    The Effect of Turmeric (Curcuma Domestica Val.) Extract on the Histopathological Lesions of Lung Induced by Cigarette Smokes in Wistar Rats

    This present study is aimed to observe and determine the effects of turmeric (Curcuma domestica Val.) extract administration towards the cytotoxicity effect of cigarette smokes in the lungs of Wistar rats. Twenty Wistar rats, male, 3 months old, were used and divided randomly into four groups, of five each. Group I is as a control (K I), Group II was given 9 cigarettes/day without turmeric extract administration (K II), Group III giventurmeric extract 1 ml 0.25%/day (K III) and Group IV was given 9 cigarettes/day and turmeric extract 0.25%/day (K IV) for 60 days. At the end of the present study, all rats were necropsied and organs, particularly the lungs were collected, processed histopatologically and stained routenely with hematoxilyn eosin. The results indicated that the lungs had macrophages infiltration in the septal alveoli and hyperplasia of bronchial associated lymphoidtissue (K II). Whereas, the lungs in the rats in Groups K I, K III and K IV were still within the normal limits. It is concluded, that turmeric extract may have a potential antipulmonary cytotoxity effect due to cigarette smoke

    Estrogen is an inhibitor of bone resorption. Although there is some disagreement, most studies indicated that bone loss is accelarated at the time of menopause wether it is induced surgically or naturally. When the condition men­tioned above is supported by imbalance diet, especially calcium to phosphor ratio, the bone loss will be acce­larated. The obyectlves of the research were to study the effect of panhisterectomized and high phosphorous intake on calcium and phosphate anorganic concentration in the blood, and bone changes in rats.Sixteen female of rats ± 30 days of age were randomly divided into four groups (A, B. C, and DJ of four each. The group A and C were unpanhisterectomized. Whereas the group B and D were panhisterectomized. The group A and B were fed diet contain Ca:P ratio = 1.5:1, whereas the ratio of calci­um and phosphorus 1.5:6 was given to group C and D. At the end of the experiment, the blood samples were taken directly from eye (plexus retroorbitalis) for calcium and phosphate analysis. The left femur were taken for histopathological analysis.From the results can be concluded that the diet con­taining the Ca:P in variation ratio, panhistcrectomized. and interaction between two of them had significant effect on the blood inorganic phosphate (P < 0,01). The diet contain­ing CaiP = 1.5:6 for unpahisterectomized caused earliest lesion of osteodistrphia fibrosa, whereas for panhisterec-tomized caused mild osteodistrophia fibrosa

    Coccidiosis is one of the problems in poultry husbandry since it causes a lot of loss. Nowadays, several ionophore coccidiostats have been used to prevent coccidiosis. However, because of some complaints for ionphore coccidios­tats usage, especially Monensin which effect to the weight of digestive tract, therefore the use of coccidiostats should be taken with care. This investigation is intended to undestand the Increase of weight and some changes in diges­tive tract because of the use of 4 ionophore coccidiosis.This investigation used 52 Hubbard broiler of 1 days age. After 2 weeks of adaption 4 broiler were taken randomly as a control group (without coccidiostat). and the remaining 46 broilers were divided randomly into A group, that is. Monensin (Mo). Karacin (Na). Salinomicin (Sal), and Madura­micin (Mad)- The Mo group was divided futher into 3 sub­groups, those are Mo 70, Mo 110, and mo 130 that were fed containing Monensin 70, 110. and 130 mg/kg feed. The other groups were also divided similarly into 3 subgroups, those are, Na 50, Na 70. Na 90; Sal 40, Sal 60, Sal 80; and Mad 4, Mad 5, Mad 6. Number after the abbreviation of coccidiostats indicated the amount of mg/kg reed. The coccidiostats were used to the broilers since 14 until 45 days old. and finally all of the broilers were Killed 4 days later.Using variance analysis with split plot pattern showed that all of the coccidiostats did not effect to the weight growth of broilers (the weighting of broilers was carried out every week since 14 until 49 days old). The use of orthogonal polynomial contrast test indicated that the Monensin coccidiostat effected to the index (the related organ weight/the total weight x 100%) of praventriculus to Cloaca by an equation of VA = 26,9736 - 0,4602 X + G,0024X^P therefore the suggested dosage is 96,53 mg/kg feed. For the Maduramicin coccidiostat. besides it effected to the index of proventriculus-to cloaca by an equation of VR = -12.790 + 7,461 X - 0.696 X*. it also effected to the inffex of duode­num to cloaca by an equation of yc = -5,910+3,585 X - 0.315 X * therefore the suggested dosage is 4 mg/kg feed. The use of Naracin and Salinomicin did not influence to the Index of tractdigestive. All of the coccidiostats did not effect on the thickness of duodenum, jejunum, and ileum. covering diameter (by macroscopic measurement), also the indexes of epithelium/villi weight, mucosa layer thickness, and mus-culer layer thickness that measured by microscopic method-From this investigation can conlcuded that the use of Monensin in 96,53 mg/kg feed. Naracin In 50-90 mg/kg feed, Salinomicin in 40-80 mg/kg feed, and Maduramicin in 4 mg/kg feed will be safe, which means that they willnot increase the weight of tractusgastrointestine

    HISTOLOGIC PICTURE OF BEEF DIGESTED BY PAYA PAPAIN ENZYME

    The study was conducted to show the histologic picture of beed digested by different concentration of PAYA papain enzyme. Twenty pieces of beed of m. femoralis, 1 x 0.5 x 0.5 cm in size, were allocated into four groups. Group I was injected with aquadest as control; group II, III, dan IV were injected with PAYA 0.25%, 0.50%, and 0.75% of the weight of meat, respectively. The samples were heated in a waterbath 65°C to activate the enzyme for 0, 15, 30, 45, and 60 minutes. Samples were then put into 10% formalin solution to stop the enzyme. The were stained with haematoxyline-eosine (HE) for histologic picture. The result shows PAYA papain enzyme starts to disintegrate myofibril in the con­centration of 0.50% for 30 minutes. The higher is the con­centration, the more do changes of myofibril take place in the form of inflamation, disintegration, and lysis

    A study was conducted to find out bacteria which probably play an important role in causing diarrhea in dog at the veterinary clinic Gadjah Mada University. Sample were collected from diarrhea cases of dog, during the periode of September to December 1997, by rectal swabbing. Samples were then sent to the laboratory for bacterial isolation and identification.The result indicated that E. coli was found from 50% of the sample, Streptococcus sp 41.9%, Staphylococcus sp 33.3% of sample, Enterobacter sp 16.7%, Bacillus sp 8.3%.Some of E. coli isolates were pathogenic to mice. The pathogenic E. coli were resistant to penicillin, ampicillin, streptomycin, erythromycin, doxycycline, besides sensitive to neomycin, gentamycin, kanamycin, polymyxin B, chloram-phenicol and sulfisoksale

    The most immunogenic and protective fraction of outer membrane proteins of Pasieurella multacida has not well understood. This study was done to find our the immunogenicity and protectivity of outer membrane protein fractions of Pasteurella multicida isolat U-6 in mice. The organism was cultured in Luria Bertani broth. The outer membrane proteins were prepared according 10 Snipes et al., (1988), Electrophoresis of the protein was carried out in 10% of sodium dodecy 1-su I ph ale poly cry 1 amide gel electrophoresis, and stained with 0.3% Coomasie brilliant blue. The result indicated that there arc many protein bands with 5 major protein bands, those are having molecular weight of 20,1 kilo Dalton (kD), 27 kD, 36 kD, 52,7kD,and75 kD. Each protein bandof molecular weight of 27 kD, 36kD,and75kD was then cut, and homogenised, and was used as antigen to immunise the mice after added with adjuvant 1:1. Two weeks after immunisation all of the mice were bled and the sera were tested by ELISA. FourtyeigM hours from bleeding, ail of the immunised and control mice were challenged with 500 colony forming unit of virulent Pasteurefla mutticida isolate U-6, and observed their mortality. The result indicated thai the outer membrane protein bands of molecular weight of 36 kD and 75 kD were immunogenic (P>0,05), while the outer membrane protein band of 27 kD was less immunogenic (P>0.05), Band with moleculer weight of 27 kD produced 0% protection, bandof 36 kD produced 60% protection, and band of 75 kD produced 40% protection of mice challenged

    Skunks experimentally infected with street virus rabies intramuscularly were used to demonstrate rabies viral antigen in paraffin-embedded tissue by immunopathology diagnosis approach, the avidin biotin complex (ABC) method. The use of ABC method provides a simple and sensitive method to localize rabies virus antigen in formalin- fixed tissues. The results of the present study also demonstrated that a procedures using formalin-fixed parafin-embeded tissue and stained immunopathologicalH by the ABC method was an excellent method for both preservation of morfological details of ceils/tissues and demonstration of rabies virus antigen

    ABSTRACT              The aim of the study was to know the influence experimental infection of Edwardsiella tarda  was isolated from sick koi fish (Cryprinus caprio) to the histologic features of liver, kidney, gill, brain, intestine and muscle in Goldfish (Carassius auratus) based on Koch Postulate. Three goldfishes (KK) were used in this reseach. KK was infected by Edwardsiella tarda as much as 0.1ml i.m.. KK1 died on the 4th day and KK2 and KK3 died on the 25th day. All the fishes were then necropsied. The livers, kidneys, gills, brains, intestines and muscles were taken for making the histopathologic preparation. Bacteriologic observation of KK1 was conducted by cultivating the bacteria into Brillliant Green Agar (BGA), Triple Sugar Iron Agar (TSI Agar), Urea Agar, Motility Agar, Peptone Broth, sugars and oksidation test.      Result of histopathologic observation showed that there was necrosis, congestion and macrofag limpocytes and neutrophisl infiltrations in the livers,colloid mass and melanomacrofages in the kidney, congestion and proliferation of secondary lamellae, limfosit and macrofag infiltration of the brain, a melanomacrofag of the muscle and the intestine showed there was limphocytes, and macrofages infiltration in the brain, and melanomacrofages in the muscles as well. It was suspected that those bacteria was  Edwardsiella tarda.  

    Validation of the Spectrophotometry method on the concetration assessment of Enrofloxacine as the Animal Drug

    Abstract Validation for analysis of enrofloxacin injection method using spectrophotometry has been conducted. Enrofloksasin in a solution of NaOH (sodium hydroxide) 0.1 N, measured at the maximum wavelength (ë) 271 nm.The parameters of validation that were carried out are precision, accuracy, linearity, limit of detection and limit of quantitation. Precision showed good results with coefficien variation (CV) results smaller than that of CV Horwitz. Accuracy parameters calculated by the method of analysis to determine levels of analyte in a known active ingredient purity and yielded 0.449%. Linearity parameters showed good results with r = 0.9996. Limit of detection value was obtained for 0.757 μg / mL and limit of quantification of 2.522 μg / mL. The results of validation test of enrofloxacin injection analysis using spectrophotometric method can be used as a good method to provide a valid test of quality assurance veterinary drugs

    The Effect of Red Fruit's (Pandanus conoideus) Extract to Eritrocytes, Hemoglobin, PCV and TPP Concentrations of Mice (Mus musculus) Infected by Toxoplasma gondii

    Toxoplasmosis is a zoonotic parasites caused by Toxoplasma gondii. The red fruit’s extract contains tocopherol and betacarotene as antioxidant and it can repair the bodies immune system. This research was aimed to study the effect of red fruit’s extract to erithrocytes, hemoglobine, the values of PCV and TPP concentration of mice infected with T. gondii, so that the red fruit extract can be used as alternative herbal medicine for eliminating Toxoplasmosis. 15 of Balb/C mice, male, three months, 30 gr of body weight were used as experimental animals. The mice were then devided into three groups. Group I was a control, group II  was tachyzoite infected and group III was tachyzoite infected and given red fruit’s extract. Group III given red fruit’s extract on first day with 0,53 g/kg BW/mice/day. Mice in group II and III were infected by injection Toxoplasma gondii tachyziote 1x103 a mice intraperitoneally. The blood sample was taken from five mice in each group at 7th and 12th day pass through cantus medialis. The blood was examinated in Clinical Pathology Laboratory, Faculty of Veterinary Medicine, Gadjah Mada University. Data were analyzed by using Factorial 2 x 3. The result showed no significant difference (P>0,05) in hemoglobin, and PCV’s value between groups I, II and III before and after infection of T. gondii’s tachyzoite. The TPP’s value between groups I, II and III before and after infection of T. gondii’s tachyzoite was significantly difference (P<0,05)
