    Isolasi dan Identifikasi Enterobacter sakazakii pada Susu Mentah dan Produk Susu Segar di Daerah Istimewa Yogyakarta

    Enterobacter sakazakii is an opportunistic pathogen that causes infections such as necrotizing enterocolitis, bacteremia, and meningitis which, in many cases, have been associated with the consumption of powdered infant formula. Raw milk has been considered to be a potential source of contamination. The aim ofthis study was to detect and identify the presence of E. sakazakii in raw milk from local farms and fresh dairy products sold by street vendors and cafés in the Special Region of Yogyakarta. A total of 80 milk samples were cultured in Enterobacterriaceae Enrichment Broth. The cell cultures were then enumerated on Tryptone Soya Agar and Chromocult Coliform Agar. For the isolation of suspect colonies, Chromogenic Druggan-Forsythe-Iversen (DFI) Agar was used. Presumptive colonies of E.sakazakii were characterized biochemically using API20E and molecularly through the amplification of the 16S rRNA gene using primer pairs 16_SUNI-L/Saka-2b and ESA-1/16_SUNI-R. Based on the α-glucosidase test of DFI Agar, only 9 isolates out of the 80 samples wereidentified as suspected E.sakazakii. Biochemical and molecular identification suggested that 5 isolates resulted positive for E.sakazakii, while the other 4 were Panthoea spp1

    The cottonseed (Gossypium hirsutum L.) contained gossypol as antifertility agent. The effect of cottonseed extract treatment could be decrease and impaired follicles development were accompanied by oocytesdamage. Damage of oocytes resulted reduction of number and viability of embryos. The objective of this study was to evaluate the effectiveness of cottonseed extract (Gossypium hirsutum L.) on the number and viability ofmice embryo (Mus musculus L.). Doses of the cottonseed extract were used consists of 0 (control), 1.5; 2.1; and 2.7 g/kg of body weight (BW) for 24 days via the oral route. This research used 24 animals healthy of femaleDDY mice 14-15 weeks old and 30-35 g BW. Embryos were collected at day 4 of pregnancy by flushing the utery cornua. The collected embryos were cultured in vitro for 48 hours in modified phosphate buffered saline (PBS) supplemented with 10% fetal bovine serum (FBS) culture medium according to the stage of its development to observe the viability of embryos. The result showed that the cottonseed extract with doses 1.5; 2.1; and 2.7 g/kg of body weight (BW) made the number of embryos which collected in D4 of pregnancy significantly lowerthan control (P<0.05). Data from embryos culture in vitro for 48 hours decreased embryos number (P<0.05) that developed in to the expanded and hatched blastocysts. At 2.7 g/kg BW, embryos only can develop to theblastocysts stage. Retardation (4-8 cells) and degeneration embryos did not develop in cultur

    Infestation of Luke Worm (Fasciola sp) and Stomach Worm (Paramphistomum sp) on Adult Bali Cattle in Tenayan Raya Pekanbaru Municipality

    This study was conducted at the Laboratory of Parasitology, Veterinary Investigation Center Regional II,Bukittinggi in January until February 2013. Bovine fecal samples of Bali cattle were taken in the District of Tenayan Raya Pekanbaru Municipality. This study was aimed to determine the prevalence of paramphistomiasis and fascioliasis of adult males and females Bali cattle and the number of eggs worm per gram of feces as well. A total of 232 fecal samples were tested by Parfit and Banks, and McMaster. Results showed that the prevalence of Fasciola spp. in males Bali cattle and cows was 49.02% and 60.71%, respectively, whereas the prevalence of Paramphistomum spp. in males Bali cattle and cows was 46.07% and 50.00%,respectively. Result count of the average number of worm eggs using Mcmaster in the fecal samples of Bali cattle in males and females was 205.88 eggs/g and 267.85 eggs/g, respectively.     Cimetidine was reported to inhibit elimination many other drugs. The research has been done to study the effect of cimetidine on elimination of sulphamethoxazole in rabbits. Twenty healthy male rabbits, 10 months of age were used as experimental animals. The rabbits were allotted into 4 groups of 5 each Group I as a control group were intramuscularly injected with 25 mg sulphamethoxazole. Group 11 were intramuscularly injected 25 sutphamethoxazole and pretreated with 12.5 mg cimetidine 30 minutes before injection. Group 111 were intramuscularly injected 25 mg sulphamethoxazole, pretreatod with 25 mg cimetidine 30 minutes before injection. Group IV were intramuscularly injected 25 mg sulphamethoxazole, pretreated with 50 mg cimetidine 30 minutes before injection. Blood sample were collecled at 0.25, 0 75, 1,75, .1.75, 10 and 24 hours after injection. Plasma sulphamethoxazole levels were determined using the Bratton-Marshall method Elimination kinetic parameters were analyzed statistically using anava. The result of the study showed that cimetidine significantly reduced total clearance of sulphamethoxazole <P<0.l). Based on the result it can be concluded that cimetidine inhibits elimination of sulphamethoxazole

    Monoclonal antibodies can be used as specific probes for molecular structure such as drugs, hormones, receptor for hormones, or any other biologycally derived or biology-cally active materials. The potential usefulness of mono­clonal antibodies is in almost every field of biology and medicine.The study was carried out to produced monoclonal anti­bodies against outer membrane protein (OMP) Pasteurella multocida isolat Dompu, which can be used to isolate imunogenic OMP fractions.Three monoclonal antibodies which specifically react with OMP fraction of Pasteurella multocida isolat Dompu were obtained previously. Those monoclonal antibodies were pro­duced in Balb/c mouse. The antibodies production were measured by ELISA and were purified by affinity chromatograPhy.The monoclonal antibody were injected intraperitoneally into group of five rats. Two additional groups of five rats were injected with immune mouse serum and normal mouse serum, which served as a positive and negative controls respectively. All rats were challenged with 1 ml lethal dose of Pasteurella multocida 24 hours after the antibody injec­tion.The outer membrane protein was separated 12,5% SDS-PAGE. The region of gel containing the appropiate antigens (PI, P2, P3) were excised. Protein antigen was mixed with incomplete Freund's adjuvant (IFA) and injected intramuscu-lary into group of five rats at dose 0,2 ml per mouse. Two additional groups were injected with gel in IFA and OMP and IFA as a positive and negative controls. Each treated mouse and control was immunized four times, at weekly interval. Two weeks after final immunization rat were bled and their antibodies productions were measured with ELISA. All mice were then challenged with lethal dose of Pasteurella multo­cida.The result indicated that eventhough immungenic the PI, P2, P3 OMP fraction individually were not able to induce protective antibody.

    This experiment was designed to study the effects of active Gumboro vaccine of high pathogenicity on the morphology of bursa Fabricius and the immune response of broiler to vaccination against Newcastle disease (ND). This experiment was conducted in 330 broilers, which were vaccinated with aviive Gumboro vaccine of high, intermediate and low pathogenicity (control) at the age of 12 days. Experimental chickens were also vaccinated against ND at the age of 4 and 18 days. Diameter of bursas were measured at day 1st through 7th, 14th, 21th, 28th, 35th post Gumboro vaccination. Evaluation of hemaggutination inhibition (HI) liters against ND virus (NDV) were done only at day 7th, 14th, 2lth, 28th and 35th. Samples of bursa were stained with the method of hematoxylin and eosin (H & E). Pathologic evaluation revealed lesions in the bursa Fabricius of chickens vaccinated with Gumboro vaccine of high pathogenicity as well as changes in the diameter of bursa of lesions in this organ. The HI titers against NDV were lower in the group of chickens vaccinated with Gumboro vaccine of high pathogenicity compared with them of other groups. Results indicated that active Gumboro vaccine of high pathogenicity caused lesions in the bursa,which were similar to lesions caused by exposure to field Gumboro virus

    The objectives of this study were to modify and develop technique for determination of 1,25-(OH)2D3 concentrations in sera and milk samples. The developed technique, used non-HPLC for extraction and purification of the samples and involved radio receptor assay which used calf thymus as 1,25-{OH)2Dj receptor for quantitation of 1,25-(OH)2D3 Results of this study were compare to that of the technique developed by Horst et at. (1981) which used HPLC for samples extraction and purification. The correlation between the two techniques was r=0,95. Using the modified technique the 1,25-(OH)2D3 concentrations in cow and sow sera were 57,42 ±5,3 pg/ml (n=20) and 75.22 ±6.05 pg/ml (n=20) respectively, whereas the laSHOH^D., concentrations in cow and sow milk were 16.62 ±2.2 pg/ml (n=20) and 21.52 + 3.125 pg/ml (n=20). Based on the results could be concluded that the technique that had been developed in this study was found satisfactory for determination the l,25-(OH)2Dj concentration either in serum or milk samples

    DYNAMICS OF NUMBER OF NEMATODE LARVAE IN GRASS IN THE PASTURE

    The existence of nematode larvae in a pasture can be used as an indication of infection or reinfection in animal grazing on it. The objective of the current study was to reveal the dynamics of the number of nematode larvae found on the grass of a pasture.Three locations of the pasture of the teaching farm of Faculty of Veterinary Medicine GMU, were chosen as sampling areas. Grass of the areas was cut just on top of the soO five times a day for ten day namely at 5, 6-9 am - 1 pm, 2-5 pm and 6 pm. Following cutting, the grass was kef t m a tube and washed with detergent solution. The washing solution was examined under a stereomicroscope, and the larvae observed were counted. Differences in the number of larvae between locations and collection times were analysed statistically by names of factorial analysis of variance.The results show that the mean number of larvae varies between 03 ± 0.67 and 6.1 ± 2.95 per grass. The number of larvae is not significantly different (F > 0,05). Cutting at however, is influenced by collection time (P < 0,05). Cutting at 5-9 am gives the largest number of larvae, while 6 pm gives the lowest count. 

    FIELD EVALUATION OF NEWCASTLE DISEASE VACCINE OF VG\GA IN STRAIN SEVERAL COMMERCIAL BROILER FARMS IN JAVA

    This field evaluation was conducted to study the level of protection, post vaccinal reactions and zootechninal performance of groups of chickens which have been vaccinated with Newcastle disease (ND) vaccine of VG/GA strain.This evaluation was done in 26.000 broilers from 4 farms, which have different condition in management practices and were located in different areas of Java.Chickens were vaccinated with VGAGA vaccine or with B-l/La Sota vaccine with/without inactivated ND. Serological tests were done at the age of 28 and 35 days, whereas challenge test with local ND virus isolates was done at the age of 45 days. Results of field evaluation indicated that in broilers, the VG/GA vaccine induced a high level of protection agamst the challenge test with a local (Indonesia) ND virus isolate and against infection with field ND virus. Statistical analysis indicated that the hemagglutination inhibition (HI) titers against NDV were significantly higher (P < 0,05) in group of chickens vaccinated with VG/GA with/without inactivated ND compared to group of chickens vaccinated with B-l/La Sota with/without inactivated ND vaccine. In certain conditions, the post vaccinal reactions were milder and the zootechnkal performance was better in groups of chickens vaccinated with VG/GA strain compared to groups vaccinated with B-l/La Sota
