Identifikasi Faktor-Faktor yang Menentukan Status Keberlanjutan Usaha Tempat Penampungan dan Potong Ayam (TPnA) di Wilayah Pondok Rumput Kota Bogor
Increased broiler production in Bogor Regency leads to the continuous development of traditional chicken collecting facilities and slaughterhouse (CCFS), especially in Bogor City. Pondok Rumput area, located in Kebon Pedes Village, as one of CCFS centers. Pondok Rumput CCFS was first established in 1971. There are currently 25 CCFS in Pondok Rumput. The CCFS in this area are located in densely populated residential area. The CCFS, are located in the middle of the residential area leads to various problems, one of which is environmental pollution.The Study aimed to identify the factors determining the sustainability status of CCFS in Pondok Rumput area, in Tanah Sareal Subdistric, Bogor City. The types of data collected included primary and secondary data. The data analysis used by Multi Dimensional Scalling (MDS) analysis (by using Rapfish software) and prospective analysis. Partial analysis of each dimension concerning the sustainability status of chicken collecting facilities and slaughterhouse in Pondok Rumput showed that ecological dimension were not unsustainable with a value of 24.66. Identifikasi Penambahan Air pada Daging Sapi dengan Metode Filter Paper Press dan Konduktivitas Listrik
Beef meat has a complete nutritional content, the high prices in the market causing some unscrupulous traders to cheat by adding water to meat to get more weight. This beef meat has a bad quality, so needs the right method to identify the quality of beef meat in the field. This study was designed to identify the addition of water to beef meat by measuring the water holding capacity (filter paper press method) and electrical conductivity (EC meter). The experimental used one hundred bovine longisimus dorsi and the samples were divided into five groups. One group as a control and four other groups were treatment group. The treatment groups were injected by water with a volume of 1%, 2%, 3%, and 4% of the sample weight and all groups were repeated for twenty times. The results were analyzed by ANOVA and Duncan Multiple Range test. The data showed that the addition of water by injection in beef meat of treatment group 2%, 3%, and 4% had a significant influence on water holding capacity and electrical conductivity (p<0.05). Pearson correlation results showed a strong to very strong correlation with value in control group (r=0.704; p<0.01), treatment group 1% (r=0.628; p<0.01), 2% (r=0.700; p<0.01), 3% (r=0.642; p<0.01) and 4% (r=0.760; p<0.01). Based on the result of the study, it is known that a water increase in beef at least 2% of the weight significantly affected water holding capacity and it can be identified by using electrical conductivity meter
ISOLASI, IDENTIFIKASI, DAN SEROTYPING Avibacterium paragallinarum DARI AYAM PETELUR KOMERSIAL YANG MENUNJUKKAN GEJALA SNOT
ABSTRAK Infectious coryza (snot) adalah salah satu penyakit pernapasan atas pada breeder, layer dan broiler yang disebabkan oleh Avibacterium paragallinarum (Av. paragallinarum). Penyakit ini sangat merugikan karena mengakibatkan penurunan produksi telur dan morbiditas yang tinggi. Kejadian snot di Indonesia sampai sekarang masih terus dilaporkan. Pemberian vaksinasi adalah salah satu tindakan pencegahan terbaik, namun laporan tentang Av. paragallinarum dan jenis serotipe yang ada di lapangan masih sedikit, sehingga kesesuaian antara jenis serotipe Av. paragallinarum di lapangan dengan yang digunakan untuk vaksinasi belum diketahui. Av. paragallinarum memiliki strain dengan antigenisitas yang berbeda dan hingga saat ini diketahui tiga serotipe yaitu serotipe A, B dan C. Serotipe A dan C bersifat patogen sedangkan serotipe B kurang patogen. Tujuan penelitian ini adalah untuk mengisolasi, mengidentifikasi dan menentukan serotipe Av. paragallinarum dari ayam petelur komersial yang menunjukkan gejala snot. Sampel diambil dari ayam yang menunjukkan gejala snot (eksudat berbau busuk dari mata dan hidung, kebengkakan infra orbital dan pial, konjungtivitis, dan adanya suara ngorok halus) dari beberapa peternakan layer. Sampel dikultur pada media agar coklat kemudian diinkubasi dalam candle jar pada suhu 37 ᴼC selama 18-24 jam. Morfologi koloni dan sel bakteri diamati dan dilakukan uji biokimia (katalase, oksidase, urease, indol, dan fermentasi karbohidrat) pada koloni yang diduga Av. paragallinarum. Uji serotipe dilakukan dengan metode plate agglutination test (PAT). Penelitian ini mendapat hasil 4 isolat Av. paragallianrum dengan 2 isolat merupakan serotipe B dan 2 lainnya serotipe C. ISOLATION, IDENTIFICATION, AND SEROTYPING Avibacterium paragallinarum FROM COMERCIAL LAYER WITH SNOT SYMPTOMS ABSTRACT Infectious coryza (snot) is one of acute respiratory disease in breeders, layers and broilers caused by Avibacterium paragallinarum (Av. paragallinarum). This disease is very harmful because its cause decrease in egg production and high morbidity. Snot incident in Indonesia still be reported. Vaccination is one of the best preventive measures, but reports about Av. paragallinarum serotype at field so lack, so that the correspondence between serotype Av. paragallinarum in the field to those used for vaccination is unknown. Av. paragallinarum has strains with different antigenicity and until now known three serotypes there are serotypes A, B and C. Serotypes A and C are pathogenic then serotypes B. The purpose of this study was to isolate, identify and to determine the serotype of Av. paragallinarum from commercial layer that showing symptoms of snot. Samples taken from layer which showed symptoms of snot (nasal foul smelling exudate, infra-orbital and wattle swelling, conjunctivitis, and snoring) from some layer farms. Samples were cultured on chocolate agar and then incubated in a candle jar at 37 ᴼC for 18-24 hours. Bacteria colony and cell morphology was observed and performed biochemical tests (catalase, oxidase, urease, indole, and fermentation of carbohydrates) in suspected colonies. Serotypes test was conducted using plate agglutination test (PAT). This study revealed 4 isolates Av. paragallinarum with 2 isolates are serotype B and 2 other are serotype C.
Penggunaan Antigen Mycoplasma gallisepticum Freeze-Thawing dalam Teknik Enzyme-Linked Immunosorbent Assay
Chronic Respiratory Disease (CRD) is a bacterial infection in chickens caused by Mycoplasma gallisepticum. CRD may result in economic loss in the livestock industry, decreasing of egg production and feed efficiency, increasing of medication costs, downgrading of carcass and egg qualities. The aim of this study was to develop the Enzyme-Linked Immunosorbent Assay (ELISA) kit for detecting antibodies of M. gallisepticum using freeze-thawing antigens. The cut-off point was determined by S/N method. Chicken antiserum againts M. gallisepticum was collected every week on 1st – 6th week post immunization of whole cell antigen of M. gallisepticum. Optimization of antigen, serum and conjugate were 10 μg/ml protein concentrations, 1:800 serum dilution and 1:10000 conjugate dilution. The validation results on developed ELISA kit (MyGELISA) analyzed by ROC curve using MedCalc statistical software revealed that developed ELISA kit (MyGELISA) had 100% sensitivity and 86.21% specificity with 95% confidence interval. The results of RSA, MyGELISA and commercial ELISA kit showed antibodies positif againts M. gallisepticum. In conclusion, MyGELISA using freeze-thawing antigen can be used to detect antibodies of M. gallisepticum
Karakterisasi dan Identifikasi Bakteri Enterobacteriaceae pada Telur Penyu Lekang (Lepidochelys olivacea) asal Lhok Pante Tibang, Banda Aceh
The population of olive ridley turtle (Lepidochelys olivacea) are currently thretened with extinction, among the contributing factors is the failure of turtle eggs to hatch. The condition can be caused by the presence of bacterial contamination, especially group Enterobacteriaceae. The purpose of this research was to isolate the group of Enterobacteriaceae bacteria from olive ridley turtle. The sample used were shells from olive ridley turtle eggs comes from natural captivity in Lhok Pante Tibang Beach, Banda Aceh. The research parameters included the number of isolates, macroscopic morphology (shape, edge, elevation, and color of isolates), cell morphology (Gram staining and cell shape), and biochemical tests. Isolation method using spread plate and quadrant method. The isolation results obtained five isolates which had varying macroscopic morphology. The five isolates were Gram negative and formed such as trunk. Identification results found that there were three genus from Enterobacteriaceae videlicet two isolates were Enterobacter, two isolates as Shigella and the other were Klebsiella.
Prima Krim sebagai Solusi Penyembuh Luka pada Penderita Diabetes Meli
Number of patients with diabetes mellitus are very large, according to International Diabetes Federation (IDF) in 2013 reaches 382 billion people between the ages of 40-59 years. Diabetic has been characterized as a chronic wounds. The animal blood waste from a slaugther house is still untapped, containing PRP (Platelet Rich Plasma) that can accelerates heal of bone and soft tissue. PRP (Platelet Rich Plasma) was made from the cattle blood waste using centrifugation. The purpose of the study is to prove that the Prima cream can heal the wounds in diabetic patients. Animals used in this study is 20 Wistar rats. Induction of diabetes was using streptozootocin 65 mg/kg and nicotinamide 235 mg/kg. The animal were divided into 4 groups: non-diabetes given placebo (ND-), diabetes given placebo (DM-), diabetic rat given commercial drug were containing chloramphenicol base 20 mg + prednisolone 2.5 mg (DM+), and diabetes given cream PRP at 20% concentration (DM PRP). Parameters used to measure the rate of wound healing is wound size and histopathologic examination. The data were analyzed using Anova One Way at 95% significant rate. PRP DM group had a wound healing faster than ND- group, DM- and DM+. Based on histopathologic examination, such as epitelialization, basal membrane, fibroblasts, collagen tissue, and presence of inflammatory cells, the best curing reaction was showed in DM PRP. Karakterisasi dan Identifikasi Bakteri Enterobacteriaceae Pada Telur Penyu Lekang (Lepidochelys olivacea) Asal Lhok Pante Tibang, Banda Aceh
Enterobactericeae bacteria often contaminating eggs, one of turtle eggs. Turtle eggs contains substances that support for the growth of bacteria. This study is aimed for characterization and isolating genus of Enterobacteriaceae bacteria that originated from turtle egg shells (Lepidochelys olivacea) that obtained at Lhok Pante Tibang, Banda Aceh. The parameters observed were colony of morphology characterization (shape, color, ledges, and elevation of isolates), morphology cells (cell shape and Gram coloring) and physiological test of Enterobacteriaceae genus. From this result we obtained five isolates of Enterobacteriaceae bacteria which have formed such as trunk. The five isolates generally have a colony morphology such as the round shape, red and pink color, wavy and smooth edges, and convex elevation kinds. The results of of physiological test indicate that the five isolates that obtained were belong to three genus of Enterobacteriaceae bacteria which were Enterobacter genus, Shigella genus and Klebsiella genus.Keywords:AbstrakBakteri Enterobacteriacea sering mengkontaminasi telur, salah satunya telur penyu. Telur penyu memiliki zat-zat yang dapat mendukung pertumbuhan bakteri. Penelitian ini bertujuan mengkarakterisasi dan mengisolasi genus bakteri Enterobacteriaceae asal cangkang telur penyu lekang (Lepidochelys olivacea) yang diperoleh di Lhok Pante Tibang, Banda Aceh. Parameter yang diamati meliputi karakterisasi morfologi koloni (bentuk, warna, tepian, dan elevasi isolat), morfologi sel, dan uji fisiologi genus Enterobacteriaceae. Hasil penelitian mendapatkan sebanyak lima isolat bakteri Enterobacteriaceae yang memiliki bentuk sel batang. Isolat tersebut secara umum memiliki morfologi koloni yang berbentuk bundar, berwarna merah dan merah muda, tepian berombak dan licin serta elevasi cembung. Hasil uji fisiologis menunjukkan bahwa lima isolat yang diperoleh terdiri atas tiga genus bakteri Enterobacteriaceae yaitu genus Enterobacter, Shigella dan Klebsiella. Kata kunci:
Identifikasi Serovar Penyebab Leptospirosis pada Anjing di Yogyakarta
Leptospirosis is a zoonotic disease of global concern, and is caused by pathogenic serovar Leptospira interrogans. Canine Leptospirososis is widespread worldwide, dogs can act as incidental hosts or maintenance hosts for various serovars. The purpose of this research was to identify leptospire serovars that infect healthy and suspected leptospirosis dogs in Yogyakarta. A total of 56 dogs (36 healthy dogs and 20 suspect leptospirosis dogs) sera were taken from cephalica vein as much as 3 ml. Sera were examined for leptospirosis with Microscopic Aglutination Test (MAT) which conducted at the Research Center for Veterinary Science, Bogor. Microscopic Aglutination Test carried out on various Leptospire serovar, namely: Ichterohaemorrhagiae, Javanica, Celledoni, Ballum, Pyogenes, Cynopeteri, Rachmati, Australis, Pomona, Canicola, Grippotyphosa, Bataviae, Hardjo, and Tarrasovi. The results showed that Celledoni serovars infected 25% of healthy dogs and 5% of suspect leptospirosis dogs, Javanica serovar infected 19% of healthy dogs, Bataviae serovars infected 15% of suspect leptospirosis dogs, Grippotyphosa serovar infected 11% of healthy dogs, Tarrasovi serovar infected 10% of suspect leptospirosis dogs, serovars Cynopteri infects 5% of healthy dogs and 5% of suspect leptospirosis dogs, serovar Pyrogenes infects 5% of healthy dogs and 5% of suspect leptospirosis dogs, and serovar Rachmati infects 5% of suspect leptospirosis dogs. Seven healthy dogs (19%) and 2 suspect leptospirosis dogs (10%) were infected with more than 2 leptospire serovars. From the results of this study it can be concluded that Celledoni serovar of Leptospira interrogans infection causes subclinical leptospirosis, while Bataviae serovar infection causes clinical leptospirosis in dogs in Yogyakarta
Efektifitas perlakuan awal Tretinoin 0,1% Topikal pada Proses Penyembuhan di Luka Insisi pada Mencit (Mus musculus)
Tretinoin has an important role in wound healing process include improve fibroplasia and collagen synthesis, maintening humoral immunity, and neutralize steroid effect.Epithelization and fibroblast proliferation were very important event in wound healing process. The aims of this study were studying epithelization and fibroblas proliferation on incision wound healing process using pre treatment of Tretinoin 0,1% topically on mice. This research used 24 male mice, 3 month of age, with 150 -180 gram of body weight. The mice were divided into 2 groups, control group (K) and treatment group (P). The control group was a group of mice with an incision wound, while the treatment group was a group of mice with an incision wound that received a pretreatment of Tretionin of 0.1% topically. Each group was divided into 2 sub groups K4, K7, P4, P7 based on time of samples collecting. K4 and P4 were examined at day 4, while K7 and P7 were examined at day 7. Epithelialization was observed by measuring the length of the epithelial incision area, whereas fibroblast proliferation was performed by counting the number of fibroblasts in five field of view on histopathologic skin preparations processed by HE staining.In this study,pretreatment of Tretinoin 0,1% topically can increase epithelialization and number of fibroblast significantly (p<0,05).This increase may be due to 1% Tretinoin in the tissues to induce keratinocyte basal proliferation including its migration to the skin surface via retinoic acid receptors ( RARδ)Pretreatment of Tretinoin 0,1% topically can increased epithelialization and number of fibroblast on incision wound healing process of albino rat
Keragaman dan Intensitas Infeksi Endoparasit Gastrointestinal pada Sapi Bali dengan Sistem Ekstensif di Kabupaten Kupang
Tanah Putih Village has the potential for the development of Bali cattle because it has extensive grazing land and the majority of farmers raise Bali cattle. The aim of the study was to obtain data on the diversity and intensity of gastrointestinal endoparasitic infection in Balinese cattle with an extensive breeding system in Tanah Putih Village, Kupang Timur District, Kupang Regency, East Nusa Tenggara Province. Gastrointestinal endoparasitic diversity was identify using sedimentation and flotation methods while the intensity of endoparasitic infection was carried out using McMaster method, while data on the diversity and rates of gastrointestinal endoparasitic infection were analyzed descriptively. The results of the study on the diversity of gastrointestinal endoparasites found there are five types of nematode worm eggs namely Haemonchus contortus, Oesophagostomum radiatum, Strongyloides papillosus, Bunostomum phlebotomum, and Trichostrongylus axei. Factors that support the diversity of endoparasites are feed sources, livestock populations, and grazing rotation. The average number of worm eggs found based on the total eggs per gram of faeces for Haemonchus contortus as many as 100 eggs and the other four types of worm eggs amounted to 50 eggs. The average number of eggs indicates that endoparasitic infections in these animals fall into the mild intensity category, is influenced by season, feed source, and age of livestock.
