    THE EFFECTIVENESS OF VITAMIN C AND E ADDITION IN VARIOUS DILUENTS OF BRAHMAN CATTLE SPERMATOZOA AFTER FREEZING

    This research aimed to determine the quality of Brahman cattle spermatozoa after freezing in various diluent ingredients (Egg Yolk Citrate, Egg Yolk Tris, Skim Milk) which added vitamin C and vitamin E. The research was carried out using 4 treatments with 3 different diluent ingredients, each of which was Vitamins C and E are added to each diluent ingredient at a dose adjusted to the type of diluent agent. The treatment given is P0: Diluent without Vitamin C and E; P1: Diluent + Vitamin C; P2: Diluent + Vitamin E; and P3: Diluent + Vitamin C and Vitamin E. Research data were analyzed using analysis of variance and followed by Duncan's test at the 5% and/or 1% level for different variables. The results showed that the addition of vitamin C and vitamin E had a significant effect (P0.05) on motility and viability in the egg yolk tris diluent and egg yolk citrate but had no significant effect (P0.05) in the skim milk diluent. The use of different diluents had no significant effect (P0.05) on motility, viability, and abnormalities. This research concludes that Vitamin C added to the diluent tris egg yolk, egg yolk citrate, and skim milk showed higher average motility, percentage of live spermatozoa, and abnormalities of post-freezing Brahman cow spermatozoa

    EFFECT OF Nigella sativa AS FEED ADDITIVE ON ORGANOLEPTIC, WATER, PROTEIN, AND CHOLESTEROL CONTENT OF BROILER CHICKEN MEAT WITH HEAT STRESS CONDITION

    This study aimed to determine the physical properties, organoleptic, proximate value, and cholesterol level of broiler meat given black cumin (Nigella sativa) as feed supplement and exposed to heat stress. This study used 15-day old chicken (DOC) broilers that were adapted for 14 days and treated on day 15 to day 28. This study used a completely randomised design (CRD) consisting of 5 treatment groups: P0 (commercial feed); P1 (commercial feed + heat stress 34-35 ); P2 (commercial feed + black cumin 4% + heat stress 34-35); P3 (commercial feed + black cumin 6% + heat stress 34-35 ); P4 (commercial feed + black cumin 8% + heat stress 34- 35 ). Feeding was given twice a day at 07.00 WIB and 17.00 WIB ad libitum. Heat stress was given from 10.00 - 15.00 WIB every day for 14 days. The chicken breast and thigh meat were collected on day 29 for analysis. The statistical analysis showed that feed supplement containing black cumin had no effect (P0.05) on the pH value, drip loss, texture, protein content, and structure of broiler meat given exposure to heat stress, but had effect (P0.05) on the organoleptic colour, smell and texture, and cholesterol level of broiler meat. Based on these results, it can be concluded that feeding with 4%, 6% and 8% black cumin did not change the pH, drip loss, texture value as well as protein and water content of broiler meat given heat stress, but produced better colour and aroma. The administration of 4% cumin can reduce cholesterol levels of broiler meat exposed to heat stress

    THE ACCURACY OF VAGINAL MUCUS CHARACTERISTICS AS CLINICAL SIGNS IN EARLY PREGNANCY DIAGNOSIS OF DAIRY CATTLE

    This study aims to observe the changes in the characteristics of vaginal mucus to diagnose early pregnancy in dairy cattle. The characteristics of vaginal mucus include pH value, consistency, volume, and Vaginal Electrical Resistance (VER) value to diagnose early pregnancy using a Draminski Estrous Detector. The sample used that collected in this research was 52 dairy cattle. The research method consists of taking samples and observing the characteristics of the mucus. Data analysis includes normality test, T-test and Pearson correlation. The results showed that the mucus volume of pregnant cattle was less (1.280.356 mL) than that of non-pregnant cattle (2.210.437 mL), the pH value in pregnant cattle tended to be acidic, while non-pregnant cattle were classified as neutral, and there were differences in the consistency of mucus in pregnant cattle (4.00.0), while non-pregnant cows tended to be liquid (2.60.204). The volume and consistency of vaginal mucus in pregnant and non-pregnant cattle were significantly different (P0.05). The VER value of pregnant cattle was higher (36563.2 ohms) than non-pregnant cattle (19055.3 ohms). The VER value was highly correlated (r= 0.819) with the pregnancy status of dairy cattle. Thus, VER can be used as an accurate indicator to determine the status of early pregnancy in dairy cattle

    THE QUALITY OF CHILLED RAM SEMEN AFTER STORAGE IN EXTENDER WITH ADDITION OF MELATONIN AT 4 C

    Semen was collected using an artificial vagina, and then examined macroscopically and microscopically. Semen with motility of 70% was diluted into different extenders: tris egg yolk-glucose (TEYG), tris egg yolk-glucose extender with 0.5 mM melatonin (TEYG + M0.5), tris egg yolk-glucose extender with 1 mM melatonin (TEYG + M1), tris egg yolk-glucose extender with 2 mM melatonin (TEYG + M2). All semen samples were stored at 4 C for 5 days (H0-H5) and examined every 24 hours. Semen was examined for spermatozoa motility, viability, abnormality, and membrane plasma integrity. The results showed that there were no significant changes of semen quality on H0 (P0.05). In general, the melatonin supplemented groups had a higher semen quality value than the control. Melatonin supplementation can protect and maintain the quality of chilled semen. In addition, the result showed that TEYG + M1 extender was the best extender treatment, which was indicated by higher values than other treatments

    ANALYSIS OF THE ACUTE IMMUNE RESPONSE OF PACIFIC WHITE SHRIMP (Litopenaeus vannamei) INFECTED BY Vibrio parahaemolyticus AND ITS METABOLITES

    This study was aimed to observe the survival rate (SR) and clinical signs, and the acute immune response of Pacific white shrimp (Litopenaeus vannamei), including total hemocyte count (THC), bacterial clearance, and prophenoloxidase (ProPO) activity due to infection with the Vibrio parahaemolyticus bacteria and its metabolites. Shrimp were infected by bacterial isolates with culture media (A), bacterial isolates without culture media (B), and metabolites suspensions from bacterial culture media (C) for 8 hours with observation every 2 hours. The results showed that experimental shrimps in groups A and B showed clinical signs and reduced SR, 24.44% and 62.22%, respectively. Experimental group C still showed the clinical signs but did not cause the mortality (SR 100.00%). The THC did not show significant changes, either based on experimental groups or duration of infection, but all groups tended to show decreasing THC at 8 hours post-infection. Bacterial clearance and ProPO activity also did not show significant differences between experimental groups or duration of infection at 8 hours post-infection. However, there was an increase in bacterial clearance activity at 6 hours post-infection in the experimental groups A and B. Based on the study results, the acute immune response in shrimp from all of the experimental groups did not show significant changes at 8 hours post-infection. However, group A significantly decreased SR compared to groups B and C

Kit production as a diagnostic tool to support the development of screening for Alzheimers disease is a strategic effort. Screening based on detection with Enzyme-Linked Immunosorbent Assay (ELISA) can work effectively and accurately to optimize a specific antibody material or monoclonal antibody to the A42 peptide.     IDENTIFICATION OF QUANTITATIVE AND QUALITATIVE TRAITS DIVERSITY IN LOCAL BUFFALO IN NAGAN RAYA DISTRICT

    b-districts, Beutong and Seunagan, with 45 buffalo samples from each sub-district. The survey method with random buffalo sampling was employed in this study. The selection of the two research sub-districts was based on the highest buffalo population numbers in Nagan Raya District. The research locations were chosen from six villages: Babah Krueng, Gunong Nagan, Blang Seunong, Sapek, Alue Dodok, and Blang Pateuk. Primary data were obtained from measuring several quantitative traits of buffaloes aged 2.5-3.0 years in the field. All data were analyzed using descriptive statistics. The research results show that the quantitative traits based on the coefficient of variation (CV) of body measurements of buffaloes in Nagan Raya vary considerably. The body measurements of male buffaloes in Beutong Sub-District showed chest circumference, shoulder height, and body length of 172.70 cm, 121.84 cm, and 111.48 cm, respectively, while the female buffalo's chest circumference, shoulder height, and body length were 180.86 cm, 122.18 cm, and 119.32 cm, respectively. The body measurements of male buffaloes in Seunagan Sub-District showed chest circumference, shoulder height, and body length of 169.23 cm, 115.91 cm, and 105.23 cm, respectively, while the female buffalo's chest circumference, shoulder height, and body length were 187.89 cm, 123.04 cm, and 117.39 cm, respectively.     THE EFFECTIVENESS OF STEM CELL THERAPY IN MYOCARDIAL INFARCTION REVIEWED FROM UREUM, CREATININE AND KIDNEY HISTOPATHOLOGY LEVELS

    This study aims to study the effectiveness of single placental stem cell therapy or cardiomyocyte combination in pigs experiencing myocardial infarction in terms of urea, creatinine and kidney organ histopathology values. A total of 9 pigs were divided int o three groups: without therapy (K1), single placental stem cell therapy (K2), and cardiomyocyte coculture combination therapy (K3). All pigs underwent circumflex artery ligation of the proximal coronary artery branch to stimulate cardiac ischemia and therapy was given after myocardial infarction and ST-elevation were formed. The pigs were then treated for eight weeks, euthanized and necropsied for kidney organ removal. The re sults showed a significant decrease in urea and creatinine values (P0.05) in K2 and K3 as compared to K1. Histopathology results showed a decrease in glomerular atrophy values (K2: 10.260.93 and K3: 12.441.21), tubular necrosis (K2: 13.673.76 and K3: 17.230.55), and tubular dilation (K2: 16.361.26 and K3: 18.031.76) and these values were significantly different than those observed in K1. It was concluded that the administration of placental stem cell therapy or combined with cardiomyocyte coculture could reduce urea, creatinine values, and improve kidney tissue histopathologically

    PHYSICO-CHEMICAL VALUE AND HYPOGLYCEMIC EFFECT OF INDUSTRIAL-GRADE ETHANOLIC EXTRACT OF PIGEON PEA (Cajanus cajan) LEAVES AND GINGER (Zingiber officinale var. amarum)

    This study aimed to analyze flavonoid and 6-gingerol content and to test the hypoglycemic effects of industrial-grade Cajanus cajan (pigeon pea) leaf and Zingiber officinale rhizome (ginger) extracts on hyperglycemic rats. Extraction utilized maceration with 70% alcohol at PT. Insular Multi Natural. Extracts were characterized and analyzed for flavonoids and 6-gingerol content. Hypoglycemic activity was assessed using the oral glucose tolerance test (OGTT) on rats with induced hyperglycemia via 90% sucrose solution. The results indicated higher flavonoid content in C. cajan leaf extract (4090.09338.23 mg/100 g) compared to Z. officinale extract (622.74203.39 mg/100 g), while gingerol content in Z. officinale extract was 701.524.42 mg/100 g. This study concluded that combination of industrial-grade pigeon pea leaves extract and ginger extract reduce the level of blood glucose in hyperglycemic model rats at all combination dose (150:150, 200:100, and 100:200 mg/kg BW). The combination dose of 200:100 mg/kg BW showed the best blood glucose level profile

    CARDIOTHORACIC RATIO IN SUNDA PORCUPINE (Hystrix javanica)

    The present study aimed to standardize cardiothoracic ratio (CTR) in clinically healthy Sunda porcupine. This study was an explorative study using four Sunda porcupines, male and female, with an estimated age of 6 months-2years. The sampled animals were radiographed in ventrodorsally projection using a digital X-ray machine with 50 kVp and 6 mAs. Each animal was injected with xylazine as sedation agent at 2 mg/kg BW, intramuscularly route in the dorsal part of animals tail. The radiogram showed that Sunda porcupine heart tends to be round and more massive, characterized as the convex edge of the heart. The cardiothoracic ratio (CTR) showed that the ratio of the broadest expansion of the heart relative to the thoracic width in a ventrodorsally projection ranged from 0.48-0.55 with an average 0.52. Standard heart size was less than half of the largest thoracic diameter (normal CTR0.5). Sunda porcupine heart tends to be round and larger characterized as the convex edge of the heart. The CTR values obtained were 0.48-0.55
