IDENTIFICATION OF SERPTICAEMIA EPIZOOTICA (SE) ON SWAMP BUFFALOES (KALANG BUFALLOES) IN SOUTH KALIMANTAN
Identification of Septicaemia Epizootica (SE) was carried out on swamp buffaloes at the districts of Hulu Sungai Tengah (HST) and Hulu Sungai Utara (HSU) and those which were slaughtered in the abbatoars of Banjarmasin, Banjarbaru and Amuntai, South Kalimantan. This study was conducted to find out whether Septicaemia Epizootica was present among the swamp buffaloes population. Thirty two samples of swab nasopharynx from the field (HSU and HST) and 60 samples from the abbatoars were collected and checked toward Pasteurella multocida. And 45 samples of sera were examined for antibody with Elisa method. Based on bacteriological examination there was no Pasteurella multocida could be identified from the samples and from Elisa examination was also negative from Septicaemia Epizootica

CALCULATION OF % PROGESTERONE IN SKIM MILK FRACTION WHEN CENTRIFUGATION TEMPERATURE AND % BUTTERFAT OF WHOLE MILK ARE KNOWN
Air susu dikumpelkan dari 70 fkor sapi perah, Departemen Animal Science, Michigan State University, USA-, sampel-sampel tersebut diperlakukan terhadop berbagai temperatur perlakuan, yaitu : 0.10 C : I 0C : 30 C : 50 C : 100 C : 200 C : 300 C dan kontrol Periode inkubasi dilakukan 10 jam. Hasil dari studi ini menunjukkan bahwa dari grafik kalkulasi persentase progesteron pada skim milk apabila temperatur sentrifugasi dan kadar lemak susu diketahui maka diperoleh hasil bahwa semakin tinggi kadar lemak ternyata persentase progesteron dalam skim milk akan menurun. Dapat diketahui pula bahwa semakin tinggi temperatur, kadar progesteron dalam skim milk akain menurun sejajar dengan peningkatan kadar lemak
North Kalimantan Is Free from EMS (Early Mortality Syndrome) in the Shrimps
The acute hepatopancreatic necrosis syndrome (AHPNS) or also known worldwide as early mortility syndrome (EMS) is very dangerous for the shrimps industry since it really strikes the shrimps by larvae size. The EMS attacks shrimps at 20 to 30 days of age and at 40 days of age as well. All the shrimps that are being infected will be dead righ away. The EMS has caused the significant economic losses in the shrimps industry due to the mass mortality of the shrimps in Vietnam (in the years of 2010- 2012). It also occurred ad spread around China (2009), Vietnam (2010), East Malaysia (2010), East of Thailand (2012) until approaching the edge of Cambodia (2013). It has been reported that the EMS is likely due to Vibrio parahaemolitycus that are infected by certain virus (phage), and then the bacteria releasing very toxic substances (toxin). Efforts has been being done by the Indonesian government, especially in the Fish Quarantine Office, Kls II, Tarakan in the form of prohibition of entry of live and/or frozen shrimps importing from the EMS-outbreak country. Besides that, monitoring and laboratory works has also been performed routinely and periodically. The results proves thatNorth Kalimantan is free of AHPNS (EMS)
Escherichia coli Surface Characters of Ettawah Cross Breed Goats Milk on the Adhession Ability of Mammary Epithelial Cells
Escherichia coli is a bacteria that may cause mastitis in goats. The ability of bacteria to infect is influenced by surface characters of each bacterial cell. Adhesion of bacteria on the udder epithelial cells plays an important role in the incidence of subclinical mastitis.The purpose of this study was to determine the surface characters of Escherichia coli colony morphology, such as the presence of polysaccharides capsule, hemagglutination activityand hydrophobicity contributes to the adhesion process. Three isolates of Escherichia coli isolated from Ettawah cross breed goats on Mandiri and Pangestu Farm at Turi, Sleman, Yogyakarta were used in the study. The ability of E. coli to fermentize lactose was done by examining its ability in fermented lactose on eosin methylene blue. Hemagglutination ability was seen by hemagglutination test using sheep erythrocytes at concentrations of 0.5%, 1%, 1.5% and 2%, respectively. Hydrophobicity expression was done by saltaggregation test using ammonium sulfate ([NH4] 2SO4) at concentrations of 1.2 M, 1.6 M, 2.0 M, 2.4 M and 3.2 M, respectively. The presence or absence of sera proteins was tested with sera soft agar, and the ability of adhesion was done by adhesion of Escherichia coli which has been marked by fluorescein isothiocyanate , with rats udder epithelial cells after birth. The results showed that isolates of E. coli with dry surface characters had athin polysaccharide capsule, positive hemagglutination , and hydrophobic, which had the ability to stick more on the udder epithelial cells after birth. Meanwhile, those with mucoid surface characters, had a thick polysaccharide capsule, negative hemagglutination, and hydrophilic which had a lower adhesion ability
HORMONAL RESPONSES OF LACTATING DAIRY COWS TO TREATMENT WITH A PROGESTERONE INTRAVAGINAL DEVICE WITH OR WITHOUT ESTRADIOL
The objective of this study was to evaluate the effect of progesterone supplementation through an intravaginal device on plasma progesterone and estradiol-17(3 profiles in lactating dairy cows. Eight lactating Friesian cows, between 3 and 5 years, in the luteal phase of a normal estrous cycle were used In the treated group A (n = 4), a CIDR intravaginal implant contains 1.9 gram progesterone, was inserted into the vagina of each cow at day 8 of the study and removed 9 days later. In the group B (n = 4), a CIDR combined with a 10 mg estradiol benzoate capsule was inserted into the vagina as in the group A Blood samples were taken daily for progesterone and estradiol-17(5 assays. Plasma concentrations of progesterone increased rapidly following the CIDR insertion, reached maximum at day 9 of insertion (6.28 ± 0.26 ng/ml and 5.62 + 0.16 ng/ml, respectively, P < 005), and then dropped abruptly after CIDR removal. Two days after the CIDR insertion, the plasma progesterone increased more rapidly in the group A than in the group B, and were significantly higher between days 11 and 17 of the study, but lower between days 20 (estrus) and 22 of the study (P < 0.05). The progesterone levels reached to basal levels within 3 days following the CIDR removal (0.64 + 0.12 ng/ml and 029 + 0.16 ng/ml, respectively, P < 0.05), when the cows were showing estrous behavior. The declines in the mean plasma concentrations of progesterone after the CIDR removal in the both treated groups were linear (R2 = 0.814 and 0 798, the rate of decline were 1.36 ng/ml/day and 1.29 ng/ml/day, respectively). Thereafter, the mean progesterone concentrations elevated steadily after the induced estrus. The plasma concentrations of estradiol-17(3 in the both treated groups also showed relatively the same pattern. They declined regularly in the luteal phase, increased slowly by the follicular phase, and reached maximum levels at estrus, 3.08 + 0.12 and 338 ±0.16 pg/ml (P < 0.05), respectively. The concentrations of estradiol-17(3 between days 6 and 9, days 20 (estrus) and 21 of the study in the group A were significantly lower than those in the group B (P < 0.05). The insertion of estradiol benzoate capsule significantly increased the plasma concentrations of estradiol-17(3. At the same time, the increase of plasma estradiol-17£ concentrations was followed by the decline of plasma progesterone levels. In conclusion, CIDR is a useful agent for continuous delivery of progesterone to enable estrus synchronization in lactating dairy cows. Its combination with estradiol benzoate resulted in a more marked decline in the plasma concentration of progesterone and more synchronous estrus following its removal than that of CIDR alone
ANALYSIS OF COLFFORM BACTERIA IN THE WELL WATER AND ITS POSSIBLE BIOPATHOLOGIC EFFECTS
Coliform is a Gram-negative, rod shape bacteria Some strains, especially Escherichia coli can cause diarrhea among animals aad humans. Contamination of water whith pathogenic E. coli has been associated with this disease in animals and humans.In the present study, coliform bacteria were determined with most probable number (MPN) method cultured on lactose broth and brillian green bile broth. Results indicated that the well water samples collected from the areas of Karangmalang, Sungai Code and Deresan had 150 coliform/100 ml, 210 coliform/100 ml and 460 coliform/l00 ml, respectively. Whereas, standard number of coliform organisms in drinking water recommended by World Health Organization (WHO) is 10 coliform/100 ml and is 1 E. coli/100 ml. Therefore, based on the present study, the coliform organisms content in the well water is for above its normal level value as recommended by WHO. EFFECTS OF SILVER INDUSTRY WASTE WATER IN KOTAGEDE, YOGYAKARTA ON HAEMTOLOGlCAL AND HISTOPATHOLOGICAL CHANGES IN EXPERIMENTAL RATS (RATTUS NORVEGICUS)
Thirty male rats, 2,5 months old with average body weight of 92 grams were used as experimental animals.The experiment rats were adapted before for one week. The animal grouping was basedon uniformity of body weight gain, Thecontrol group consist of 15 animals and 15 animals for treated group. The treated group was given silver industry waste water from Kotagede, Yogyakana orally ad libitum the control group was supplied drinking tap water ad libitum. Blood samples via plexus retroorhitalis vein at medial cantus of the eye were taken for routine heamiolafiical examinations, clinical symptoms and body weight gain were also recorded. After treatment, for about 12 weeks, all experimental animals were sacrificed, necropsied, then the samples of brain, spinal cord, lung, tesiis. liver, and kidney were taken for histopathological preparations. The concentration of metals (especially Ag and Cu) in the liver, silver industry waste water, well water, Tap water, and soil samples (which [he silver industry waste water was discarded) were analyzed. The results of the experiment indicated thai the silver industry waste water administration in long period caused decreasing of the rat growth rate with clinical symptoms e.g. thinness,dullnessof the fur, anaemic, depression and nerve symptoms including foot paralysis. Statistical calculation showed there were not significantly changes on hematological profile between the control and treated groups, it could be due to severe dehydratation level occured, so there were reducing of blood volume. This finding was similar to leucocyte and differential counts, but there tended to decrease at week-12. There could be due 10 bone marrow depression which were caused by heavy metal intoxication. Microscopical examinaton showed there were thickness of alveoli septa, congestion, enlargement of sinusoids and infiltration of mononuclear cells around the liver centralvein, degeneration and necrosis of tubule cells, enlargement of tubule lumen and Bowman capsule space, infiltration of monoclear cells and haemorrhage performance. There were a few Purkinje cells showed more denses and the sTain more absorpted intensively (the same findings occuredin the nerve cells of spinal cord), it could be possible that there were cell structural changes, degeneration and necrosis of The nerve cells especially in the spinal cord tissues. The silver, copper and chrome concentrations in the silver industry waste water and soils samples were too high, it could be as potential sources for environmental pollution. The concentration of these metals in the tap water and well water were not detected yet
Stability of gag-ca gene of Jembrana Virus in pcDNA-ca recombinant plasmid vector in Escherichia coli DH5α which has been stored for eight years
The aim of this study was to determine the stability of gag-ca gene in a pcDNA-ca recombinant plasmid vector in Escherichia coli DH5α which has been stored for eight years using Polymerase Chain Reaction method (PCR). Gag-ca gene which codes mayor protein has been characterized as immunodominan antigen, and it also has positive reaction to animal antibody which infected by Jembrana virus, so it usually used as vaccine resources and serologic detection for Jembrana disease. The research was done by analyzing gag-ca gene of Jembrana virus in pcDNA-ca recombinant plasmid in Escherichia coli DH5α which has been stored for eightyears. The pcDNA-ca recombinant plasmid was purified from cultured Escherichia coli DH5α recombinant bacteria using "High Pure Plasmid Isolation Kit", then amplificated with “Pure Taq-Ready To Go PCR Kit” using specific primers. PCR products showed a positive result that pcDNA-ca recombinant plasmid contains gag-ca gene. PCR products identification using electrophoresis on agarose gel 1% showed that DNA stand apparently carried gag-ca gene at approximate amplicon 210 base pair (bp)
Comparison of Toxoplasma gondii Cysts Predilection in the Heart and Diaphragm of Pig in Bali
A study was conducted on Toxoplasma gondii in the heart and diaphragm of pigs in Bali. The study was aimed to compare the predilection of Toxoplasma gondii cysts in the hearts and diaphragms of pigs and compared to the number of T. gondii cysts per gr from the hearts and diaphragm sof infected pigs. Pigs were examined as many as 171. Each organ was examined by digestion method in order to identify the presence of T. gondii cysts in these organs. The parameters measured were the percentages and numbers of T. gondii cysts which werefound in each organs. The results showed that 9 of pigs were infected with T. gondii (5,26%) which in the hearts and diaphragms were 4.09% and 1.17%, respectively. The average number of T. gondii cysts per gr of the heart and diaphragm were 30.86 ± 21.38 and 24, respectively
