Mapping Local Manifestations of the Strain Mediated Magnetoelectric Effect in Composites
The magnetoelectric effect has gained immense impetus in the last century, mainly due to its interesting potential applications into the technologies of the era. The fact that the two important space/matter permeating fields, the electric and the magnetic field, could be in a material, gives rise to key applications like high sensitivity magnetic field sensors, and voltage controlled non-volatile memory storage. As the various materials exhibiting magnetoelectric effect are being continually explored, the composites with combination of a ferroelectric and a ferro/ferrimagnetic material have emerged as favorite candidates with exceptional performance under ambient conditions. Quite often the magnetoelectric effect in composite is mediated via strain, requiring a complex constitutive model for their representation. Very recently, attempts have been made to construct robust models for understanding the strain mediated magnetoelectric effect in composites with various morphologies. However, such models lack the necessary experimental support. In general, there is a lack of proper understanding about the behavior of the strain mediation in the vicinity of the interface between the constituent phases. The present study is aimed to tackle the lack of information surrounding the local manifestations of the magnetoelectric effect in composites, with a focus on pushing the boundaries of microscopic techniques like Scanning Probe Microscopy (SPM), and Confocal Raman Microscopy. Various SPM modes like Piezoresponse Force Microscopy (PFM), Magnetic Force Microscopy (MFM), and Kelvin-probe Force Microscopy (KPFM) were synergistically utilized. As a subject of study, bulk polycrystalline composites based on combinations of ferroelectric BaTiO3 and various ferrimagnetic ferrites (viz. BaFe12O19, SrFe12O19, CoFe2O4, and NiFe2O4) were investigated. The choices of the magnetic components served as a wide spectrum of materials with different magnetocrystalline symmetries. In addition, the constituent morphologies favorably allowed longitudinal investigations in the vicinity of the ferroelectric-ferrimagnetic interface.
Both, direct and converse magnetoelectric effects were locally studied using in-situ MFM and PFM respectively. As a part of the converse effect the application of instantaneous electric poling under the SPM tip resulted in interesting micromagnetic fluctuations which were observed in MFM, and later analyzed using a self-developed image processing algorithm. The results suggest an intricate role of defects and microstructure in inducing the observed changes in magnetic domain configurations. As a part of the direct magnetoelectric effect, the in-situ application of magnetic field during PFM imaging results in a corresponding variation in PFM amplitude. This was rationalized as modulation of the local electromechanical coupling in BaTiO3 by the magnetoelectrically induced stress via the magnetic phase. This was further studied in a systematical manner, using Principal Component Analysis (PCA) of PFM image sequences. The results revealed interesting magnetic field modulations of PFM amplitude, the extent of which followed a distinct spatial distribution. These patterns and their corresponding spatial distributions, indicated existence of various stress regimes which was mainly attributed tothe constituent morphologies. The distributions of the effect also suggested certain degrees of closeness to the classical micromechanical problem of inclusion in a matrix (Eshelby‟s solution). Later, the localized ferroelectric switching using the Switching Spectroscopy PFM (SSPFM) mode was also studied under and applied in-situ magnetic field. Maps of nucleation biases of the local switching were generated, suggesting presence of in-built electric fields localized at the interfaces. In addition, the application of magnetic field apparently modulated these fields, which could be attributed to the appearance of magnetoelectrically induced fields that could influence the balance of fields during the switching process. In order to corroborate the indirect observations of the effects of induced stress, a direct observation of the stress was carried out by the means of in-situ Confocal Raman Microscopy. A self-developed algorithm, based on a conjunction of PCA and Self Modeling Curve Resolution (SMCR) was utilized to generate 2D distributions of least-square fitted peak positions of BaTiO3 Raman modes. On application of magnetic field the Raman modes manifest spatially consistent frequency shifts, which were considered as a direct consequence of the magnetoelectrically induced strain in the BaTiO3 phase.
The outcomes, in totality, suggest a greater role of microstructure, as compared to that of the constituent material characteristics, in determining the persistence of the stress and hence the magnetoelectric effect within the composite.Der magnetoelektrische Effekt hat grosses Gewicht um vergangene Jahrhundert gewonnen, hauptsächlich durch seine interessantes Potential in technologischen Anwendungen. Die Tatsache, dass elektrische und magnetische Felder durch magnetoelektrische Effekte gekoppelt werden können, eröffnet die Möglichkeit für Anwendungen wie hochsensitive Magnetfeldsensoren und Spannungskontrolle in nicht flüchtigen Schreibprozessen für Datenspeicher.
Kompositmaterialien mit getrennten ferroelektrischen und magnetischen Phasen sind für Anwendungen vielversprechender. Normalerweise, ist der Kopplungsmechanismus zwischen diesen Phasen spannungsvermittelt und benötigt komplexe Modelle um beschrieben zu werden. Neuerdings wurden robuste theoretische Modelle erstellt mit verschiedenen Morphologien. Diese benötigen jedoch weitere experimentelle Unterstützung. Es gibt generelle einen Bedarf an genauem Verständnis über die Spannungsvermittlung an Grenzflächen.
Die vorliegende Arbeit, zielt darauf das Verständnis auf diesem Gebiet auszuweiten, mit dem Fokus, die Grenzen mit Techniken wie Rasterkraftmikroskopie und konfokaler Raman Mikroskopie zu erweitern. Ausserdem sollen piezoresponse force microscopy (PFM), magnetic force microscopy (MFM) und Kelvin-probe force microscopy (KPFM) synergetisch eingesetzt werden.
Untersucht werden insbesondere polikrsitalline Systeme aus farroelektiscfhem BaTiO3 und verschiedenen ferrimagntischen ferrites (BaFe12O19, SrFe12O19, CoFe2O4 und NiFe2O4).
Sowohl direkter als auch der konverse magnetoelektrische Effekt wurde lokal durch in-situ PFM und MFM untersucht. Durch die Effekte konnten interessante mikromagnetische Fluktuationen bzw. Variationen in den PFM Amplituden beobachtet werden und mit selbst entwickelten Algorithmen, teilweise auf Basis von Principal Component Analysis (PCA) untersucht werden. Die Ergebnisse enthüllten interessante Muster der Magnetfeldmodulation der PFM Amplitude einhergehende mit einer speziellen räumlichen Verteilung.
Später kam switching spectroscopy zum Einsatz unter in-situ Magnetfeld um Karten von lokalen Keimbildungsspannungen aufzunehmen. Diese wiesen auf intrinische elektrische Felder an den Grenzflächen hin, die durch Magnetfelder moduliert werden konnten. Diese wurden wieder mithilfe von Algorithmen analysiert und mit den Raman Moden aus Raman Mikroskopiemessungen verglichen. Durch Einschalten des Magnetfelds, wurden diese Moden konsistent freqeunzverschoben was auf die magnetoelektrische Spannungseinwirkung zurückzuführen ist.
Die Ergebnisse weisen im Endeffekt auf eine größere Rolle der Mikrostruktur hin, als es bei den Einzelkomponenten der Fall wäre
Emergence, development and configuration of (trans-)national care arrangements of the elderly – Italy and Germany compared
In recent years there has been an increasing academic interest in how welfare states solve the care deficit resulting from the growing populations of the elderly on the one hand and significant changes in traditional family care structures on the other hand: increasing female employment rate, changes in family and household structures and cultural attitudes, but still widespread traditional imbalances between partners‘ commitment to reproductive work.
Whereas mainstream research devotes its attention above all to the interpersonal configuration of care regimes, there has been a shift in feminist research to the growing importance of transnational care work within the emergence of global labor markets. It was stated that nearly all western welfare states depend to various degrees on migrant care work both in service provision due to staff shortages and in private households due to the changes mentioned above. In particular, the welfare states belonging to the so-called conservative-corporate regimes such as Italy and Germany recourse to privately paid migrant female carers often living in their employer’s household and being strongly vulnerable to exploitation.
For this doctoral thesis, Italy and Germany have been chosen as interesting cases for a comparison because of their rapid demographic and social development, which should have provoked political pressure towards new and adequate caring arrangements. For all structural similarities between the countries of this regime, the Mediterranean countries represent a discrete regulation type of welfare state due to specific features of their socio-economic development, labor market, migration regime and a strong familialism.
Italy has a long tradition in employing migrant care workers in private households whereas in Germany this is quite a new development. The question therefore is, if Italy displays an extraordinary case resulting from its specific features and history or whether both countries show similar reasons for and characteristics of transnational care arrangements.
Deriving from theories of welfare regimes, of concepts of social care as well as of migration this doctoral thesis applies an explicit binary analysis approach using secondary sources, survey data, policy documents and legislations to analyse interconnections of social developments, the care regime and the related labor market as well as the migration regime.
The main finding concerning the Italian situation is that the country has undergone a radical and mostly uncoordinated marketization of care quite early and within this framework has developed a strong dependence on migrant care work in service delivery as well as in private households. These developments were based on a temporally rising economic prosperity, the beginning of irregular migration, the attraction of existing ex-post regularization measures, which consistently have been boosting both the demand for new irregular migration and the shadow economy. The state not only supported these trends but endowed families with generous cash of free use so that even the economically not well-off were able to hire migrant care work. The latter were also enabled to purchase private care on the market due to the political strengthening of an enduring familialism that goes far beyond a lack of public services in-kind and further measures of defamilialisation that are generally taken into account when it comes to comparing social policies.
Concerning the German situation on the other hand, the implementation of the statutory long-term insurance scheme has led to a certain degree of defamilialisation and the social right of the individual (person) to opt for services in-kind which are not dependent on the public annual budget at hand or the place of living as in Italy. The benefits deriving from the cash-for-care scheme are mostly low and therefore not sufficient for paying a migrant carer, so that privately hiring migrant care workers is restricted to quite well-off people. The latter also results from the involvement of recruitment agencies whose emergence is connected to the german labour migration policy in the course of the EU enlargement whereas the employment of irregular third nationals – the main source a cheap labour force in Italy – has to be neglected in Germany
Untersuchungen zum Einfluss von Melatonin, Citalopram und Sarpogrelate auf die Hämostase in der sub-akuten Endotoxinämie in männlichen Wistar-Ratten
Bei Patienten mit einer schweren Sepsis stellt die Disseminierte intravasale Gerinnung (DIG) einen starken Prädiktor für die Mortalität dar. Die DIG ist gezeichnet durch eine systemische Aktivierung der Gerinnung mit einem simultanen Verlust von Kompensationsmechanismen. Inhibitorische Effekte von exogenem Melatonin (MLT) auf die plasmatische Gerinnung sowie die Thrombozytenaggregation wurden bereits in vivo bzw. in vitro unter Normalbedingungen nachgewiesen.
In dieser Arbeit sollte untersucht werden, ob exogenes MLT auch die Lipopolysaccharid (LPS)-induzierte DIG während der sub-akuten Endotoxinämie vermindert. Darüber hinaus sollte aufgrund der strukturellen Ähnlichkeit von MLT zu Serotonin und der bereits in vitro nachgewiesenen Hemmung des Transports von Serotonin in die Thrombozyten durch MLT, der Einfluss von MLT auf die DIG in vivo sowie auf die Serotonin-induzierte Thromboyztenaktivierung in vitro mit denen der selektiven Serotoninantagonisten Citalopram (CIT), einem Serotoninwiederaufnahmehemmer, und Sarpogrelate (SAR), einem 5-HT2-Rezeptorantagonisten, verglichen werden.
Die sub-akute Endotoxinämie wurde in männlichen Wistar-Ratten durch eine intravenöse (i.v.) Dauerinfusion von LPS (0,5 mg/kg x h) induziert. MLT (3 x 3 mg/kg x 15 min) und SAR (0,28 mg/kg x 15 min) wurden als Kurzzeitinfusion 15 min vor und dann 120 min und 240 min nach dem Beginn der LPS-Infusion i.v. verabreicht. CIT wurde (0,46 mg/kg x 15 min) lediglich 15 min vor Beginn der LPS-Infusion i.v. infundiert. Zur Untersuchung der Gerinnungskinetik mittels der Thromboelastometrie und zur Bestimmung der Thrombozytenzahl wurde jede Stunde über die rechte Arteria femoralis der Tiere Blut entnommen. Für die Untersuchung der Serotonin-induzierten Thrombozytenaktivierung in vitro wurde gesunden Probanden venöses Vollblut abgenommen und dieses mittels der Impedanzaggregometrie untersucht.
Die Dauerinfusion von LPS verursachte zuerst eine Hyperkoagulobilität und später eine Hypokoagulobilität des Blutes sowie eine signifikante Abnahme der Thrombozytenzahl. Die Gabe von MLT vor und während der sub-akuten Endotoxinämie hatte keinen Einfluss auf die LPS-induzierten Veränderungen der Parameter der Thromboelastometrie. Allerdings bewirkte MLT eine signifikante Reduktion der LPS-induzierten Abnahme der Thrombozytenzahl. Die Applikation von CIT vor der sub-akuten Endotoxinämie bewirkte eine anfänglich stärkere Aktivierbarkeit der Gerinnung und gegen Ende des Versuchs eine signifikant verminderte Hypokoagulobilität des Blutes im Vergleich zu den LPS-Tieren. Die Infusion von SAR vor und während der sub-akuten Endotoxinämie reduzierte ebenfalls die LPS-bedingten Hypokoagulobilität des Blutes. Die Thrombozytenzahl war sowohl bei den LPS+CIT-Tieren als auch bei den LPS+SAR-Tieren im Vergleich zu den LPS-Tieren stark vermindert. In vitro bewirkten MLT, CIT und SAR eine Dosis-abhängige Hemmung der Serotonin-induzierten Thrombozytenaktivierung (IC50= ~1413 μM MLT, ~58 μM CIT, ~48 μM SAR).
Den Ergebnissen zufolge vermindert exogenes MLT die LPS-induzierte Thrombozytopenie jedoch nicht die LPS-induzierte DIG während der sub-akuten Endotoxinämie. Im Gegensatz zu MLT bewirkten die selektiven Serotoninantagonisten CIT und SAR eine signifikante Reduktion der hypokoagulablen Phase der LPS-induzierten DIG. Sowohl der Transport von Serotonin in die Thrombozyten als auch die Aktivierung der Thrombozyten über den 5-HT2A-Rezeptor scheinen, den Ergebnissen nach, essentiell für die Thrombozytenhomöostase, da eine Hemmung dieser Prozesse durch CIT bzw. SAR eine Reduktion der Thrombozytenzahl verursachte. In den in vitro Versuchen wurde im Gegensatz zu den in vivo Versuchen eine serotoninantagonistische Wirkung von Melatonin auf die Thrombozyten gezeigt, die jedoch erst bei hohen Konzentrationen von MLT (mM) erreicht wurde und demzufolge in vivo nicht von Bedeutung ist
Craving and decision making in individuals with pathological buying
Das pathologische Kaufen gilt als klinisch relevantes Phänomen, bei dem Betroffene die Kontrolle über ihr Kaufverhalten verlieren. Das Phänomen ist aktuell nicht psychiatrisch klassifiziert und obwohl Parallelen zu Suchterkrankungen gezogen werden, fehlen experimentelle Studien, die diese Parallelen untersuchen und in ein Gesamtmodell der Entstehung und Aufrechterhaltung der Störung integrieren. Diese Forschungslücke soll mit der vorliegenden kumulativen Dissertation geschlossen werden. Schrift 1 des Kumulus adressiert die Nähe zu Suchterkrankungen. Unter Anwendung eines Cue-Reactivity Paradigmas mit kaufbezogenen Bildern wurden subjektive und physiologische Cravingreaktionen (Suchtmittelverlangen) bei Personen mit pathologischem Kaufverhalten festgestellt. Schrift 2 widmet sich dem pathologischen Kaufen im Online-Kontext und fand ebenfalls Cravingreaktionen bei Personen mit einer Tendenz zum pathologischen Online-Kaufen. Die Suchtkonzeption aufgreifend konnte in Analogie zu einem Modell für spezifische Formen einer Internetsucht gezeigt werden, dass der Zusammenhang zwischen einer prädisponierenden Erregbarkeit durch Einkaufen und der Symptomschwere bezüglich des Online-Kaufens durch die Nutzungserwartung mediiert wurde. Die Befunde aus Schrift 1 und 2 legen nahe, dass Cravingreaktionen, Belohnungsantizipation und Gratifikationsmechanismen in Analogie zu Suchterkrankungen einen wesentlichen Faktor in der dysfunktionalen Verhaltenssteuerung darstellen können. Schrift 3 untersucht Entscheidungsverhalten im Kontext von Suchterkrankungen bei pathologischem Kaufen. Es konnte gezeigt werden, dass Personen mit pathologischem Kaufverhalten Entscheidungsdefizite aufwiesen, wenn die Eintrittswahrscheinlichkeit und Konsequenzen einer Alternative unbekannt waren (Ambiguität), wohingegen sie keine Schwierigkeiten zeigten, wenn diese explizit vorlagen (Risiko). Die Ergebnisse legen nahe, dass dysfunktionales Entscheidungsverhalten durch ein Defizit in der emotionalen Feedbackverarbeitung bei intakten kognitiven Funktionen erklärt werden kann. Die vorliegenden Befunde aus den Schriften 1-3 des Kumulus werden in Bezug auf ein Gesamtmodell diskutiert, welches im Einklang mit Dual-Prozess-Theorien der Selbstregulation davon ausgeht, dass Cravingreaktionen und ein dysfunktionales Entscheidungsverhalten kovariieren und damit die Entscheidungen für die kurzfristig belohnende Alternative des Einkaufens ungeachtet der langfristig negativen Konsequenzen (Überschuldung, etc.) erklärt werden können. Zusammenfassend werden Parallelen des pathologischen Kaufens und Suchterkrankungen aufgezeigt und eine mögliche Klassifikation als Verhaltenssucht diskutiert.Pathological buying is a clinical relevant phenomenon, in which affected individuals lose control over their buying behavior. Currently, the phenomenon is not psychiatrically classified, and although parallels to addictive behaviors have been considered, there is a lack of experimental studies which investigate this relationship and integrate these findings with respect to a comprehensive model for the development and maintenance of the disorder. This cumulative dissertation aimed to address this research gap. Scripture 1 of the cumulus addresses the potential proximity to addictive behaviors and found craving reactions on a subjective and physiological level in a group of individuals with pathological buying by applying a cue-reactivity paradigm with shopping related pictures. Scripture 2 investigates pathological buying patterns in the online context and found craving in individuals with a tendency to pathological online buying. With respect to the addiction concept, in accordance with a current model for specific Internet addiction, it was demonstrated that the relationship of a predisposing excitability from shopping and the symptom severity of online shopping was mediated by specific online shopping expectancies. Taken together, the findings of scripture 1 and 2 emphasize the important role of craving, reward anticipation, and mechanisms of gratification with regard to the dysfunctional behavioral control of pathological buying. Scripture 3 addresses the role of decision making in the context of addictions for pathological buying. The results emphasize that individuals with pathological buying showed decision-making impairments when the probability of occurrence and consequences were unknown (ambiguity), whereas non-impairments were observed when they were explicitly given (risk). Potential correlates included emotional feedback-processing deficits, rather than executive dysfunctions. The findings of the scriptures 1-3 will be discussed in the context of a comprehensive model, which emphasizes in line with dual-process theories of self-regulation that craving and dysfunctional decision making may be related, explaining the preference for the short-term rewarding option of buying despite long-term negative consequences (such as financial difficulties, etc.). Altogether, parallels of pathological buying with addictive disorders are revealed and a potential classification with respect to behavioral addictions is discussed
Biophysical and functional characterization of human tyrosine-sulfated peptides
Die Sulfatierung von Tyrosinen ist eine weit verbreitete post-translationale Modifikation, welche in den intrazellularen Transport sekretierter Peptide, in die Regulierung von extrazellulären Protein-Protein-Interaktionen, in die Regulierung verschiedenen Rezeptoren und in die Förderung proteolytischer Prozesse einiger Peptid-Hormone involviert ist. Innerhalb dieser Arbeit wurden mit dem N-Terminus des CCR5-Rezeptors und dem Speichel-Peptid Histatin1 zwei humane Vertreter Tyrosin-sulfatierter Peptide auf unterschiedliche Weise charakterisiert.
Als erstes sollten für eine Sequenz aus dem N-Terminus des CCR5 Rezeptors Binder mithilfe einer kombinatorischen Protein-Bibliothek identifiziert werden. Dazu wurde zunächst eine Bibliothek auf Basis der Phosphat-bindenden IRS1-PTB-Domäne entwickelt und das Ziel-Peptid CCR512-18 mittels Festphasen-Peptid-Synthese hergestellt. Für die Selektion bindender Mutanten wurde ein Ribosomen-Display etabliert. Die daraus hervorgehenden Mutanten wurden exprimiert und gereinigt sowie zur Bestimmung ihrer Bindungs-Affinität gegenüber der sulfatierten und der nicht-sulfatierten Variante des Ziel-Peptids in einem Fluoreszenz-Anisotropie-Assay untersucht. Dabei konnte eine Triple-Mutante der IRS1-PTB-Domäne identifiziert werden, welche zwischen dem Tyrosin-sulfatierten Peptid und der Kontrolle diskriminiert. Zudem zeigt ein mögliches Bindungs-Modell des sulfatierten CCR512-18 an die Mutante ein analoges Bindungsverhalten wie bei der gp120-Corezeptor-Bindung.
Als zweites sollte das im Gegensatz zu Histatin5 kaum untersuchte Histatin1 strukturell und funktionell charakterisiert werden. Dazu wurde dieses zunächst in einen geeigneten Expressionsvektor kloniert und die Expression in E. coli sowie die zugehörige Reinigung etabliert. Anschließend wurde die dreidimensionale Struktur des Peptids in hydrophiler Umgebung aufgelöst. Diese beinhaltet drei definierte Helices, welche zwei mögliche Grenz-Konformationen zueinander einnehmen. Außerdem wurden sowohl die antifungale Aktivität als auch die Wundheilungs-Eigenschaften von Histatin1 unter Beachtung der funktionellen Domänen untersucht. So kam ein N terminales Fragment (Histatin14-15) in einem Toxizitäts-Assay gegen S. cerevisiae zum Einsatz und zeigte volle antifungale Aktivität gegenüber Histatin1 in voller Länge. Des Weiteren wurde ein Wundheilungs-Assay laborintern etabliert, mit welchem ein C terminales Fragment (Histatin120-32) als sulfatierte und nicht-sulfatierte Variante verglichen werden konnte. Dabei wurde ein aktivierender Effekt des sulfatierten Histatin120-32 nachgewiesen, welcher jedoch vermutlich auf die eingebrachte negative Ladung in der C-terminalen Helix zurückzuführen ist. Bei Übertragung der jeweils minimal aktiven Fragmente auf die Struktur, entspricht der für die antifungale Aktivität verantwortliche Bereich dem Gelenk zwischen Helix 1 und 2 und das für die Wundheilung zuständige Fragment dem Gelenk zwischen Helix 2 und 3.Tyrosin sulfation is a widespread post-translational modification implicated in the intracellular trafficking of secreted peptides, in the modulation of extracellular protein-protein interactions, in the modulation of several receptors and in the promotion of proteolytic processing of some peptide hormones. Here with the N-terminal CCR5 receptor and the salivary peptide histatin 1 two representatives of human tyrosine sulfated peptides were characterised differently.
First, artificial binders should be identified for a sequence of the N-terminus of the CCR5 receptor using a combinatorial protein library. For that, a library based on the phosphate binding IRS1 PTB domain was developed and the target peptide CCR512-18 was prepared by solid phase peptide synthesis. For the selection of binding mutants a ribosome display was established. The resulting mutants were expressed, purified, and examined in a fluorescence anisotropy assay to determine their binding affinity with respect to the sulfated and non-sulfated variant of the target peptide. Thereof a triple mutant of IRS-1 PTB domain has been identified which discriminates between the tyrosine sulfated peptide and control. In addition, a potential binding model of sulfated CCR512-18 to the mutant indicates analogous binding behavior as the gp120 coreceptor binding.
Second, compared with histatin 5, histatin1 ist hardly examined. Thus it should be structurally and functionally characterized. For this purpose, it was cloned into a suitable expression vector and expression in E. coli as well as purification has been established. Subsequently, the threedimensional structure of the peptide was determined in hydrophilic environment. It includes three defined helices, which occupy two possible border conformations. Furthermore, both the antifungal activity and the wound-healing properties of histatin 1 in compliance with the functional domains have been investigated. An N-terminal fragment (histatin 14-15) was used in a toxicity assay against S. cerevisiae and showed full antifungal activity in comparison with full length histatin 1. A wound healing assay has been established in which a sulfated and non-sulfated variant of a C-terminal fragment (histatin 120-32) could be compared. An activating effect of the sulfated histatin 120-32 was detected, which is, however, probably due to the negative charge introduced in the C-terminal helix. Untersuchungen zur Adsorption und Desorption von Aldehyden und Aminen im Spurenbereich
Ziel der Arbeit ist es, das Adsorptions- und Desorptionsverhalten von Aldehyden und Aminen in feuchter und trockner Stickstoffatmosphäre an handelsüblichen Aktivkohlen und neuartigen Adsorbentien wie (nicht-)funktionalisierten periodisch mesoporösen Organosilikas (PMO), Carbon Mesoporous Korea (CMK), mikroporösen Kohlenstoffadsorbentien und Controlled Pore Glas (CPG) im Spurenbereich von 10 – 1200 ppmV systematisch zu untersuchen und zu beschreiben. Dazu werden aus der Stoffgruppe der Aldehyde das kurzkettige Acetaldehyd und das langkettige Hexanal sowie aus der Stoffgruppe der Amine das kurzkettige Methylamin als Adsorptive eingesetzt. Aufgrund der gesundheitsschädigen Eigenschaften der Adsorptive finden die Messungen in einem komplexen vakuumdichten Messaufbau statt, der zusätzlich von einer Glovebox umgeben ist.
Mikroporöse kohlenstoffbasierte Adsorbentien und Amin-funktionalisierte Adsorbentien stellen die höchsten Adsorptionskapazitäten für Hexanal bereit. Im Falle der mikroporösen Adsorbentien überwiegen die Dispersionswechselwirkungen in den Mikroporen mit einem Durchmesser in der Größenordnung des minimalen Moleküldurchmessers des Hexanals. An den Amin-funktionalisierten Adsorbentien findet eine Schiffsche-Base Reaktion zwischen dem Hexanal und den primären Aminogruppen der Funktionalisierung unter Freisetzung von Wasser statt. Im Gegensatz zu den Amin-funktionalisierten Adsorbentien kann das Hexanal von den nicht-funktionalisierten Adsorbentien vollständig desorbiert werden. Zur Beschreibung der Kinetik der Adsorption von Hexanal an den Amin-funktionalisierten Adsorbentien wird ein Modell entwickelt, das für die Adsorption ein Zusammenwirken von Physisorptions- und Chemisorptionsvorgängen annimmt.
Bei der Ad- und Desorption von Acetaldehyd kann in bestimmten Fällen an verschiedenen kohlenstoffhaltigen Adsorbentien eine ausgeprägte chemische Reaktion mit der Porenoberfläche festgestellt werden. Vermutlich oligomerisiert Acetaldehyd an der Oberfläche zu Paraldehyd.
Im Rahmen der Adsorptionsmessungen von Methylamin an den kohlenstoffbasierten Adsorbentien und einem nicht-funktionalisierten PMO können keine erheblichen Kapazitätsunterschiede beobachtet werden. Vermutlich überwiegen Wechselwirkungen des Typs Dipol – Dipol zwischen dem Methylamin und den polaren Gruppen der Adsorbentien. Die Phosphorsäure-imprägnierte Aktivkohle weist aufgrund einer Säure-Base Reaktion zwischen dem Methylamin und der Imprägnierung eine etwas höhere Kapazität auf.In this work the adsorption and desorption characteristics of aldehydes and amines on commercially available adsorbents as well as newly developed (non-)functionalized adsorbents like peridodic mesoporous organosilicas (PMO), carbon mesoporous Korea (CMK), microporous carbon-based adsorbents and controlled pore glass (CPG) are systematically investigated and discussed. For this purpose the short-chained acetaldehyde and the long-chained hexanal as exemplary aldehydes as well as the short-chained methylamine as exemplary amine are used as adsorptives at trace concentrations from 10 to 1000 ppmv in a dry or humid nitrogen atmosphere. Due to the adverse health effects of the adsorptives measurements are done in a complex vacuum-tight experimental set-up which is enclosed in a glovebox.
The results show that microporous carbon-based adsorbents and amine-functionalized adsorbents have the highest adsorption capacity for hexanal. For microporous adsorbents the dispersion interactions are predominant in micropores with a diameter in the dimension range of the minimal molecular diameter of hexanal. On the amine-functionalized adsorbents a Schiff base reaction takes place between the primary amines of the functionalization and the hexanal forming imines and water. In contrast to the amine-functionalized adsorbents the hexanal can be completely desorbed from the non-functionalized adsorbents. A kinetic model based on the interaction of physisorption and chemisorption is derived to describe the adsorption kinetic of hexanal on amine-functionalized adsorbents.
During the ad- and desorption of acetaldehyde on various carbon-based adsorbents a distinct chemical reaction with the pore surface can be observed at certain conditions. Acetaldehyde probably oligomerizes to paraldehyde on the pore surface.
The adsorption capacity for methylamine on various non-functionalized carbon-based adsorbents and PMOs is very similar because the dipole-dipole interactions between methylamine and the polar groups on the surface of the adsorbent are probably predominant. A phosphoric acid-impregnated activated carbon shows the highest capacity due to an acid-base reaction between the methylamine and the phosphoric acid impregnation
Skills development in the training of "media scouts" as media educational approach to peer education.
Hier wurde der Erwerb von Medienkompetenz einer definierten Gruppe von 20 Schülerinnen und Schülern (acht Mädchen und zwölf Jungen) der Jahrgangsstufe 8 (später 9) im Alter von etwa 13 Jahren (später 15 Jahren) in der als Peer-Education-Ansatz angelegten Ausbildung zu „Medienscouts“ im Rahmen formaler Bildung in der Schule in Form eines „Wahlpflichtkurses“ und in ihrer Tätigkeit als Peer-Multiplikatoren sowie ihre Selbst-Einschätzung als Peer-Multiplikatoren über einen Zeitraum von etwa zwei Jahren (von September 2011 bis Juli 2013) empirisch untersucht. Dazu wurden als Methodentriangulation zu Beginn halbstandardisierte Einzelinterviews geführt, im Verlauf vier schriftliche Befragungen mit Fragebögen durchgeführt und insgesamt 18 Texte analysiert. Die Fragebögen wurden zu Beginn (als Fragebogen „A“ bezeichnet) im Oktober 2011, nach einem halben Jahr (Januar 2012, „B1“), zur Halbzeit (September 2012, „C“) und in Form eines Re-Tests von „B1“ als „B2“ am Ende (Juni 2013) erhoben. Die Texte sind kontinuierlich über den gesamten Zeitraum entstanden. Als Kontrollgruppe der schriftlichen Befragungen fungierten über 60 Mitschülerinnen und Mitschüler des gleichen Jahrgangs der gleichen Schule. Die Unterschiede zwischen den Gruppen wurden auf statistische Signifikanz geprüft, die bei einzelnen Variablen gegeben war.
Im theoretischen Teil wurde der Kompetenzbegriff im Kontext von Schule, so durch die OECD und KMK, und die schulrelevanten Theorie-Modelle von Medienkompetenz und Beispiele für die Messung von Medienkompetenz erörtert. Nach den Grundlagen und Beispielen der Erkenntnisse der Peer-Education-Forschung wurde die peer-basierte Medienkompetenzförderung in Deutschland bis Ende 2014 betrachtet. Darin sind 19 Peer-Programme dargestellt und miteinander verglichen. Bei aller Unterschiedlichkeit der Programme bezüglich der Reichweite von bundesweit bis lokal und der Organisationsformen gibt es deutliche Gemeinsamkeiten: Peer-basierte Medienkompetenzförderung ist von Institutionen initiiert und eine Reaktion auf Probleme der Mediennutzung. Aus diesem Grund werden nahezu durchgängig Themen des Jugendmedienschutzes behandelt. Die Zielgruppe bilden Kinder und Jugendliche ab einem Mindestalter von 12 Jahren bis hin zu einem Alter von 16 Jahren. Sie werden von erwachsenen Expertinnen und Experten ausgebildet (je nach Programm aus Medienpädagogik, Suchtberatung, Polizei, Jugendschutz, Datenschutz etc.), aber nicht durch die (eigenen) Lehrerinnen / Lehrer. Die Themen der Ausbildung sind in allen Programmen vergleichbar und die bekannten Jugendmedienschutz-Themen, die man in Schlagworten bezeichnen kann mit: Soziale Netzwerke, Datenschutz, Cyber-Mobbing, Urheberrecht, Handy, exzessive Mediennutzung und jugendgefährdende Inhalte. Diese Themen werden in einigen Programmen ausdrücklich ergänzt durch die Förderung sozialer und persönlicher Kompetenzen (wie bspw. Teamfähigkeit oder Empathiefähigkeit etc.).
Zur Beschreibung der Entwicklung von Medienkompetenz der Jugendlichen diente als theoretische Folie das Medienkompetenzmodell nach Groeben (2002b), das darüber hinaus hier für schulische Zwecke operationalisiert und auf Grundlage von Modellen zu Bildungsstandards in Form eines fünfstufigen Modells zur Beurteilung der Performanz von Schülerinnen und Schüler im Alter von 13 bis 15 Jahren gebracht wurde. Die Hypothesen 1 bis 7 dieser Untersuchung sind entlang der Dimensionen des Medienkompetenzmodells formuliert und postulieren eine positive Performanzentwicklung in „Medienwissen“ (Hypothese 1), „Medienspezifischen Rezeptionsmuster“ (2), „Genussfähigkeit“ (3) , „Kritikfähigkeit“ (4) , „Orientierungsfähigkeit“ (5), „Produktive Partizipationsmuster“ (6) und „Anschlusskommunikationen“ (7). Außerdem wurde eine verbesserte Medienkompetenzentwicklung im Vergleich zur Kontrollgruppe (Hypothese 8) und eine positive Selbsteinschätzung ihrer Tätigkeit als Peer-Multiplikatoren (Hypothese 9) angenommen. Alle Hypothesen konnten bestätigt werden. Die Medienscouts zeigten im Mittel eine positive Entwicklung ihrer Medienkompetenz, auch im Vergleich mit der Kontrollgruppe und schätzten ihre Arbeit als Peer-Multiplikatoren positiv ein mit einem positiven Selbstbild, einer positiven Einschätzung ihrer Selbstwirksamkeit und anhand sozialer Peer-Aspekte.
Im Einzelnen erwarben die Medienscouts ein größeres „Medienwissen“ und zeigten am Ende der zweijährigen Ausbildung / Tätigkeit als Peer-Multiplikator ein besseres „Medialitätsbewusstsein“. Die Effekte waren dort besonders stark, wo Inhalte und Themen der Ausbildung berührt waren, so beispielsweise in dem Wissen um die Mechanismen der Werbung in der Suchmaschine Google. Die „medienspezifischen Rezeptionsmuster“ und insbesondere darin die technologisch-instrumentellen Fertigkeiten wurden von den Medienscouts weiterentwickelt. Dies zeigte sich besonders stark bei den Fähigkeiten / Fertigkeiten, wie sie in der alltäglichen Mediennutzung der Jugendlichen benötigt wurden, d.h. sie lernten das besonders gut, was sie konkret für ihren Medienalltag benötigten. Dies zeigte sich zum Beispiel in ihren gesteigerten Fähigkeiten in der Benutzung von Social-Media-Angeboten, von Software zur Textverarbeitung oder in der produktiven Bedienung von Video-Portalen („YouTube-Uploads“). Bei einer individuellen Auswertung der Problemlösestrategien der Medienscouts zeigten sich große Unterschiede in Quantität und Kombination verschiedener Strategien. Eine „Genussfähigkeit“ als Teil einer Medienkompetenz nach Groeben war schwierig zu beschreiben. Trotzdem zeigten die Medienscouts, dass sie „ihre“ Medien genießen, auch wenn es ein oberflächlicher Genuss ist, der von den Jugendlichen – mit einer Ausnahme - nicht weiter reflektiert wird. Die Ausnahme bildet die Fähigkeit der Medienscouts zu erläutern, warum sie gerne Computerspiele spielen. Es konnte gezeigt werden, dass die Jugendlichen „Universal-Medien“ haben, wie das Smartphone, die sie für jede Stimmung / jede Gefühlslage nutzen. Darüber hinaus schätzen die Medienscouts ihre „Genussfähigkeit“ als hoch ein. Das gilt auch für die untersuchte „Kritikfähigkeit“ als eine der Dimensionen von Medienkompetenz nach Groeben. Die Selbsteinschätzung dazu war hoch, allerdings konnte die „Kritikfähigkeit“ über den Untersuchungszeitraum, ausgehend von einer relativ unkritischen Mediennutzung, verbessert werden, aber nicht generell über alle medialen Inhalte, sondern nur zu den Themen / Inhalten der Ausbildung. So konnte beispielsweise die Kritikfähigkeit zu den Herausforderungen „Geschlechterrollen in medialen Angeboten“ und „Firmenwerbung auf Facebook“ nicht verbessert werden, da sie nicht explizit Teil des Ausbildungskonzepts der Medienscouts waren. Ganz anders bei anderen Themen, wie „Recherche-Fähigkeiten über Suchmaschinen“ und „Quellenanalyse“. In der Dimension „Selektion / Kombination der Mediennutzung“ zeigte sich erwartungsgemäß, dass alle befragten Jugendlichen (Medienscouts wie Kontrollgruppe) intensive Mediennutzer sind. Die Medienscouts konnten ihre Performanz in dieser Dimension steigern, wenn auch zu sehen war, dass die Jugendlichen kaum alternative mediale Angebote nutzen. So wird bspw. „Wikipedia“ wenig reflektiert als einzige Quelle zur Recherche für ein Referat ausgewählt, was sich auch im Untersuchungszeitraum über zwei Jahre kaum änderte. Daran zeigte sich ein wiederkehrendes Muster, dass die Jugendlichen Medien nicht als Selbstzweck sehen, sondern gezielt für ihre Zwecke nutzen. Der Verfasser interpretierte im Beispiel „Recherche-Quellen“ kein Unvermögen, sondern eine fehlende Einsicht in die Notwendigkeit von Alternativen. Anders ausgedrückt: „Wikipedia“ als Quelle für Referate reichte den Jugendlichen. Wie oben schon erwähnt, nutzen die Jugendlichen das Smartphone als „Universalmedium“, das bezüglich des Gerätes keine weitere „Selektion / Kombination“ erforderlich macht. Unerwartet fielen die Ergebnisse der Dimension „Produktive Partizipationsmuster“ aus. Nur wenige Jugendliche nutzten trotz der Verfügbarkeit der erforderlichen Hardware und Software Medien produktiv-kreativ, dabei zu einem Drittel auf Social-Media-Anwendungen wie „Facebook“. Dabei sehen sie sich in der Selbsteinschätzung realistisch und schätzen sich in dieser Dimension selbst schlechter ein als in den anderen sechs Dimensionen nach Groeben. Dabei sind es weniger die fehlenden Fertigkeiten / Fähigkeiten der Schülerinnen und Schüler, sondern eine fehlende Motivation zur produktiv-kreativen Nutzung von Medien. Anders ausgedrückt: Sie könnten Medien in dieser Weise nutzen, wollen es aber nicht. Die Anschlusskommunikationen der Jugendlichen waren mit einer Ausnahme erwartungsgemäß hoch. Sie kommunizieren viel über Medien, sowohl medial vermittelt als auch im persönlichen Gespräch. Dabei sind – wie erwartet – vor allem Gleichaltrige die Gesprächspartner, auch und insbesondere im Sinne von Vertrauenspersonen. Doch nicht alle Jugendlichen zeigen die von Groeben beschriebenen „Anschlusskommunikationen“, sondern nur etwa drei Viertel kommunizieren über mediale Erlebnisse. Vom Verfasser war nicht erwartet worden, dass Lehrerinnen / Lehrer von nahezu keinem Jugendlichen als Gesprächspartner über mediale Erfahrungen benannt wurden.
Im Vergleich der Gruppen Medienscouts und Nicht-Medienscouts zeigte sich zu Beginn, dass sie sich kaum voneinander unterscheiden, außer in ihrer Technik-Affinität und in ihrer etwas höheren Mediennutzung. Für alle hier befragten Jugendlichen sind trotz einer sehr intensiven Mediennutzung die Themen Freunde / Freundschaft das Wichtigste und ihr Interesse an Themen ist nicht ausschließlich an Medien orientiert.
Die hier untersuchten 20 Medienscouts haben ein zumeist positives Selbstbild und ein hohes Selbstvertrauen. Sie schätzten ihre Selbstwirksamkeit zurückhaltend, aber nicht grundsätzlich negativ ein. Die Jugendlichen fühlten sich sehr sicher im Umgang mit Geräten wie dem Computer.
In dieser Untersuchung konnten die sozialen Aspekte der Peer-Education in Form eines verbesserten Selbstbildes und der Einschätzung ihrer Selbstwirksamkeit ebenso bestätigt werden wie die Förderung von Medienkompetenz im „Medienscouts“-Ansatz der peerbasierten Medienkompetenzförderung.Subject of the study was the acquisition of media literacy in a defined group of 20 8th grade students (later 9th grade, eight girls and twelve boys) aged approximately 13 years (later 15 years) during "media scout" elective courses using peer education approach methods within a formal school education, and their function as peer multipliers as well as their self-assessments as peer multipliers over a period of almost two years (September 2011 - July 2013). Methodological triangulation consisted of semi-structured individual interviews conducted at the start of the study period, four written surveys with questionnaires taken during the study, and the analysis of a total of 18 texts. The questionnaires were completed at the beginning (questionnaire "A") in October 2011, after half a year (January 2012 "B1"), at the half-way point (September 2012 "C") and a re-test of "B1" as "B2" at the end of the study (June 2013). Texts were written during the entire period. The control group for the written surveys consisted of over 60 classmates attending the same year in the same school. The differences between the groups were tested for the statistical significance of the single variables assigned.
The theoretical part investigates the concept of competence within the context of schools, as defined by the OECD and the German Standing Conference of the Ministers of Education and Cultural Affairs (KMK), and the school-relevant theory models of media literacy and examples for the measurement of media literacy. After looking at the fundamentals and examples of peer education research findings, peer-based media-competence promotion in Germany until the end of 2014 was considered. Here 19 peer programmes are presented and compared. There are significant similarities despite a diversity of programmes with regard to reach, from local to nationwide, and the forms of organization: Peer-based promotion of media competence is initiated by institutions and in response to problems of media use. For this reason, themes treated are almost exclusively those involving youth media protection. The target group is children and young people of a minimum age of 12 up to 16 years. They are trained by adult experts (from media education, substance abuse counselling, police, youth protection, data protection etc. in accordance with the programme requirements), but not by (their own) teachers. The training topics are similar in all programmes and involve the usual youth media protection topics as described in the keywords: social networks, privacy, cyber-harassment, copyright, cell phones, excessive media use and adult content. These topics are expressly supplemented in some programmes through the promotion of social and personal skills (such as teamwork or empathy ability, etc.).
The media literacy model from Groeben (2002b) was used as the theoretical framework to describe the development of media literacy in young people, further operationalised here for educational purposes and on the basis of models of educational standards in the form of a five-stage model for the assessment of the performance of students between the ages of 13 to 15 years. The hypotheses 1 to 7 of this investigation are formulated along the dimensions of the media competence model. They posit a positive performance development in "Media knowledge" (hypothesis 1), "Media specific reception pattern" (2), "Ability to enjoy" (3), "Critical abilities" (4), "Orientation abilities" (5), "Productive participation pattern" (6) and "Communication connections" (7). In addition, an improved media literacy development in comparison to the control group (hypothesis 8) and a positive self-assessment of their activities as peer multipliers (hypothesis 9) were assumed. All hypotheses were confirmed. The media scouts showed on average a positive development of their media literacy, even in comparison with the control group. They saw their functions as peer multipliers positively through a positive self-image, a positive evaluation of their self-efficacy and in response to social peer aspects.
The media scouts as individuals acquired a wider "media knowledge" and demonstrated an improved "media awareness" at the end of their two-year training/work as peer multipliers. The effects were particularly strong when content and themes were relevant to the school education programme, such as in the knowledge of the mechanisms of advertising on the search engine Google. The media scouts demonstrated a further development of "media-specific reception patterns" and in particular the technological-instrumental skills involved. This was particularly strong in the skills/abilities applied by young people in their everyday use of media. That is to say, they were particularly proficient at learning the skills needed for their daily media needs. This was evident, for example, in their increased skill in the use of social media services, software for word processing or in the productive use of video portals (YouTube uploads). In an individual evaluation of problem-solving strategies employed by media scouts, wide differences in quantity and combinations of different strategies was observed. An "ability to enjoy" as part of a media literacy as defined by Groeben was difficult to describe. Nevertheless, the media scouts demonstrated that they enjoyed "their" media, even if only as a superficial pleasure and one which the young people did not reflect on except in one case. The exception is the media scouts' capacity to explain why they like to play computer games. It was shown that the youth have "universal media", such as smartphones, which they use for every mood or feeling. Moreover, the media scouts evaluated their "ability to enjoy" as high. This also applies to the investigation category "critical abilities", one of Groeben's dimensions of media literacy. Self-assessment was high for this category, however "critical abilities", if an initial relatively non-critical use of the media is assumed, may have been improved during the study period. This is not valid for all media content in general, but only for the topics/content forming part of the education programme. For example, critical abilities regarding the challenges "gender roles in media offers" and "companies advertising on Facebook" could not be improved as they were not explicitly part of the media scout training concepts. Other topics, such as "search capabilities through search engines" and "source analysis", revealed quite different results. The dimension of "selection/combination of media use" showed, as expected, that all young people surveyed (media scouts as well as the control group) are intensive media users. The media scouts were able to increase their performance here, even if it was obvious that young people take little advantage of the alternatives offered. For example, "Wikipedia" is selected, without much thought, as the sole research source for written research tasks, This hardly changed over the two years of the investigation period. A recurring pattern was seen whereby young people view the media not as an end in itself, using it instead specifically for their purposes. The authors did not interpret a lack of ability in the dimension "search sources" here, but rather, a lack of understanding of the necessity of alternatives. In other words, for young people, "Wikipedia" is sufficient as a source for a research paper. As mentioned above, the youth use their smartphone as a "universal medium" which as a device requires no further "selection/combination". Unexpectedly, results for the dimension "productive participation patterns" were not available. Few young people took advantage, despite the availability, of the necessary media productive-creative hardware and software, whereby one third used social media applications such as "Facebook". However, they assess themselves realistically in this dimension and evaluate themselves as worse than in the other six of Groeben's dimensions. Important here is not so much the lack of skills/abilities as the lack of a motivation to use the media productively and creatively. In other words, they were capable of using the media in this manner, but did not want to. The communication connections used by the youth were as high as expected, with one exception. They communicate frequently over the media, both media mediated and in personal conversations. Here - as expected - others of the same age were the main interlocutors, and in particular in the role of confidant. But not all the young people demonstrated Groeben's "communication connections". Only about three-quarters communicate through media experiences. The authors did not expect that no young person mentioned using the media named a teacher as an interlocutor.
The media scout and control groups showed hardly any difference from one another at the beginning of the study, other than in their technical affinities and slightly higher media usage. Despite a very intensive use of media, the themes friends/friendship are the most important for all surveyed adolescents, and their interest in topics is not based exclusively on the media involved.
The 20 media scouts in the study have a mostly positive self-image and high levels of self-confidence. They are restrained in their estimates of their self-efficacy, but are in the main not negative. Young people feel very confident in their handling of devices such as computers.
The study confirmed an improved self-image and assessment of self-efficacy through the social aspects of peer education, as well as a promotion of media literacy in the media scout approach of peer based media literacy promotion
Soot diagnostics in high pressure laminar premixed flames: Combination of laserinduced incandescence (LII) with optical and conventional diagnostics
Im Rahmen dieser Arbeit wird die Anwendung des berührungslosen Partikel-diagnostischen Verfahrens der laserinduzierten Inkandeszenz (LII) in vorgemischten Hochdruckflammen (1–30 bar) untersucht. Dabei werden im Wesentlichen zwei Fragen beantwortet: Ist der von Mel-ton [1] postulierte lineare Zusammenhang zwischen der maximalen LII-Signalintensität und dem Rußvolumenbruch auch für Hochdruck-Anwendungen gültig und wie stark abhängig sind die durch zeitaufgelöste LII (TiRe-LII) bestimmten Partikelgrößen vom angenommen Wärmeleitungsmodell, der Aggregation und der Partikelgrößenverteilung?
Zur Beantwortung der ersten Frage wird der durch LII bestimmte Rußvolumenbruch mit Ex-tinktionsmessungen verglichen. Um die Vergleichbarkeit beider Verfahren zu gewährleisten werden beide Messverfahren zunächst an der nicht-vorgemischten Flamme eines Gülder-Brenners bei Atmosphärendruck verglichen. Dabei wird die Extinktion bei verschiedenen Wellenlängen angewendet und in mehreren Bereichen der Flamme untersucht. Da selbst die längste verwendete Extinktionswellenlänge (825 nm) noch Absorption an Rußvorläufern zeigt, wird als Schlussfolgerung für die Hochdruck-Messungen ein Bereich in der Flamme ausgewählt, in dem keine Rußvorläufer mehr vorhanden sind. Die Ergebnisse zeigen eine starke Abhängigkeit des durch LII bestimmten Rußvolumenbruchs von der Abschwächung des Inkandeszenz-Signals durch den Flammenruß auf dem Weg zum Detektor. Nach der Korrektur bezüglich Reabsorption stimmen beide Verfahren jedoch gut überein, sodass ge-zeigt werden kann, dass der beschriebene lineare Zusammenhang zwischen LII-Signalintensität und Rußvolumenbruch auch im Hochdruck gültig ist. Zur Anwendung der Absorptions-Korrektur ohne zusätzliche Messverfahren ist jedoch Wissen über den generellen Trend der Rußverteilung im untersuchten Objekt notwendig. Es wird jedoch auch gezeigt, dass der beobachtete Effekt der Signal-Reabsorption nur relevant wird, wenn zugleich hohe Rußkonzentrationen und große Dicken der Rußzone (Weg zum Detektor) vorhanden sind. So bewirkt beispielsweise eine Rußkonzentration von 10 ppm bei einer Dicke der Rußzone von 1 mm lediglich eine Signal-Reabsorption von 10%. Jedoch tritt Rußbildung in der Regel an nicht-vorgemischten Flammen auf und diese besitzen deutlich weniger dicke Rußzonen.
Zur Beantwortung der zweiten Frage werden die zeitaufgelösten Signale der Laserinduzierten Inkandeszenz (TiRe-LII) ausgewertet und die resultierenden Partikelgrößen mit TEM-Messungen an Rußproben verglichen. Hierfür wurde ein neuartiges Probenahme-System für Hochdruckflammen entwickelt und umfassend charakterisiert. Für die Bestimmung der Parti-kelgrößen durch TiRe-LII werden verschiedene Wärmeleitungsmodelle mit und ohne Aggre-gation bzw. polydisperser Größenverteilung angenommen. Der Vergleich zeigt eine fast per-fekte Übereinstimmung zwischen den Ergebnissen von LII und TEM für 16–20 bar unter An-nahme des Fuchs-Modells (inkl. Aggregation und polydisperser Verteilung) und einer sehr guten Übereinstimmung für 1–10 bar unter Annahme des freimolekularen Modells.

Opt. 23, 2201-2208 (1984).In this work, the application of the non-intrusive particle diagnostics method laser-induced incandescence (LII) is investigated in premixed high-pressure flames at 1–30 bar. Two ques-tions will be answered: Is the linear relationship between the maximum LII signal intensity and the soot volume fraction postulated by Melton [1] valid also for high pressure applications and how strongly does the particle-size measurement with time-resolved LII (TiRe-LII) depend on the heat conduction model, aggregation, and the particle-size distribution?
To answer the first question, the soot volume fraction determined with LII is compared to the result of extinction measurements. To ensure comparability, both methods are first applied to measurements in a non-premixed flame of a Gülder burner at atmospheric pressure. Extinc-tion measurements are carried out with various wavelengths and in several regions of the flame. Since even the longest extinction wavelength used (825 nm) is also affected by ab-sorption of soot precursors, for the high-pressure measurements an area in the flame is se-lected, where no soot precursors are present. The results show a strong dependence of the soot volume fraction determined by LII on the attenuation of the incandescence signal by soot on the way to the detector. After correction for reabsorption, both methods are in good agreement and the described linear relationship between LII signal intensity and soot volume fraction is fund to be valid even at high pressure. For the application of the absorption correc-tion without a complementary measurement, knowledge of a general trend of the distribution of the soot volume fraction in the investigated object is necessary. It is also shown that the observed effect of the signal attenuation is only relevant if both high soot concentrations and an extended area of the soot zone (along the signal path towards the detector) are present. For example, a soot concentration of 10 ppm at a soot zone thickness of 1 mm results in sig-nal reabsorption of only 10%. However, soot occurs mostly in non-premixed flame and these have significantly less thick soot zones.
To answer the second question, time-resolved signals of laser-induced incandescence (TiRe-LII) are investigated and the resulting particle size is compared to results from soot samples investigated with TEM. For this purpose, a new sampling system for high-pressure flames was developed and characterized. For the analysis of TiRe-LII signals, different heat-conduction models are assumed with or without aggregation or polydisperse size distribution. The comparison shows an almost perfect agreement between the LII and TEM results for pressures between 16 and 20 bar assuming the Fuchs model (incl. aggregation and polydis-perse distribution) and a very good agreement for pressures at 1 to 10 bar assuming the free molecular model.

The impact of hypoxia-inducible factor-1 in myeloid cells on inflammatory bowel disease
Chronisch entzündliche Darmerkrankungen (CED) stellen weiterhin eine Krankheit dar, von der mehr als 400.000 Menschen in Deutschland betroffen sind (Stand 2010) und für die es bislang keine Heilungsmöglichkeit gibt.
Es ist bereits bekannt, dass Entzündungen immer mit einem Mangel an Sauerstoff (Hypoxie) verbunden sind, und dass der Hypoxie-induzierbare Faktor-1 (HIF-1) eine protektive Rolle in verschiedenen Immunzellen (T-Zellen, DC) und Epithelzellen während CED spielt. Außerdem konnte bereits gezeigt werden, dass HIF-1 für verschiedene Funktionen von Makrophagen essenziell ist. Der Einfluss von HIF-1 in Makrophagen während CED wurde jedoch bisher noch nicht untersucht. Daher war es Ziel dieser Arbeit die Rolle von myeloischem HIF-1 während DSS-induzierter Colitis in der Maus zu untersuchen.
Nach erfolgreicher Verifizierung der Knock-out Effizienz von ca. 80 % wurden Tiere mit und ohne funktionelles HIF-1α in Makrophagen für 4-6 Tage mit 2,5 % DSS behandelt um eine Colitis zu induzieren. Hier zeigten die HIF-1α defizienten Tiere ein schwächer ausgeprägtes Krankheitsbild der DSS-induzierten Colitis mit reduziertem Gewichtsverlust und DAI und abgemilderter Colon-Gewebeschädigung. Einhergehend waren reduzierte Anzahlen verschiedener inflammatorischer Immunzellen wie Makrophagen, Neutrophile, DC und Th, wie auch verminderte Mengen inflammatorischer Zytokine wie TNFα und IFNγ.
Da HIF-1α bekanntermaßen an der Motilität und Invasion von Makrophagen beteiligt ist, ist für die hier vorliegenden Ergebnisse anzunehmen, dass es in den HIF-1α defizienten Tieren zu einer reduzierten Rekrutierung von Makrophagen ins entzündliche Colongewebe in Folge der DSS-induzierten Epithelschädigung gekommen ist. Folgend wurden dementsprechend reduzierte Mengen an Zytokinen ausgeschüttet und somit weniger weitere Immunzellen rekrutiert und aktiviert.
Des Weiteren konnte eine erhöhte Anzahl anti-inflammatorischer Treg in HIF-1α defizienten Tieren identifiziert werden. Dies könnte zu einem begünstigenden Verhältnis von anti-inflammatorischen Treg zu inflammatorischen Th1/Th17 in den HIF-1α defizienten Tieren beigetragen haben, wodurch in diesen Tieren ein abgemildertes Krankheitsbild zustande kam.
Die Ergebnisse dieser Arbeit erweitern bereits vorliegende Daten zur Rolle von HIF-1α in Epithelzellen und Immunzellen wie T-Zellen und DC während Colitis. Während eine HIF-Induktion in diesen Zellen eher protektive Effekte im Tiermodell vermuten ließ, deuten unsere Daten auf eine krankheitsfördernde Rolle von HIF 1α in myeloischen Zellen hin. Eine Verabreichung von globalen HIF-Stabilisatoren zur therapeutischen Behandlung von CED ist daher kritisch zu prüfen, da HIF-1 durchaus konträre Rollen in den unterschiedlichen Zelltypen einnehmen kann.Inflammatory bowel disease (IBD) still remains incurable and affects more than 400,000 people in Germany (as in 2010).
It is known that inflammation is related to an inappropriate oxygen supply (hypoxia) and hypoxia inducible factor-1 (HIF-1) has a protective role in various immune cells (T cells, DC) and epithelial cells during IBD. Additionally it has been shown that HIF-1 is essential for various functions of macrophages. The impact of HIF-1 on macrophages during IBD still remains unknown. This prompted us to investigate the role of myeloid HIF-1 during DSS induced colitis.
Knockout of HIF-1α in myeloid cells showed an efficiency of nearly 80 %. Animals with and without myeloid HIF-1α were treated with 2.5 % DSS for 4 6 days to induce colitis. Mice deficient in myeloid HIF-1α showed a milder DSS induced colitis with reduced weight loss and DAI and milder colon tissue damage. This was accompanied by reduced numbers of various inflammatory immune cells like macrophages, neutrophils, DC, and Th. This was accorded by diminished amounts of inflammatory cytokines like TNFα and IFNγ.
HIF-1α is known to be involved in macrophage motility and invasion. The results of this study might rely on the fact that mice deficient in HIF-1α showed a reduced recruitment of macrophages to sites of DSS-induced inflammation. Accordingly, reduced amounts of cytokines were released and therefore diminished numbers of further immune cells were recruited.
Furthermore HIF-1α deficient mice showed increased numbers of anti-inflammatory Treg. This could contribute to a beneficial balance between anti-inflammatory Treg and inflammatory Th1/Th17 in HIF-1α deficient mice and an attenuated DSS induced colitis in these mice.
The results of this study underline the pivotal role of HIF-1α in epithelial cells and immune cells like T cells and DC during colitis. Whereas induction of HIF in these cells reveals a protective role during colitis, our results indicate a disease promoting role of myeloid HIF-1α. Thus the systemic application of HIF-stabilizing PHD inhibitors in IBD should be considered with great care, because HIF-1 might act diversely in various cell types
Analyses of the impact of DNA methylation and methylation-modifying enzymes in different cell systems
Im Rahmen der vorliegenden Arbeit wurde die Bedeutung der DNA-Methylierung und methylierungsmodulierender Enzyme in verschiedenen Geweben untersucht.
Ein charakteristisches Merkmal der Alzheimer-Krankheit sind reduzierte Somatostatin- und Somatostatin-Rezeptor-Level im Gehirn, die mit dem Ausbruch der sporadischen Form der Krankheit in Verbindung gebracht werden. Um zu klären, ob die Reduktion des Peptidhormons Somatostatin (SST) und des im Kortex von Alzheimer-Patienten am stärksten reduzierten Rezeptor-Subtyps 4 (SSTR4) auf eine Hypermethylierung der entsprechenden Promotorregion zurückzuführen sein könnte, wurden diese Regionen mittels Bisulfit-Sequenzierung in Zellen von Alzheimer-Patienten und nicht-dementen Normalpersonen analysiert. Die ähnlichen Methylierungslevel dieser Promotorregionen in verschiedenen Gehirnregionen von Patienten und nicht-dementen Normalpersonen unterstützen jedoch nicht die Hypothese eines Zusammenhangs von DNA-Hypermethylierung und den reduzierten SST- und SSTR4-Leveln einer Alzheimer-Erkrankung. Es fand sich jedoch ein Trend zu erhöhter Methylierung des SST-Promotors mit erhöhtem Alter.
In einem weiteren Teil dieser Arbeit wurden drei unabhängige transgene Linien eines Mausmodells zur ubiquitären Überexpression der somatischen Form der DNA Methyltransferase 1, Dnmt1s, untersucht. Anhand dieses Modells sollten ursprünglich die Konsequenzen einer solchen Überexpression analysiert werden. Es stellte sich heraus, dass transgene Tiere zwar eine bis zu 229-fach erhöhte Dnmt1-mRNA-Expression aufwiesen, jedoch keine erhöhten Dnmt1-Proteinlevel besaßen. Reportergenversuche weisen auf eine reduzierte Translationseffizienz des Transgenkonstrukts hin, die jedoch nicht allein das Ausbleiben erhöhter Proteinlevel im Mausmodell erklären kann. Es bleibt offen, ob die statischen Proteinlevel auf spezifische Eigenschaften des Transgens oder auf eine stringente Kontrolle der Dnmt1-Level in vivo zurückzuführen sind.
Im Hauptteil der vorliegenden Arbeit wurde der Einfluss moderat veränderter TET-Dioxygenase-Level auf HEK293-Zellen untersucht. Die Überexpression von TET1 führte zu erhöhten globalen 5hmC-Leveln und verursachte eine leichte Hypomethylierung von Promotoren, Genen und CpG-islands. Im Gegensatz dazu führte der simultane Knockdown von TET1, TET2 und TET3 zu verringerten globalen 5hmC-Leveln und einer leichten Hypermethylierung der genannten Regionen. Auch wenn die Überexpressions- bzw. Knockdown-Experimente entgegengesetzte Methylierungsveränderungen hervorriefen, gab es nur eine kleine Anzahl reziproker Veränderungen. Die Methylierungsveränderungen beider Experimente geschahen mit unterschiedlicher Präferenz in Abhängigkeit vom endogenen Methylierungsgrad und der Stärke der Genexpression. Sowohl nach der TET1-Überexpression als auch nach dem TET-Knockdown wurden geringfügig hoch- und herunterregulierte Gene identifiziert. Die Expressionsveränderungen konnten jedoch nicht mit Methylierungsveränderungen der Genpromotoren in Verbindung gebracht werden und sind womöglich auf nicht-katalytische Funktionen der TET-Enzyme zurückzuführen. Die Kartierung der 5hmC-DNA-Modifikation im genomweiten Maßstab zeigte, dass die TET1-Überexpression die genomweite 5mC-Oxidation ohne einen erkennbaren Verteilungsschwerpunkt induzierte. Detailliertere Analysen offenbarten den Trend einer unterschiedlichen Suszeptibilität für eine Oxidation durch TET1, der abhängig vom endogenen 5hmC-Vorkommen war und in allen untersuchten genomischen Elementen und Regionen vorlag. Zusammengenommen weisen die in dieser Arbeit durchgeführten Untersuchungen auf eine ausgedehnte Rolle der TET-Familie bei der Regulation der DNA-Methylierungslevel hin.This thesis focused on elucidating the relevance of DNA methylation and methylation-modulating enzymes in different tissues.
Reduced levels of somatostatin (SST) and its receptors represent pathological characteristics of Alzheimer’s disease (AD) brains, and deregulated somatostatin signaling has been proposed to play a major role in the development of late-onset sporadic AD. To investigate whether the reduced levels of somatostatin and its most affected receptor subtype 4 (SSTR4) could be caused by DNA hypermethylation of the respective promoter regions, these regions were analyzed in cells from the neocortex of AD patients and non-demented controls using bisulfite sequencing. Tissue samples from two areas of the neocortex were analyzed, each separated into cortical gray and infracortical white matter. However, the similar methylation levels observed in the SST and SSTR4 promoter regions in all samples from AD patients and non-demented controls are not supportive of a causal role of DNA hypermethylation with respect to altered somatostatin signaling in AD. But there was a trend toward increased SST promoter methylation with increasing age.
Consequences of ubiquitous overexpression of the somatic form of the DNA methyltransferase 1, Dnmt1s, were supposed to be studied in a transgenic mouse model. Three independent lines of this model were analyzed. Transgenic mice ubiquitously overexpressed Dnmt1 mRNA up to 229-fold, but were lacking increased protein levels. Although reporter gene assays indicated a reduced translation efficiency of the transgene construct, this reduction was not strong enough to solely account for the lack of protein increase in the mouse model. It remains unclear whether the static protein levels are due to intrinsic features of the transgene or a stringent control of Dnmt1 levels in vivo.
The main part of this thesis shows that altering TET dioxygenase levels within physiological range can affect DNA methylation levels of HEK293 cells. TET1 overexpression led to increased global 5hmC levels and mild hypomethylation of promoters, gene bodies and CpG islands. Conversely, the simultaneous knockdown of TET1, TET2 and TET3 led to decreased global 5hmC levels and was accompanied by mild hypermethylation of above-mentioned regions. Although the overexpression and knockdown studies caused opposite methylation changes, most changes were non-reciprocal and occurred with different preference depending on endogenous methylation and gene expression levels. Both, TET1 overexpression and TET knockdown, led to slight transcriptional up- and downregulation. However, the expression changes could not be linked to methylation changes of the corresponding gene promoters and were probably caused by non-catalytic functions of the TET enzymes. 5hmC profiling performed on a genome-wide scale showed that TET1 overexpression induced 5mC oxidation without a distribution bias. Detailed analyses revealed a trend of different susceptibility to TET1-induced oxidation that was linked to endogenous 5hmC content and found in all genomic elements and regions. Taken together, the experiments conducted point to a widespread role of the TET family in regulating DNA methylation levels
