Crisis situations and needs of mothers caring for an adult child with experience of schizophrenia
This master's thesis focuses on the needs and experiences of mothers caring for an adult child with schizophrenia in crisis situations. The literature section of thesis describes the concept of informal caregivers and the role of the family in the recovery process of individuals with schizophrenia. The core chapters of the theoretical section explore stress models in caregivers and their experiences with crisis situations. Additionally, models of needs and findings from studies on parental needs are discussed. The final chapter concentrates on available forms of support and services designed for families. The research part has a qualitative character and aims to identify crisis situations and the needs of mothers caring for an adult child with schizophrenia. Main focus is given to crises in social relationships, adaptation crises following the onset of the illness, and mothers' self-care needs. Thematic analysis identified three key themes related to experiences with crisis situations: the early stages of caregiving, access to healthcare during acute psychiatric episodes, and family challenges along with social isolation of mothers. The impact of Immune distribution cellular patterns on clinical outcome in immunologically cold and hot carcinomas
Intratumorální terciární lymfoidní struktury (TLS) představují klíčové regulátory protinádorové imunity. Jejich funkční a prognostická role se však výrazně liší mezi jednotlivými typy nádorů a v závislosti na charakteru nádorového mikroprostředí (TME) jednotlivých pacientů. Cílem této práce bylo hodnotit vliv prostorového uspořádání, aktivace a buněčného složení TLS ve dvou imunologicky odlišných typech nádorů: v nemalobuněčném karcinomu plic (NSCLC), charakterizovaném vysokou nádorovou mutační náloží (TMB) a obecně příznivou odpovědí na terapii inhibitory imunitních kontrolních bodů imunitních reakcí (ICI), a v metastazujícím high- grade serózním karcinomu ovaria (mHGSOC), typicky provázeném nízkou TMB a imunitně vyčerpaným mikroprostředím. Multimodální přístup využívající multiparametrickou imunofluorescenci, pokročilou digitální patologii, prostorovou transkriptomiku a single-cell RNA-seq analýzu identifikoval klíčové rozdíly v četnosti, fenotypech a vlivu TLS na CD8⁺ T-lymfocyty mezi sledovanými nádory. Vzorky NSCLC s intratumorálně lokalizovanými a aktivovanými TLS (mTLS) vykazovaly vyšší zastoupení progenitorových CD8⁺PD1⁺TOX+ TCF1⁺ T buněk, které jsou známé zvýšenou odpovědí na terapii ICI. Atomic-scale study of Protein-DNA interfaces using Probability Density Maps and Local Energy Decomposition
This thesis presents a novel approach to the study of protein-DNA interfaces using probabil- ity density maps constructed by analyzing a large structural dataset. This analysis is enabled by using local conformation classification for nucleic acids. The aim was to gain a detailed understanding of the relationship between the two molecules, which could be used to identify binding modes or to identify additional restrictions for macromolecular docking. The results of the analyses, visualized in the form of probability density maps, allow interpretation of the data and finding of the most probable interactions between proteins and DNA. Furthermore, interaction sites, identified using the maps, were investigated using ab initio calculations and energy decomposition analysis, to describe the energy landscape of the interaction and validate the results from the probability density maps
Enzyme Kinetics of the HelD Protein
Mycobacterial HelD is a protein responsible for the dissociation of RNA polymerase from nucleic acids during stalled transcription. Subsequent release of HelD from RNAP can be achieved through ATP and GTP hydrolysis by HelD. Notably, HelD exhibits catalytic activity independent of RNA polymerase. This thesis identifies optimal reaction condi- tions to maximize the hydrolysis rate and characterizes the enzyme kinetics under these conditions. Furthermore, a modified substrate inhibition equation is proposed to describe the observed substrate inhibition, providing a model for the inhibitory effects at high substrate concentrations. Professional Competence of Police Officers in Working with Persons with Paraphilic Preference: Stigmatization and Evaluation of the Effect of the Parafilik Educational Program
The subjectively perceived level of professional competence and the level of stigmatization of people with paraphilic preferences play a key role in the willingness of professionals to work with this group, with insufficient knowledge and negative attitudes representing the main barriers to the use of care by paraphilic individuals. The purpose of the Parafilik educational program is precisely to develop specific work competence of police officers. The aim of this study is to 1) determine the perceived level of professional competence and willingness to work with paraphiles, 2) determine the level of stigmatization of non-offending paraphiles, and 3) evaluate the effect of the Parafilik educational program in a cohort of police officers (N = 62),. For this purpose, data from the original evaluation questionnaires administered as part of this educational program were analyzed. The research showed that after completing the program, police officers experienced a statistically significant increase in their subjective understanding of paraphilias and a subjective increase in their professional competence and willingness to work with paraphilic persons, although these changes were not statistically significant. Two Ways of Relating to Doubt in Kierkegaard's Existential Anthropology
The presented dissertation seeks to philosophically interpret the motif of one doubt but two ways in which one can relate to it. The two possible existential outcomes of doubt are considered within the Kierkegaardian framework of becoming oneself or becoming Christian. The dissertation's main premise stems from one of the key literary sources for interpretation, i.e. the Eighteen Upbuilding Discourses in which Kierkegaard explicitly states that there are two doubts-one that is subjected to education ("saving doubt") and one that deceives ("false doubt"). Importantly, the relating to corresponds with an act of will, meaning that it is up to the individual how she will decide to doubt. The doubt can be either targeted objectively outwards or subjectively inwards. While false doubt perpetuates a vicious cycle of existential despair from which it is extremely difficult to emancipate, and which drowns the individual in the multiplicity of knowledge, saving doubt leads one to a vulnerable state of "being capable nothing at all" and "needing God". Effect of deep transcranial magnetic stimulation on inhibitory control processes
5 Abstract Background: Inhibitory control (IC) is a core executive function impaired in obsessive- compulsive disorder (OCD), often linked to hyperactivity in the anterior cingulate cortex (ACC). Repetitive transcranial magnetic stimulation (rTMS), particularly deep TMS (dTMS) targeting the ACC, may modulate inhibitory control, providing therapeutic potential. This study investigates the effects of anterior cingulate double cone (ACDC) TMS on inhibitory control in healthy individuals and patients with OCD. Aims: The primary aim was to assess the impact of ACDC TMS on inhibitory control in healthy participants and patients with OCD using the interference Stroop effect. The secondary aim was to compare other behavioral and electrophysiological (EEG) effects of ACDC TMS across groups, including changes in reaction times (RT), number of errors, and error-related negativity (ERN) amplitude. Methods: The study comprised three parts: (1) ACDC TMS in 30 healthy participants, (2) ACDC TMS in 30 OCD patients, and (3) a comparative analysis of ACDC TMS effects between the two groups. Results of comparative damage detection case study
A significant amount of attention has been devoted to rapid bridge testing in recent years at the Institute of Theoretical and Applied Mechanics. New methods, such as the Vehicle Scanning Method and Moving Impulse Load Testing, have been developed and/or tested on a scaled laboratory bridge model. This article summarizes all conducted tests, discusses the evaluation methods used, presents the results obtained, and explores possibilities for their improvement. Among the tested methods, the application of Moving Impulse Load (MIL) moving at slow speed, with an excitation frequency around the first natural frequency of the tested bridge, appears to be the most promising. Two passages of the MIL allow for an approximate estimation of the dominant natural frequencies, while repeated passages at small velocity increments provide an excellent means of detecting changes in structural stiffness
Comparing salaries and wages in the Czech Republic: a bigger problem than it seems
Comparing public sector Salaries in the Czech Republic, where remuneration is based on tariffs (salary sphere), and Wages (wage sector employment, dominated by private entities) is a surprisingly complicated agenda. Our study explains the methodological difficulties of these comparisons and proposes a simple but methodologically sound approach. We discuss its shortcomings and apply it. Further, we offer recommendations for continued development of the data base and complementary analyses
