    HEPATOPROTECTIVE ACIVITY OF THE ETHANOLIC EXTRACT OF DOG FRUIT RIND (Pithecellobium lobatum Benth.)

    The present study was conducted to investigate the hepatoprotective activity of ethanolic extract of dog fruit rind (Pithecellobium lobatum benth.) against carbon tetrachloride (CCl4) induced hepatotoxicity in rats. Male Wistar albino rats were randomly divided into four groups and administered orally with 50 mg/200 g body weight of dog fruit rind extract (K1), 100 mg/200 g body weight (K2) of dog fruit rind extract, 5 mg/200 g body weight of silymarin (K3/positive control), and 0.4 mL/200 g body weight of distilled water (K4/negative control), for seven days The levels of alanine transaminase (ALT) of each K1; K2; K3; and K4 were 143.4083.75 U/L, 94.8093.77 U/L, 130.2058.54 U/L and 147.25107.97 U/L, respectively, while the aspartate transaminase (AST) levels were 304.20128.67 U/L; 213.2088.93 U/L; 333.00128.31 U/L; and 239.25 94.90 U/L, respectively (P0.05). Group K2 showed better histological pattern than other groups with 60% of mild and 40% of moderate liver damage.     REPEATED FREEZE-THAW CYCLES BUT NOT SHORT-TERM STORAGE OF FECAL EXTRACTS AT AMBIENT TEMPERATURE INFLUENCE THE STABILITY OF STEROID METABOLITE LEVELS IN CRESTED MACAQUES

    The objective of this study was to examine the effects of repeated freeze-thaw cycles and short-term storage of fecal extracts at ambient temperature on the stability of fecal glucocorticoid (fGCM) and estrogen metabolite (fEM) levels from crested macaques.In total 100 aliquots of fecal extracts from fecal samples collected from female crested macaques (Macaca nigra) living at the Tangkoko-Batuangus Nature Reserve, North Sulawesi were used. We performed two different experiments: (1) An experiment to investigate if levels of fGCM and fEM measured from fecal extracts that were exposed to two, four, six and eight repeated freeze-thaw cycles (test groups) differ to control samples (i.e. fecal extracts always stored frozen); (2) An experiment to evaluate whether storing fecal extracts at ambient temperature for two, four, six, and eight days (test groups)affects the levels of fGCM and fEM compared to the control group (i.e. fecal extracts frozen immediately).Results showed that hormone levels were significantly increased (P0.05) after four freeze-thaw cycles for fGCM and after eight freeze-thaw cycles for fEM. By contrast, there was no significant difference (P0.05) in levels of fGCM and fEM between the test groups and the control group in fecal extracts stored at ambient temperature. In conclusion, our data show that more than two and six repeated freeze-thaw cycles should be avoided when measuring fGCM and fEM in crested macaque fecal extracts, respectively. We also demonstrate that storing fecal extracts at ambient temperature is possible for at least 8 days without taking a risk of affecting the stability of fGCM and fEM levels

    DETECTION OF FIVE VIRUS INFECTIONS IN THE LAYER FARM WITH RUNTING-STUNTING SYNDROME IN SUKABUMI AND TANGERANG USING POLYMERASE CHAIN REACTION TECHNIQUE

    The objective of this study was to identify the five different viral infections including avian influenza, Newcastle disease, avian reovirus, avian encephalomyelitis, and Mareks disease in the runting-stunting syndrome outbreak in several commercial layer farms in Sukabumi and Tangerang in November 2014 using polymerase chain reaction technique. As results, this study identified mix infection of three viruses in the field samples, including Newcastle disease, reovirus, and avian encephalomyelitis; however, it was negative for avian influenza and Mareks disease viruses. Subsequently, the inoculation of several samples into embryonated chicken eggs confirmed the growth of these three viruses. As a consequence, disease control management should be conducted in the affected farms by implementing effective biosecurity and vaccination program

    PECTORALIS THORACICUS MUSCLE PERFORMANCE OF HYBRID CHICKEN (F1) DERIVED FROM CROSSBREED BETWEEN BROILER AND PELUNG (Gallus gallus gallus)

    This research purpose was to study the phenotype characters of body weight and pectoralis thoracicus muscle performance of hybrid chicken (F1) derived from crossbreed between parent stock broiler strain Cobb-500 and Pelung (Gallus gallus gallus). The chicks were divided into 3 groups and each group consists of 5 days-old chicks (DOC). Group 1 (T1) was used as control involving DOC derived from crossbreed between female and male of broiler Cobb-500 strain. Group 2 (T2) involving DOC derived from crossbreed between female and male Pelung, while Group 3 (T3) was DOC derived from crossbreed between female broiler strain Cobb-500 and male Pelung. The chicken was grown up to 49 days to evaluate the body weight, the pectoralis thoracicus weight, the pectoralis thoracicus area, and myofiber area. Data were statistically analyzed using one way Anova. The results showed that the average of body weight in T1; T2; and T3 were 2,17086.02; 506.220.02; and 1,23868.25 g, respectively (P0.05). The average of pectoralis thoracicus muscle weight in T1; T2; and T3 were 165.0411.97; 21.262.44; and 68.741.94 g, respectively (P0.05). The average of pectoralis thoracicus muscle area in T1; T2; and T3 were 97.09,7; 616,4; and 63,41,8 cm2 , respectively (P0,05). The average of pectoralis thoracicus myofiber area in T1; T2; and T3 were 14.721.04; 4.662.1; and 13.131.3 m2 , respectively (P0.05). It was concluded that crossbreed between broiler strain Cobb-500 and Pelung improved the body weight and pectoralis thoracicus muscle performance of hybrid chicken (F1)

    PROFILE OF ALLERGY HYPERPLASMA PATHOLOGIC ANTIBODY AND IMMUNOGENIC CHARACTERISTIC

    The objective of this research was to determine the allergy hyperplasma pathologic antibody and to evaluate the immunogenic characteristics. Blood and serum were collected from human suffering rhinitis, skin, eye and asthma allergies. To obtain a cloned allergen plasma protein (protein allergen that had been induced with the serum from human with allergic rhinitis, skin, eye and asthma), the goat was used as an intermediate animal. Hematological analysis showed that the leukocyte cell such as neutrophil, lymphocyte, monocyte, and eosinophil increase in allergic-suffered human. The blood smear test exhibited that the mastocyte cell was dominant which contributes to allergy activities in human body. The sodium dodecyl sulphate-polyacrylimide gel electrophoresis (SDS-PAGE) assay detected protein allergens with molecular weight of 188 kDa (IgE) and 60-62 kDa (mastocyte cell). The reactivity assay using enzyme linked immunosorbent-assay (ELISA) revealed that cloned-allergens (whole hyperplasma allergen from goat isolates) express the best reactivity at various concentrations of IgE than the leukocyte cells. This research concluded that the clones of protein allergen have better immunogenic characteristic and those proteins can be recommended as the candidate of allergen to induce the humoral immunity on host and deliver specific product of anti-allergy such as milk

    DIAGNOSTIC IMAGING AND ENDOSCOPY OF THE SCHNAUZER DOG WITH UPPER GASTROINTESTINAL TRACT DISORDERS

    The aim of this study was to describe the role of diagnostic imaging and endoscopy to define the diagnose of the upper gastrointestinal tract disorder of the Schnauzer. The information from the owner stated that the dog has been vomiting for a year, sometimes containing blood. There might also be a possibility that it had eaten a corpus alienum. This case study covered physical examination, hematology and blood chemical analysis, diagnostic imaging radiography and ultrasonography as well as endoscopy performed. Physical examination showed weight loss and anorexia. Hematology and blood chemical analysis showed an increase in hemoglobin, hematocrit, lymphocyte, ureum, creatinine, alanine aminotransferase (ALT), and alkaline phosphatase (ALP) values. Abdominal radiograph showed no abnormality in the abdominal organs. Abdominal ultrasonography showed a hiperechoic elongated mass attached on the stomach mucosal surface which forms an acoustic shadowing at the ventral. Endoscopy showed pathological lesions that is inconsistency of stomach mucosa surface, foamy fluid in the stomach, ulcers and erosion of the stomach mucosal surface. Based on the diagnostic imaging and endoscopy performed, the animal was clearly diagnosed with chronic gastritis accompanied by stomach ulcer and erosion

    PENGARUH PROBIOTIK TERHADAP KOLESTEROL DARAH PADA AYAM PETELUR (LAYER) (Effect of Probiotik on Blood Cholesterol in Laying Hens)

    The aim of this research was to find out the effect of probiotic on blood cholesterol in laying hens. Materials used were laying hens (Isa Brown strain) 43 weeks old and ration using probiotics. This research was conducted by using completely randomized design (CRD). The advanced research was done with testing the local probiotic with various concentration treatment, they are P0 (ration without probiotic), P1 (ration + 1% local probiotic), P2 (ration + 2% local probiotic), and P3 (ration + 3% local probiotic) with 5 time repetitions and in every repetitions consists of 2 chicken. Variables that were observed in the advance research were laying henss blood cholesterol that consists of total cholesterol, high density lipoprotein (HDL), low density lipoprotein (LDL), triglycerides, and total leucocyte. The level of total cholesterol on P0, P1, P2, and P3 were 129.0038.26, 116.00 26.53, 121.0033.72, and 137.0052.53 mg/dL (P0.05) respectively; the HDL level were 50.6016.47, 35.8011.28, 59.4019.74; 56.8030.56 mg/dL (P0.05); the LDL level were 100.2049.53, 39.2017.14, 104.6051.23, and 96.8066.22 mg/dL (P0.05), respectively; the trigycerides level were 862.00355.5, 533.00146.1, 829.00242.6, and 810.00304 mg/dL (P0.05), respectively; and leucocyte count were 22.001.72, 21.003.79, 31.003.99, and 34.003.55 x103 mm3 (P0.01), respectively. In conclusion, the administrtion of local probiotic do not effect the blood cholesterol of layer chicken

    PENERAPAN METODE PENCUCIAN DENGAN AIR MENGALIR UNTUK MENURUNKAN KADAR NITRIT PADA SARANG BURUNG WALET (Application of Washing Method under Running Water to Reduce Nitrit Level of Edible Birds Nest)

    This study was aimed to determine the influence of the washing method under running water on nitrite levels of edible birds nest (EBN). Total of 40 samples of EBN were divided into four groups with different washing frequency, control group without washing treatmet (P0), once, twice, and three times washing treatment (P1, P2, and P3) respectively. Each washing was performed for 30 seconds under running water. Nitrite levels assessment was carried out by spectrophotometry at 540 nm of wavelength. The results showed that the average nitrite levels of EBN in P0, P1, P2, and P3 were 93.124.40 ppm, 65.243.38 ppm, 63.603.81 ppm, and 30.872.11 ppm, respectively.     The aims of the research were to evaluate resistance of Staphylococcus aureus against several antibiotics and to detect the gene encoding ofmethicillin resistant Staphylococcus aureus (MRSA) in broiler. Twenty three Staphylococcus aureus isolated from infected broiler in Yogyakartawere used. Genotypic identifications were done based on the occurrence of gene nuc and 23SrRNA. The resistance assay to several antibioticsrevealed that Staphylococcus aureus were resistant to penicillin 78% (18/23), doxycyclin 56% (15/23), gentamicin 26% (6/23), tetracyclin 22% (5/23), erythromycin 13% (3/23), and methicillin 9% (2/23).     KUALITAS SPERMATOZOA AYAM PERANAKAN SENTUL DALAM PENGENCER RINGER LAKTAT KUNING TELUR DENGAN BERBAGAI MONOSAKARIDA (Quality of Sentul Crossbreed Chicken Spermatozoa in Ringer Lactate-Egg Yolk Diluents Supplemented with Various Monosaccharide)

    The aim of this study was to examine the preservation of sentul crossbreed chicken semen in ringer lactate egg yolk diluent supplemented with various monosaccharide. Semen was collected from three roosters using massage method. Immediately after collection, the semen was evaluated macroscopically and microscopically. Semen with more than 70% motility was divided into four tubes. Each of them diluted with ringer lactate egg yolk glucose (RLEYG), ringer lactate egg yolk fructose (RLEYF), ringer lactate egg yolk xylose (RLEYX) and ringer lactate egg yolk mannose (RLEYM). Semen was stored in refrigerator (5o C) for sixty hours and evaluated every twelve hours for spermatozoa motility and viability. Results showed that no significant difference (P0.05) among diluents used on spermatozoa quality parameters after dilution and during preservation. Semen quality decrease during storage and at sixty hours of storage, the motility and viability of spermatozoa ranging from 48.332.56 to 55.422.26% and 58.592.87 to 64.832.42%, respectively. This research conclude that glucose, fructose, xylose and mannose can be used as energy source for roosters semen during preservation
