Steering protein and salt ad- and desorption by an electrical switch applied to polymer-coated electrodes
Although solid-phase chromatography is a well-established method for protein separation, chemically intensive and often costly regeneration steps are needed to make reuse of the adsorbent possible. Here, we demonstrate the use of electrochemical principles as sustainable alternative. We make use of spontaneous adsorption of proteins to solid electrodes and reverse this process by applying an electric potential to regenerate the interface. This allows for adsorption of proteins to take place at 0 V difference between the electrodes, due to electrostatic interactions between the protein and the electrode surface. The desorption is then triggered by applying a potential difference (−1.2 V) between the electrodes.It is demonstrated that the incorporation of negatively charged polystyrene sulfonate (PSS) or positively charged polydiallyldimethylammonium chloride (PDMAC) in or on top of the respective activated carbon electrodes increases the amount of exchanged protein from 1 to 10 mg g−1, as compared to simple activated carbon electrodes. Interestingly, salt ad- and desorption occurs in opposite cycles compared to protein ad- and desorption, resulting in simultaneous concentration and desalting of the protein when 0 V is applied. On top of that, we also found that an enrichment in β-lactoglobulin could be achieved starting from whey protein isolate. These results clearly demonstrate that electrochemical technologies can be used not only for protein separation (including removal of salt), but also for protein fractionation, while not requiring solvent use
Electroresponsive polyelectrolyte brushes studied by self-consistent field theory
End-grafting of polyelectrolyte chains to conducting substrates offers an opportunity to fabricate electro-responsive surfaces capable of changing their physical/chemical properties (adhesion, wettability) in response to applied electrical voltage. We use a self-consistent field numerical approach to compare the equilibrium properties of tethered strong and weak (pH-sensitive) polyelectrolytes to applied electrical field in both salt-free and salt-containing solutions. We demonstrate that both strong and weak polyelectrolyte brushes exhibit segregation of polyions in two populations if the surface is oppositely charged with respect to the brush. This segregation gives rise to complex patterns in the dependence of the brush thickness on salt concentration. We demonstrate that adjustable ionization of weak polyelectrolytes weakens their conformational response in terms of the dependence of brush thickness on the amplitude of the applied voltage.</p
The future of phenomics in dairy cattle breeding
Increasingly complex dairy cattle production systems require that all aspects of animal performance are measured across individuals' lifetimes. Selection emphasis is shifting away from traits related to animal productivity toward those related to effcient resource utilization and improved health and welfare/ resilience. The goal of phenomics is to provide information for making decisions related to on-farm management, as well as genetic improvement.</p
Genome-wide identification of small G protein ROPs and their potential roles in Solanaceous family
Small GTPases function as molecular switches to active or inactive signaling cascades via binding or hydrolyzing GTP. A type of plant specific small GTPases, the ROPs are known to be involved in plant growth, development and immunity. We determined whether ROPs are conserved in Solanaceous species and whether they are involved in plant growth, development and resistance against Phytophthora capsisi. In genome-wide screening, a total of 66 ROPs in six Solanaceous species (SolROPs) were identified, including 16 ROPs in Solanum tuberosum L. (potato), 9 in Solanum lycopersicum L. (tomato), 5 in Solanum melongena L. (eggplant), 9 in Capsicum annuum L. (pepper), 13 in Nicotiana benthamiana Domin and 14 in Nicotiana tabacum L. (tobacco). Phylogenetic analysis revealed that 11 AtROPs and 66 SolROPs fall into five distinct clades (I-V) and hence a novel and systematic gene nomenclature was proposed. In addition, a comprehensive expression analysis was performed by making use of an online database. This revealed that ROP genes are differentially expressed during plant growth and development. Moreover, gene expression of SlROP-II.1 in S. lycopersicum could be significantly induced by P. capsici. Subsequently, SlROP-II.1 and its homologues in N. benthamiana and C. annuum (NbROP-II.1 and CaROP-II.1) were selected for functional analysis using virus-induced gene silencing. Infection assays with P. capsici on silenced plants revealed that SlROP-II.1, NbROP-II.1 and CaROP-II.1 play a role in P. capsici resistance, suggesting conserved function of ROP-II clade across different Solanaceous species. In addition, NbROP-II.1 is also involved in regulating plant growth and development. This study signified the diversity of Solanaceous ROPs and their potential roles in plant growth, development and immunity.</p
Mitigating eutrophication nuisance: in-lake measures are becoming inevitable in eutrophic waters in the Netherlands
In the Netherlands, legacies and diffuse nutrient pollution continue to fuel recurrent cyanobacterial blooms in mostly shallow and relatively small surface waters. A survey in peer-reviewed literature and Dutch grey-literature was performed to gain insight into the physical-, chemical- and biological in-lake interventions used to bring these waters towards their desired state. A critical overview is presented on efficacy of different measures to counteract cyanobacterial blooms directly via targeting the cyanobacteria or indirectly via reduction of nutrient availability. Many actions have no or limited effects on minimising cyanobacterial blooms (air-bubble- or oil screens, surface mixers, low-energy ultrasound, effective micro-organisms, fish introduction), while others are more effective, but may vary in longevity and costs (dams, excavation or dredging, hydrogen peroxide, phosphorus inactivation agents), meet legislation restrictions (copper-based algaecides, herbicides, dreissenids), or are not currently implemented (hypolimnetic withdrawal). The selection of promising interventions requires a proper diagnosis of each problem lake, based on water- and nutrient fluxes, the biology of the lake (plants, fish), the function of the lake and the characteristics of the method, such as efficacy, costs, safety and ease of implementation. In the Netherlands, ongoing diffuse loads and legacies necessitate repetitive in-lake interventions.</p
Digest: Climate change and agricultural intensification influence the strength and direction of natural selection in tree swallows
How does environmental heterogeneity affect natural selection on tree swallow nestlings? Houle et al. (2020) show that more precipitation and higher temperatures result in stronger selection on body mass and wing length and that agricultural intensity can affect the direction of selection. These findings raise the question of how genetic diversity changes under strong selection pressures, which will be especially important under ongoing agriculture intensification and climate change.</p
Smart farming technology trends: Economic and environmental effects, labor impact, and adoption readiness
Farming faces challenges that increase the adverse effects on farms’ economics, labor, and the environment. Smart farming technologies (SFTs) are expected to assist in reverting this situation. In this work, 1064 SFTs were derived from scientific papers, research projects, and industrial products. They were classified by technology readiness level (TRL), typology, and field operation, and they were assessed for their economic, environmental, and labor impact, as well as their adoption readiness from end‐users. It was shown that scientific articles dealt with SFTs of lower TRL than research projects. In scientific articles, researchers investigated mostly recording technologies, while, in research projects, they focused primarily on farm management information systems and robotic/automation systems. Scouting technologies were the main SFT type in scientific papers and research projects, but variable rate application technologies were mostly located in commercial products. In scientific papers, there was limited analysis of economic, environmental, and labor impact of the SFTs under investigation, while, in research projects, these impacts were studied thoroughly. Further, in commercial SFTs, the focus was on economic impact and less on labor and environmental issues. With respect to adoption readiness, it was found that all of the factors to facilitate SFT adoption became more positive moving from SFTs in scientific papers to fully functional commercial SFTs, indicating that SFTs reach the market when most of these factors are addressed for the benefit of the farmers. This SFT analysis is expected to inform researchers on adapting their research, as well as help policy‐makers adjust their strategy toward digitized agriculture adoption and farmers with the current situation and future trends of SFTs.</p
Rapid Identification of Mycoplasma bovis Strains from Bovine Bronchoalveolar Lavage Fluid with Matrix-Assisted Laser Desorption Ionization-Time of Flight Mass Spectrometry after Enrichment Procedure
Mycoplasma bovis is a leading cause of pneumonia in modern calf rearing. Fast identification is essential to ensure appropriate antimicrobial therapy. Therefore, the objective of this study was to develop a protocol to identify M. bovis from bronchoalveolar lavage fluid (BALf) with matrix-assisted laser desorption ionization-time of flight mass spectrometry MALDI-TOF MS and to determine the diagnostic accuracy in comparison with other techniques. BALf was obtained from 104 cattle, and the presence of M. bovis was determined in the following three ways: (i) rapid identification of M. bovis with MALDI-TOF MS (RIMM) (BALf was enriched and after 24, 48, and 72 h of incubation and was analyzed using MALDI-TOF MS), (ii) triplex real-time PCR for M. bovis, Mycoplasma bovirhinis, and Mycoplasma dispar, and (iii) 10-day incubation on selective-indicative agar. The diagnostic accuracy of the three tests was determined with Bayesian latent class modeling (BLCM). After 24 h of enrichment, M. bovis was identified with MALDI-TOF MS in 3 out of 104 BALf samples. After 48 and 72 h of enrichment, 32/104 and 38/100 samples, respectively, were M. bovis positive. Lipase-positive Mycoplasma-like colonies were seen in 28 of 104 samples. Real-time PCR resulted in 28/104 positive and 12/104 doubtful results for M. bovis The BLCM showed a sensitivity (Se) and specificity (Sp) of 86.6% (95% credible interval [CI], 69.4% to 97.6%) and 86.4% (CI, 76.1 to 93.8) for RIMM. For real-time PCR, Se was 94.8% (CI, 89.9 to 97.9) and Sp was 88.9% (CI, 78.0 to 97.4). For selective-indicative agar, Se and Sp were 70.5% (CI, 52.1 to 87.1) and 93.9% (CI, 85.9 to 98.4), respectively. These results suggest that rapid identification of M. bovis with MALDI-TOF MS after an enrichment procedure is a promising test for routine diagnostics in veterinary laboratories.</p
From Extra Virgin Olive Oil to Refined Products : Intensity and Balance Shifts of the Volatile Compounds versus Odor
To explore relationships between the volatile organic compounds (VOCs) of different grades of olive oils (OOs) (extra virgin olive oil (EVOO), refined olive oil (ROO), and pomace olive oil (POO)) and odor quality, VOCs were measured in the headspace of the oils by proton transfer reaction quadrupole ion guide time-of-flight mass spectrometry. The concentrations of most VOCs differed significantly between the grades (EVOO>ROO>POO), whereas the abundance of m/z 47.012 (formic acid), m/z 49.016 (fragments), m/z 49.027 (fragments), and m/z 115.111 (heptanal/heptanone) increased in that order. Although the refined oils had considerably lower VOC abundance, the extent of the decline varied with the VOCs. This results in differences in VOCs proportions. The high VOC abundance in the EVOO headspace in comparison to ROO and POO results in a richer and more complex odor. The identified C5-C6 compounds are expected to contribute mainly to the green odor notes, while the identified C1-C4 and C7-C15 are mainly responsible for odor defects of OOs. Current results reveal that processing strongly affects both the quantitative and relative abundance of the VOCs and, therefore, the odor quality of the various grades of OOs.</p
