    Molecular characterization of H10 subtype avian influenza virusesisolated from poultry in Eastern China

    AbstractIn recent years, avian-origin H10 influenza viruses have proved capable of infecting human beings, and they pose a potentialpublic health threat. Seven H10 avian influenza viruses (AIVs), H10N3 (n = 2), H10N7 (n = 1), and H10N8 (n = 4), wereisolated from chickens in Zhejiang Province, Eastern China, during surveillance of AIVs in live poultry markets in 2016and 2017. Phylogenetic analysis indicated that Zhejiang H10 strains received gene segments from H10, H3, and H7 virusesfrom birds in East Asia. Animal inoculation tests showed that these isolates have low pathogenicity in mice and can replicatein this species. Our findings suggest these H10 AIVs have the ability to adapt to chicken or other poultry, and highlight theneed of long-term surveillance

    AbstractTumor growth, invasion and metastasis are dependent on angiogenesis. The Vascular endothelial growth factor (VEGF)/VEGF receptor 2 (VEGFR2) signaling pathway plays a pivotal role in tumor angiogenesis and therefore represents a reasonabletarget for anti-angiogenesis/anti-tumor therapy. In the present study, we generated T4 recombinant phages expressingthe extracellular domain of VEGFR2 (T4-VEGFR2) and investigated their anti-angiogenic activity. The T4-VEGFR2 phageswere able to bind to VEGF specifically and inhibit VEGF-mediated phosphorylation of VEGFR2 and its downstream kinasessuch as extracellular signal-regulated kinase (ERK) and p38 mitogen activated protein kinase (MAPK). The in vitro experimentsshowed that the T4-VEGFR2 phages could inhibit VEGF-stimulated cell proliferation and migration of endothelialcells. Finally, administration of T4-VEGFR2 phages was able to suppress tumor growth and decrease microvascular densityin murine models of Lewis lung carcinoma and colon carcinoma, and prolong the survival of tumor bearing mice. In conclusion,this study reveals that the recombinant T4-VEGFR2 phages generated using T4-based phage display system caninhibit VEGF-mediated tumor angiogenesis and the T4 phage display technology can therefore be used for the developmentof novel anti-cancer strategies

    AbstractChronic kidney disease of unknown aetiology (CKDu) reported in Sri Lanka and other countries is a mysterious and seriousdisease. Recently, we reported a high seroprevalence of antibodies to a hantavirus antigen among CKDu patients inGirandurukotte, Badulla district, Sri Lanka. However, the type of hantavirus with which the residents were infected was notdetermined. In this study, a total of 89 seropositive sera were examined to identify their serotypes using an indirect immunofluorescentantibody assay, a truncated-N-protein-based enzyme-linked immunosorbent assay, and a cross-neutralizationtest. These results indicated that the residents in this area were frequently infected with Thailand orthohantavirus or anantigenically related virus

    The complete genome sequence analysis of West Nile virus strainsisolated in Slovakia (central Europe)

    AbstractThe present study reports the first complete genome sequence analysis of West Nile virus (WNV) strains isolated from brainsamples from raptors. The results prove the circulation of closely related WNV lineage II strains in central Europe and geneticanalysis revealed seven amino acid substitutions in structural (PrM3, E159and E231)and in non-structural (NS1109, NS5259,NS5310and NS5600)proteins. Observed amino acid substitutions Phe3and Ser231were common only within the lineage VIIKoutango strain isolated from Rhipicephalus guilhoni tick in Senegal. Further research could reveal whether these substitutionsinfluence the biological properties of WNV, including virulence and neuroinvasiveness

    End-to-middle-to-end solution for IMS media plane security

    Abstract IP multimedia subsystem (IMS) is becoming the prevailing candidate for managing future mobile multimedia communications, including critical communications such as public safety, emergency professionals and corporate networks. IMS security and privacy has gained much attention in the few last years. The review of recent IMS security activities stresses the inclusion of intermediate nodes in the media path of secured communications as an open issue. This paper presents an endto-middle-to-endsolutionwhichenablestheusageofIMSmediaplaneelementssuch asrecorders,transcodersandnovelcross-cipheringfunctionsinasecureway.Theproposed solution, which is fullycompliant withIMS, includes the network architecture, the signaling plane for session signaling and key management, and the media-plane security characteristics. Experimental results demonstrate that the proposed solution can provide media interoperability (both transcoding and cross-ciphering) with a cost of 17 % overhead to a standard IMS call setup in the signaling plane

    Genomic characterization and pathogenicity of porcinedeltacoronavirus strain CHN?HG?2017 from China

    AbstractPorcine deltacoronavirus (PDCoV) was first detected in Hong Kong and has recently spread to many countries around theworld. PDCoV causes acute diarrhea and vomiting in pigs, resulting in significant economic losses in the global pork industry.In this study, a Chinese PDCoV strain, designated CHN-HG-2017, was isolated from feces of a suckling piglet with severewatery diarrhea on a farm located in central China. Subsequently, the virus was identified by an indirect immunofluorescenceassay and electron microscopy. A nucleotide sequence alignment showed that the whole genome of CHN-HG-2017 is 97.6%-99.1% identical to other PDCoV strains. Analysis of potential recombination sites showed that CHN-HG-2017 is a possiblerecombinant originating from the strains CH/SXD1/2015 and Vietnam/HaNoi6/2015. Furthermore, the pathogenicity of thisrecombinant PDCoV strain was investigated in 5-day-old piglets by oral inoculation. The challenged piglets developed typicalsymptoms, such as vomiting, anorexia, diarrhea and lethargy, from 1 to 7 days post-inoculation (DPI). Viral shedding wasdetected in rectal swabs until 14 DPI in the challenged piglets. Interestingly, high titers of virus-neutralizing antibodies insera were detected at 21 DPI. Tissues of small intestines from CHN-HG-2017-infected piglets at 4 DPI displayed significantmacroscopic and microscopic lesions with clear viral antigen expression. Our analysis of the full genome sequence of arecombinant PDCoV and its virulence in suckling piglets might provide new insights into the pathogenesis of PDCoV andfacilitate further investigation of this newly emerged pathogen
