    Predation Intensity in Mangrove Ecosystem in Marine Protected Area, North Sulawesi

    This study aims to get an overview of the intensity of predation on mangrove ecosystem in five marine protected areas (MPA), namely Tumbak, Basaan, Blongko, Bahoi and Tambun. The research method was carried out by installing Squidpops bait within one hour and calculating the number of lost bait during the exposure of baits in high tide. Fish species that migrate in the mangrove area are obtained through visual census; Mega Bentos is recorded.  The result of this study indicates the intensity of predation in the mangrove ecosystem in the five North Sulawesi DPLs are varied in each location, which has the possibility of being influenced by local condition, predatory fish population, the level of disturbance at observation, method and level of preference for the bait provided.Keywords: Predation, Predator, Mangrove, Fish Community, Squidpops ABSTRAKPenelitian ini bertujuan untuk mendapatkan gambaran intensitas predasi pada ekosistem mangrove di lima daerah perlindungan laut (DPL), yaitu Tumbak, Basaan, Blongko, Bahoi dan Tambun. Metode penelitian dilakukan dengan pemasangan umpan Squidpops dalam waktu 1 jam dan menghitung jumlah umpan yang hilang selama umpan terpapar pada saat air pasang. Jenis ikan yang bermigrasi di daerah mangrove diperoleh melalui sensus visual; mega bentos dicatat. Hasil dari penelitian ini menunjukkan intensitas predasi di ekosistem mangrove pada 5 DPL Sulawesi Utara bervariasi pada tiap lokasi yang memiliki kemungkinan dipengaruhi oleh kondisi lokal, populasi ikan predator, tingkat gangguan saat pengamatan, metode dan tingkat kesukaan pada umpan yang disediakan.Kata Kunci: Predasi, Predator-Mangsa, Mangrove, Komunitas Ikan, Squidpop

    The Survival Rate and Growth of Juvenile Crayfish (Cherax quadricarinatus) With Different Types of Shelter

    An experiment was designed to assess the relative performance of three shelter types on the survival and growth of red-claw crayfish (Cherax quadricarinatus).  A 13 days old juvenile crayfish with an average length of 10 mm were cultured in 400 ml water of glass bowl and were provided with one of 3 types of shelter over 19 days.  The various shelter types assessed were dried coconut leaves, bamboo and Hydrilla.  Juveniles in each bowl were fed with Artemia twice a day morning and in the afternoon. There was no significant effect (P>0.05) of shelter type on the survival rate; however, there was a significant effect (P<0.05) on growth with dried coconut leaves (5.33%) performing significantly better than Hydrilla (2.33%) and bamboo (2.00%).  This experiment showed that the shelter types provided had a significant effect on the growth but not in their survival.Keywords: Survival rate, Growth, juvenile Cherax quadricarinatus, ShelterABSTRAKTujuan penelitian ini adalah untuk menganalisis sintasan hidup dan  pertumbuhan  juvenil lobster air tawar (C. quadricarinatus) dengan shelter yang berbeda. Juvenil lobster yang berumur 13 hari dengan panjang rata rata 10 mm dipelihara dalam wadah toples kaca dengan diberikan ketiga perlakuan selama 19 hari.  Ketiga jenis perlakuan adalah daun kelapa kering, bambu dan Hydrilla.  Juvenil diberi pakan Artemia 2 kali sehari yaitu pada pagi dan sore hari.  Hasil penelitian menunjukkan ketiga perlakuan tidak berbeda nyata terhadap sintasan hidup (P>0.05), namun berbeda nyata (P< 0.05 ) untuk pertumbuhan dengan daun kelapa kering (5.33%), Hydrilla (2.33%) dan bambu (2%).     Penelitian ini menunjukkan bahwa jenis shelter memberikan pengaruh nyata  terhadap pertumbuhan namun tidak berbeda nyata untuk sintasan hidup.Kata kunci: Sintasan hidup, Pertumbuhan, Juvenil Cherax quadricarinatus, Shelte

    The biodiversity of macroalgae in the coastal waters of Bahoi Village, West Likupang Sub-District, North Minahasa Regency

    This study was carried out in coastal waters of Bahoi Village, West Likupang Sub-District, North Minahasa Regency with an objective of knowing the taxa composition of macroalgae through morphological studies. Data collection used exploring survey. Results found 8 species of macroalgae, consisting of 3 divisions, 3 classes, 5 orders, 6 families, and 6 genera.Keyword: Macroalgae, Bahoi Village.  ABSTRAKPenelitian ini dilakukan di perairan pesisir Desa Bahoi, Kecamatan Likupang Barat, Kabupaten Minahasa Utara dengan tujuan untuk mengetahui komposisi taksa makroalga melalui pendekatan morfologi. Pengambilan data dilakukan dengan menggunakan metode Survei Jelajah.     In Vitro Culture of Seaweed Kappaphycus alvarezii under Different Formulation of Growth Stimulating Substances and Culture Media

    This study aims at obtaining a sustainably superior seed stock following the characteristics of the parent plant, determining the best formulation of the growth stimulating substance. In general, cytokinin and auxin combination was used, but this study also added with the combination of cytokinin and giberelin and cytokinin and abscisic acid (AA).Parameters measured were bud length, number of buds, and survival rate. Bacterial Vibrio sp test was also done as a cause of the explant mortality. Results showed that the longest bud was recorded in treatment C (S+A 1:2.5) cultured in a jar, 1.343 mm long, 38% of survival, while the highest number of buds was found in treatment B (S+A 1 : 2) 8.86. The shortest bud was recorded in treatment J (S + AA 1:2.5) cultured in a jar, 0.093 mm long, 2.64 buds, 10% of survival, while the explant cultured in the bottle had a length of 0.051 mm long, 1.50 buds, 4% of survival. As conclusion, the best growth stimulating substance was found in the treatment C for the bud length and the survival rate, while the best number of bud was recorded in the treatment B. The best culture tank was topless bottle (aerated). In vitro culture could also use S + G formulation. The explant mortality was caused by Vibrio charchariae. The use of S + AA formulation had lower growth than that of control treatment.Keywords :in vitro, growth stimulating substance, culture media, Kappaphycus alvarezii, Vibrio charchariae ABSTRAKPenelitian ini bertujuan untuk memperoleh benih unggul secara berkelanjutan yang mengikuti karakteristik dari tanaman induk, menentukan formulasi terbaik dari substansi pertumbuhan merangsang. Secara umum, kombinasi sitokinin dan auksin digunakan, tetapi penelitian ini juga menambahkankombinasi sitokinin, giberelin, sitokinin dan asam absisat (AA). Parameter yang diukur adalah panjang tunas, jumlah tunas, dan tingkat kelangsungan hidup. Bakteri Vibrio Uji sp juga dilakukan sebagai penyebab kematian eksplan . Hasil penelitian menunjukkan bahwa tunas terpanjang terdapat pada perlakuan C (S + A 1: 2,5)  kultur dalam toples, 1,343 mm, 38% hidup, sementara jumlah tertinggi tunas ditemukan pada perlakuan B (S + A 1: 2) 8.86 . Jumlah tunas paling sedikit terdapat pada perlakuan J (S + AA 1: 2,5) yang dikultur dalam toples, 0,093 mm, 2,64 tunas, 10% hidup, sedangkan eksplan yang dikultur dalam botol memiliki panjang 0.051 mm, 1. 50 tunas , 4% bertahan hidup. Sebagai kesimpulan, pertumbuhan terbaik merangsang zat ditemukan dalam perlakuan C untuk panjang tunas dan tingkat kelangsungan hidup, sementara jumlah tunas terbanyak ditemukan pada perlakuan B. Penggunaan wadah budaya terbaik adalah topless yang diaerasi. Kultur in vitro juga dapat menggunakan formulasi S + G. Kematian eksplan disebabkan oleh Vibrio charchariae . Penggunaan formulasi S + AA memiliki pertumbuhan yang lebih rendah dari pada pengobatan kontrol .Kata kunci : in vitro, zat perangsang tumbuh, media kultur, Kappaphycus alvarezii, Vibrio charcharia

    The Distribution of Genus Favia (Oken, 1815) at the Reef Flats of Likupang Kampung Ambong Village Minahasa Utara

    Six species of Favia were found in this study i.e, Favia pallida (Dana, 1846), Favia speciosa (Dana, 1846), Favia favus (Forskål, 1775), Favia truncatus (Veron, 2000), Favia rotundata (Veron and Pichon, 1977) and Favia matthaii (Vaughan, 1918). The species with the highest percentage of coral cover was Favia pallida (0,386 %). The values of Dominance Index showed that there were no dominant Favia species on Kampung Ambong coral reef flats. Diversity Index (H’) of Favia was 1,47. There are two distribution patterns shown by Favia on Kampung Ambong coral reef flats area, which are clumped and uniformed.Keywords: Favia, Distribution  ABSTRAKDalam penelitian ini ditemukan 6 spesies karang genus Favia, yaitu Favia pallida (Dana, 1846), Favia speciosa (Dana, 1846), Favia favus (Forskål, 1775), Favia truncatus (Veron, 2000), Favia rotundata (Veron and Pichon, 1977) dan Favia matthaii (Vaughan, 1918). Spesies dengan persentase tutupan karang tertinggi adalah Favia pallida (0,37 %). Nilai Indeks Dominasi menunjukkan tidak ada spesies karang genus Favia yang dominan pada rataan terumbu Kampung Ambong. Indeks keanekaragaman karang genus Favia menunjukkan nilai H’ = 1,47. Terdapat dua Pola distribusi yang diperoleh dari karang genus Favia di rataan terumbu Kampung Ambong, yaitu mengelompok dan seragam.Kata Kunci : Favia, Distribus

    Bacillus sp. As a Decomposition Agent in The Maintenance of Brachionus rotundiformis Which Uses Raw Fish As a Source of Nutrition

    The research was conducted at the Laboratory of Marine Molecular Biology and Pharmacy, Faculty of Fisheries and Marine Sciences, Sam Ratulangi University. This research aims to isolate and analyze the morphology and molecular types of bacteria associated in rotifer’s culture media that use fisheries waste.The research was begin by culturing bacteria in rotifer maintenance media using Nutrient Broth media. After bacterial isolates were obtained, morphological characterization and DNA extraction was carried out. extraction was done using DNeasy Blood and Tissue Kit (Qiagen). After DNA was obtained, DNA was amplified through the Polymerase Reaction Chain (PCR) machine using a 16S RNA primer, followed by the separation of PCR products through electrophoresis and detection through UV Transluminator. The target PCR product was determined by comparing the 100 bp ladder DNA, with a yield of around 1400 bp, which was measured using ladder DNA available in the laboratory. The DNA that was successfully amplified was sent to be sequenced to determine the species of each microbe obtained.Based on the results of the research conducted, obtained Bacillus sp. bacteria associated with rotifer maintenance media.Keywords: Bacteria, Culture Media, DNA Extraction, PCR, Sequencing ABSTRAKPenelitian ini dilakukan di Laboratorium Biologi Molekuler dan Farmasetika Laut, Fakultas Perikanan dan Ilmu Kelautan, Universitas Sam Ratulangi.  Penelitian ini bertujuan untuk mengisolasi dan menganalisis morfologi dan molekuler jenis-jenis bakteri yang berasosiasi dalam media pemeliharaan rotifer yang menggunakan limbah perikanan.Penelitian dilakukan dengan cara mengkultur bakteri yang ada pada media pemeliharaan rotifer menggunakan media Nutrient Broth. Setelah isolat bakteri didapatkan, dilakukan karakterisasi morfologi dan dilakukan ekstraksi DNA.  ekstraksi dilakukan menggunakan DNeasy Blood and Tissue Kit (Qiagen).  Setelah DNA didapatkan, DNA diamplifikasi melalui mesin Polymerase Reaction Chain (PCR) menggunakan primer 16S RNA, diikuti dengan pemisahan produk PCR melalui electrophorisis dan deteksi melalui UV Transluminator. Produk PCR target ditentukan dengan membandingkan ladder DNA 100 bp, dengan hasil sekitar 1400 bp, yang diukur menggunakan ladder DNA yang tersedia di laboratorium.  DNA yang berhasil diamplifikasi, dikirim untuk dilakukan sekuensing untuk mengetahui spesies dari setiap mikroba yang didapatkan.  Berdasarkan hasil penelitian yang dilakukan, diperoleh bakteri Bacillus sp. yang berasosiasi pada media pemeliharaan rotifer. Kata Kunci: Bakteri,  Media Pemeliharaan, Ekstraksi DNA, PCR, Sekuensin

    The use of Sponge Crude Extract to Increase Growth and Immune Response of Nile Tilapia Oreochromis niloticus

    This study aims to examine the effect of sponge crude extract addition to the fish feed on growth and non-specific immune response of nile tilapia (O. niloticus) and determine the most effective dose of the extract in raising the growth and the non-specific immune response of nile tilapia (O. niloticus). The sponge sample was macerated and then evaporated using a rotary vacuum evaporator until the sample became a paste and mixed with feed. The fish were fed for 14 days respectively as much as 5% of body weight per day with feeding frequency of two times a day. The results showed that addition of sponge crude extract to the fish feed gave good effect on the growth and the non-specific immune response of nile tilapia (O. niloticus) with best dose of  40 g / kg feed.Keywords: Sponge extract, fish immune response, fish growth, immune response, immunostimulant. ABSTRAKPenelitian ini bertujuan untuk menguji pengaruh penambahan ekstrak kasar spons pada pakan ikan terhadap pertumbuhan dan respon imun  non spesifik ikan nila (Oreochromis niloticus) serta menentukan dosis ekstrak kasar spons yang paling efektif dalam meningkatkan pertumbuhan dan respon imun non spesifik ikan nila (Oreochromis niloticus). Sampel spons dimaserasi kemudian dievaporasi menggunakan rotary vacuum evaporator sehingga diperoleh ekstrak kasar berbentuk pasta yang kemudian dicampurkan pada pakan. Ikan uji diberi pakan perlakuan 5% dari bobot biomassa selama 14 hari dengan frekuensi pemberian pakan dua kali sehari. Hasil penelitian menunjukan bahwa penambahan ekstrak kasar spons pada pakan ikan memberikan pengaruh yang baik terhadap pertumbuhan dan respon imun non spesifik ikan nila (Oreochromis niloticus), yang mana dosis yang terbaik adalah 40 gr / Kg pakan.Kata Kunci : Ekstrak spons, respon imun ikan, pertumbuhan ikan, respon imun, imunostimulan

    A Study On Biological Parameters Of Aquaculture Area In Bahoi Village Northern Minahasa Regency, North Sulawesi

    The aims were to know the biological parameters, identify and analysis of plankton density and diversity in aquaculture area of Bahoi Village. The plankton were sampled from July to September, 2018. The study was carried out monthly in 2 stations by using plankton net. The results showed that the most types of plankton in the waters of Bahoi village were the phytoplankton Bacillariophyceae class with (63 genera) while the type of zooplankton dominated by the genus Acartia sp. Crustaceae class. The highest density of plankton is found at Station 2 (outside the cultivation area) at noon time with a density of 3.18 ind/L, the lowest density is at Station 2 with the morning retrieval time of 1.64 ind/L. Index value of diversity (H '), which is 2.5-2.9, shows the waters of Bahoi village including the medium fertility category and diversity of the moderate plankton. The water quality conditions at the time of overall study can be said to be good and plankton can grow and develop.Keywords : Biological parameters, Plankton, Water quality, Bahoi VillageABSTRAKTujuan penelitian ini adalah untuk mengetahui parameter biologi dan mengidentifikasi jenis-jenis plankton serta menganalisis kepadatan dan keanekaragaman plankton di lokasi budidaya Desa Bahoi.  Sampel plankton diambil sejak bulan Juli sampai September 2018. Pengambilan sampel plankton dengan planktonnet selama tiga bulan pada dua stasiun yang telah ditentukan. Hasil penelitian menunjukkan bahwa jenis plankton terbanyak yang ada di perairan Desa Bahoi adalah fitoplankton kelas Bacillariophyceae dengan (63 genera) sedangkan untuk jenis zooplankton didominasi oleh genus acartia sp kelas Crustaceae. Kepadatan tertinggi plankton terdapat pada stasiun 2 (di luar area budidaya) 3,18 ind/l, kepadatan yang terrendah terdapat pada stasiun 2 dengan waktu pengambilan pagi hari sebesar 1,64 ind/l. Nilai indeks keanekaragaman (H’) yaitu 2.5-2.9, menunjukkan bahwa perairan Desa Bahoi termasuk dalam kategori kesuburan sedang dan keanekaragaman jenis plankton yang sedang. Kondisi kualitas air pada saat penelitian secara keseluruhan dapat dikatakan baik dan plankton dapat bertumbuh dan berkembang.Kata kunci : Parameter Biologi, Plankton, Kualitas Air, Desa Baho

    The use of fish silage in different composition for the growth of carp Cyprinus carpio L.

    This study aims to examine the fish silage for total gain weight, growth rate, and the specific growth rate of common carp (Cyprinus carpio L.), which is cultivated in 12 nets (1x0,5x1m), with the density of 10 fish (3-5 cm) on each net. The experimental design used was a completely randomized design (CRD) with 4 treatments and 3 replications; treatment A with 10% fish silage, treatment B with fish silage 20%, treatment C with 30% fish silage and treatment D were commercial pellet. The ANOVA test showed that the treatment effect among feed A, B, C, and D tested did not give a significant difference to total gain weight, growth rate and the specific growth of carp fry.Keyword: common carp, fish silage, fish feed, growth ABSTRAKPenelitian ini bertujuan untuk menelaah pemberian silase ikan bagi pertumbuhan mutlak, pertumbuhan nisbi, dan pertumbuhan harian ikan mas (Cyprinus carpio L.), yang dipelihara dalam petak-petak jaring berukuran 1x 0,5 x 1 m, dengan padat penebaran 10 ekor ikan berukuran  3-5 cm. Rancangan percobaan yang digunakan adalah rancangan acak lengkap (RAL) dengan 4 perlakuan dan 3 ulangan; perlakuan A pelet berkomposisi silase ikan 10%, perlakuan B pelet berkomposisi silase ikan 20 %, perlakuan C pelet berkomposisi silase ikan 30 % dan perlakuan D adalah pelet komersil. Uji ANOVA menunjukkan bahwa pengaruh perlakuan di antara pakan A, B, C, dan D yang diujicobakan tidak memberikan perbedaan yang nyata terhadap pertumbuhan mutlak, pertumbuhan nisbi dan pertumbuhan harian benih ikan mas.Kata kunci : ikan mas, silase ikan, pakan ikan, pertumbuha

    Antibacterial Screening Activity of Several Sponges Against Staphylococcus aureus, Escherichia coli, Staphylococcus saprophyticus, dan Pseudomonas aeruginosa

    Sponge samples collected around Manado waters were obtained 30 species and their crude extracted have been tested in vitro for their activity in inhibiting bacterial growth. Based on the results of antibacterial screening in 30 sponge extracts, there were 23 sponge extracts which had bioactivity in inhibiting the growth of S. aureus, E. coli, S. saprophyticus and P. aeruginosa, Sponge extract No. 43 (of 30 sponge extracts tested) was the most active in inhibiting bacterial growth and had the widest inhibition zone diameter.Keywords: screening, sponge, crude extract, antibacterial, ManadoABSTRAKSampel spons dikoleksi di sekitar Perairan Manado sebanyak 30 jenis/spesies, dimana ekstrak kasarnya telah diuji secara in vitro aktivitasnya dalam menghambat pertumbuhan bakteri.
