(ENG) Charles University Faculty of Pharmacy in Hradec Králové Department of Pharmaceutical Chemistry and Pharmaceutical Analysis Researcher: Dominika Puchírová Thesis supervisor: doc. PharmDr. Jan Zitko, Ph.D. Consultant: Mgr. Andrea Bachtíková Title of thesis: Synthesis and evaluation of pyrazinamide-containing derivatives 1,2,3-triazole linker as an inhibitor of prolyl-tRNA synthetase This thesis focuses on the design, development and synthesis of novel compounds with potential antimycobacterial activity, while the target structure is mycobacterial prolyl-tRNA synthetase, an enzyme essential for proteosynthesis, growth and cell viability. Basic structure of the proposed compounds is based on previously published 3-aminopyrazinamides with confirmed antimycobacterial activity, with the amide bond partially replaced by 1,2,3-triazole linker in order to increase the affinity for prolyl-tRNA synthetase and thus biological efficacy. The basic structure of the molecules of pyrazinamide with a 1,2,3-triazole linker and simple substituents at aromatic core. Basic structure of pyrazinamide compounds with a 1,2,3-triazole linker and simple substituents on the aromatic core Structural modifications were designed using in silico molecular modeling methods. The synthesis of the resulting compounds was carried..

Revision of the communication strategy of the Czech Sailing Association

    No full text
This bachelor thesis deals with the revision of the communication strategy of the Czech Sailing Association (CSA) for the period 2021-2024. The aim of the thesis was to identify the strengths and weaknesses of the current communication practice of the association, to assess its consistency with the strategic goals of the organization and the preferences of target groups, and to propose specific measures for its improvement based on the findings. A mixed research approach combining quantitative and qualitative content analysis was chosen to meet the set objectives. The quantitative part focused on traditional media and looked at the scope, topics, emotional colouring and impact of the outputs. The qualitative analysis focused on the content and style of communication on social media, the web and in newsletters, while basic quantitative metrics were also tracked for social media. Findings were then validated through semi-structured interviews with four representatives of the sailing community. The research revealed that CSA emphasizes sporting topics and representation, which is generally perceived positively. At the same time, however, poor communication towards internal stakeholders, low emphasis on the community level and little publicity of recruitment and sponsorship activities persist. The need..

Thematization of suffering as a tool of narrative authenticity in the work of Atiq Rahimi

is bachelor's thesis explores the theme of suffering in two novels by Afghan-French writer Atiq Rahimi, in the context of Persian exile literature in France. Atiq Rahimi has lived in France since the 's, where he first published all his novels. His very first novel, Khâkestar-o khâk ( ), written in Persian, and his first novel written in French, Syngué sabour: Pierre de patience ( ), are the focus of this analysis. e plots of both novels take place in war-torn Afghanistan. e protagonist of Khâkestar-o khâk suffers directly as a consequence of war, in which he loses part of his family. In contrast, the heroine of Syngué sabour: Pierre de patience is liberated from enforced silence as a result of the war's outcome and finds her own voice. e themes of war trauma and female narrative of liberation directly connect to both modern Afghan literature and Persian exile literature in France. Within exile literature, a New Orientalist narrative emerges, which this thesis adopts. is narrative functions between the literary work itself and the society that receives it. erefore, this thesis focuses on the analysis of peritexts, Second-person narrative, and translingual literature. rough these approaches, I conclude that Syngué sabour: Pierre de patience represents a New Orientalist narrative. Like the protagonists of..

The needs of male foster parents in long-term foster care

    No full text
The text of the thesis is focused on the issue of male foster parents' needs in long-term foster care within the Ústí nad Labem region. The main aim is to identify the conditions and requirements for obtaining foster parent status and to discover the key aspects of a foster parent's role. The theoretical part of the thesis describes substitute family care, family and its functions, the needs of parents and children, foster parents' needs, and their possible saturation by the state and related organizations. Everything is focused on the foster parent in long-term foster care- who they are, what the decision to care for a child in foster care involves, and what rights and obligations the foster parent has. In the practical part, I learned through qualitative research using thematic analysis of the foster care stories of the men addressed, including the joys, hardships, and difficulties they encountered. I also explored what they need to manage their roles and whether they have an overview of the possibilities of saturation of needs and take advantage of these opportunities. Based on the data received, I developed recommendations and suggestions on how and with what to approach or attract new people interested in substitute family care, as well as how to improve or expand support for foster parents...

Evaluation of the Success of selected game skills in mens Volleyball Nations League (VNL)

Title: Evaluation of the Success of selected game skills in mens Volleyball Nations League (VNL) Objectives: The goal of this thesis was to compare spin and floating serves and their passing. Methods: To construct the analysis, data was collected and imputed to table using strokes. The data was obteined by closely monitoring games from match recordings. Furthermore, in order to analyse the data correctly, the game had to be split into individual sections of serves, passes and subsequent sets. The last method that was used in this work was the evaluatin scale, both for serves and passes. Results: The results were noted for clarity in graphs and tables, where they were commented. Keywords: volleyball, serve, pass, success, player, tea

The impact of physical activity in natural environments on fatigue

    No full text
Title: The impact of physical activity in natural environments on fatigue Objectives: This thesis employs an empirical-theoretical approach and utilizes a quantitative method of data collection through a questionnaire survey. For the purpose of the research, an original questionnaire was developed consisting of a total of 24 questions-23 closed-ended and 1 open-ended. The target group comprised 12 adult respondents, regardless of gender, who have personal experience with physical and mental fatigue in various life situations. The questionnaire was distributed and completed via the online platform Google Forms, enabling efficient and clear data collection. Methods: The individual results are presented in the form of graphs, accompanied by commentary highlighting key trends in responses. The study's findings indicated affirmative answers to research questions 1 and 2, thus supporting the hypothesis that walking in natural environments contributes to regeneration and improved mental well-being. The third research question could not be verified within this sample, as all respondents reported a positive relationship with the natural environment. Results: The individual results are presented in the form of graphs, accompanied by commentary highlighting key trends in responses. The study's findings..

    Stochastic methods in crystallography

    No full text
We introduce a statistical framework for analyzing and testing dependencies among crystallographic orientations in polycrystalline materials. Motivated by applications in materials science, we address the problem of testing pairwise independence within r-tuples of orientations of symmetric objects in three-dimensional space. We propose a novel co- variance measure between random orientations and construct multivariate asymptotic tests for pairwise independence based on U-statistics. The performance of these tests is assessed through simulation studies under various models. Complementing this, we present a general methodology for modelling the distribution of grain orientations in polycrystalline microstructures represented by three-dimensional Laguerre tessellations. Conditional on the tessellation, we introduce stochastic interaction models that capture dependencies between neighbouring grain orientations. Model parameters are estimated via maximum pseudolikelihood and model adequacy is evaluated using simulation-based graphical tools. The proposed methodology is applied for analyzing a dataset repre- senting a nickel-titanium shape memory alloy, demonstrating the practical relevance for understanding material behaviour through orientation statistics.
