Prevalensi dan Serovar Penyebab Leptospirosis pada Domba di Kabupaten Kulon Progo
Leptospirosis is a zoonotic disease, caused by Leptospira interrogans. A source of transmission in human leptospirosis are rodents, livestock, pets and wild animals. The prevalence of leptospirosis in cattle soon after theoutbreak of leptospirosis in humans that occurs in Kulon Progo district in 2011, reached 3.4% by the various serovar Leptospira. Breeding conditions of the people who still puts cattle and sheep in a single environment which enables transmission of leptospirosis in cattle. The purpose of this study was to determine the prevalence of leptospirosis and identify serovar caused of leptospirosis in sheep in Kulon Progo. A total of 60 sheep were done blood collection from the jugular vein 5 ml, serum was separated for leptospirosis examination with Microscopic Aglutination Test (MAT) which conducted at the Research Center for Veterinary Science, Bogor. Microscopic Aglutination Test carried out on various Leptospira serovar, namely: chterohaemorrhagiae, Javanica, Celledoni, Ballum, Pyogenes, Cynopeteri, Rachmati, Auatralis, Pomona, Canicola, Grippotyphosa, Bataviae, Hardjo, and Tarrasovi. Leptospirosis prevalence rate was calculated by dividing the result by the number of MAT positive samples examined. Serovar types that give a positive agglutination result was serovar that caused leptospirosis in sheep. The results showed that two samples were positive against antigen serovarIchterohaemorrhagiae. Based on these results can be concluded that the prevalence of leptospirosis in sheep in Kulon Progo district were 3.3%. Kemampuan Reagen Curcumax Mendeteksi Boraks dalam Bakso yang Direbus
Borax is a harmful compound to human health but sometimes was used illegally as preservative in meatball. Curcumax reagent is a compound which developed from the previous borax detector base on tumeric extract. This reagen was developed as an easy kit to apply in the field. This study aims were to determine accuracy of sight obesrvation and spectrophotometer to detected borax in boiled meatballs which tested by Curcumax and to determine the effect of boiling on the borax presence in the meatballs. This study was using meatballs groupas a control and another meatballs group preserve with borax 0.5%. Each treatment group was boiled in 0, 5, 10, and 15 minutes and each treatment unit had three replications which consist five meatballs each of them. Boraxdetection was done by mixing 1ml Curcumax reagent into 1 ml meatball extract. Qualitative examination of borax presence in the meatballs was known based on the color changing of tested meatball’s extract from yellow became orange (direct sight with eyes). The Quantitaive analysis was done by measured using spectrophotometer base on wave-length (λ) at 570 nm. Data was analyzed with ANOVA and continued by Least Significant Difference (LSD) analysis. Borax detection using curcumax based on sight sense against color change giving an inconsistence result. Meanwhile quantitative measurement by spectrophotometer more accurate than one. Curcumax had been detected borax in meatball which boiled for 15 minutes
Optimalisasi Waktu Pengomposan dan Kualitas Pupuk Kandang dari Kotoran Kambing dan Debu Sabut Kelapa dengan Bioaktivator PROMI dan Orgadec
Animal manures can be used as organic fertilizer because the high nutrient content such as nitrogen (N), phosphorus (P), and potassium (K). The nutrients are needed by plants and soil for fertility. One of the animal manuresthat can be used for organic fertilizer is goat manure. Goat manure is used as organic fertilizer it contains relatively more balanced nutrients than other natural fertilizer. In addition, the goat manure is mixed with the goat urine that alsocontains high nutrients. Coconut coir dust contains high potassium so that the addition of coconut coir dust in organic fertilizer will increase the potassium content (K) in organic fertilizer. The purpose of this research was to find out theinfluence of effective Orgadec and PROMI bioactivators on the optimal time of composting and the quality of the organic fertilizer. The organic fertilizer was analyzed to determine the content of N, P, K, organic-C, C/N ratio, watercontent, and pH. The result of analysis is compared with SNI 19-7030-2004. Organic fertilizer analysis was conducted on day 0, 10, 20, 30, 40, and 50. The result of the organic fertilizer analysis with Orgadec bio-activator more effectiveand faster on the decomposition organic matter in the compost compared PROMI (<20 days). The quality of organic fertilizer with Orgadec bioactivator on day 10 and 20, while organic fertilizer with PROMI on day 20, 30, 40, and 50days was in accordance with the quality stated in SNI 19-7030-2004 (C/N ratio, levels of N, P, K, water, and organic-C)
Optimalisasi Pembekuan Sperma Limbah Kauda Epididimis Kambing Lokal dengan Metode Bertahap dan Stabilisasi
Buck slaugthering produce waste such as testicles including epididymis which contain fertile sperm. Utilization of cauda epididymis as the sources of sperm for producing goat frozen sperm was not reported yet. The aims of this study were improving the frozen-thawed sperm using stabilization and multistep methods which recovered from the waste of buck slaughtering as the source of sperma. Cauda epididymis spermatozoa which was washed then diluted using extender 1 (Tris-citrate-antibiotics) and extender 2 (extender 1- glycerol-egg yolk). The extender 2 addition was performed by single or multistep methods then freezed. Modification in the pre freezing proces were performed by comparing the conventional equilibration and stabilization methods. The sperm suspension was incubated in 4°C for 2 hours after filling-sealing into straws on the equilibration group whether the stabilization group was cooled in tube 15 mL. All cooled straws from both groups were placed 4 cm horizontally on liquid nitrogen surface for 10 minutes and then plunged into liquid nitrogen for storage. The evaluation of motility parameters such as pattern of the movement and motility percentation were done followed the standard methodology. The student t-test, correlation and one-way ANOVA were used for data analysis with P<0.05. The results showed that multistep dilution method could increase the motility (25.0 ± 1.8 %) compared with single step (18.3 ± 1.7 %). Pre freezing method with stabilization also resulted higher motility (24.2 ± 2.0 %) than equilibration method (17.5 ± 2.8 %). The pattern of the movement were not different between all methods and its combination. The multistep dilution method and stabilization cooling method as well as its combination seems could increase the quality of frozen-thawed cauda epididymis spermatpzoa of local buck
Variasi Morfologi dan Deteksi Leucocytozoon caulleryi dengan Metode PCR pada Ayam Ras di Wilayah Endemis Indonesia
The purpose of this study was to analyze the variation in morphology and detection Leucocytozoon caulleryi by PCR in mtDNA genes cty b that attacking bred chickens in endemic areas of Indonesia. This study was divided into three stages, data collection of Leucocytozoonosis cases in endemic areas of Indonesia; identification of parasites Leucocytozoon caulleryi morphologically through microscopic examination; and the identification of Leucocytozoon caulleryi cyt b genes that attack bred chickens in endemic areas by PCR. The results showed that there are variations in morphology of Leucocytozoon caulleryi that attacking chicken in endemic areas of Indonesia. Gamete morphometry of L. caulleryi had an average length and width 18.233 ± 4.672 dan 12.934 ± 3.349 µm. Nested PCR clearly showed positive reaction of Leucocytozoon infections by amplicons in 600 bp and 03 bp length.
Penambahan Boraks dalam Bakso dan Faktor Pendorong Penggunaannya Bagi Pedagang Bakso di Kota Bengkulu
The research of using borax in meatballs which was undertaken in the Bengkulu City in 2011 showed that 10% from 100 samples of meatballs containing borax. Considering the dangers of borax for the consumer if consume continuously and increasing the number of meatball traders until 2013, it is necessary to study with more meatballs samples and widely area. The aim of the study was to detect the presence of borax in meatballs, borax content after heating, storability meatballs which containing borax and determine the factors that support the meatball traders to use borax. The study was designed using cross sectional study using two types of data, they were data of meatball sample test in the laboratory and interview data to meatball traders in the BengkuluCity. Total of 160 meatball samples from traders and five meatball samples from grinders were collected and examined with qualitative test using turmeric paper. The qualitative test showed that 165 samples (100%) takenfrom meatball traders and grinders didn't contain borax. Knowledge and attitudes level of traders were at a good level. There were significant relationship between education level with knowledge, information source fromtelevision with attitude, and knowledge with attitude of traders in Bengkulu City (p < 0,05)
Karakterisasi Faktor-faktor Virulensi Staphylococcus aureus Asal Susu Kambing Peranakan Ettawa secara Fenotip dan Genotip
Staphylococcus aureus is a major cause of mastitis in large or small ruminants, and often manifested by subclinical mastitis in Peranakan Ettawa (PE) goats. Staphylococcus aureus in human can cause food borne disease. The research aimed to characterize the virulence factors of Staphylococcus aureus isolated from milk PE goats, phenotypic- and genotypically. Phenotypically characterization were determined through the pigmen assay as well as hydrophobicity, haemolysin, and hemaglutinin reaction. Polymerase chain reaction (PCR) analysis was used to detect 4 virulen genes including coa, clf, fnbA, and fnbB genes. The results of research showed that Staphylococcus aureus abled to produce white pigmen (35,7%), yellow pigmen g (57,1%), andorange pigmen (7,2%). Staphylococcus aureus showed α-hemolysis zone (35,7%), β-hemolysis (35,7%), dan γ-hemolysis (28,9%). Hydrophobicytic test revealed 14,3% Staphylococcus aureus isolates were hydrophobe and85,7% hydrophil. Staphylococcus aureus (85,7%) isolates abled to aglutinated sheep blood cells. Based on genotypic analysis of Staphylococcus aureus could be detected coa gene (92,8%), clf gene (64,3%), fnbA gene (78,6%), and fnbB gene (64,3%). Based on the phenotypic and genotypic characters, it can be concluded that Staphylococcus aureus are virulent strains. This information can be used as the basis for control mastitis in PE goat
Gambaran Histopatologi Rumen dan Retikulum Sapi Bali Akibat Adanya Benda Asing
Rumen and reticulum hold an important role in the ruminant’s digestive tract. During the meal, Bali cattle accidentally ingest foreign objects because they can’t distinguish between foreign body object and feed. Cattles do not really chew food before swallowing. This study aims to determine the existence of a foreign body in the stomach and histopathological overview of Bali cattle rumen and reticulum due to a variety of foreign objects. This study used 10 samples of cattle Bali consisting of nine samples of rumen and reticulum contained foreign objects (plastic, metal, wood and stone) and one normal sample of the rumen and reticulum Bali cattle. Samples were obtained from Slaughter House at Mambal, Abian Semal District, Badung regency, Bali. Based on the results of research conducted on the rumen and reticulum Bali cattle, there are a variety of foreign objects such as plastic, metal, wood and stone. Histopathological changes in the mucosa of the rumen and reticulum form of inflammatory cell infiltration from mild to severe. Conclusions from this research that found a foreign body object in the rumen and reticulum Bali cattle, and foreign body objects cause tissue destruction
Respon Imun Mencit terhadap Protein 24 dan 71 kDa Toxocara vitulorum dalam Membentuk Antibodi dan Protektifitasnya terhadap Infeksi Buatan
This study aimed to get 24 and 71 kDa protein of T. vitulorum that have a high antigenicity and imunogenicity on ELISA and to get the protein which able to protect mice against artificial infection of L2 T. vitulorum. This study using mice Balb/c aged 6 to 8 weeks. Proteins isolated were 24 and 71 kDa. Proteins 24, 71 kDa and intestinal homogenates immunized in mice with the addition of adjuvant (1: 1) for 3 times with period of 2 weeks. Two weeks after the last booster, serum drawn from mice tested by Indirect ELISA to determine the value of optical density (OD). The next stage, mice were infected L2 with a dose of 10-17 larvae / g of body weight. The results showed the average OD value by ANOVA Factorial antigen P24 was not significantly different with antigen P71 T. vitulorum. Antigen 24 kDa and 71 kDa with different immunization, both were showed P0 significantly different with P1, P2 and P3. Based on percentage of L2 in the somatic tissue of mice, P0 were showed 79.1% of total number of L2 early infection, whereas the treatment of P1 were showed 0.04%, P2 and P3 showed as much as 0.02% and 0.04%. 24 and 71 kDa protein of T. vitulorum that have a high antigenicity and imunogenicity
Kualitas Oosit Kerbau dari Status Reproduksi Ovarium yang Berlainan
In vitro embryo production relies on the quality of oocytes, however the quality is subjected to ovaries reproduction cycle. This study was conducted to observe the potency of buffalo ovaries from various reproductive cycle in producing quality oocytes. Collected pairs of ovaries from slaughter house were weighed and grouped of 5 according to the cycle. Oocytes were collected by slicing techniques, then selected based on quality. The results showed the average weight of buffalo ovaries of (+CL, +FD); (+CL, -FD); (-CL, +FD); (-CL, -FD) are 7.2 g; 5.5 g; 4.1 g; and 4.5 g respectively. No significant quality difference of produced oocytes between ovaries cycles (P>0.5). Good quality of collected oocytes were only 40-55%. Approximately 2-5 oocytes of grade A and 1-5 oocytes of grade B can be collected per pair of ovaries.
