spectroscopic applications. In this way, new routes for the preparation and modification of nanoporous materials are shown

    How women change medicine: a case study in a German hospital

    In the last decade, Germany has seen a significant increase in the number of female physicians. Literature on the topic suggests that this so called “feminization of medicine” will engender changes in the profession’s structure and the practice of medicine, resulting in an augmenting demand for part-time work, more women in high level positions and a stronger patient-centered and humane care. In order to analyze how feminization has influenced medicine, I conducted qualitative, semi-structured interviews with 21 male and female physicians holding diverse positions in a department of Surgery, a Breast Center and an Internal and Integrative Medicine department of a hospital in the Ruhr area in Germany. Using qualitative content analysis as an analytical framework, this thesis explains that an increase in the number of women doctors has taken place in the departments, but that this has not yet caused for major transformations to come about. Due to the gendered substructure of the departments, part-time work, with the exception of the complementary and alternative medicine unit in the Breast Center, is difficult to accomplish as is the compatibility of work and family life. Furthermore, clear guidelines on the provision of care leave little room for change in the practice of medicine. Most physicians do not see room to alter matters, with which organizational structures, cultures and practices in the distinct departments are in many facets reproduced instead of transformed. It is, however, not the case that the feminization process has not engendered any change at all. But modifications are only allowed when consonant with the dominant organizational culture, structures and prevalent gender stereotypes. As this thesis shows, the increase in female physicians has thus not automatically caused for qualitative changes to take place. It is rather the interplay between the agency of the physicians and organizational cultures and structures that determines how the increase of female physicians is affecting the organization and practice of care.In den letzten Jahren hat Deutschland einen deutlichen Zuwachs an weiblichen Ärzten erlebt. Gemäß der Literatur zur „Feminisierung der Medizin“ wird der Zuwachs an weiblichen Ärzten die Strukturen im Krankenhaus und die Praxis der Medizin verändern und zu mehr Teilzeitstellen, zu mehr Frauen in Führungspositionen und zu einer humaneren und stärker patientenzentrierten Versorgung führen. Um den Einfluss der Feminisierung auf die Medizin zu analysieren, führte ich qualitative, semi-strukturierte Interviews mit 21 Ärzten und Ärztinnen in verschiedenen Positionen in einer chirurgischen Abteilung, einem Brustzentrum sowie einer Abteilung für Innere und Integrative Medizin in einem Krankenhaus im Ruhrgebiet in Deutschland durch. Unter Verwendung von qualitativer Inhaltsanalyse erläutert diese Thesis, dass es einen deutlichen Anstieg des Ärztinnenanteils in den Abteilungen gegeben hat, aber dass dieser nicht zur großen Transformationen führte. Mit Ausnahme der Sektion der komplementären und alternativen Medizin des Brustzentrums, ist Teilzeitarbeit sowie die Vereinbarung von Arbeit und Familie wegen der gendered Substruktur der Abteilungen schwierig zu verwirklichen. Außerdem lassen klare Leitlinien in der medizinischen Versorgung die praktische Anwendung betreffend wenig Raum für Veränderungen. Dieses führt dazu, dass die Organisationsstrukturen und -kultur und die medizinischen Anwendungsverfahren der Abteilungen in den meisten Fällen reproduziert statt geändert werden. Es ist jedoch nicht so, dass der Feminisierungsprozess gar keine Änderungen nach sich gezogen hat. Die Modifikationen sind allerdings nur erlaubt, wenn sie mit der dominanten Organisationskultur und den generellen Stereotypen harmonieren. Die gestiegene Zahl an Ärztinnen selbst hat also nicht direkt für qualitative Änderungen in die Abteilungen gesorgt, sondern vielmehr bestimmt das Zusammenspiel zwischen ärztlichem Agens und der Organisationsstruktur und –kultur den Effekt der Feminisierung auf die Organisation und die Praxis der Medizin

    Stufenkinetik vizinaler Si(001)-Oberflächen unter dem Einfluss hoher Temperatur und Au-Adsorption

    Inhalt dieser Dissertationsschrift ist die Untersuchung der Stufenkinetik auf leicht vizinalen Si(001)-Oberflächen bei sehr hohen Temperaturen mittels LEEM. Zunächst wird der lagenweise Zerfall isolierter Si-Hügel und Sattelpunkte in deren Umgebung detailliert analysiert. Aufgrund des Gibbs-Thomson-Effekts zerfällt ein solcher Hügel für Temperaturen zwischen 850°C und 1060°C immer schneller als der Sattel. In diesem Temperaturbereich können Hügel folglich vollständig von der Oberfläche entfernt werden. Für höhere Temperaturen werden die Stufenfluktuationen so groß, dass der Gibbs-Thomson-Effekt überall gleich stark wirkt; Sattel und Hügel zerfallen entsprechend gleich schnell. Weiterhin kann gezeigt werden, dass die Zerfallsrate der obersten Hügellage nicht konstant ist, sondern jede zweite Lage schneller zerfällt. Dieser als “Pairing” bezeichnete Effekt beruht auf einer Brechung der Symmetrie. Detaillierte Untersuchungen machen deutlich, dass auf der selben Probe der Pairing-Effekt bei allen Hügeln gleich groß ist und immer derselbe Terrassentyp bevorzugt zerfällt. Beides variiert jedoch für unterschiedliche Proben. Die Brechung der Symmetrie muss daher in einer makroskopischen Verspannung der Probe begründet sein. Dadurch wird schließlich der Zerfall eines der beiden Terrassentypen bevorzugt ablaufen. Darüber hinaus wird gezeigt, dass die Deposition von Gold auf der Oberfläche zu einer Beschleunigung des Hügelzerfalls führt. Gold und Silizium bilden ein Oberflächeneutektikum, wodurch der Schmelzpunkt lokal herabgesetzt wird. Dies geschieht zunächst an den Stufenkanten, was eine deutlich erhöhte Stufenfluktuation und den beschleunigten Hügelzerfall zur Folge hat. Eine noch höhere Goldbedeckung führt schließlich auch zum Aufrauen der Terrassen, sodass dieser Effekt im Wesentlichen wie eine Reduktion der kritischen Aufrauungstemperatur wirkt. Auf unverspannten, gleichmäßig gestuften Oberflächen führt dieser Anlagerungsmechanismus darüber hinaus zu zwei goldinduzierten Domänendominanzen. So wird gezeigt, dass die Adsorption von Gold zunächst zu einer deutlichen Verbreiterung des Terrassentyps führt, auf dem die Dimerreihen parallel zu den Stufen verlaufen. Weiteres Gold kehrt diese Dominanzsituation um, sodass kurz vor dem Aufrauungsphasenübergang der Terrassentyp dominiert, auf dem die Dimerreihen gerade senkrecht zu den Stufen liegen. Erklärt wird dieser Effekt durch eine Minimierung der veränderten Energie des Verspannungsfeldes. Eine extrinsische Verspannung verändert die Situation und treibt das System zunächst in die Dominanz, des bereits ohne Gold vorherrschenden Terrassentyps

    Progranulin is a novel prognostic marker for chronic lymphocytic leukemia

    Progranulin ist ein 88 kDa großes sezerniertes Protein dem eine wichtige Funktion bei der Regulation des Zellwachstums sowohl unter physiologischen Bedingungen als auch bei der Tumorentstehung zukommt. In einer vorangegangenen Genexpressionsstudie konnte mit Hilfe der DNA-Microarray-Technologie gezeigt werden, dass die Progranulin-mRNA-Expression in den Tumorzellen von Patienten mit einer chronischen lymphatischen Leukämie (CLL) mit einem ungünstigen klinischen Verlauf korreliert. In der hier dargestellten Arbeit sollte die potentielle prognostische Bedeutung von Progranulin auf Proteinebene weiter untersucht werden. Zu diesem Zweck wurde die Progranulinkonzentration im Plasma von 131 CLL-Patienten mittels enzyme-linked-immunosorbent-assay (ELISA) quantifiziert und mit etablierten Prognosemarkern und dem klinischen Verlauf der Patienten korreliert. Die Ergebnisse zeigen signifikant erhöhte Progranulinkonzentrationen im Plasma von CLL Patienten im Vergleich zu einem Vergleichskollektiv gesunder Blutspender. Bei den CLL-Patienten korrelierte Progranulin zudem mit der Ausprägung prognostisch ungünstiger Risikofaktoren wie z.B. einer Deletion 17p und 11q in der Fluoreszenz in-situ Hybridisierung, einem unmutierten IgVH-Mutationsstatus, CD38 und ZAP-70 Expression sowie einem fortgeschrittenen Binet-Stadium. Außerdem war eine hohe Progranulinkonzentration mit einer kürzeren Zeit bis zur ersten Therapiebedürftigkeit und einem reduzierten Gesamtüberleben der Patienten assoziiert. Diese im Essener Patientenkollektiv erhobenen Ergebnisse wurden in einer unabhängigen, mehr als 10 Jahre prospektiv beobachteten Kohorte von 163 Patienten im Stadium Binet A der deutschen CLL-Studiengruppe validiert. In der multivariaten Analyse konnte Progranulin in beiden Kohorten als unabhängiger Prognosemarker bestätigt werdenProgranulin is a 88 kDa secreted protein with pleiotropic functions including regulation of cell cycle progression, cell motility, wound repair and tumorigenesis. Using microarray based gene expression profiling it has been recently demonstrated that the gene for Progranulin, granulin (GRN), is significantly higher expressed in tumor cells of aggressive as compared to indolent chronic lymphocytic leukemia (CLL) cases. Here, Pgrn plasma concentrations were measured by enzyme-linked immunosorbent assay (ELISA) of 131 CLL patients from Essen and examined for association with established prognostic markers and clinical outcome. Progranulin was significantly higher expressed in the plasma of CLL patients compared to healthy donors . High Progranulin plasma levels in CLL patients were strongly associated with adverse risk factors including unmutated IGHV status, expression of CD38 and ZAP-70, poor risk cytogenetics (11q-, 17p-) as detected by flourescence in situ hybridization (FISH) and high Binet stage. Progranulin concentration was prognostic for time to first treatment and overall survival in this series. Importantly, these results could be confirmed in the independent multicentric CLL1 cohort of 163 untreated Binet stage A patients.     Pointer state dynamics of dissipative quantum systems

    In the present work, I identify the pointer states of quantum Brownian motion as rotated Gaussian states in phase space. By means of a particular piecewise deterministic unraveling, the pointer state unraveling, I determine the trajectories of the position and momentum expectation of the pointer states. They turn out to be described by a diffusion process in phase space and, in the semiclassical limit, they turn into the Langevin equation of classical Brownian motion. For collisional decoherence, the non-dissipative limit of the quantum linear Boltzmann equation, which describes the dynamics of a marker particle in an ideal gaseous environment, I examine the width of the pointer states and thereby extend previous results to more than one dimension. Crucially, it follows that pointer states do not exist for all parameter values in that case. By expanding the Lindblad operators of the quantum linear Boltzmann equation, I derive a new Lindblad master equation, dissipative collisional decoherence, which shows a decoherence behavior similar to that of collisional decoherence and also incorporates frictional as well as diffusion effects. Moreover this equation exhibits soliton-like solutions as candidates for the pointer states.In der vorliegenden Arbeit identifiziere ich die Zeigerzustände der quantenbrownschen Bewegung als rotierte gaußsche Zustände im Phasenraum. Mithilfe des Zeigerzustandsunravelings, ein bestimmtes stückweise deterministisches Unraveling, leite ich die Trajektorien der Orts- und Impulserwartungswerte der Zeigerzustände ab. Es stellt sich heraus, dass diese durch einen Diffusionsprozess im Phasenraum beschreibbar sind, und dass sie im semiklassischen Grenzfall die Langevingleichung der klassischen brownschen Bewegung ergeben. Für Stoßdekohärenz, den nichtdissipativen Grenzfall der quantenlinearen Boltzmanngleichung, welcher die Dynamik eines in einem idealen Gas eingetauchten Markerteilchens beschreibt, untersuche ich die Breite der Zeigerzustände und erweitere dabei frühere Ergebnisse auf mehr als eine Dimension. Als entscheidender Unterschied folgt, dass in diesem Fall die Zeigerzustände nicht für alle Parameterbereiche existieren. Durch eine Entwicklung der Lindbladoperatoren der quantenlinearen Boltzmanngleichung leite ich eine neue Lindbladmastergleichung her, dissipative Stoßdekohärenz, die ein Dekohärenzverhalten ähnlich dem der Stoßdekohärenz hat, und darüberhinaus auch Reibungs- und Diffusionseffekte enthält. Außerdem zeigt sie solitonartige Lösungen, welche Kandidaten für die Zeigerzustände darstellen

    Mutual adaptation in differently evolved host-parasite systems

    Wirt-Parasit-Systeme bieten ideale Voraussetzungen zur Erforschung von koevolutionären Prozessen, da die beteiligten Individuen in kontinuierlicher Interaktion stehen. Insofern sind die entsprechenden Adaptationsprozesse gezielt darauf ausgerichtet, die Wirkungen des Gegenspielers abzuschwächen. Invasive Parasitenarten sind in diesem Zusammenhang von großer Bedeutung, denn sie lassen Vergleiche zwischen etablierten und neu entstandenen Wirt-Parasit-Verbindungen zu. Die invasive, ursprünglich im Japanischen Aal (Anguilla japonica) parasitierende Schwimmblasennematoden-Art Anguillicola crassus hat sich seit ihrer Einführung in den frühen 1980er Jahren über Populationen des Europäischen Aals (Anguilla anguilla) in ganz Europa ausgebreitet. Die vorliegende Studie basiert auf Infektionsexperimenten mit diesen beiden Wirtsaalarten und A. crassus sowie der nahe verwandten Parasitenart A. novaezelandiae, die endemisch im Kurzflossenaal (Anguilla australis) vorkommt und dementsprechend in beiden untersuchten Aalarten nicht heimisch ist. Daraus resultieren vier unterschiedlich angepasste Wirt-Parasit-Systeme, die eine Gegenüberstellung von langfristig, kurzfristig und nicht-koevolvierten Beziehungen ermöglichen. Die Untersuchungen beziehen sich auf den Infektionserfolg, die morphologischen Eigenschaften und die Stressreaktionen, gemessen an den Expressionsniveaus von Hitzeschockproteinen (hsp70), der beiden Anguillicola-Arten in den verschiedenen Wirtsarten sowie die jeweilige im Wirt hervorgerufene Schwimmblasenschädigung, um wirtsspezifische Unterschiede herauszustellen, die in Zusammenhang mit koevolutionären Prozessen stehen. Die Wiederfindungsraten beider Parasitenarten waren in Japanischen Aalen signifikant niedriger als in Europäischen Aalen, was auf die Etablierung einer effektiven Genus-spezifischen Immunantwort auf Infektionen mit Anguillicola-Arten im Japanischen Aal hindeutet, der in der Lage ist, eindringende Larven einzukapseln und zu töten. Diese Einkapselungen gingen mit einer deutlichen Verdickung der Schwimmblasenwände einher. Detaillierte morphometrische Analysen konnten zeigen, dass beide Parasitenarten in Japanischen Wirten auch eine deutlich geringere Körpergröße als in Europäischen Wirten erreichten. Stark erhöhte Hitzeschockproteinwerte konnten in Individuen der Art A. crassus in Japanischen Aalen festgestellt werden, während A. crassus im Europäischen Aal leicht erhöhte Werte aufwies, was darauf hinweist, dass leicht bis stark an die jeweilige Infektion angepasste Wirtsarten eine höhere Stressreaktion in den Parasiten hervorrufen als unangepasste Arten. Anguillicola novaezelandiae zeigte weder im Japanischen noch im Europäischen Wirt gesteigerte hsp70-Niveaus. Dennoch deuten die Ergebnisse der vorliegenden Arbeit zusammen betrachtet deutlich auf eine beeinträchtigte Entwicklung beider Anguillicola-Arten in Reaktion auf die ausgeprägte Immunantwort eines stark angepassten Wirtes.Host-parasite systems are an ideal field of research for studying co-evolutionary processes since the two involved individuals are continuously interacting. In this respect, adaptation processes are directly targeted at coping with the antagonistic species. Invasive parasite species are of particular importance when comparing data on established and new host-parasite associations. The invasive swim bladder nematode Anguillicola crassus, originally parasitizing the Japanese eel (Anguilla japonica), has spread throughout populations of the European eel (Anguilla anguilla) since its introduction in the early 1980s. The present study is based on infection experiments of these two eel host species with A. crassus and its close relative A. novaezelandiae, which is endemic to the Short-finned eel (Anguilla australis) and thus non-native to both analyzed host species. Accordingly, four host-parasite associations differing in terms of mutual adaptation were investigated. With these it was possible to contrast systems featuring long-term, short-term, and no co-evolutionary relationship at all. Infection success, swim bladder pathogenicity, morphological features, and stress response based on hsp70 levels of both Anguillicola species in each eel species were analyzed in order to reveal host-specific differences, which could be related to co-evolutionary processes. Recovery rates of both parasite species proved to be significantly lower in Japanese eels than in European eels indicating that Japanese eels developed a strong genus-specific immune response against nematode infections with the ability to encapsulate and kill invading larvae. Those encapsulation processes proved to implicate a distinct thickening of the swim bladder walls. Detailed morphological measurements showed that both nematode species attain a reduced maximum body size in Japanese eel hosts compared to European eels. Highly increased hsp70 responses could be detected in A. crassus individuals taken from Japanese eels, slightly increased levels were observed for A. crassus taken from European eels indicating that adapted hosts induce a higher stress response in the parasites. Anguillicola novaezelandiae showed neither elevated hsp levels in Japanese nor in European eel hosts. However, taken as a whole, the present results strongly hint at an impaired development of both Anguillicola species when confronted with the strong immune response of a well-adapted eel host species
