pH-Controlled Coacervate-Membrane Interactions within Liposomes
Membraneless organelles formed by liquid-liquid phase separation are dynamic structures that are employed by cells to spatiotemporally regulate their interior. Indeed, complex coacervation-based phase separation is involved in a multitude of biological tasks ranging from photosynthesis to cell division to chromatin organization, and more. Here, we use an on-chip microfluidic method to control and study the formation of membraneless organelles within liposomes, using pH as the main control parameter. We show that a transmembrane proton flux that is created by a stepwise change in the external pH can readily bring about the coacervation of encapsulated components in a controlled manner. We employ this strategy to induce and study electrostatic as well as hydrophobic interactions between the coacervate and the lipid membrane. Electrostatic interactions using charged lipids efficiently recruit coacervates to the membrane and restrict their movement along the inner leaflet. Hydrophobic interactions via cholesterol-tagged RNA molecules provide even stronger interactions, causing coacervates to wet the membrane and affect the local lipid-membrane structure, reminiscent of coacervate-membrane interactions in cells. The presented technique of pH-triggered coacervation within cell-sized liposomes may find applications in synthetic cells and in studying biologically relevant phase separation reactions in a bottom-up manner.</p
Within reach? Habitat availability as a function of individual mobility and spatial structuring
Organisms need access to particular habitats for their survival and reproduction. However, even if all necessary habitats are available within the broader environment, they may not all be easily reachable from the position of a single individual. Many species distribution models consider populations in environmental (or niche) space, hence overlooking this fundamental aspect of geographical accessibility. Here, we develop a formal way of thinking about habitat availability in environmental spaces by describing how limitations in accessibility can cause animals to experience a more limited or simply different mixture of habitats than those more broadly available. We develop an analytical framework for characterizing constrained habitat availability based on the statistical properties of movement and environmental autocorrelation. Using simulation experiments, we show that our general statistical representation of constrained availability is a good approximation of habitat availability for particular realizations of landscape-organism interactions. We present two applications of our approach, one to the statistical analysis of habitat preference (using step-selection functions to analyze harbor seal telemetry data) and a second that derives theoretical insights about population viability from knowledge of the underlying environment. Analytical expressions for habitat availability, such as those we develop here, can yield gains in analytical speed, biological realism, and conceptual generality by allowing us to formulate models that are habitat sensitive without needing to be spatially explicit.</p
Responses of soil microbial biomass and enzyme activity to herbicides imazethapyr and flumioxazin
The use of herbicides is important for controlling weeds in crops. However, they can present impacts on soil properties, such as biological properties. In this study, we evaluated the responses of soil microbial biomass and enzymes activity to the application of the herbicides imazethapyr and flumioxazin and their mixture in an experiment under laboratory conditions, using soils with a different history of use. Soil microbial biomass C (MBC) decreased, while microbial biomass N (MBN) was not affected after the application of the herbicides as compared to the control. Soil respiration, respiratory quotient, and dehydrogenase (DHA) activity increased significantly after the application of the herbicides compared to the control. The hydrolysis of fluorescein diacetate (FDA) was not significantly different between the control and the herbicide treatments. The principal response curve showed the largest initial effects for the flumioxazin, followed by imazethapyr and their mixture. Flumioxazin had a different influence on soil respiration and respiratory quotient than imazethapyr and their mixture. Finally, the effects of herbicides on soil microbial biomass and enzymes are short-term as we observed recovery in the biological parameters over time.</p
Ghost Introgression: Spooky Gene Flow in the Distant Past
Evolution is a continuous trial and error process in which most lineages go extinct without leaving fossil remains. Many of these lineages would be closely related and occasionally hybridized with lineages that gave rise to extant species. Hence, it is likely that one can find genetic signatures of these ancient introgression events in present-day genomes, so-called ghost introgression. The increasing availability of high-quality genome assemblies for non-model organisms and the development of more sophisticated methods for detecting introgression will undoubtedly reveal more cases of ghost introgression, indicating that the Tree of Life is even more reticulated than assumed. The presence of ghost introgression has important consequences for the study of numerous evolutionary processes, including adaptation, speciation, and macroevolutionary patterns. In addition, detailed studies of introgressed regions could provide insights into the morphology of the extinct lineage, providing an unexpected link between genomics and the fossil record. Hence, new methods that take into account ghost introgression will need to be developed.</p
Evaluación hidrogeológica de la recarga gestionada de un acuífero en un complejo de morena glacial mediante el análisis a largo plazo de datos de aguas subterráneas
Managed aquifer recharge (MAR) is a strategy to address dwindling water availability that is used to recharge stressed groundwater systems for recovery or adaptation purposes. Glacial moraine complexes can host large groundwater volumes in thick coarse-grained sandy and gravelous aquifers, and therefore are often suitable for MAR. In this research, the impact of 20 years of MAR (infiltration) in the Veluwe glacial moraine complex in the Netherlands is evaluated through time-series analysis and water quality modeling. The research enhances the understanding of hydrological processes in a glacial moraine complex and thus supports effective MAR design. The results show that MAR in the Veluwe area has raised nearby groundwater heads, and that 20 years of infiltration did not significantly deteriorate the groundwater quality. In addition, the analysis revealed an unexpected hydraulic interaction between the glacial complex and the underlying aquifer. The results suggest that MAR in a glacial moraine complex can be an effective strategy for storage of surplus surface water, compensation of groundwater abstraction, or water quality improvement. Monitoring and evaluation of groundwater heads and quality will improve the understanding of the hydrology and hydrochemistry of the water system and the used water resource, which is essential to design effective MAR systems.</p

Een kritisch-pragmatische bestuurskunde : Oxymoron of gelukkig huwelijk?
A pragmatic criticaster or a critical pragmatist is considered a schizophrenic in daily life: it seems impossible to be solution oriented and critical at the same time. You are either an optimist or a pessimist. This schism also seems to run between public administration and political scientists. Public administration is focused on (positive) problem solving, whereas political scientists – especially in a tradition of critical theory – examine the exertion of power. This essay proposes a combination of the two extremes: a critical-pragmatist approach for public administration.In this approach, critical political theory goes hand in hand with pragmatist reconstruction and design. This design is impossible without normative and procedural principles, for example ideas about sustainability, justice and democracy. This is illustrated with an example for designing just governance arrangements in the Dutch regional energy strategies. The article shows that public administration that is relevant, reflective and democratic builds on a critical-pragmatist approach
Is nutrition science ready for the twenty-first century? Moving towards transdisciplinary impacts in a changing world
Malnutrition in an obese world was the fitting title of the 13th Federation of European Nutrition Societies (FENS) conference held in October 2019. Many individuals do not eat a healthy, well-balanced diet, and this is now understood to be a major driver of increased disease risk and illness. Moreover, both our current eating patterns and the food system as a whole are environmentally unsustainable, threatening the planetary systems we depend on for survival. As we attempt to feed a growing global population, food systems will increasingly be confronted with their environmental impacts, with the added challenge of climate change-induced threats to food production. As we move into the third decade of the twenty-first century, these challenges demand that the nutrition research community reconsider its scope, concepts, methods, and societal role. At a pre-meeting workshop held at the FENS conference, over 70 researchers active in the field explored ways to advance the discipline’s capacity to address cross-cutting issues of personal, public and planetary health. Using the world cafe method, four themed discussion tables explored (a) the breadth of scientific domains needed to meet the current challenges, (b) the nature and definition of the shifting concepts in nutrition sciences, (c) the next-generation methods required and (d) communication and organisational challenges and opportunities. As a follow-up to earlier work [1], here we report the highlights of the discussions, and propose the next steps to advance responsible research and innovation in the domain of nutritional science.</p
