Characterizing intensity of a non-contact boxing session for individuals with Parkinson's disease: a cross-sectional pilot study
TRPV4 and DISC1: regulators of cytoskeletal control in microglia
Microglia are the primary immune cells of the central nervous system, and their functions are essential for proper brain development, maintenance, and health. During embryogenesis, these cells colonize the brain and adopt a ramified, surveillant phenotype in the healthy adult brain. Throughout life, microglia are uniquely positioned to respond dynamically to their environment, allowing them to contribute to key physiological processes such as synaptic pruning, neurogenesis, angiogenesis, and maintenance of brain cell homeostasis. These roles rely on microglial motility, ranging from cell migration to the dynamic remodeling of their processes. Impairments in microglial function are increasingly associated with brain malformations and neurological disorders, including Alzheimer’s disease, autism spectrum disorder, and schizophrenia. Developmental disruptions in microglial function can lead to long-term consequences that alter the brain’s ability to respond to infection and process cognitive information in adulthood. This highlights the importance of understanding the mechanisms that regulate microglial motility and function throughout life. Despite the established importance of microglial motility in brain development and homeostasis, the cellular and molecular mechanisms underlying their migration, morphology, and immune functions remain incompletely understood. This doctoral dissertation aims to address this gap by focusing on two proteins that govern cytoskeletal remodeling in microglia, particularly in pathological conditions
Global impoverishment of natural vegetation revealed by dark diversity
Anthropogenic biodiversity decline threatens the functioning of ecosystems and the many benefits they provide to humanity(1). As well as causing species losses in directly affected locations, human influence might also reduce biodiversity in relatively unmodified vegetation if far-reaching anthropogenic effects trigger local extinctions and hinder recolonization. Here we show that local plant diversity is globally negatively related to the level of anthropogenic activity in the surrounding region. Impoverishment of natural vegetation was evident only when we considered community completeness: the proportion of all suitable species in the region that are present at a site. To estimate community completeness, we compared the number of recorded species with the dark diversity-ecologically suitable species that are absent from a site but present in the surrounding region(2). In the sampled regions with a minimal human footprint index, an average of 35% of suitable plant species were present locally, compared with less than 20% in highly affected regions. Besides having the potential to uncover overlooked threats to biodiversity, dark diversity also provides guidance for nature conservation. Species in the dark diversity remain regionally present, and their local populations might be restored through measures that improve connectivity between natural vegetation fragments and reduce threats to population persistence.Acknowledgements

Experimental and analytical assessment of the combined in-plane bending-shear behavior of timber frame walls
As timber buildings grow taller, understanding the mechanical behavior and robustness of multi-story timber structures is increasingly important. In Europe, Eurocode 5 governs their design but offers little guidance for failure scenarios. In particular, it lacks provisions for diaphragm walls under combined bending and shear forces that can occur during events like wall loss. This study investigates the behavior of such walls through experimental and analytical methods. An experimental campaign was conducted using eleven timber frame walls with OSB sheathing, fastened with either screws or staples. Five walls underwent four-point bending tests, two were tested under shear, and four experienced combined bending-shear loading. In the latter, a vertical preload was applied before monotonic shear loading to failure. Walls fastened with staples showed ductile failure with significant yielding, whereas walls fastened with screws exhibited brittle failure via screw tear-out. Shear failures primarily occurred at sheathing-to-framing connections in the lower rail. Under combined loading, increased vertical preload reduced shear capacity, displacement at failure, and force at yielding. The experiments supported an analytical model developed to estimate in-plane bending strength and maximum allowable shear load in combined loading scenarios. The model conservatively predicted the behavior of walls fastened with screws in bending but overestimated the strength of walls fastened with staples. For combined loading, the model provided a conservative estimate of maximum shear resistance. The authors acknowledge the work of Jan Leuraers, Dan Dragan, and Ricky Pereira, who helped perform the experiments. Health impacts of extreme heat in medically at-risk populations: a space-time stratified case-crossover analysis in Belgium
BackgroundThe impact of heat on a vulnerable population, particularly those with pre-existing chronic diseases, is a growing public health concern. However, the risks for those with specific conditions and their variation across geographic, demographic, and socioeconomic (SES) status remain underexplored. This study aimed to investigate the association of extreme heat with morbidity and mortality among individuals with pre-existing chronic diseases in Flanders, Belgium.MethodologyWe analysed 14 years (2005-2019) of general practitioners (GPs) data from Flanders, northern Belgium, assessing both morbidity and mortality. Morbidity was defined as GP-recorded general and heat-related illnesses among individuals with pre-existing chronic diseases, while mortality included overall mortality and mortality among people with pre-existing chronic diseases. A space-time-stratified case-crossover design was employed, with a distributed lag non-linear model (DLNM) applied in quasi-Poisson regression. Various subgroup analyses were conducted to identify the most at-risk population. We quantified the relative risk (RR) at the 99th percentile of the daily minimum temperature relative to the minimum morbidity/mortality temperature (MMT).ResultsWe found a strong association of heat with morbidity and mortality. A substantially increased risk of morbidity was observed among individuals with pre-existing heart failure (RR = 2.79 [95% CI: 1.84-4.24]) and a high risk of mortality was found among those with pre-existing hypertension (RR = 2.01 [95% CI: 1.23-3.30]). We also observed a rise in heat-related morbidity risks for individuals with pre-existing Chronic Obstructive Pulmonary Disease (COPD) (RR = 2.09 [95% CI: 1.53-2.85]), hypertension (RR = 1.37 [95% CI: 1.08-1.74), chronic kidney disease (CKD) (RR = 1.74 [95% CI: 1.25-2.42]), and chronic mental health disorders (RR = 1.41 [95% CI: 1.06-1.89]). There was an increased risk of overall mortality (RR = 1.29 [95% CI: 1.02-1.62]) and mortality with pre-existing COPD (RR = 1.80 [95% CI: 1.19-2.73]). Urban populations and low-SES groups had increased heat-related risks for some health outcomes, and the highest vulnerability was observed in those aged 85+ for several chronic diseases.ConclusionOur findings indicated that chronic diseases are associated with higher vulnerability to adverse health effects due to extreme heat. We observed notable variations across geographic, demographic, and socioeconomic subgroups, emphasizing the need for targeted public health strategies.Endale Alemayehu Ali is funded by the Partner Organization Environmental Health Care, the Department of Health/Flemish Ministry on Welfare, Public Health and Family Matters. Metal selection for pitting corrosion resistant layer in geological containment of radioactive waste under anaerobic conditions
