    Isolation and Antibacterial Activity assay of Endophytic Symbiont Bacteria on Seaweed Gracilaria verrucosa originated from Batu Meja Tongkaina Beach, North Sulawesi

    Seaweed can produce bioactive compounds such as antibacterial. Seaweed co-exists with bacteria endophytes. Entophytic bacteria are bacteria that live in host tissues and have the ability to protect the host itself by producing antibacterial compounds against pathogens. Therefore, the endophytic bacteria of seaweed symbionts can be utilized to produce antibacterial compounds. Bacteria can be mass-cultured because of their fast-growing characteristic. The aim of this study was to determine the antibacterial activity from endophytic symbiont bacteria on seaweed Gracilaria verrucosa originated from Batu Meja Tongkaina Beach, North Sulawesi. Isolation of bacteria was carried out by dilution steps from 101 to 10־3 times, then cultivation on Nutrient Agar media to obtain a single colony of the bacteria. Antibacterial activity was tested used paper disc diffusion method. The pathogens used were bacterial strains of Escherichia coli, Staphylococcus aureus, Streptococcus mutans, and Salmonella typhi. Moreover, antibiotics were used as positive controls. The antibacterial ability of the symbiotic bacteria was measured based on inhibition zones around the paper disc containing the isolates of endophytic bacteria. A total of 6 endophytic bacterial isolates were found, namely isolate B, C, D, E, F and G, characterized by different morphological features. The results showed that isolates B and C having inhibition zone of 0.5 – 1.0 mm against S. typhi, indicating that these two isolates produce antibacterial compounds with a weak ability  against S. typhi

    Identification And Diversity Of Crab In Pondang And Lopana Beach Waters, South Minahasa

    There are crab species that live in the forest and supratidal areas, on sandy, rocky, and muddy coastal areas. The aim of this research is to identify the morphology, of meristic crabs and the effect of the moon phase on species diversity. Sampling locations were on the coast of Pondang Village and Lopana Village, sampling using the roaming method and carried out at the two lowest low tide phases during the dead moon and full moon phases, during the lowest low tide crabs were generally found with walking legs. Based on the identification of the morphology of crabs found on the coast of Pondang Village and Lopana Village, East Amurang District, South Minahasa Regency, there are six species of them Ocypode ceratophthalmus (Stimpson, 1858), Grapsus albolineatus (Latreille in Milbert, 1892), Eriphia sebana (Shaw and Nodder, 1803), Atergatis floridus (Linnaeus, 1767), Pilumnus vespertilio (Fabricius, 1793), Episesarma mederi (Edwards, 1853). Keywords: Coastal; Habitat; Crabs; Morphology; meristic AbstrakTerdapat jenis kepiting yang hidup di daerah hutan dan supratidal, di daerah pesisir pantai berpasir, berbatu dan berlumpur. Tujuan penelitian mengidentifikasi secara morfologi, meristik kepiting dan pengaruh fase bulan terhadap keanekaragaman spesies. Lokasi pengambilan sampel di pesisir pantai Kelurahan Pondang dan Desa Lopana, pengambilan sampel menggunakan metode jelajah serta dilakukan pada dua fase surut terendah saat fase bulan mati dan purnama, saat surut terendah umumnya dijumpai kepiting yang memiliki kaki jalan.     Utilization Of Geographic Information System (Gis) For Selection Of Idle Pond For Vannamei Shrimp Cultivation

    Land suitability is one aspect that determines the success of vannamei shrimp cultivation in coastal pond areas. Cultivation of vannamei shrimp in ponds has a spatial component as well as differences in the biophysical and socio-economic characteristics of each location. Many ponds that are intensively managed have not taken advantage of the advantages of Geographical Information Systems (GIS) in selecting locations and managing vannamei shrimp culture, which is important to do to avoid business failure. This study used a survey method to collect data on water quality parameters (acidity / pH, temperature, salinity, dissolved oxygen / DO, nitrate, and phosphate). Analysis of land suitability for vannamei shrimp cultivation in ponds using the scoring method, each water quality parameter is weighted and a score is then divided into 4 land suitability classes, namely classes S1 (Very Suitable), S2 (Sufficiently Suitable), S3 (Conditional Appropriate), and N (Unsuitable). The results showed that the vannamei shrimp pond land suitability map produced a thematic map of the suitability of the vannamei shrimp pond land which had a total area of 13.65 ha of vannamei shrimp and had 4 levels of suitability, namely very suitable class (S1) with an area of 3.96 ha, a class that appropriate (S2) has an area of 7.8 ha, a moderately suitable class (S3) has an area of 0.85 ha, and an unsuitable class (N) has an area of 1.04 ha.Keywords: Litopenaeus vannamei; Water quality; Land Suitability; Geographical Information SystemsAbstrakKesesuaian lahan merupakan salah satu aspek yang menentukan keberhasilan kegiatan budidaya udang vannamei di wilayah tambak pesisir. Budidaya udang vannamei di tambak memiliki komponen keruangan serta perbedaan karakteristik biofisik dan social ekonomi dari setiap lokasi. Banyak tambak yang dikelola secara intensif belum memanfaatkan kelebihan Sistem Informasi Geografis (SIG) dalam melakukan pemilihan lokasi dan pengelolaan budidaya udang vannamei, dimana hal tersebut penting dilakukan untuk menghindari kegagalan usaha. Penelitian ini menggunakan metode survei untuk pengambilan data parameter kualitas air (keasaman/pH, suhu, salinitas, oksigen terlarut/DO, nitrat, dan fosfat). Analisis kesesuaian lahan budidaya udang vannamei di tambak menggunakan metode skoring, parameter kualitas air masing-masing diberi bobot dan skor yang kemudian dibedakan menjadi 4 kelas kesesuaian lahan yaitu kelas S1 (Sangat Sesuai), S2 (Cukup Sesuai), S3 (Sesuai Bersyarat), dan N (Tidak Sesuai). Hasil penelitian didapatkan peta kesesuaian lahan tambak udang vannamei menghasilkan peta tematik kesesuaian lahan tambak udang vannamei memiliki total luas tambak udang vannamei seluas 13,65 Ha serta memiliki 4 tingkat kesesuaian yaitu kelas sangat sesuai (S1) dengan luasan sebesar 3,96 Ha, kelas yang sesuai (S2) memiliki luasan sebesar 7,8 Ha, kelas yang cukup sesuai (S3) memiliki luasan sebesar 0,85 Ha, dan kelas yang tidak sesuai (N) memiliki luasan sebesar 1,04 Ha.Kata Kunci: Litopenaeus vannamei, Kualitas Perairan; Analisa Kesesuaian; Sistem Informasi Geografi

    Dissolved Oxygen in the East Bolaang Mongondow Waters, North Sulawesi

    Oxygen in seawater comes from the air through diffusion and photosynthesis. This study aims to determine the dissolved oxygen content, NAEC (Normal Atmospheric Equilibrium Concentration), and AOU (Apparent Oxygen Utilization) in East Bolaang Mongondow waters. The analysis showed that the dissolved oxygen content in the surface layer ranged from 6.01-7.05 mg/l; 5.91-6.98 mg/l (5 m depth); and 5.75-6.90 mg/l (near bottom depth). NAEC is closely related to water temperature and implicitly increases with depth. The highest NAEC value of 5.90 mg/l (t=28.0ºC) was found at the bottom depth, and the lowest NAEC of 5.63 mg/l (t=30.0ºC) was found at the surface layer. The dissolved oxygen saturation level of more than 100% and the average positive AOU value at 0.59-0.81 mg/l describe the amount of oxygen available in the surface layer.Keywords: Dissolved Oxygen; Normal Atmospheric Equilibrium Concentration (NAEC); Apparent Oxygen Utilization (AOU); East Bolaang Mongondow.AbstrakOksigen dalam air laut bersumber dari udara melalui proses difusi dan hasil fotosintesis. Penelitian ini bertujuan untuk mengetahui kandungan oksigen terlarut, NAEC (normal atmospheric equilibrium concentration) dan AOU (apparent oxygen utilization) di perairan Bolaang Mongondow Timur. Hasil analisis menunjukkan bahwa kandungan oksigen terlarut di lapisan permukaan berkisar antara 6,01-7,05 mg/l; kedalaman 5 meter 5,91-6,98 mg/l dan dekat dasar 5,75-6,90 mg/l. NAEC sangat berhubungan dengan suhu air dan secara implisit konsentrasinya akan semakin tinggi dengan bertambahnya kedalaman. Nilai NAEC tertinggi 5,90 mg/l (t=28,0ºC) ditemukan pada kedalaman dekat dasar dan  NAEC terendah 5,63 mg/l (t=30,0ºC) ditemukan pada lapisan permukaan. Tingkat kejenuhan oksigen terlarut >100 % dan rata-rata nilai AOU positif (0.59-0.81 mg/l) menggambarkan banyaknya kandungan oksigen yang tersedia pada lapisan permukaan.Kata kunci: oksigen terlarut; normal atmospheric equilibrium concentration (NAEC); apparent oxygen utilization (AOU); Bolaang Mongondow Timur

    Benefit Value of Mangrove Forest in Sarawet Village, Likupang Timur, Minahasa Utara

    The purpose of this study is to: 1) identify various types of direct and indirect benefits of mangrove forests, and 2) estimate the total economic value of the mangrove forest in Sarawet Village. This type of research is descriptive and quantitative using survey methods. Data analysis in this study uses descriptive and quantitative analysis. Descriptive analysis is used to explain the activities of utilizing natural resources of mangrove ecosystems, while quantitative analysis is used to calculate the total economic value of mangrove ecosystems. The selection of respondents used the purposive sampling method, with a total of 76 respondents. Analysis of the total economic value for the value of direct benefits using market prices, and the value of indirect benefits using replacement costs. The results showed that the people of Sarawet Village use the mangrove forest ecosystem directly in the form of mangrove wood used as firewood and fence stakes, leaves Nipah are made with roofs, and the community carries out fishing and crab activities around mangrove waters.Analysis of the total economic value of the mangrove forest in Sarawet Village amounted to Rp. 6.884.344.178,-/year. The total economic value is obtained from the value of direct benefits including utilization as firewood Rp. 4,160,000,- /year; fence pegs Rp. 87.500,-/year; roof of the house Rp. 330.000,-/year; fishing Rp. 769.174.000,-/year; crab fishing is Rp. 190.320.000,-/year, and the value of indirect benefits is as a barrier to abrasion and seawater waves Rp. 5.920.272.678,-/year.     Morphology of Crabs in Minanga Beach, Malalayang Satu, Manado City

    The coastal area in North Sulawesi which has a length of about 1.985 km, is a potential area with biodiversity supported by the associated biota in it. Order Decapoda, infraorder Brachyura is one of the many biotas found in coastal areas. It is estimated that about 5,400 species of Brachyura live in coastal areas. The purpose of the study was to identify the crabs caught at Minanga Beach, Manado Bay Waters, Malalayang Satu Village Malalayang District Manado City based on the morphological characteristics of crabs. The sample was collected in February 2022. The method used in this research is the cruising survey method, which has been done by tracing the marked location and then taking all the crabs species found. Identification is done by observing the morphology of the crab, such as carapace shape, claws, walking legs, presence of spines on the carapace, carapace size, abdomen shape, sexual dimorphism that occurs in crabs, presence of hair/setae, and color/pattern of crabs. Based on the research conducted, as many as five species of crabs were found in Minanga Beach, namely: Thalamita prymna, Thalamita sp., Thalamita admete, Ozius rugulosus, Ozius tuberculosus.Keywords: Brachyura; Identification; Thalamita; Ozius; Manado Bay.AbstrakWilayah pesisir di Sulawesi Utara yang memiliki panjang sekitar 1.985 km, merupakan wilayah yang potensial dengan keanekaragaman hayati yang didukung oleh biota yang berasosiasi di dalamnya. Ordo Decapoda, infraordo Brachyura merupakan satu dari banyak biota yang ditemukan di wilayah pesisir. Diperkirakan sekitar 5.400 spesies brachyura hidup di wilayah pesisir pantai. Tujuan penelitian adalah untuk mengidentifikasi kepiting yang ditangkap di Pantai Minanga, Perairan Teluk Manado, Kelurahan Malalayang Satu, Kecamatan Malalayang, Kota Manado berdasarkan ciri morfologi kepiting. Pengambilan sampel dilakukan pada bulan Februari 2022. Metode yang digunakan dalam penelitian adalah metode survei jelajah, yaitu dengan menyusuri lokasi yang telah ditandai lalu mengambil semua spesies kepiting yang ditemukan. Identifikasi dilakukan dengan memperhatikan morfologi kepiting, seperti: bentuk karapas, capit, kaki jalan, keberadaan duri pada karapas, ukuran karapas, bentuk abdomen, seksual dimorfisme yang terjadi pada kepiting, keberadaan rambut/setae, dan warna/corak kepiting. Berdasarkan penelitian yang dilakukan, sebanyak lima spesies kepiting ditemukan di Pantai Minanga, yaitu: Thalamita prymna, Thalamita sp., Thalamita admete, Ozius rugulosus, Ozius tuberculosus.Kata Kunci: Brachyura; Identifikasi; Thalamita; Ozius; Teluk Manado

    Crabs are organisms that have been found moving actively in tidal areas. One is in the coastal area of Mokupa, Whare the study boasts a great diversity of poorly informed crabs. this study aims to identify the morphology and diversity of crab species during the full moon and late nephew phase. The location for sampling is on the coast of Mokupa Beach, Minahasa Regency, North Sulawesi.  This research was conducted using the roaming method and captured directly at night at the lowest low tide based on the phases of the full moon and late nephew. Furthermore, the collected crabs were identified by morphology, body length, body color, carapace shape, pair of claws, walking legs, and abdomen, and morphometric calculations were performed. Based on the morphological identification found, namely: Thalamita crenata, Geograpsus crinipes, Selatium brocki, and Sesarmops impressus.Keywords: Coastal beaches, Morphology, Crab type. AbstrakKepiting merupakan organisme yang ditemukan bergerak aktif di daerah pasang surut. Salah satunya di daerah pesisir Pantai Mokupa, lokasi ini memiliki banyak keanekaragaan jenis kepiting yang belum terinfomasi dengan baik. Penelitian ini bertujuan mengidentifikasi morfologi dan keanekaragaman jenis kepiting saat fase bulan purnama dan perbani akhir. Lokasi penangkapan sampel di pesisir Pantai Mokupa, Kabupaten Minahasa.Sulawesi Utara, dilakukan mengunakan metode jelajah dan ditangkap langsung pada ­waktu malam hari saat surut terendah berdasarkan fase bulan purnama dan perbani akhir. Selanjutnya, kepiting yang yang terkumpul diidentifikasi morfologi diamati ukuran panjang tubuh, warna tubuh, bentuk karapas, sepasang capit, kaki jalan dan abdomen, serta dilakukan perhitungan morfometrik.     Early Warning of Heavy Metal Pollution in the Waters of Obi Island Based on Plankton Elements

    Industrial activities that are around the waters will donate dangerous heavy metals and enter the waters through rivers. Heavy metal pollution is very dangerous for animals, plants, and humans. Exposure to heavy metals in humans occurs through the food chain. Pollution observation methods using living organisms such as plankton as bioindicators are more effective. As an indicator of biological, composition, and abundance of plankton can provide a guide for monitoring the occurrence of pollution by using the saprobic index. This research aims to determine the level of pollution in the waters of Obi Island. The results of identification of plankton samples found 17 genera phytoplankton in 2 different classes, Diatom and Dinoflagellate, and 3 genera of zooplankton class. The average abundance of phytoplankton from the results of the study was 5.253 individual/L which means that the water conditions were mesotrophic. Meanwhile, the average abundance of zooplankton is 57 individuals/m3. The saprobic index and trophic saprobic index analysis tend to show that the waters are in the mild to moderate polluted category, where the waters are at the β-Mesosaprobic level.Keywords: Phytoplankton; Zooplankton; SI index; Obi island AbstrakKegiatan industri pertambangan di Kawasi merupakan pertambangan untuk memperoleh biji nikel (Ni). Biji Ni umumnya berasosiasi dengan log am berat lainnya seperti tembaga (Cu), arsenik (As), besi (Fe), dan platina (Pt) sehingga berpeluang besar masuk ke perairan di sekitar kawasan pertambangan melaluli aliran sungai. Interaksi yang terjadi dalam ekosistem perairan sungai dan beban masukan yang tidak terkendali dapat mempengaruhi ketersediaan unsur hara dan fitoplankton. Fitoplankton merupakan parameter biologi yang dapat di jadikan indikator untuk mengevaluasi kualitas dan tingkat kesuburan perairan. Sebagai indikator biologi komposisi dan kelimpahan fitoplankton dapat memberikan petunjuk untuk memantau terjadinya pencemaran dengan menggunakan indeks saprobitas. Hasil identifikasi sampel plankton yang dijumpai berjumlah 17 genus fitoplankton dalam 2 kelas berbeda, yaitu diatom dan Dinoflagellate, serta 3 genus zooplankton kelas copepoda. Dari hasil penelitian ini, kelas fitoplankton yang paling banyak tercacah adalah diatom atau Bacillariophyceae. Rata – rata kelimpahan fitoplankton (Diatom dan Dinoflagellata) dari hasil penelitian sebesar 15.503 ind/L. Hal ini, berarti bahwa kelimpahan perairan pada lokasi penelitian dalam kondisi tinggi (eutotrof). Jenis zooplankton yang tercacah merupakan zooplankton kelas copepoda yang terdiri atas Oithona sp.; Corycaeus sp.; dan Paracalunus sp. Analisis SI dan TSI cenderung menunjukan perairan berada pada kategori tercemar ringan sampai sedang, dimana perairan berada pada tingkat β- Mesosaprobik.Kata Kunci: Biji Nikel; Fitoplankton; Zooplankton; Indeks SI dan TSI

    Analysis of Mangrove Vegetation and Distribution Using Landsat 8 Images In Bolaang Mongondow East, North Sulawesi

    Mangrove is one of the objects that can be identified by remote sensing technology using satellite imagery. Analysis of the distribution and density of mangrove vegetation using Landsat 8 imagery was carried out in Bolaang Mongondow Timur, North Sulawesi in September 2020. This study aims to map the distribution of mangroves and determine the correlation between NDVI values, canopy cover, and mangrove density. The data analysis used Landsat 8 images with ENVI 5.3 and ArcGIS 10.1 software. Maximum likelihood classification is used to separate mangrove and non-mangrove features. The calculation of mangrove vegetation density using the NDVI algorithm and single-channel classification using the density slice method to divide mangrove density based on the range of pixel values of the NDVI image. Next, to test the accuracy of the classification results using an error matrix (confusion matrix) and the NDVI vegetation index correlation test compared with canopy cover and density data. The classification resulted in four different land cover classes with an overall accuracy of 97.70% and a kappa coefficient of 0.9688. The mangrove vegetation distribution from the classification results is 524.75 ha. The NDVI correlation with the percentage of canopy cover is very significant with a correlation coefficient (r) = 0.9516, while the NDVI correlation with density resulted in moderate correlation (r = 0.5315).Keywords: density; mangrove; Landsat 8; NDVI                                             AbstrakMangrove merupakan salah satu objek yang dapat diidentifikasi menggunakan teknologi penginderaan jauh yakni memanfaatkan citra satelit. Analisis sebaran dan kerapatan vegetasi mangrove menggunakan citra Landsat 8 telah dilakukan di Bolaang Mongondow Timur, Sulawesi Utara pada bulan September 2020. Penelitian ini bertujuan untuk memetakan sebaran mangrove dan mengetahui hubungan korelasi antara nilai NDVI dengan tutupan kanopi dan kerapatan mangrove. Pengolahan data citra Landsat 8 dengan perangkat lunak ENVI 5.3 dan ArcGIS 10.1. Klasifikasi maximum likelihood digunakan untuk memisahkan fitur mangrove dan non mangrove. Perhitungan kerapatan vegetasi mangrove dengan algoritma NDVI dan klasifikasi saluran tunggal menggunakan metode density slice untuk membagi kerapatan mangrove berdasarkan rentang nilai piksel citra NDVI. Uji akurasi hasil klasifikasi menggunakan matriks kesalahan (confussion matriks) dan uji korelasi indeks vegetasi NDVI dengan data tutupan kanopi dan kerapatan. Hasil klasifikasi mendapatkan empat kelas tutupan lahan yang berbeda dengan overall akurasi sebesar 97,70 % dengan kappa coefisien sebesar 0,9688. Luas sebaran vegetasi mangrove dari hasil klasifikasi adalah 524,75 ha. Korelasi NDVI  dengan persentase tutupan kanopi termasuk korelasi sangat kuat dengan koefisien korelasi r = 0,9516 sedangkan korelasi NDVI  dengan kerapatan termasuk korelasi sedang (r = 0,5315).Kata kunci: kerapatan; mangrove; Landsat 8; NDVIÂ

    Fishing Season Estimation of Mackerel Scad (Decapterus spp) with purse seines in the Sulawes Sea based on catch data

    This study aims to determine the pattern or fluctuations in the fishing season of mackerel scad (Decapterus spp) and suspected months of the fishing season. Data analysis is made in the form of cross-tabulation and mapped into graphs. Fishing season is determined by the average percentage methods based on time series analysis. The results indicated that the fishing season of mackerel scad in the Sulawesi Sea fluctuates. The fishing season occurs from May to November with the highest season/seasonal index occurring in July, while non-seasonal conditions occur from December to April with the smallest seasonal index/non-seasonal peak occurring in January.Keywords:  mackerel scads, fishing season, fluctuation, Sulawesi SeaAbstrakPenelitian ini bertujuan untuk mengetahui mengetahui pola atau bentuk fluktuasi musim  penangkapan  dan pada bulan apa saja diduga menjadi kondisi musim penangkapan ikan layang (Decapterus spp). Untuk analisis musim penangkapan ikan menggunakan Metode Presentasi Rata-rata (The Average Percentage Methodes) yang didasarkan pada Analisis Runtun Waktu (Time Series Analysis). Hasil analisis menunjukan bahwa musim penangkapan ikan layang (Decapterus spp) di Laut Sulawesi berfluktuasi. Musim penangkapan terjadi pada Mei sampai November dengan puncak musim/indeks musim tertinggi terjadi pada Juli sedangkan kondisi bukan musim terjadi pada Desember sampai April dengan indeks musim terkecil/puncak bukan musim terjadi pada Januari. Kata-kata Kunci: ikan layang, musim penangkapan, fluktuasi, laut Sulawesi
