Neuroprotective effects of FTO inhibition in the animal model of Alzheimer's disease
Alzheimer's disease is a serious neurodegenerative disorder that is one of the most common causes of dementia, and there is currently no sufficiently effective treatment. A promising ther- apeutic target appears to be the α-ketoglutarate-dependent demethylase FTO, as FTO partici- pates in several pathophysiological processes within the m6 A pathway, such as pro-inflamma- tory activation of microglia and ferroptosis, with ferroptosis also being shared with other neu- rodegenerative diseases such as Parkinson's disease, amyotrophic lateral sclerosis, and glau- coma. In this thesis, the inhibitor MO-I-500, which inhibits FTO, was used and tested in vivo using behavioural tests in a streptozotocin model of sporadic Alzheimer's disease. The open field test found no changes in this model between the control model and the streptozotocin- treated control and there was also no effect of the inhibitor. A statistically significant deficit was found in the Morris water maze, which was mitigated by the administration of the MO-I- 500 inhibitor. Despite the numerous limitations of this experiment, the FTO inhibitor can be considered a potentially suitable therapeutic agent, but further research is needed. S uznáním nedostatku relevantního výzkumu v oboru ohledně intersekcionality tvorby digitálních identit, tato diplomová práce zkoumala, jak jedinci konstruují své digitální identity, obousměrný přenos identity mezi internetem a realitou, proces diferenciace mezi "autentickou osobností" a tou tvořenou pomocí "internetového vlivu", shody a neshody mezi online a off-line znázornění identity stejně jako faktory, které usnadňují změnu napříč těmito kontexty. Tato práce využívá smíšený metodologický přístup, což zahrnuje kvantitativní analýzu obsahu sociálních médií generovaného uživateli z Tik Toku a Redditu se zaměřením na obsah původem z Tradwife and Alpha male komunit a šest semistrukturovaných pohovorů se členy generace Z, kteří se identifikují jako členové těchto komunit nebo s nimi mají zkušenost. Introduction of the concept of angle in Grade 6
The thesis deals with the engaging introduction of the concept of angle in the 6th grade of elementary school. The theoretical part is based on the theory of generic models as a framework for describing the process of student learning. For effective lesson planning and evaluation, it draws on the principles of responsive teaching, in particular the backward design approach. The thesis examines what various contemporary authors consider to be stimulating teaching. It presents the constructivist-transmissive spectrum and describes how its poles can be combined and used to benefit students. The concept of angle is then introduced. The work describes its curricular anchoring in the Czech edu- cational system: in the Rámcové vzdělávací programy, Standardy pro základní vzdělávání, the entry exams to secondary schools, and textbooks. This anchoring is compared with the historical development of the concept of angle and current research on effective didac- tic approaches. It addresses the fact that curricular documents and some of the examined textbooks do not reflect recent research. In the practical part, the thesis presents one possible formulation of what a teacher may understand as engaging teaching. Neighbourhood Residential Changes of the Prague Inner City: Trajectories and Consequences
1 Abstract The dissertation examines neighbourhood residential changes within Prague's inner city. Despite extensive research on inner-city transformations in Central and Eastern Europe, comprehensive analyses of inner-city neighbourhood change remain notably scarce. This thesis thus addresses this gap by analysing the evolving inner differentiation of the Prague inner city and its neighbourhoods over time and across various spatial levels. Specifically, the research investigates shifts in socio-spatial patterns, identifies occurring neighbourhood change processes and their driving forces, and explores their consequences, including impacts on the daily lives of long-term residents. A mixed-methods approach was employed, combining diverse data sources and quantitative and qualitative research methods. The dissertation contributes to empirical and conceptual debates concerning inner-city neighbourhood change and gentrification in post- socialist countries, the outcomes of such changes, and the complex relationship between gentrification and other neighbourhood change processes
Systematic development of map work in upper secondary school students
The diploma thesis deals with the issue of developing map skills in upper secondary school students through a systematic educational intervention based on the principle of scaffolding, i.e., the gradual cognitive increase of tasks. The aim of the thesis was to determine whether targeted systematic work with maps contributes to improving map skills in reading, analysis, and interpretation of maps and how students perceive this process. Methodologically, the research was based on a combination of quantitative and qualitative approaches. It included pre-instruction testing (pretest), the implementation of three thematically distinct worksheets focused on physical, socio-economic, and regional geography, and subsequent testing (posttest) supplemented by a questionnaire survey. The research sample consisted of students of a grammar school and students of the Ecology and environmental protection study programme. The results showed that after completing the intervention, most respondents achieved demonstrable improvement in the level of their map skills. The questionnaire survey confirmed predominantly positive evaluations of the benefits of working with worksheets, both in terms of skill development and motivation to engage actively in lessons. Fluid flows with random factors
This thesis concerns the existence of solutions to the magnetohydrodynamics (MHD) system with stochastic perturbation introduced via random initial data and a stochastic integral solely in the induction equation, while the fluid equations remain deterministic. We focus on the case where the adiabatic exponent exceeds three, although more general results can be achieved. The existence proof is carried out using the penalization method. We begin by defining the notion of a martingale solution and establishing sufficient con- ditions for its existence. The proof then proceeds by means of a stochastic compactness method. Using an energy inequality, we derive a priori estimates in expectation. Due to the stochastic nature of the problem, we demonstrate convergence in law of the penalized solutions.
