    Prevalence, genotype distribution and mutations of hepatitis B virus and the associated risk factors among pregnant women residing in the northern shores of Persian Gulf, Iran

    Considering perinatal transmission and the high rate of chronic hepatitis B virus (HBV) infection in infants, diagnosis of HBV infection during pregnancy and timely interventions are of great importance. Therefore, this study was performed to investigate the prevalence and genotype distribution of HBV infection and the associated risk factors among pregnant women in the northern shores of the Persian Gulf, South of Iran. Methods Serum samples of 1425 pregnant women were tested for the presence of HBsAg and HBcAb by ELISA (HBsAg one—Version ULTRA and HBc Ab ELISA kits, DIA.PRO, Milan, Italy). The seropositive samples were tested for the presence of HBV DNA by nested PCR, targeting S, X, pre-core (pre-C), and basal core promoter (BCP) regions of the HBV genome. The amplified fragments were sequenced by Sanger dideoxy sequencing technology to evaluate the genotype distribution and mutations of HBV infection by using the MEGA 7 software. The HBV seropositive pregnant women were tested for HCV and HIV coinfections by ELISA (HCV Ab and HIV Ab/Ag ELISA kits, DIA.PRO, Milan, Italy). Results Of 1425 participants, 15 pregnant women (1.05%, 95% CI: 0.64%-1.73%) were positive for HBsAg, 41 women (2.88%, 95% CI: 2.10%-3.88%) were positive for HBcAb, and 5 women (0.35%, 95% CI: 0.15% –0.82%) had HBV viremia with genotype D, sub-genotype D3 and subtype ayw2. One of the viremic samples was positive for HBcAb but negative for HBsAg, which is indicative of an occult HBV infection. HBsAg seroprevalence was higher among pregnant women aged 20 to 29 years, women in the third trimester of pregnancy, residents of Khormuj city, Afghan immigrants, illiterate women, and pregnant women with a history of tattoo and HBV vaccination. The highest rate of HBcAb seroprevalence was observed in residents of Borazjan city, Turk ethnicity, the age group >39 years, and those women with more parities and a history of abortion. Nevertheless, HBV seroprevalence among pregnant women was not statistically associated with these variables. In contrast, HBcAb seropositivity was significantly associated with the history of tattoo (P = 0.018). According to mutations analyses, seven amino acid substitutions in the HBsAg, one point mutation in the pre-C region, and five points mutations in the BCP region were detected. Besides, the BCP mutations caused amino acid substitutions in the X protein. Of note, the conversion of Ala → Val at amino acid 168 (A168V) and Thr → Pro at amino acid 127 (T127P) were detected in HBsAg of the occult HBV strain. Conclusion These results indicate a relatively low prevalence of HBV infection among pregnant women in the South of Iran, while tattooing is a risk factor for exposure to HBV infection. Moreover, all of the HBV-positive pregnant women were asymptomatic and unaware of their infection. Therefore, routine screening for HBV markers during pregnancy, appropriate treatment of HBV-infected women, and HBV vaccination are recommended to decrease mother-to-child transmission of HBV

    OBJECTIVES: Osteoporosis is a major public health problem that is appeared with increasing age. This study evaluated the effect of the algae Dichotomaria obtusata methanol extract on osteogenic differentiation of the cultured bone marrow mesenchymal stem cells (BMMSCs) in vitro and analyzed the algae methanol extract to find out the potent beneficial components. METHODS: Dichotomaria obtusata were collected from the coastal area of Bushehr City in the Persian Gulf, Iran. The expression of osteogenesis-related genes was examined using real-time PCR. The formation of calcium deposits in differentiated MSCs was examined by Alizarin R staining. Analyses of algae extract ingredients were performed by gas chromatography-mass spectrometry (GC-MS). KEY FINDINGS: Methanol extract of the algae caused the up-regulation of osteogenic genes that were significant for Osteopontin and Osteocalcin (P < 0.05) and also led to an increase in calcium deposits and matrix mineralization in BMMSCs. The GC-MS analyses of the algae extracts resulted in the identification of steroids and essential fatty acids. CONCLUSION: The results of the study indicated that the methanol extract of D. obtusata may possess significant potentials for the prevention of osteoporosis in vitro

    Removal of amoxicillin from wastewater in the presence of H2O2 using modified zeolite Y- MgO catalyst: An optimization study

    In this paper, Zeolite-MgO was generated using alkali-thermal method and was utilized as a catalyst to decrease amoxicillin (AMX) concentration in the presence of H2O2 from wastewater. Different tests like Fourier-transform infrared (FTIR), Brunauer-Emmett-Teller (BET), field emission scanning electron microscopy-energy dispersive X-ray analysis (FESEM-EDX), thermogravimetric analysis (TGA), and X-ray diffraction (XRD) were done to determine catalyst properties. Active groups of C-S–C, C[dbnd]O, C[dbnd]C, C–N, C–O, N–O, and N–H were identified in catalyst frame. According to XRD results, lower crystallinity of nanoparticles after modification of zeolite by MgO can lead to improvement of AMX removal. Active surface of zeolite (2.32 m2/g) was increased after optimization by MgO to 2.96 m2/g, indicating an increase in the catalyst capacity for activation of H2O2. In addition, furnace temperature (200–500 °C), residence time in the furnace (1–4 h), and Mg(NO3)2: zeolite ratio (0.25: 2, 0.5:2, 1:2 w/w) were studied to achieve the optimized catalyst for AMX removal. Different parameters like pH (5–9), H2O2 concentration (0–6 mL/100 mL), dose of catalyst (0–10 g/L), AMX concentration (50–300 mg/L), and reaction time (10–130 min) were also studied. The best efficiency (97.9%) of AMX removal was achieved at acidic pH with the lowest amount of H2O2 (0.1 mL/100 mL) and 7 g/L of catalyst.     Capsule: Comparison of polycyclic aromatic hydrocarbons levels of maternal and cord serum samples. © 2021 Elsevier Ltd The presence of polycyclic aromatic hydrocarbons (PAHs) in the fetal environment is a high-priority concern due to the fetus being more sensitive than adults to these ubiquitous xenobiotics. The aim of the present study was to compare the maternal and fetal serum levels of ΣPAHs and their effects on fetal growth in an industrial and an urban area in Southwest Iran. The industrial area was the petrochemical and gas area (PGA) of the Central District of Asaluyeh County and the urban area (UA) was the Central District of Bushehr County, Ninety-nine maternal serum (MS) and 99 cord serum (CS) samples from the PGA and 100 MS and 100 CS samples from the UA were collected during May 2018 to February 2019. The mean concentrations of ΣPAHs were significantly (p < 0.05) higher in the PGA than the UA in both MS (157.71 vs. 93.56 μg/L) and CS (155.28 vs. 93.19 μg/L) samples. Naphthalene (NAP) was the predominant PAH detected in all the studied samples. Significant negative associations were found between birth weight and anthracene (ANT) level in MS (β = −22.917, p = 0.032; weight decrement = 22.917 g for a 1 μg/L increase in ANT); head circumference and chrysene (CHR) level in MS (β = −0.206, p = 0.023; head circumference decrement = 0.206 cm for a 1 μg/L increase in CHR); and birth height and NAP level in CS (β = −0.20, p = 0.005; height decrement = 0.20 cm for a 1 μg/L increase in NAP). Maternal diet had a significant effect on the serum levels of PAHs. The results of this study showed that transmission of PAHs from mother to fetus through the cord blood is an important issue and mothers who live in industrial areas and consume PAH-containing foodstuffs, and their fetuses, are more at risk than those living in a non-industrial urban area

    Three-Dimensional Imaging in Stem Cell-Based Researches

    Stem cells have an important role in regenerative therapies, developmental biology studies and drug screening. Basic and translational research in stem cell technology needs more detailed imaging techniques. The possibility of cell-based therapeutic strategies has been validated in the stem cell field over recent years, a more detailed characterization of the properties of stem cells is needed for connectomics of large assemblies and structural analyses of these cells. The aim of stem cell imaging is the characterization of differentiation state, cellular function, purity and cell location. Recent progress in stem cell imaging field has included ultrasound-based technique to study living stem cells and florescence microscopy-based technique to investigate stem cell three-dimensional (3D) structures. Here, we summarized the fundamental characteristics of stem cells via 3D imaging methods and also discussed the emerging literatures on 3D imaging in stem cell research and the applications of both classical 2D imaging techniques and 3D methods on stem cells biology

    Genotyping and phylogenetic analysis of hydatid cysts isolated from livestock in Bushehr province, Iran

    Hydatid cyst is one of the parasitic zoonoses caused by infection with the larval stage of Echinococcus granulosus tapeworm. The spread of this parasite is global and is of great importance in terms of public health. To date, ten different species of this parasite have been identified that differ in characteristics such as life cycle, epidemiology and pathogenesis. The purpose of this study was to determine the genotype and phylogenetic relationship of hydatid cysts isolated from livestock of Bushehr province, Iran. About 62 samples of hepatic and pulmonary hydatid cysts were collected from slaughtered animals. DNA extracted by phenol–chloroform method was amplified by PCR using primers specific for the cox1 gene. The PCR products of 50 samples were sequenced and analyzed using BioEdit software and compared with sequences in the GenBank. The phylogenetic tree was drawn using Neighbor Joining tree-NJ method, and its reliability was evaluated. Sequencing results showed that out of 50 sequenced samples, 43 samples had the genotype of Echinococcus granulosus and 7 samples had the genotype of Taenia hydatigena. By drawing a phylogenetic tree, all 43 hydatid cyst samples belonged to G1 strain. The predominance of G1 strain of hydatid cyst in livestock of Bushehr province shows the main role of this genotype in establishing the life cycle of parasite in this region and if the genotype of the parasite in dogs and humans is determined, then these findings can be used to disrupt the life cycle of the parasite and reduce the human infections

    Toxic heavy metals and nutrient concentration in the milk of goat herds in two Iranian industrial and non-industrial zones

    This work aimed to explore the concentration of nickel, manganese, iron, copper, chromium, and lead in the milk of goat herds in the industrial area of Asaluyeh (southern Iran) and the non-industrial area of Kaki. The milk of 16 goat herds (each herd had at least ten goats) was collected in several villages in each area, and at the same time, the drinking water and forage of goats were sampled. The concentration of elements in the samples was determined by ICP-OES. The mean concentrations of chromium, copper, iron, manganese, lead, and nickel in milk samples of the Asaluyeh area were 16.423 ± 0.349, 0.146 ± 0.118, 6.111 ± 0.501, 0.239 ± 0.016, 0.141 ± 0.030, and 1.447 ± 0.101 mg/kg, respectively. Concentrations of heavy metals (except for copper) in the milk of goats in the industrialized area of Asaluyeh were significantly higher than that of Kaki (P < 0.05). Also, the content of heavy metals was significantly correlated with lactose levels (P < 0.05). The hazard index for drinking the goat milk was computed to be 0.444 and 0.386 for the Asaluyeh and Kaki area, respectively, which shows a minimal effect of this exposure pathway

    Dicentracin-Like from Asian sea bass Fish and Moronecidine-Like from Hippocampus Comes: Two Candidate Antimicrobial Peptides Against Leishmanina major Infection

    Anti-Leishmanial drug therapy faces significant challenges related to cytotoxicity and drug resistance. Thus, new and efficient anti-Leishmanial drugs need to be identified. Due to their broad-spectrum antimicrobial and also immunomodulatory activities, antimicrobial peptides (AMPs) have attracted considerable attention. In this study, we comparatively assessed the anti-Leishmanial activities of two recently identified AMPs (dicentracin-like and moronecidine-like) and the well-known AMP piscidin from the hybrid striped bass. AMPs were first assessed against Leishmania major promastigotes using MTS. Subsequently, macrophages were infected with L. major and treated with AMPs to evaluate anti-amastigotes activity of AMPs, and non-infected macrophages were treated with AMPs to determine cytotoxicity against mammalian cells using MTS. The induction of factors limiting L. major growth (IL-12, TNF-α and reactive oxygen species (ROS)) by AMPs was measured by ELISA and dichlorofluorescin-diacetate (DCFH-DA) assay, respectively. Piscidin was more efficacious against L. major promastigotes as compared to dicentracine-like or moronocidin-like peptides, whereas, dicentracine-like and moronocidin-like peptide exhibited a higher activity against L. major amastigotes compared to piscidin. In turn, piscidin was most cytotoxic in non-infected macrophages compared to the other two AMPs. A direct association was observed between hydrophobicity of AMPs and their anti-promastigote and cytotoxic activities. Dicentracine-like or moronocidin-like peptides induced higher levels of IL-12, TNF-α and ROS in macrophages compared to piscidin. Collectively, our results suggest that dicentracine-like and moronocidin-like peptides represent potentially promising multi-functional therapeutic agents that might not only directly kill L. major but also induce anti-Leishmania factors that can limit L. major growth and intracellular survival

    Investigation of in vitro antifungal susceptibility testing and genetic diversity of clinical isolates of Trichophyton benhamiae and Trichophyton eriotrephon in Iran

    Background: Trichophyton benhamiae is a zoophilic dermatophyte, known as one of the causative agents of dermatophytosis. Objectives: The purpose of this study was to explore the genotypes of T. benhamiae strains isolated from geographically different areas of Iran and also to evaluate in vitro antifungal susceptibility profile of these strains against seven antifungal drugs. Methods: Twenty-two strains of T. benhamiae and two strains of T. eriotrephon were isolated from patients with distinct types of dermatophytosis. DNA extraction and amplification of rDNA regions using ITS1 and ITS4 primers were conducted on the isolates. The in vitro antifungal susceptibility of posaconazole (PSC), voriconazole (VRC), itraconazole (ITC), ketoconazole (KET), caspofungin (CAS), terbinafine (TRB) and griseofulvin (GRZ) was evaluated according to CLSI M38-A2 protocol. Results: The multiple alignment of the ITS-rDNA sequences of T. benhamiae indicated a mean similarity of 99.5%, with 0–3 interspecies nucleotide difference. The geometric mean (GM) values of minimum inhibitory concentrations (MICs) and minimum effective concentrations (MECs) across the all isolates were respectively: TRB: 0.025 mg/L, PSC: 0.032 mg/L, ITC: 0.050 mg/L and VRC: 0.059 mg/L with lower values and CAS: 0.31 mg/L, KTZ: 0.56 mg/L and GRZ: 0.76 mg/L with higher values. Conclusion: Diverse ITS sequence types of T. benhamiae were shown in different geographical regions of Iran. The TRB, PSC and ITC were the most effective drugs against T. benhamiae strains, respectively.     Microplastics accumulation in sediments and Periophthalmus waltoni fish, mangrove forests in southern Iran

    This investigation was aimed to identify microplastics in the sediment and mudskipper fish (Periophthalmus waltoni) in mangrove forests in southern Iran. Sediments and mudskipper samples were collected at high, mid, and low tidal points of five stations. A total of 2657 plastic particles in different size, color, shape, and genera were identified from sediment samples and 15 microplastic were isolated from mudskippers. The highest and lowest abundance of isolated microplastics from sediments was observed in mangrove forests of Bidkhoun (urban area) and Bordkhon, respectively while no microplastics were found in the fish tissue in those stations. The black (60%) and white (7%) color microplastics in the mudskipper had the highest and the lowest frequency. The highest and lowest polymers in mangrove forest sediments were corresponded to polystyrene (26%) and polycarbonate (3%), respectively. Raman and Fourier transform infrared spectroscopy (FT-IR) techniques were used to identify the type of the polymer. Most of the microplastics found were made of polystyrene, polypropylene, and polyethylene terephthalate. The type of studied area and texture of sediment separately affected the frequency of microplastic and mesoplastic (P-value <0.05) in the sediment samples. The abundance of microplastics in the sediment samples of the Bidkhoun mangrove forest was higher than other studied stations due to proximity to urban and industrial areas. The findings of this study raised concerns about microplastic pollution in the mangrove forests of southern Iran, a threat to the ecosystem and public health, which requires careful actions to prevent and diminish its adverse effects
