Identifikasi Ektoparasit pada Benih Ikan Mas (Cyprinus carpio) di Balai Benih Ikan Kabat, Kabupaten Banyuwangi
Diseases caused by parasitic groups get less serious attention from the farmers because it is chronic morbidity and mortality rates and small. However, if observed further, parasitic diseases may trigger secondary infection by bacteria, viruses and fungi that can cause death and high losses. Thus this study aims to identify the types of ectoparasites that infected the seed goldfish in BBI Kabat, Banyuwangi which the results will be used as an early warning to prevent the death of fish in bulk. The research sample in the form of seeds 3-5 cm carp amounted 150 obtained from BBI Kabat, Banyuwangi. Ectoparasites inspection method performed by scraping the surface of the body, fins and gills of fish, and then observed under a microscope with a magnification ranging from 40-400x. The observation of ectoparasites are further identified accordance morphology based on the keys of identification. Identified ectoparasites infect seed goldfish in BBI Kabat, Banyuwangi consists of 8 genus is Zoothamnium sp., Trichodina sp., Oodinium sp., Vorticella sp., Argulus sp., Lernaea sp., Dactylogyrus sp., and Gyrodactylus sp. Further research is needed to determine the prevalence and intensity of parasites and appropriate measures to prevent the spread of parasites in healthy fish in BBI Kabat, Banyuwangi.
Analisis Cemaran Staphylococcus aureus pada Gelas, Darah Segar, dan Jamu dengan Ramuan Darah Ular Kobra Jawa (Naja sputatrix)
Jamu yang mengandung ramuan darah ular kobra merupakan salah satu jamu tradisional yang dipercaya memiliki banyak khasiat bagi kesehatan tubuh manusia. Di sisi lain belum diketahui seberapa besar cemaran bakteri yang terdapat pada ramuan darah tersebut. Staphylococcus aureus sebagai bakteri yang dapat menyebabkan keracunan pangan dan umumnya terisolasi dari produk makanan. Penelitian ini mengungkap keberadaan Staphylococcus aureus pada jamu darah ular kobra. Penelitian ini menggunakan 10 sampel darah segar ular kobra dan 10 sampel ramuan jamu tradisional dengan darah ular kobra. Isolasi Staphylococcus aureus menggunakan media Vogel Jhonson Agar (VJA). Hasil penelitian menunjukkan cemaran Staphylococcus aureus terdeteksi pada swab gelas, darah segar, dan ramuan darah ular kobra. Analisis variansi (ANOVA) darirerata cemaran Staphylococcus aureus menunjukkan tidak ada perbedaan antara swab gelas, darah dan ramuan darah ular kobra. Walaupun demikian, jamu yang mengandung darah ular kobra berpotensi mengandung Staphylococcus aureus yang membahayakan konsumen
Daya Vermisidal Ekstrak Lima Jenis Etnofarmakologi terhadap Cacing Haemonchus contortus Secara In-vitro
Research aimed to identify the most effective concentration of five extract types (EKBMCP, EKPAS, EDMSP, EBJCP, dan EDMCO) as vermicidal anthelmintic against H. contortus in-vitro. Research was consisted of seven groups, among them five groups were considered as treatment groups that named based on the extract type (EKBMCP, EKPAS, EDMSP, EBJCP, dan EDMCO). Each treatment group was consisted five subgroups of etnopharmacological concentration : 0,5%, 1,5%, 2,5%, 3,5% dari 0,2 g/mL. Other two group were regarded as a negative control by distilled water treatment, and a positive control by pyrantel pamoat 5%. Every single treatment group was quadruplet and exposure each concentration were performed in quadruplet with variation in exposure times i.e. : 1, 3, 5, and 7 hours. The research focused on several variables that were the percentage of live and dead H. contortus in the treatment groups, extract concentration, and treatment period by using selected etnopharmacological extract. The effective vermicidal anthelmentic concentration of each treatment was analyzed descriptively. The results showed that five etnopharmacological extract types that were EKBMCP, EKPAS, EDMSP, EBJCP, dan EDMCO had a potency as vermicidal anthelmentic. Among those treatment, EKPAS and EBJCP had the lowest concentration (2,5%) to produce optimum vermicidal effect (100%) with period five hours exposure. Other treatment groups with EKBMCP and EDMSP needed 3,5% concentration with exposure period seven hours to produce optimum vermicidal effect, respectively. Another treatment group with EDMCO needed 3,5% concentration with exposure time seven hours to produce optimum vermicidal effect.
Tingkat Kejadian Escherichia coli Penghasil Extended Spectrum Β-Lactamase yang Diisolasi dari Feses Broiler di Kota Bogor
The use of antibiotics in food-producing animals may encourage the occurence of bacterial resistance named Escherichia coli that produces Extended Spectrum β-lactamase (ESBL). Antibiotical resistance is an important issue in the animal health and human health. This study focused on the presence of ESBL-producing E.coli in chicken feces. The number of sample feces was 100. E. coli isolates were obtained from broiler chicken feces in Bogor. Isolates obtained were identified using API 20E kit. Confirmation of ESBL in this study used Double disc method using antibiotical disc namely cefpodoksim, ceftazidime and cefotaxime. The existence ofESBL-producing E. coli which isolated from the feces of broiler chickens in the city of Bogor is 25% (4/16)
Deteksi Gen Penyandi Sifat Resistensi Metisilin, Penisilin dan Tetrasiklin pada Isolat Staphylococcus aureus Asal Susu Mastitis Subklinis Sapi Perah
Detection of gene encoding resistance of bacteria could be used as an accurate method to determine resistance of Staphylococcus aureus which is causing mastitis in dairy cows to the several antibiotics. This research aimed to detect the gene encoding resistance of methicillin, penicillin and tetracycline from identified S. aureus. Sixty milk samples were collected from subclinical mastitis of cows from various dairy farming in Yogyakarta. Isolation and identification of S. aureus based on the culture, Gram staining and biochemical test. Phenotypes of S. aureus resistances against antibiotics were carried out by disc diffusion method, meanwhilespecies specific gene of S. aureus and the gene encoding methicillin, penicillin and tetracycline were confirmed by PCR method. The results showed 11 isolates representing of Methicillin Susceptible Staphylococcus aureus (MSSA) could be identified, wherein 5 isolates were harboring both of penicillin and tetracycline resistant genes respectively
Identifikasi Virus Avian Leukosis Sub Grup J pada Peternakan Ayam Petelur Komersial di Kabupaten Tangerang Tahun 2015
Avian leukosis virus (ALV) is one of causing agents for neoplastic syndrome in commercial chicken farms. There are six sub groups of ALV in chicken including A, B, C, D and E. However, the sub group J (ALV-J) is the most alarmed and anticipated because of its severe economic repercussion. The objective of this study was to identify the presence of ALV-J in the commercial chicken farms in Tangerang District. Two identification methods were utilized including antigen capture enzyme-linked immunosorbent assay (ac-ELISA) technique and nested reverse transcriptase polymerase chain reaction (nRT-PCR) test. As results, the ac-ELISA test detected that most of 150 serum samples from 5 commercial layer farms were positive for ALV in about 86,67%. On the other hand, none of the 60 cloacal swab and 60 albumen samples was positive for presence ALV. Subsequently, testing of 45 serum samples from 5 farms using nRT-PCR showed that in about 20% of these samples were positive of ALV-J. These evidences indicated the circulation of ALV-J with other sub groups in thecommercial layer farms in Tangerang District
Korelasi Berat Badan dan Umur Sapi terhadap Berat, Volume Cairan dan Konsentrasi Prostaglandin F2α pada Vesikula Seminalis
Seminal vesicles were collected from 60 heads of Bulls which butchered in slaughter house (RPH) Yogyakarta. The aims of this study are knowing the relationship between body weight, age, fluids volume and concentration of prostaglandin F2 α (PGF2α) in seminal vesicle of beef cattle. Those seminal vesicles were gathered from bulls which recorded its body weight and age then measured its seminal vesicles for weight, fluids volume and PGF2α levels. The PGF2α level was measured by Enzyme-Linked Immunosorbent Assay. Statistical analysis was performed using one way – analysis of varian, regression and correlation with P<0.05. Body weight of bulls showed positive correlation with the weight of seminal vesicle and its fluids volume. However, PGF2α levels were not correlated with the body weight of cattle. Weight of seminal vesicles also exhibited positive correlation with volume of vesicle fluids but no correlation with PGF2α levels. Based on the age of bulls, there were no correlation withthe weight of seminal vesicles, seminal fluids volume and PGF2α levels. Those result indicated that the weight and fluids volume were affected by the body weight of bulls altough the PGF2α levels have a standard of developmentwhich seems affected by other factors such as concentration of androgen hormone
Ekspresi IL – 1β pada Proses Kesembuhan Fraktur Os Femur Tikus Putih (Rattus norvegicus) Ovariektomi yang Diterapi dengan Ekstrak Cikal Tulang (Cissus quadrangularis)
This study aims to determine the potential of the Cissus quadrangularis extract to decreased the expression of IL-1 β so that it can to inhibit bone resorption and osteoporosis fracture healing is not delayed union. Forty rats adapted for 1 week, and then divided into four groups with 10 replications. P0 (sham-operated + Osteotomy + CMC Na); P1 (ovariectomy + Osteotomy + CMC Na); P2 (ovariectomy + Osteotomy + Raloxifene 5.4 mg /kg); P3 (ovariectomy + Osteotomy + CQ 750 mg / kg). Osteoporosis induced is made by bilateral ovariectomy in rats, then to make sure that osteoporosis is already happened, 8 weeks post-ovariectomy performed radiology examinations on the femur, then the osteotomy action. IL1-β expression observations performed in at the 2nd week and 6th week after osteotomy through the immunohistochemistry examination. The results of immunohistochemistry examination in the 2nd week showed an increased expression of IL1-β in all groups of rat that did ovariectomy (P1, P2 and P3) were significantly different (p <0.05) with a rat group that didn’t ovariectomy, as well P2 dan P3 were significantly different with P1. The Results of immunohistochemistry examination in the 6th week showed the samepattern with the 2nd week and there is increased IL1-β expression in all groups of rat compared with the results of the examination in the 2nd week. Based on these results it can be concluded that Cissus quadrangularis extract andRaloxifen can decreased the expression of IL-1 β, and the results that don’t significantly different, so that these both materials can to inhibit bone resorption and osteoporosis fracture healing don’t get delayed union
Pendekatan Diagnosis Avian Influenza Virus dan Newcastle Disease Virus pada Kasus Lapangan Ayam Petelur: Imunopatologis Streptavidin Biotin
Poultry can be infected not only by one type of virus, but also by more than one type of virus. Avian influenza virus (AIV) and Newcastle disease virus (NDV) are two important pathogens in poultry that can cause recurrent outbreaks and often lead to clinical symptoms and pathological lesions similar in poultry with high morbidity and mortality causing significant economic losses to the poultry industry. This study aims to detect and differentiate pathogenic viruses in commercial laying chickens with similar clinical symptoms, especially torticolis and curled toe paralysis, and anatomic pathological lesions, such as foci necrotic hemorrhages in the digestive tract by applying streptavidin-biotin immunohistochemical immunological test (IHC SB). Samples (chickens) were taken from the cases of the disease in poultry in several commercial poultry farms. The layer chickens in which showed clinical symptoms of torticolis and curled toe paralysis, and hemorrhagic lesions in the lungs and / or digestive system are suspected of having AIV and NDV infections. After being necropsied, then lungs and gastrointestinal tract were tested IHC SB. The IHC SB results were examined with a digital microscope camera system. The results of the present study indicated that AIV antigen was detected in the lungs, but not in the digestive tract, whereas, NDV was detected in the digestive tract, but not in the lungs. It was concluded, that the IHC SB can be applied for confirmation of the diagnosis of AIV and NDV and indicated, that the commercial laying chickens with clinical symptoms of torticolis and curled toe paralysis, as well as pathological hemorrhagic lesions in the gastrointestinal tract could be infected by both viruses
Kondisi Biosekuriti Tempat Penjualan Burung Terkait Avian Influenza di Wilayah Jakarta
The aims of this study were to identify biosecurity condition and to analyse the relation of bird vendors characteristics toward biosecurity practices and biosecurity condition related to Avian influenza in Jakarta bird markets. Data were collected from four bird markets in Jakarta using randomly interview method of 75 respondents and observe the biosecurity practices in bird markets. Data were analysed using chi-square test and gamma test to analyse the relation between bird vendors’ characteristics and biosecurity condition. The result showed that 69,3% of bird vendors did the biosecurity practice in moderate level and only 10,7% did it well, meanwhile, most of bird kiosks (53,5%) in Jakarta got the poor category of biosecurity level. Beside that, bird vendors’ characteristics which has the significant relation (p<0,05) were age (p=0,003), education level (p=0,007), training (p=0,047), knowledge (p=0,000) and attitude (p=0,000) toward biosecurity condition related to Avian influenza in Jakarta bird markets.
