Isolation, Identification and Characterization of the Surface Glycoprotein of Myxobolus koi
Myxobolus koi is considered to be one of the pest diseases of fish quarantine class I, which has to be destroyed if it is found. Technique that is regularly used for diagnosis M. koi, such as histopathological examination, polymerase chain reaction (PCR) and sequencing always done in dead fishes. The objective of this study was to isolate the protein of M. koi as the initial step for producing antibody that can be used as a material for immunology-based diagnostic method. In the present study. identification of surface protein of M. koi involved three different stages. The first stage was to isolate and identify M. koi from koi fish using wet mount, and histopathologic, PCR and sequencing techniques. In the second stage, purification and isolation of the surface protein of M. koi was done by Percoll gradient ultracentrifugation technique. Purification using the technique produced 5 different layers on the surface of the tube . Myxobolus koi spores were found in the 2nd, 3rd and 4th layers. The third stage was to identify and characterize the surface protein of M. koi using SDS PAGE. The results of SDS PAGE analysis showed that spores purified from 3 different layers all have the same profiles. Molecular sizes of the surface protein of M. koi that could be isolated in this study were 12 Kd, 16 Kd, 25 Kd, 27 Kd, 40 Kd and 45 Kd. Optimization of Tissue Decellularization Method Based on Macroscopic and Microscopic Observation in The Sheep Peripheral Nerves
Animal graft tissue (xenograft) was developed to replace the limited supply of human graft tissue (allograft). Peripheral nerve graft tissue is needed to replace the damage. Swine is the most developed source of tissue donor for the preparation of acellular tissue because a lot of livestocks population and its extracellular matrix components similar to human collagen. Swine xenograft development would be an obstacle in Indonesia because of socio-cultural so that sheep used as swine donor replacement. Sheep cartilage tissue acellular induced less human inflammatory mediators than swine. Xenograft is necessary to decellularizebefore implantation into human that results in only extracellular matrix. However, decellularization process varies depending on the species and methods so that it is needed a preliminary study to get the best decellularization method for sheep peripheral nerve specimens. Five samples of sheep Ischiadicus nerves were conducted decellularization various processes and one sample of fresh nerve (control). Decellularization methods were 24 hours shaking, tissue soaking and perfusion for both 14 days and 17 days, respectively. Decellularization solution was 0.1% SDS-EDTAin PBS. Post decellularization, samples were observed macroscopically, fixed, HE staining of histopathologic examination for microscopic examination. The data were analyzed descriptively. At the macroscopic observation of post decellularization tissue showed white, and samples of soaking method showed softer consistency than that of shaking and perfusion. Microscopic examination showed that samples were not being completed decellularization at shaking method. There was destruction of collagen fibers of perineurium and endoneurium in 17 days of soaking and perfusion methods, while 14 days soaking and perfusion methods showed that samples were being completed decellularization and both perineurium and endoneurium collagen were still good. Conclusion of this study is that 14 days soaking and perfusion decellularization methods are better than that of 24 hours shaking method and 17 days of soaking and perfusion methods for sheep decellularization peripheral nerve.
Reduction of Intestinal Motility and Contractility in The Cats Anethetized with Tiletamine-Zolazepam Through Contrast Radiography Study
Intestinal motility and contractility in cats can be affected by many factors that originate from inside and outside the body. Anesthesia is one of the factors that could influence it.This research was conducted to figure the effect of tiletamine-zolazepam on the intestinal motility and contractility in the cats through contrast radiography study. Three male cats weighing between 2,5-3,5kg were used in this present study. Radiography procedure was performed twice on each cat: without anesthesia and one week later with anesthesia (0,02 mg/BW atropine sulphate and 10 mg/BW Zoletil®).The cats were given a positive contrast, barium sulphate (BaSO4) 30% w/v (12 ml/kg BWper oral) followed by radiography study. Radiography was performed at 5, 30, 60, 120, and 180 minutes after administration of BaSO4. The cats were positioned on laterolateral view. Data were analyzed descriptively using zone division and quantitatively by measuring the difference between the intestines diameter during contraction and relaxation. The intestinal diameter was measured and statistically tested using ANOVA. Result showed that in minute 180, BaSO4 reached zone four in the unanesthetized cat while in the anesthetized cat, it was still empty. The diameter differences of the unanesthetized cat was higher compared to the anesthetized meaning that the intestinal contraction in the unanesthetized cat was higher. DISTRIBUSI Culicoides spp. (DIPTERA: CERATOPOGONIDAE) PADA PETERNAKAN AYAM PETELUR DI KABUPATEN SLEMAN YOGYAKARTA = A DISTRIBUTION OF Culicoides spp. (Dag bRA: CERATOPOGONIDAE) ON THE LAYER POULTRY FARMS IN SLEMAN REGEN
Telah diteliti distribusi spesies Culicoides yang ada di kitar peternakan ayam petelur di Kabupaten Sleman Daerah Istimewa Yogyakarta. Penelitian ini ditujukan untuk mengetahui spesies-spesies Culicoides yang mempunyai peranan dalam menyebarkan penyakit leukositozoonosis pada ayam petelur. Sebanyak 762 ekor Culicoides (Diptera: Ceratopgonidae) dikumpulkan dari dua belas peternakan ayam petelur di wilayah Kabupaten Sleman dengan menggunakan perangkap serangga Pirbright type rniniatur light trap. Culicoides yang diperoleh dikelompokkan dan diidentiflkasi berdasarkan karakter morfologis menurut Wirth den Hubert. Basil penelitian menunjukkan tidal( ada perbedaan populasi Culicoides dari petemakan ayam petelur yang ada di wilayah Kabupaten Sleman pada dataran rendah maupun dataran tinggi. Ada delapan spesies Culicoides yang diperoleh dari petenakan ayam petelur di Kabupaten Sleman yang berhasil diidentifikasi yaitu: C. lye, C. guttifer, C. arakawae, C. oxystoma, C. parahumeralis, C. peregrinus, C. sumatrae dan C. clavipalpis. Spesies Culicoides yang dicurigai sebagai penyebar penyakit leukositozoonosis di Kabupaten Sleman adalah C. huffi, C. guitifer , dan C.arakawae
HUBUNGAN KONSENTRASI MALONDIALDEHIDA, GLUKOSA DAN TOTAL KOLESTEROL PADA TIKUS PUTIH YANG DIINJEKSI DENGAN STREPTOZOTOCIN
Diabetes mellitus is a major source of morbidity in developed countries. The morbidity and mortality areincreased in patients with diabetes mellitus compared with the general population. Relationship between highsugar or fat consumption with the increasing of glucose or blood cholesterol concentration has been wellunderstood, but the mechanism of diabetes mellitus as a result of streptozotocin injection and its relation withglucose and cholesterol concentration still need to be investigated. So far, many research mention thatstreptozotocin causes only the damage of a cell of pancreas which that directly relates to metabolism of bloodsugar. The purpose of the research is to investigate the mechanism of pancreas damage in its relation with lipidperoxidation. Sixteen male Wistar rats, 200 grams of bodyweight were used as animal models. Rats weredivided into two groups of eight animals each. Group I was not injected with streptozotocin, while Group II isinjected intraperitoneally with streptozotocin 50 mg/kg BW.Three days after the injection, blood samples weretaken intracardially for Malondialdehida (MDA), glucose and total cholesterol analysis. Kajian Penanda Genetik Gen Cytochrome B Pada Tarsius sp. =Study of Genetic Marker on Cytochrome B Gene of Tarsius sp.
Tarsius merupakan salah satu satwa endemik Indonesia yang keberadaannya mulai memprihatinkan. Konservasi sebagai salah satu cara untuk pelestarian satwa ini akan lebih terarah dan berhasil guna apabila karakteristik dan keragaman sumber genetiknya diketahui dengan pasti. Tujuan dari penelitian ini adalah mengkaji penanda genetik spesifik gen cyt b pada Tarsius sp. Pengurutan hasil PCR menggunakan primer H 15149 pada gen cyt b didapatkan urutan basa sebesar 276 pb (menyandi 92 asam amino. Fragmen cyt b hash! pengurutan disejajarkan berganda dengan primata lain dari data Genbank dengan bantuan perangkat lunak Genetyx-Win versi 3.0 dan Clustal W, kemudian dianalisis dengan menggunakan program MEGA versi 3.1. Dari hasil analisis diperoleh 14 situs asam amino yang berbeda. Tarsius dianae memiliki 12 situs asam amino (asam amino ke 2, 6, 9, 22, 23, 29, 39, 41, 42, 45, 55 dan 85), T. spectrum memiliki 7 situs asam amino (asam amino ke 2, 6, 9, 41, 45, 55 dan 85) dan T bancanus memiliki 2 situs asam amino ( ke 23 dan 45) yang dapat digunakan sebagai penanda genetik. Lima asam amino unik ditemukan pada T dianae, yaitu pada situs asam amino ke 6 (valina), ke 22 (alanina), ke 29 (alanina), ke 39 (serina) dan ke 42 (valina). Jarak genetik berdasar nukleotida cyt b yang dihitung menggunakan model 2 parameter Kimura ditemukan nilai paling kecil sebesar 0,7%, nilai paling besar 22,3% dan rata-rata 13,1%. Filogram menggunakan metode neighbor joining berdasar hasil urutan nukleotida dan asam amino cyt b tersebut dapat dijadikan pembeda masing-masing spesies Tarsius

Penelitian ini bertujuan untuk mengetahui pengaruh pemberian rumput laut terhadap kualitas sperma tikes akibat pencemaran timah hitam. Dalam penelitian ini digunakan 12 ekor tikus putib jantan dewasa, dibagi secara acak menjadi 4 kelompok masing-masing 3 ekor. Kelompok I, tikus-tikus hanya diberi akuades secara oral. Kelompok11,111 dan IV, semua tikus diberi Pb asetat 0,5 g/kg bb secara oral setiap hart selama 120 hart. Kelompok DI dan IV masing-masing disertai dengan pemberian larutan rumput laut dosis 8 g/kg dan 16 g/kg bb secara oral pe, hari selama 120 hart_ Pada akhir penelitian masing-masing films dart semua kelompok dikoleksi spermanya untuk dilakukan pemeriksaan terhadap motilitas, viabilitas dan konsentrasi apermanya. Semua tikus percobaan kemudian dietanasi dan dinekropsi, organ testis diambil dan difiksasi dalam ICJ% formalin untuk preparasi histopatologis.
