Molecular Amplification and Cell Culturing Efficiency for Enteroviruses’ Detection in Cerebrospinal Fluids of Algerian Patients Suffering from Meningitis
Get PDFEnteroviruses (EVs) represent a major cause of viral meningitis, being responsible for nearly 1 billion infections each year worldwide. Several techniques were developed to obtain better diagnostic results of EV infections. Herein, we evaluated the efficiency of EV detection through isolation on both Rhabdomyosarcoma (RD) and Vero cell line cultures, conventional reverse transcription-polymerase chain reaction (RT-PCR) and real-time RT-PCR. Thus, 50 cerebrospinal fluid (CSF) samples belonging to patients suspected to have viral meningitis in northern Algeria were collected, anonymously numbered from 1 to 50 and subjected to the above-mentioned techniques for EV detection. Using real-time RT-PCR, 34 CSF samples were revealed to be positive for viral origin of meningitis (68%). Thirteen of them were positive when the conventional RT-PCR was used (26%), and only three samples gave positive results when the cell culture technique was used (6%). Surprisingly, two cell culture-positive CSF samples, namely, 31 and 39, were negative using RT-PCR directly on the original samples. However, they turned to be positive when amplification was carried out on their corresponding cell culture supernatant. The cell-cultured viral isolates were then identified by sequencing their viral genome’s VP1 regions. All of them were revealed to belong to the echovirus 27 strain. This investigation demonstrates that RT-PCR techniques are often more sensitive, accurate and much faster, providing reliable results within a clinically acceptable timeframe. Ce travail de fin d'étude s'inscrit dans le cadre de l'obtention du diplôme de Licence Professionnalisante en Génie Formulation. Il est basé sur un stage effectué au sein de l'unité SAIDAL-CHERCHELL, où l'on a eu l'opportunité de découvrir le milieu industriel et de consolider ses connaissances théoriques sur la fabrication et le contrôle de la qualité du médicament LAMIDAZ® 250 mg. L'objectif du rapport est de détailler le processus de fabrication du médicament ainsi que les contrôles effectués pour garantir sa qualité.
Le contrôle de qualité repose sur la conformité du produit avec la Pharmacopée Européenne, notamment les analyses physico-chimiques et les méthodes d'appareillage de laboratoire utilisées. L'étude a démontré que LAMIDAZ® 250 mg respecte les normes de sécurité et de conformité exigées, et les résultats obtenus montrent que ce médicament répond aux critères de qualité définis par la Pharmacopée Européenne 2014.

This thesis is part of the final project for obtaining a Bachelor's degree in Professional Formulation Engineering. It is based on an internship at the SAIDAL-CHERCHELL unit, where the opportunity was provided to explore the industrial environment and consolidate theoretical knowledge on the manufacturing and quality control of the drug LAMIDAZ® 250 mg. The objective of the report is to detail the manufacturing process of the drug and the controls implemented to ensure its quality.
Quality control relies on the product's compliance with the European Pharmacopoeia, particularly the physicochemical analyses and the laboratory equipment methods used. The study demonstrated that LAMIDAZ® 250 mg meets the required safety and compliance standards, and the results show that this drug meets the quality criteria defined by the European Pharmacopoeia 2014.

يعد هذا العمل جزءًا من مشروع التخرج للحصول على درجة البكالوريوس في الهندسة الصيدلانية. ويستند إلى فترة تدريب قضاها المؤلف في وحدة SAIDAL-CHERCHELL، حيث أتيحت له الفرصة لاكتشاف البيئة الصناعية وتعزيز معرفته النظرية حول تصنيع ومراقبة جودة الدواء LAMIDAZ® 250 ملغ. يهدف التقرير إلى توضيح عملية تصنيع الدواء والضوابط المتبعة لضمان جودته.

Au total, Dix-huit (18) éprouvettes de sections droites de (10×15) cm2 et une longueur de 60 cm, (03) éprouvettes non renforcées (poutre témoin), et (15) éprouvettes renforcées de fibres métalliques,
Cinq (05) éprouvettes renforcées avec la même fraction volumique de 1,5% de fibres, et avec différentes dispositions par couches sont considérées. Les différentes configurations considérées sont :
H : Poutre en BAF (P-HT)
H/2 : Poutre mi-hauteur (PH/2)
a 70%&b30% : Poutre multi couche (P- a70%b30%)
a 80%&b20% : Poutre multi couche (P- a80% b20%)
a100%&b0% : Poutre multi couche (P- a100%b 0%)
Les résultats de l'essai de flexion 4- points ont été présentés sous forme de graphes et courbes et analysés. Une confrontation est menée pour mettre en évidence l’intérêt de notre proposition de répartition des fibres métalliques par couches considérées de la poutre
Conception et réalisation d’une serre agricole intelligente
Get PDFCe mémoire de notre projet a pour objectif de concevoir et de mettre en oeuvre un système d’automatisation et de contrôle à distance via Blynk pour les serres agricoles, dans le but de réduire les efforts des agriculteurs et d'améliorer la productivité, tant en quantité qu'en qualité. Le projet est structuré en trois parties. La première partie consiste à élaborer un système intelligent capable de reconnaître les conditions climatiques à l'intérieur de la serre grâce aux données des capteurs, et de prendre les décisions nécessaires pour activer les moteurs afin de maintenir des conditions optimales pour les plantes. La deuxième partie se concentre sur le développement d'une interface homme-machine permettant de visualiser en temps réel les paramètres climatiques et de définir des seuils à ne pas dépasser dans la serre. Enfin, la troisième partie a permis la création d'un prototype de serre, nous permettant de tester et de valider les performances et le bon fonctionnement du systèm
