    AbstractTransmissible gastroenteritis virus (TGEV) infection causes severe diarrhea in piglets and imposes a significant economicburden on pig farms. Single-chain fragment variable (scFv) antibodies effectively inhibit virus infection and could be apotential therapeutic reagent for preventing disease. In this study, a recombinant scFv antibody phage display library wasconstructed from peripheral blood lymphocytes of piglets infected with TGEV. The library was screened with four roundsof biopanning using purified TGEV antigen, and scFv antibodies that bound to TGEV were obtained. The scFv gene wassubcloned into the pET-28a(+), and the constituted plasmid was introduced into Escherichia coli BL21 (DE3) for proteinexpression. All three scFv clones identified had neutralizing activity against TGEV. An immunofluorescence assay andwestern blot analysis demonstrated that two scFv antibodies reacted with the spike protein of TGEV. These results indicatethat scFv antibodies provide protection against viral infection in vitro and may be a therapeutic candidate for both preventionand treatment of TGEV infection in swine

    AbstractDengue disease is characterized by a marked decrease in platelet count, which is life threatening. In the present study, weinvestigated the antiviral activity of an aqueous extract of Carica papaya leaves (PLE) against dengue virus (DENV) and itseffect on platelet augmentation. The anti-dengue activity of PLE in DENV-infected THP-1 cells was examined by immunoblottingand flow cytometry. The effect of PLE on erythrocyte damage was investigated using hemolytic and anti-hemolyticassays. Virus-infected THP-1 cells were assayed for IFN-? secretion. The effect of PLE on platelet augmentation in rats withcyclophosphamide-induced thrombocytopenia was also investigated. The platelet count of blood from the retro-orbital plexusof rats was determined on the 1st,4th,7th,11thand 14thday of study. On the 14thday, the rats were sacrificed for histopathologicalexamination of the liver, kidney and spleen. Plasma of thrombocytopenic rats was tested for thrombopoietin (TPO)and IL-6 secretion. The data suggest that PLE significantly decreases the expression of the envelope and NS1 proteins inDENV-infected THP-1 cells. A marked decrease in intracellular viral load upon PLE treatment confirmed its antiviral activity.This also resulted in a significant decrease in erythrocyte damage and hydrogen-peroxide-induced lipid peroxidation. Asignificant increase in the number of platelets was found in thrombocytopenic rats treated with PLE, along with an increasein IL-6 and TPO levels. These findings suggest that PLE can potentially be used as an antiviral agent, as it helps in plateletaugmentation and exhibits antiviral activity against DEN

    AbstractIn this study, we isolated a novel virulent Escherichia phage, SRT7. Its genome is a double-stranded linear DNA moleculecontaining 39,883 bp. Direct terminal repeats with a length of 175 bp, are present at both ends of the genome. The G+Ccontent is 50.54%. Forty-seven putative protein coding genes were identified. No tRNA or rRNA genes were identified.Comparative genomic analysis revealed that phage SRT7 is a novel member of the T7-like phage cluster, but it forms asingleton subcluster

    Viral metagenomics reveals significant viruses in the genital tractof apparently healthy dairy cows

    AbstractThe virome in genital tract secretion samples collected from 80 dairy cattle in Shanghai, China, was characterized. Virusesdetected included members of the families Papillomaviridae, Polyomaviridae, Hepeviridae, Parvoviridae, Astroviridae,Picornaviridae, and Picobirnaviridae. A member of a new species within the genus Dyoxipapillomavirus and six circularRep-encoding single-stranded DNA (ssDNA) (CRESS-DNA) viral genomes were fully sequenced and phylogeneticallyanalyzed. The prevalence of bovine polyomaviruses 1 and 2 was measured by PCR to be 10% (8/80) and 6.25% (5/80),respectively. PCR screening also indicated that the novel papillomavirus ujs-21015 and bovine herpesvirus 6 were presentin three and two out of the 80 samples, respectively
