Pengaruh Ekstrak Buah Delima Terstandar 40% Ellagic Acid terhadap Profil Darah Tikus Putih Yang Mengalami Nefrotoksisitas akibat Induksi Gentamisin
The purpose of this study was to determine the effect of pomegranate extract of 40% ellagic acid on blood profile of albino rats that had nephrotoxicity after included with Gentamicin. a total of 32 white rats were divided into four treatment groups, each consisting of eight rats. the four treatment groups were healthy rats (P0), Gentamicin induced rats (P1), Gentamicin induced rats and ellagic acid treatment (P2) and Gentamicin induced rats and standardized Pomegranate extract therapy 40% ellagic acid treatment (P3). After seven days of treatment, blood was taken for blood profile tests. The results showed that in the Gentamicin-induced group (P0), a decrease in blood profile allegedly caused by Gentamicin that could cause damage and impaired renal tubular function. Givibg ellagic acid and pomegranate extracts of 40% ellagic acid proved to improve the blood profile of Gentamicin induced rats. This is due to inhibition of renal damage and improved renal function due to antioxidant activity possessed by ellagic acid and pomegranate extract of standardized 40% ellagic acid
Newcastle Disease Virus Detection from Chicken Organ Samples Using Reverse Transcriptase Polymerase Chain Reaction
Newcastle disease (ND) is a systemic, viral respiratory disease that is acute and easily transmitted which affects various types of poultry, especially chickens. Diagnosis of ND which generally involves virus isolation and subsequent identification with serological assays has limitations that needs more time. This research was aimed to detect Newcastle Disease virus (NDV) in chickens suspected with ND using the Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR) technique. Nine chicken organ samples such as lien, trachea, and lungs were collected from chicken farms diagnosed with ND. The organ samples were processed and the targeted viral RNA was extracted using the RNA extraction kit. Genome amplification was performed with RT-PCR using specificprimers to target the F gene. Amplification results produced an amplicon product of 565 base pairs (bp). PCR product samples were then visualised using agar gel electrophoresis and viewed using the unified gel documentation system. Amplification results show nine samples positive for the DNA bands corresponding to the targeted band of the NDV F gene fragment. The results of this research confirm that the RT-PCR method is applicable for NDV detection from chicken organ samples
Infeksi Virus Peste de Petits Ruminants (PPR) pada Kambing dan Domba di Indonesia
Peste des petits ruminants (PPR) merupakan salah satu penyakit virus pada kambing dan domba yang ditandai dengan peradangan pada saluran pencernaan dan pernafasan. Penyakit ini masuk dalam “daftar penyakit” yang dibuat oleh OIE. Keberadaan penyakit ini di Indonesia belum pernah dilaporkan sehingga perlu dilakukan surveilan secara serologis pada kambing dan domba untuk mendapatkan informasi awal keberadaan antibodi terhadap virus PPR. Sebanyak 680 sampel serum kambing dan domba yang berasal dari Rumah Potong Hewan (RPH) Jakarta dan pedagang/pengumpul kambing dan domba di Jakarta pada tahun 2014 telah dikoleksi. Hasil pengujian dengan uji kompetitif ELISA yang menggunakan antigen inaktif (recombinant nucleoprotein- NP), menunjukkan bahwa telah terdeteksi antibodi terhadap virus PPR dengan prevalensi 0,2% pada kambing dan 1,7 % pada domba asal RPH Jakarta. . Berdasarkan lokasi asal ternak yang masuk ke RPH Jakarta, prevalensi tertinggi diperoleh dari Indramayu (2,8%), Solo 0.75%. Serum yang positif, beberapa negatif dan dubius tersebut kemudian diuji untuk dikonfirmasi di Australian Animal Health Laboratory (AAHL) menggunakan uji ELISA dan menunjukkan hasil yang konsisten yaitu tetap positif atau negatif. Selanjutnya dilakukan pengambilan sampel serum dari domba dan kambing di beberapa kabupaten Indramayu pada tahun 2016. Hasil menunjukkan bahwa seluruh sampel sebanyak 326 sera telah diuji dengan uji ELISA hasilnya negatif. Hasil penelitian ini memperlihatkan bahwa infeksi PPR masih sangat rendah, namun hasil ini dapat menunjukkan indikasi awal adanya infeksi PPR di Indonesia. Oleh karena itu, kewaspadaan terhadap meningkatnya kejadian infeksi PPR di Indonesia perlu mendapat perhatian
Efektivitas Terapi Multivitamin, Obat Cacing dan Premiks pada Sapi Terdiagnosa Hipofungsi Ovarium di Wilayah Kecamatan Prambanan, Yogyakarta
Ovarian hypofunction in cattle is reproduction disorder related to massive economic loss. Etiology of this ovarium disfunction are low quality feed, minimum health concern, poor sanitation and pen problem. Eighty-five (85) Peranakan Ongole (PO), Peranakan Limosin (PL) and Peranakan Simental (PS) cows from Prambanan Sub District of Daerah Istimewa Yogyakarta were diagnosed with ovarian hypofunction by rectal palpation and physical examination. The cows were then programmed for ovary condition recovery by administration of vitamine A,D,E (IM), per oral anthelminthic bolus (Klosantel) and premix mixture in feed given daily for 4 weeks. After 4 weeks, the cows were re-examined. Sixty-two (72,9%) cows were recovered from ovary hypofunction, however ovaries of 23 (21,9%) cows were palpated abnormal, which require another administration of A,D,E (IM). There were no significant difference between the type of cows (PO,PL,PS) with recovery percentages after one programme therapy (P>0.05). It can be concluded that single injection of vitamin A,D, E , per oral bolus of Klosantel, and addition of premix into feed daily for 4 weeks were able to reduce 72,9% ovary hypofunction in Peranakan Ongole, Peranakan Limosin and Peranakan Simental cows at Prambanan sub district, Daerah Istimewa Yogyakarta.
Karakterisasi Staphylococcus aureus Isolat Susu Sapi Perah Berdasar Keberadaan Protein-A pada Media Serum Soft Agar terhadap Aktivitas Fagositosis Secara In Vitro
Staphylococcus aureus merupakan salah satu bakteri penyebab utama mastitis. Protein-A berperan penting dalam adesi dan kolonisasi bakteri pada sel inang. Penelitian ini bertujuan untuk mengetahui hubungan aktivitas fagositosis S. aureus berdasarkan keberadaan protein-A pada media serum soft agar. Sebanyak 19 isolat S. aureus susu sapi perah asal Jawa Barat dan Jawa Tengah digunakan pada penelitian ini. Seluruh isolat tersebut direidentifikasi dengan dipupuk pada media plat agar darah (PAD), koloni bakteri kemudian diidentifikasi dengan pewarnaan Gram, uji mannitol salt agar (MSA), katalase dan uji koagulase. Karakterisasi S.aureusdilakukan dengan menanam bakteri pada media serum soft agar (SSA) yang mengandung serum kelinci untuk mengetahui keberadaan protein-A, kemudian dilakukan uji fagositosis dengan menggunakan sel polimorfonuklear. Dari 19 isolat tersebut seluruhnya teridentifikasi sebagai S. aureus yang ditunjukkan dengan Gram positif, sel berbentuk kokus bergerombol, mampu memfermentasi manitol pada media MSA, positif pada uji katalase, 15,79% sampel menunjukkan hasil koagulase negatif, sedangkan 84,21% menunjukkan hasil koagulase positif. Pertumbuhan pada media SSA menunjukkan hasil 12 isolat (63,16%) koloni berbentukkompak dan 7 isolat (36,84%) koloni berbentuk difus. Koloni kompak menunjukkan bakteri tersebut memiliki protein-A, koloni difus menunjukkan bakteri tersebut tidak memiliki protein-A atau memiliki protein-A tetapi tertutup oleh kapsul. Hasil uji fagositosis menunjukkan S. aureus yang memiliki protein-A lebih sedikit difagosit oleh leukosit polimorfonuklear (2,99 bakteri/sel) dari pada S. aureus yang tidak memiliki protein-A, atau mempunyai protein-A tetapi tertutup oleh kapsul (3,85 bakteri/sel). Staphylococcus aureus yang memiliki protein-A lebih patogen daripada S. aureus yang tidak memiliki protein-A. Super Javanese chicken is the result of a cross breeding between male domestic chicken and female layer chicken. . There has not been much research aboutSuper Javanese chicken embryonic development. This study attempts to provide information about embryonic development and sexing on Super Javanese chicken. This study used Super Javanese chicken fertile eggs from Java Farm, Maguwoharjo, Depok, Sleman. These eggs were incubated at temperature of 37o-38oC. Samples were observed with Neutral Red and Hematoxilin Eosine (HE) staining. The measurement of the Super Javanese chicken embryo were about length of the third finger, beak, and the length of the embryo's body. This observation obtained embryonic development of the super Javanese chicken spesifically. Gambaran Histopatologi Otak Tikus Akibat Injeksi Trimetyltin sebagai Model Penyakit Alzheimer
Trimethyltin chloride (TMT) is an organotin compound which neurotoxic at limbus system and hippocampus in human and animal. Pathology changes that caused by the induction of TMT is a neurodegenerative disorder such as nerve cell death and cognitive impairment. This study was aimed to observe brain pathology induced by TMT with multiple doses for 14, 21 and 28 days after treatment. Twenty seven of Wistar rats, at 2 months of age with weight ranging between 200-300 grams were used and divided randomly into 3 groups (n=9). Group I were injected by trimetyiltin with a dose of 6 mg / kg, group II were injected bytrimetyltin with a dose of 8 mg / kg and group III as control without injection. Observation of brain pathology was done by euthanasia on day 14, 21 and 28 after treatment, three rats each. Cortex and hippocampus of the brainwere observed using Hematoxilin and Eosin staining (HE). All of the research procedure was done with the approval and supervision of Animal Ethics Committee LPPT UGM No. 300/KEC-LPPT/VII/2015. The observation of histopathology of the brain's neuron cells injected by trimetyltin dose of 6 mg/kg and 8 mg/kg body weight was showed increasing cell death of brain neurons in the cortex and hippocampus compared to the control group. The highest cell death was on day 14 in the hippocampus and cortex cerebral on day 21after TMT injection. The neuron cell death characterized by the shrink of brain neurons as well as colored eosinophilic cytoplasm. One way ANOVA statistical analysis showed a significant difference number of neurons cell deathbetween control and treatment groups. Based on this research, it can be concluded that the trimetyltin injection dose of 6 mg / kg and 8 mg / kg of body weight caused neuron cell death in the brain rats from fourteen day aftertreatment, especially in the hippocampus and cortex
Mozart KV 448 Menurunkan Densitas dan Aktivitas Neuroglia Hipokampus Mencit (Mus musculus) Selama Stres Prenatal No. 416-KE
The aim of this research was to explore the influence of Mozart KV 448 classical music therapy to the neuroglia cells of mice’s hippocampus that were exposed to stress during prenatal. This research were employing twenty female mices and twenty male mices. Female mices were estrus synchronized with PMSG and hCG then monomating to the males. Pregnant females were then divided into four groups (P0, P1, P2, P3). P0 was as control, P1 was treated by one minute forced swim test, P2 was treated by one minute forced swim test followed by thirtyminutes classical music Mozart KV 448 and P3 was treated by one minute forced swim test followed by sixty minutes classical music Mozart KV 448. This research was carried out for twenty-one days during gestation period. The neuroglia density result was analyzed using ANOVA and Duncan test. The neuroglia activity result wasanalyzed using Kruskal wallis test and Z test. The histology reading showed degradation of density and activity of hippocampus neuroglia
Daya Ovicidal Ekstrak Kulit Buah Muda (Calotropis procera) terhadap Haemonchus contortus secara in vitro
Penelitian ini bertujuan untuk mengetahui konsentrasi efektif ekstrak kulit buah muda Calotropis procera (C. procera) dalam menghambat perkembangan telur cacing Haemonchus contortus secara in-vitro. dari 0,2g/ml sediaan larutan ekstrak stok, albendazole konsentrasi 0,055%. Data hasil penelitian dianalisis secara deskriptif. Konsentrasi efektif ekstrak kulit buah muda C. procera (EKBMCP) dalam menghambat perkembangan daya tetas telur cacing Haemonchus contortus secarain-vitro adalah perlakuan EKBMCP konsentrasi 4,5% dengan daya hambat 88% sedangkan perlakuan EKBMCP konsentrasi 2,5% dan 3,5% daya hambatnya secara berturut-turut adalah 70,5% dan 81%
Perbandingan Aktivitas Linimentum Ekstrak Koral Kelimutu dan Linimentum Ekstrak Daun Lamtoro (Leucaena leucochepala) Terhadap Penyembuhan Scabies Pada Kelinci (Oryctolagus cuniculus)
Tujuan penelitian ini adalah untuk mengetahui perbandingan aktivitas linimentum ekstrak koral kelimutu dan ekstrak daun lamtoro (Leucaena leucochepala) sebagai terapi penyembuhan scabies pada kelinci (Oryctolagus cuniculus). Jenis penelitian ini eksperimental dengan menggunakan Rancangan Acak Lengkap (RAL). Sampel hewan yang digunakan adalah kelinci jantan umur 4-6 minggu sebanyak 24 ekor yang dibagi menjadi empat kelompok, yaitu kelompok kontrol negatif /normal, tanpa diobati (P0), kontrol positif : diobati linimentum sulfur ppt (P1), diobati linimentum ektrak daun lamtoro (P2), diobati linimentum ekstrak coralkelimutu (P3). Semua bulu kelinci dicukur pada daerah punggung anterior sepanjang 3x3 cm dan diinfestasikan parasit Sarcoptes scabiei. Semua kelinci dibiarkan selama 7 hari agar parasit dapat tumbuh dan berkembang biak tersebar merata ke seluruh permukaan kulit. Setiap hari kulit kelinci yang terkena scabies diolesi dua kali dengan linimentum yang diuji dengan dosis 25%. Pengamatan makroskopis dilakukan pada hari ke- 3, 5, 7, dan pengamatan mikroskopis dilakukan pada hari ke- 3 setelah dilakukan masa pengobatan selama 7 hari. Semuadata kuantitatif diuji secara statistik menggunakan Analisa Sidik Ragam (ANOVA) dengan uji post hoc metode Turkey dan metode LSD dengan taraf kepercayaan 95% (α = 0,05). Hasil penelitian menunjukkan bahwa pada kelinci yang diobati dengan linimentum ekstrak coral kelimutu (P3) memberikan hasil yang lebih baik daripada kelompok P0, P1 dan P2. Berdasarkan analisis statistik uji Post hoc metode Turkey dan metode LSD menunjukkan p = 0,000 ; R squared = 0,88 dan hitung (7,11) > F tabel (1,74), maka disimpulkan bahwa terdapat perbedaan yang signifikan antara kelompok P3 dengan P0, P1 dan P2 terhadap perlekatan penutupan luka, pengurangan keropeng dan peningkatan jumlah kematian Sarcoptes scabiei. Hal ini menunjukkan aktivitaslinimentum ekstrak koral kelimutu paling baik dan efektif digunakan untuk terapi penyembuhan scabies pada kelinci
