    Identifikasi resistensi ragam antibiotik terhadap Escherichia coli yang diisolasi dari karkas sapi Rumah Potong Hewan di Banyuwangi

    Tujuan penelitian ini adalah mengidentifikasi resistensi antibiotik dari Escherichia coli pada karkas sapi yang diambil dari rumah potong hewan di Kabupaten Banyuwangi. Sampel dikoleksi dari Kecamatan Banyuwangi, Rogojampi, Genteng dan Kalibaru. Penelitian ini menggunakan kajian cross sectional dengan prevalensi diasumsikan 50% setiap cemaran bakteri, tingkat kepercayaan 95% dan tingkat kesalahan 10%, sehingga didapatkan 96 sampel yang harus diambil untuk diuji. Hasil penelitian menunjukkan tujuh sampel (7.3%) positif E. coli. Hasil resistensi E. coli terhadap antibiotik golongan sefalotin (42,9%), trimethoprim (14,3%) dan eritromisin (42,9%) sedangkan ampisilin, enrofloksasin, kloramfenikol dan tetrasiklin masih dinyatakan sensitive

    Paratuberculosis or Johne’s Disease is a granulomatous enteritis chronic disease of domestic and wild ruminants caused by infection of Mycobacterium avium subspecies paratuberculosis. The disease commonly infects dairy cattle with clinical signs of chronic diarrhea, decreasing body weight, low milk production, oedema, anemia and occasionally infertility. The basic procedure in order to control Paratuberculosis in farms is to do a good and proper handling of animal faecal. Disinfection of animal environments such as pens, faecal, sewerage and sewage are important in prevention of transmission of this disease. The purpose of this research is to determine specific disinfectan and dosage for Mycobacterium avium subspecies paratuberculosis decontamination in cattle feces so  it can be applied as disease control measures. Cow's feces were contaminated with MAP 105CFU/ml and treated with ammonium quartener, phenolic and formaldehyde disinfectant doses 10%, 15% and 20%. The effectiveness of the disinfectant was tested based on MAP identification using Löwenstein-Jensen culture medium and nested Polymere Chain Reaction(PCR). The results showed 15% and 20% doses of formaldehyde disinfectants efective to decontaminate Mycobacterium avium subspecies paratuberculosis  in catle feces

    Deteksi Hemaglutinin, Hemolisin dan Koagulase Secara Fenotipik dan Genotipik pada Staphylococcus aureus Isolat Asal Broiler

    Staphylococcus aureus is a bacterium causing diseases in animals and human. Staphylococcus aureus in broilers cause septicemia, tendosinovitis, dermatitis, endocarditis, wound infections, arthritis and bumblefoot. In this research, 24 isolates of Staphylococcus aureus from broiler were characterized of its virulent factors including the presence of haemagglutinin, ability to agulate the plasma in tubular coagulase test as well presipitate formation in clumping factor test, and haemolysis types. By polymerase chain reaction (PCR) were genotypically detected genes coa, clf, hlaA, and hlaB. All of the isolates (100%) had haemagglutinin, capable to agglutinate and precipitae of rabbit plasma. All isolates could lyse sheep red blood cells with the type of α-hemolysis (45.8%),  β-hemolysis (50.0%) and γ-haemolysis (4.2%). Genotypically, all isolates (100%) had coa and clf genes,  hlaA gene 7(0.8%) and hlaB gene (29.2%)

    Calcitriol supplementation in ovariectomized rats decreased bone resorption and increased bone formation, however, it depend on dose. The objective of the research was to study the response of femur bone in osteoporosis rats consuming calcitriol. Thirty female Wistar rats at 8 weeks of age were randomly divided into six groups (sham operated rats as normal control rats/group N and NK, ovariectomized control rats/group Ov and OvK, ovariectomized rats/group OvDand OvE) of five each. All rats were fed standard diet for 8 weeks. At 16 weeks  of age, group N and Ov  were euthanized, the right femur were taken for histopathological  examination. Group NK and OvK were fed a standard diet, group OvD was fed a standard diet +40ng calcitriol;  and group OvE was fed a standard diet+25µg ethynil ethyl estradiol. Treatments were done for six weeks. At the end of study, blood samples were taken from plexus orbitalis medialis for estrogen analysis. All rats were euthanized using ketamine10% and xylazine 2%. Right femur was taken for histopathological examination  using hematoxylin and eosin stain, and immunohistochemistry using monoclonal antibody anti TRAP5b which was detected with streptavidin-biotin.         The results showed that estradiol level of the rats in group OvD was not significantly different compared with the rats in OvK group, however, it was significanly lower compared to the rats in group OvE. Histopathologic figure of right distal femur metaphysis in group OvD was shown lesser adipocyte in the bone marrow and more trabecular bone speculum compared to group  OvK, however, there was more adipocyte in the bone marrow and lesser trabecular bone speculum compared to group OvE. Immunohistochemistry of distal femur metaphysis in group OvD and OvE were revealed tartrate resistant alkaline phosphatase 5b (TRAP5b) expression in trabecular bone, which was located in bone marrow space and trabecular speculum surface as well. Based on the results, it can be concluded that calcitriol 40ng/day supplementation in osteoporosis rats for 6 weeks decreased bone resorption and increased bone formation distal femur metaphysis

    Aeromonas hydrophila causes a disease that often infects fish and is known as Motile Aeromonas Septicaemia (MAS), Hemorrhagi Septisemia, Ulcer disease or Red-Sore disease. The   aims of this study were to develop polyclonal antibody of  Aeromonas hydrophila in the rabbits to   confirm the diagnosis of Aeromonas hydrophila  in the fish by immunohistochemistry staining method. Preparation of polyclonal antibodies was performed on the rabbits used to Aeromonas hydrophila bacteria that have been tested biochemically by intravenous and intraperitoneal injection. Doses of Aeromonas hydrophila  bacteria were 109 CPU/ml  of 0.5 ml at first injection, 1 ml at second injection, 2 ml at thirth injection and 3 ml at fourth injection. Blood serum collection was performed at week 5 after injection from  an  ear and intracardial vein. The result of antibody titer was 28 = 1024 which measured by   tube test. Furthermore, polyclonal   antibody was used to immunohistochemistry  staining with 400x dilution. The results of the staining showed that an immunopositive reaction in the liver, skin,lien,  gill, kidney, and heart of fish to Aeromonas hydrophila antibody. The research conclution was polyclonal antibody from rabbit can be used to accurately confirm the diagnosis of Aeromonas hydrophila  based on antigen and antibody reaction.

    Investation of Fasciola sp. On several species of cattle in Indonesia has occurred since a longtime ago. Fasciolosis caused a high economical loss such as decreasing of carcass and organs,sometimes caused death of calves. The objectives of the research were to evaluate the effect of fasciolosis on Serum Glutamic Pyruvic Transaminase (SGPT), and the obstruction of bile duct by Alkaline Phosphatase (ALP) in Bali cattle suffered from fasciolosis. Faeces of Bali cattle raised in endemic areas of fasciolosis were examinated by sedimentation test to identify eggs per gram of faeces (EPG) of Fasciola sp. The blood samples of cattle which positive to fasciolosis were use to study the hematology by autoanalyser, SGPT and ALP. The results of research showed that from 89 cattle, 28 were infected by Fasciola sp. whithin of 7-115 EPG (min-max). From 20 fasciolosis cattle were indicated that MCHC of 90%, RBC of 30%, HB of 20%, and eosinophils of 2 % of cattle were lower than that of normal standard, while lymphocytes of 60%, MCV of 40%, ALP of 10% and SGPT of 5% of cattle were higher than that of normal standard. hematological appearances of cattle suffered from fasciolosis had macrocytichypochromic anemia, eosinopenia, and lymphocytosis. The increase of SGPT may be caused byhepatic cells destruction by young liver flukes, and the increase of ALP was caused by the obstruction of bile duct by mature liver flukes

    Sea Cucumber (Stichopus sp) belongs to Holothuroidea family. Sea Cucumber has lot of uses in the medicine sector, such as: for wound healing, anti-biotic, anti-bacteria, anti-tumor, and anti-coagulation. Diabetes mellitus is a disease characterized by high blood sugar levels with metabolic disorders. This disease can cause wounds called gangrene This research has purpose to prove the influence of giving nano spray of sea cucumber extract within 14 days. In the in vitro examination found 40% concentration of sea cucumber extract which is effective to resist the growth of Methicillin Resistant Staphylococcus aureus bacteria and examined to wistar mouse which are already diabetic inducted with Streptozotocin (STZ) and hurt with punch biopsy with the diameter of 0,8 cm on its back and the wound is infected with bacteria which is intradermal injected. Divided into 3 groups : (K-) no medical treatment, nano spray of sea cucumber extract (T), Nano spray penicillin (K+) to wistar mouse towards the speed of diabetic mellitus wound healing which is infected by Methicillin Resistant Staphylococcus aureus bacteria. Parameter used to measure the rate of wound healing is wound size and hitopathology examination.After it’s regularly given the medicine, the result of nano spray sea cucumber extract 40 % concetration category treatment on wound healing process the diameter of wound on the 3rd day 0.8 cm , on the 7th day 0.4 cm and on the 14th day  wound fully recover according to macros (wound size) and histopathology examination shows faster and better healing compared to treatment category of nano spray penicillin and control marked by score cell and lymphocyte in a small quantity

    Antimicrobial resistance is amongst the primary concern in the field of veterinary medicine worldwide. The research was conducted to evaluate the resistance of Escherichia coli due to inappropriate antibiotic treatment. Escherichia coli in this research was cultured in vitro using 5-ml Tryptone Soya Broth (TSB), which was mixed with an antibiotic, then was incubated at 37 0C for four days (stage I). Subsequently, a part of the culture was transferred  into another 5-ml TSB and incubated at 37 0C for four days (stage II). This procedure was undertaken continuously for stage III, IV, V,and VI (until day 24). Antibiotics in this research was oxytetracycline with three various doses, which are lower dose (1 mg/ml), normal dose (2 mg/ml), and higher dose (8 mg/ml). The development of the antrimocobial resistance was evaluated every four days, using disk diffusion method and the data were analysed descriptively. The results showed that the normal and higher doses of oxytetracycline has the same rate (day 16) in causing E. coli resistant to oxytetracycline. Therefore, the treatment of oxytetracycline with a normal and higher doses continuously could accelerate the emergence of antimicrobial resistance in Escherichia coli

    Deteksi Molekuler Virus Infectious Bursal Disease (IBD) pada Samp l Bursa Fabrisius yang Diperoleh dari Ayam Terdiagnosa Penyakit IBD

    Kasus penyakit Infectious Bursal Disease (IBD) dewasa ini masih sering ditemukan pada peternakan ayam komersial baik layer maupun broiler di Indonesia. Diagnosis penyakit IBD sejauh ini mengandalkan lesi patologik spesifik dan kultur in ovo dengan mengamati lesi makroskopis embrio, serta diidentifikasi dengan uji agar gel presipitasi (AGP). Penelitian ini bertujuan menerapkan diagnosis dengan teknik reverse transcriptase polymerase chain reaction (RT-PCR) dari sampel Bursa Fabrisius (BF) sebagai konfirmasi pada kasus terdiagnosa IBD. Deteksi serologis virus IBD dengan uji AGP dengan sumber antigen chorioallantoic membrane (CAM) dan embrionya, untuk melihat potensinya sebagai sumber antigen uji AGP. Sampel BursaFabrisius sebanyak 5 yang diperoleh pada kasus terdiagnosa IBD, dikoleksi dari peternakan ayam komersial di Yogyakarta. Konfirmasi diagnosis dilakukan dengan metode RT-PCR. Sampel positip uji RT-PCR yang mengamplifikasi fragmen gen VP2. Isolasi virus IBD yang dilakukan kultur in ovo pada telur ayam berembrio (TAB) antibodi negatif terhadap virus IBD, berumur 11 hari. Desposisi materi inokulasi dilakukan pada (CAM), diinkubasi selama lima hari. Panen virus dilakukan dengan mengkoleksi membran korioalantois dan embrio, selanjutnya diamati lesi makroskopis yang timbul akibat infeksi virus IBD. Membran korioalantois dan embrioselanjutnya digerus dan diproses sebagai suspensi antigen yang digunakan dalam uji AGP. Hasil uji RT-PCR terhadap lima sampel Bursa Fabrisius yang dikoleksi dari peternakan ayam terdiagnosa penyakit IBD, tiga sampel menunjukkan hasil positif teramplifikasi fragmen gen VP-2 virus IBD dengan produk amplifikasi sebesar 440 bp, sedangkan dua sampel sisanya menunjukkan hasil negatif. Uji AGP dengan sumber antigen CAM menunjukkan hasil positip 2 dari 3 sampel yang diuji, sedangkan sumber antigen embrio menunjukkanhasil negatif. Berdasarkan data yang diperoleh dalam penelitian ini dapat disimpulkan bahwa uji RT-PCR dapat digunakan dalam mendeteksi virus IBD dari sampel BF terdiagnosa IBD. Uji AGP dengan sumber antigen CAM menunjukkan hasil lebih baik dari pada embrionya

    Pengembangan Deteksi Aeromonas hydrophila pada Ikan Nila (Oreochromis niloticus) dengan Metoda Agar Gel Presipitasi di Yogyakarta

    Aeromonas hydrophila telah diidentifikasi sebagai agen penyebab penyakit pada ikan Nila.Mikroorganisme ini diketahui patogen terhadap manusia terutama sebagai penyebab gastroentritis, meskipun demikian, belum dilaporkan adanya infeksi pada manusia di Yogyakarta. Uji karakteristik biokimiawi dapat mengarahkan pada diagnosa yang kurang akurat, oleh karena itu terdapat kebutuhan untuk mengembangkan metode yang lebih spesifik dan akurat, yaitu dengan metode agar gel presipitasi (AGP). Dua sampel yang dikoleksi berasal dari lokasi budidaya ikan Nila di Yogyakarta yang terjadi outbreak A. hydrophila dengan mortalitas lebih dari 50%. Antigen terlarut yang tahan terhadap panas dipersiapkan dari 4 kultur isolat murni yang diperoleh untuk uji AGP. Antiserum yang digunakan untuk uji pada sumuran adalah antiserum A. hydrophila (ATCC 35654) yang diproduksi dengan menginokulasikan whole-cell antigen (heat-stable) dan antigen flagellar (formalin-killed) pada kelinci. Kontrol positif yang digunakan untuk uji ini berasal dari A. hydrophila (ATCC 35654), dan kontrol negatif berasal dari Edwardsiella tarda (E. tarda) ATCC 15947 dan Aeromonas salmonicida (A. salmonicida). Antiserum yang digunakan terhadap kultur isolat murni dan juga kontrol positif menunjukkan adanya reaksi positif saat diujikan menggunakan metode AGP. Hasil reaksi positif adalah dengan adanya pembentukan garis presipitasi diantara sumuran antiserum dan antigen, sedangkan hasilnegatif tidak menunjukkan adanya garis presipitasi tersebut. Hasil dari uji ini menunjukkan bahwa AGP dapat dianggap sebagai metode yang dapat digunakan dan diandalkan (reliable) untuk mengidentifikasi A. hydrophila. Hasil yang sama ditunjukkan melalui uji AGP terhadap antiserum pada isolat A. hydrophila yang berasal dari isolat sampel ikan Nila dari Yogyakarta serta A. hydrophila sebagai kontrol positif (ATCC 35654)
