    RELATIONSHIP BETWEEN SURFACE HYDROPHOBICiTY, HAEMAGGLUTTNATION AND ADHERENCE MECHANISM OF STREPTOCOCCUS SUIS

    Streptococcus suis is well known as the causative agent of disease in various animal including pigs and human. The streptococcal cultures isolated from pigs and human have been characterized and identified biochemically and serologically as Streptococcus suis. The Cultures could be serotyped with 28 different monospecific antisera. Tne result of tins study indicated that the predominant antigen types of the S. suis cultures belonged to serotypes 2, 1/2, 1. The occurrence of the type specific microcapsule appeared to be realed with the surface hydrophobicity of the bacteria. Toe surface hydrophobicity could be demonstrated by hexadecan adherence tests. The S. suis cultures with hydrophobic surface agglutinated erythrocytes of humans, pigs and rabbits. The relationship between surface hydrophobicity, haemagglutinating and adherence properties of cultures could be demonstrated with 3 cell models. The haemagglutinating cultures adhered in high number to HeLa cells, ephitelial cells and to alveolar macrophages from rabbits. The non haemagglutinating cultures with hydrophobic surfaces adhered only to ephitelial cells and to alveolar macrophages S, suis cultures with hydrophilic surfaces showed no comparable reaction with 3 cell models

    Chemistry and Microbiology Qualities of Hard Candy-Green-Sirih Leaves (Piper betle L.) As Cattle Feed Additive

    Cattle diseases caused by bacterial infection and unqualified feed can decrease production of beef which people consume more in Indonesia. This research objective is to know quality of hard candy as cattle feed additive including chemistry and microbiology characteristics. Research steps consist of material preparation test, making sirih leaf extract, making hard candy, chemistry tests (The levels of water, ashes, reduction sugar, sacarose and atsiri oil, respectively) and microbiology test (Total count test, and yeast and Fungi Test). Theresults showed that hard candy made fulfill SNI (Indonesia National Standar) considered of water level, ashes level, sacarose level, total count, and yeast fungi level. Atsiri oil level before sirih leaves are made into hard candy is 0,405% and it still exists in hard candy with increasing atsiri oil level, 0,1333%

    Crab is one of the most abundant marine animals in Indonesia. Crabs not only rich of nutrient, but also have commercial value. Today, there is an innovation in crab aquaculture use ecdysis mechanism, known as soft shell crab. However, the limited studies about biochemical and nutritional composition of this crab remain to be explored. The aims of this research were to understand total fat, calcium, iron, and zinc contents of crab (Scyllaserrata) during ecdysis process until being soft shell crab comparing to the hard shell crab from the same species. This research was done using crab Scylla serrata, male, weighing ± 150-180 gr, completely randomized design with four variations of treatment, during 8 days, 16 days, soft shell crab, and control, repeated 3 times. Samples from each treatment were grown and were prepared for the chemical analysis. Total fat was measured usingSoxhlet method with chloroform and methanol (1:2, v/v) as solvent. Atomic absorption spectrometer was used to measure calcium, iron, and zinc contents. Then, the result were analyzed using one way Anova and Duncan 5 %. The results showed that the total fat, calcium and zinc contents of soft shell crab Scylla serrata were higher and significantly different with three other treatments. It could be concluded that the ecdysis process, raised total fat, calcium and zinc contents of crab Scylla serrata. The total fat, calcium, and zinc contents of soft shell crab Scylla serrata was higher than that of hard shell crab from the same species

    A Biological Study of Larvae and Adult Hemonchus contortus in Goat

    A biological study of larvae and adult Hemonchus contortus was carried out in goat. The aims of this study were to know the capability of the infective larvae (L3) development to adult, and the prepaten period of H. contortus. A number of 14 goat abomasum collected from Ngampilan Slaughterhouse, Yogyakarta Province was used as source of H. contortus. Eggs worm were collected from direct surgery of the H. contortus in physiological saline. The egg development of H. contortus to the L3 stadium was carried out by a modification Harada-Morito, whereas the development of the L3 to adult and the prepaten period were studied in the goat asexperimental animal in vivo. Before the goat necropsied, the diagnosis of H. contortus egg was done every two day post infection and started two weeks after infection. The data was analyzed descriptively. The results showed that the capability of the egg development of the worm to the L3 stadium was 0,33%, the capability of the L3 development to adult was 32,42%, and the prepaten period of H. contortus was 21 days

    Disease was a major obstacle in the cultivation of orrnamental fish, one of them is botia. The many types of diseases that infected botia can cause huge losses. The efforts that could minimize these losses was prevented the spread of disease. The purpose of this study was to determine the types of pathogen that infected botia (Chromobotia macracanthus Bleeker) in fish quarantine Supadio, Pontianak. The observations were identification of parasites, bacteria and fungi. The results showed that the agent of disease affecting botia are Gyrodactylus sp., Myxobolus sp., Argulus sp., Vorticella sp. from parasite agent, Neisseria sp., Aeromonashydrophila, Chromobacterium violaceum, Pasteurella hemolytica from bacteria agent and Saprolegnia sp. from fungi agent. All of agent diseases are not belonged to fish disease quarantine

    THE EFFICACY OF DORAMECTIN FOR TREATMENT OF Ancylostomiasis IN DOGS

    A study was done to evaluate the efficacy of doramectin injection for Ancylostomiasis treatment in 10 dogs. All of the dogs were heavily and naturally infected with Ancylostoma Sp. the heavy infection criterion (25 egg per gram) was based on the flotation method. At the beginning of the study the dogs were injected doramection subcutaneously (200 ,wg/kg of body weight). The pathogenesis of the disease was observd through the clinical sign and feses examination during 2 weeks after treatment. Dogs were considered recovery when they have a good response to the treatment and clinically showed normal of apetite, no sign of diarrhea and no egg was found laboratorically. Results of the study showed that all of the dogs had positive response to the treatment and therefore it can be concluded that doramectin effective against Ancylostoma Sp. in dogs.

Clinical epidemiology revolves around the use and interpretation of tests.

    The Accuracy of Transrectal Ultrasonography Diagnostic for Examination of Ovarian Structures in Cattle

    The study aimed to determine the accuracy of ovarian follicle and corpus luteum measurements in vivo using ultrasonography diagnostic and in vitro after slicing of the ovaries. A total of 15 cows were examined using transrectal ultrasonography just before being slaughtered. Ultrasonographic examination was performed using 7.5 MHz transrectal tranducer on ovaries, measured dominant follicle and corpus luteum diameters. Follicleappeared echogenic, black with round shape. Corpus luteum, appeared as low echogenicity structure and grayish in color.Ovaries were taken out soon after animals slaughtered, soaked in 10% buffered formalin for 12 hours,then cut in thin slices and measured follicle and corpus luteum diameters. Results of follicle and corpus luteum diameter measurements in vitro and in vivo compared using two sample or paired t-test, correlation and regression analyses. The two methods of follicle and corpus luteum measurements had linear orrelation (follicle measurement, R2 = 0.933 , r = 0.966, n = 15, P < 0.05) (corpus luteum, R2 = 0.912, r = 0.955, n = 15, P < 0.05).Measurements of dominant follicle diameter and corpus luteum size manifested the real size of the two ovarian structures.     Microscopic Agglutination Test for Diagnosis of Leptospirosis in Beef Cattle from Bantul and Kulon Progo

    Leptospirosis is a zoonotic disease caused by bacterial infection, Leptospira interrogans. The bacteria usually infect many kinds of wild animals and humans as well. There was an extraordinary outbreak of leptospirosis in humans in Yogyakarta in 2011. Therefore, it is necessary to investigate the seroprevalences of leptospirosis in cows, especially in the districts of Bantul and Kulon Progo in order to determine the possible role(s) of the cows in the spread of leptospirosis among other animals and humans. In this research, the diagnostic method was conducted by using microscopic agglutination test (MAT). Bovine sera samples were obtained from several farms in some areas of the districts of Bantul and Kulonprogo. The MAT was conducted on 150 sera samples of cows from districts of Bantul and was on 200 samples from Kulonprogo. Seroprevalences of leptospirosis in the cows were 18.67 % ( 28/150 ) and 14.5% (29/200) from Bantul and Kulonprogro districts, respectively. Mostly, serovar tarassovi, hardjo, icterohemorrhagiae and batavia were detected in cows from both Bantul and Kulonprogo districts. Antibody titers were obtained ranged from 1:100, 1:400 and 1:1600

    Levels of Blood Glucose and Total Protein of Repeat Breeding Dairy Cows From Daerah Istimewa Yogyakarta

    Repeat breeding in dairy cows is a reproductive disorder characterized by a length of calving interval.This research was aimed to determine levels of blood glucose and total protein as one of the causes of repeat breeding. This research used ten repeat breeding Friesian Holstein cross breeds at the age of 3-8 years old from Daerah Istimewa Yogyakarta. They have had birth at least once, normal reproductive cycle, and in a healthy condition. Ten ml blood samples of each cow from jugular vein were collected and analyzed at LPPT UGM. Blood glucose and total protein plasm were examined using a photometer Microlab 300 with a spectrophotometer. The data ware analyzed descriptively. The results showed that dairy cows with repeatedbreeding case had lower blood glucose and total protein plasm concentrations than that of the normal. The average concentrations of blood glucose and total protein plasm were 48.58±6.675 mg/dl and 6.815±821 g/dl, respectively
