Body Weight, Physiological Status and Volatile Fatty Acid on Kacang and Etawah Crossbreed Goat by Reduction and Refeeding of Feed Quantity
The aimed of this study was to observe the effect of feed restriction and refeeding on body weight, physiological status and blood VFA on Kacang and Etawah Crossbreed Goat. Eight male goats consisting of 4 Kacang Goats and 4 Etawah Crossbred goats were used as experiment objects in this study. Both types of goats were subjected to the same treatment (three phases). First was adaptation, feed was given gradualy until reach the intended amount. Secondly, the feed was reduced by giving as much as 1.7% dry matter of body weight for each; thirdly, both goats were fed on ad libitum (refeeding phase). The variables observed were body weight changes, physiological status and blood VFA. The data obtained were analyzed by Various Random Test using Completely Randomized Design (RAL) on 2x2 factorial patterns. Duncan Multiple Range Test was used to test treatments that significantly different. The results showed that the pattern of changes in body weight on both goats breeds were relatively similar. Physiological status (pulse, rectal and respiratory temperature) on both breed of goats were not significantly different. However, decrease in the amount of feed impacted in the decrease in body temperature, pulsus and respiration, although it was still within normal range. The same also happened on blood VFA levels. Based on the results of the study, it can be concluded that breed differences breed of goat did not affect the physiological condition of goat. Differences in the amount of feed affect the physiological conditions and the performance of goat production
Deteksi Johne’s Disease pada Sapi Potong di Kabupaten Kebumen Berdasarkan Uji Elisa
Kebumen Regency have been set as the source of the beef cattle breeds by the Ministry of Agriculture of the Republic of Indonesia. As the parent stock of beef cattle, these should be free from any infectious disease. Base on Balai Besar Veterinary Wates survey at 2015, some beef cattle in some districts at Kebumen indicated Johne's Disease. This research was carried out with the epidemiologic approach to detect Johne's Disease. About 173 serum specimens were taken from 173 heads of cows who have clinical symptoms of Johne's Disease. Sera were analyzed by ELISA. Base on ELISA result, 38 from 173 serum were seropositive. These cows sample come from local ancestor and there were not Johne’s disease vaccination program in Kebumen. This research proved that Johne's Disease have occurred in this populations which dedicated for parent stock of beef cattle in the District of Kebumen. Disease control program for Johne’s disease should be undertaken in this area to prevent the disease transmition
Prevalensi Strain Avian Pathogenic Escherichia coli (APEC) Penyebab Kolibasilosis pada Burung Puyuh
Avian Pathogenic Escherichia coli (APEC) is a pathogen that causes colibacillosis in poultry, including salpingitis, omphalitis, cellulitis, swollen head syndrome, coligranuloma yolk sac inflammation, and air sacs inflammation. APEC is a zoonotic strain which spread through raw meat and processed meat products of animals and birds. In this research, the isolation and identification of Escherichia coli were done by using selective media MacConkey, Kligger Iron Agar, and Gram staining. Polymerase chain reaction (PCR) was used to analyse genetopically to detect 16SrRNA genes, vt1 genes, and vt2 genes. Thirty one (55,36%) isolates of 56 specimens collected from quail were detected as Escherichia coli. The detection of APEC strains towards 31 Escherichia coli isolates were done by using polymerase chain reaction (PCR) with vt1 and vt2 specific primer. The results showed that 32,26% (10/31) was APEC strains and 67.74% was non-APEC strains. From 10 isolates, 90% had vt1 gene and 10% had vt2 gene. Escherichia coli isolates were found in eyes (32,26%), infraorbital sinus fluid (32,26%), nasal fluid (16,20%), also in lungs, air sacs, ascites, and heart for 3,2% each. The isolates could not be found in the specimens from the skull. As a zoonotic agent, the isolates have an impact on human health.
Mycotoxin Binders Effect on Ovaries of Pregnant Mice Exposed to Zearalenone
Mycotoxin, a secondary metabolite produced by mushroom, has proven to cause various cytotoxic effects to animals. Zearalenone is one type of mycotoxin which is produced by Fusarium graminearum mushroom and has a cytotoxic effect to the reproduction system and cattle productivity by inducing apoptosis in the animal's ovaries, uterus, and placenta. This research tested the potentials of mycotoxin binders in eliminating the effect of zearalenone which can be seen from the expression of caspase 3, the content of malondialdehyde (MDA), and the quantity of corpus luteum in the ovaries of pregnant mice (Mus musculus). Twenty mice were grouped into five different treatments, namely K+ (not exposed to zearalenone and mycotoxin binders), K- (0.1 mg/mouse/day of zearalenone dosage), P1 (0.1 mg/mouse/day of zearalenone dosage and 0.5 mg/mouse/day of mycotoxin binder dosage), P2 (0.1 mg/mouse/day of zearalenone dosage and 1 mg/mouse/day of mycotoxin binders), and P3 (0.1 mg/mouse/day of zearalenone and 2 mg/mouse/day of mycotoxin binders), with 10-days treatment time. The results showed that the potentials of mycotoxin binders could be found on the gradual decline in the expression of caspase 3 in P1, P2 and P3 compared to that in K- (negative control). The results also showed a significant difference in the content of malondialdehyde (MDA) in K- compared to that in K+, P1, P2, and P3. Meanwhile, the quantity of corpus luteum in K- was also starkly different to that in K+, P1, P2, and P3. It can then be concluded that mycotoxin binders are very effective in eliminating the exposure of zearalenone.
MENURUNNYA KANDUNGAN KOLESTEROL TELUR ITIK DENGAN PEMBERIAN BAWANG PUTIH SEGAR (ALLIUM SATIVUM L) DENGAN WAKTU ANALISIS YANG BERBEDA
Telur itik merupakan bahan makanan yang mengandung zat-zat makanan yang lengkap dan seimbang serta mempunyai kandungan protein yang lebih tinggi, akan tetapi bahan makanan ini memiliki kandungan kolesterol yang cukup tinggi. Untuk itu perlu dilakukan inovasi-inovasi tertentu dalam pakan itik agar dapat menurunkan kadar kolesterol, salah satu cara yang dapat dilakukan adalah dengan pemberian bawang putih (Allium sativum L.) segar. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pemberian bawang putih segar terhadap penurunan kadar kolesterol telur itik dengan waktu analisis yang berbeda. Untuk mencapai tujuan tersebut, maka dilakukan suatu penelitian dengan menggunakan 50 ekor sebagai kontrol dengan pemberian pakan lokal tanpa diberikan bawang putih dan 50 ekor diberikan 25 gram/ekor/hari bawang putih segar. Data dianalisis menggunakan T-Test (Steel dan Torrie, 1993), Parameter yang diamati dalam penelitian ini meliputi: kadar kolesterol, trigliserida kuning telur, konsumsi pakan, konversi pakan, produksi telur, bobot telur, penyakit dan mortalitas. Analisa kadar kolesterol dan trigliserida kuning telur dilakukan pada umur penelitian I minggu, III minggu, dan V minggu dengan metode ekstrak ether dan selanjutnya dengan prosedur CHOD-PAP. Hasil penelitian menunjukkan bahwa terjadi penurunan kandungan kolesterol sebesar 152,24 mg per dl (72,19 %) dan trigliserida sebesar 130,56 mg per dl (23,42 %) yang sangat tajam sekali dari minggu pertama ke minggu ketiga. Konsumsi pakan, bobot telur, produksi telur dan konversi pakan tidak dipengaruhi oleh waktu analisis yang berbeda. Hasil penelitian ini menunjukkan bahwa untuk mendapatkan kandungan kolesterol dan lemak yang rendah, cukup diberikan bawang putih segar selama tiga (3) minggu saja
Single Parent Nectarinia jugularis
Both of Nectarinia jugularis parents, male and female, usually raise their offsprings together. They feed the chicks, carry away faecal sac and keep their offspring safe from any threats such as predators. On one day the female did not appear, it was not clear why she did not come back home. Since then, the male took care of the chicks by feeding them, disposing the faecal sac and keep the chicks save all day, even though he had never slept in the nest at night as the female did. The male tried to increase the feeding rate and faecal sac removal to compensate the absence of the female. Parental care by single parent male continued until they leave the nest. The male still fed the chicks after they flew off from the nest
Frekuensi Nafas dan Pulsus Ular Sanca Batik (Malayopython reticulatus) Lokalitas Jawa
Reticulated python (Malayopython reticulatus) java locality is highly demand as pet animal. Respiratory and pulse rate were important physiological data needed for diagnostic references. The aim of this research was to study the repiratory and pulse rate on Java M. reticulatus. Thirthy healthy adult Java M. reticulatus were used in this study. The respiratory rate recorded by counting inspiratory and expiratory movement of the thorax for one minute and pulse rate recorded by heart palpation for one minute, measured at 6-8 AM and at 4-6 PM for five consecutive days. Temperature and humidity were record as secondary data. The data were analyzed using independent t-Test. The respiratory rate in this study recorded in the morning ranged 5-48 breaths/minute averaging 20 breaths/minute and in the evening ranged 7-49 breaths/minute averaging 24 breaths/minute. The pulse rate in the morning ranged 21-68 beats per minute (bpm) averaging 45.5 bpm and in the evening ranged 40-82 bpm averaging 56 bpm. Totally, M. reticulatus having the respiratory rate 5-49 breaths/minute and pulse rate 21-82 bpm. The morning temperature ranged 25-27oC and the evening 26-27.5oC. The range of morning and evening air humidity is 66-75% and 65-70% respectively. The results showed that the comparison of respiratory rate (R) and pulse rate (P) were in normal range namely 1:2.25 in the morning and 1:2.33 in the evening. There were no significant differences (P>0.05) between morning and evening respiratory rate and no significant effect (P>0.05) of temperature and humidity on respiratory and pulse rate. It could be concluded that healthy adult Java M. reticulatus has a respiratory rate 5-49 breaths/minute dan pulse rate 21-82 bpm
PERBANDINGAN GAMBARAN DARAH ULAR SANCA BATIK (Malayophyton reticulatus) LOKAL JAWA DAN KALIMANTAN
Reticulated python (M. reticulatus) Java and Kalimantan locality highly demand as pet animal. Blood profile has an important role on animal health status. This research was aim to study the blood profile comparison of Java and Kalimantan M. reticulatus. Ten blood samples of Java M. reticulatus and 10 samples of Kalimantan M. reticulatus were used in this study. As much as 1 ml blood sample was collected from ventral coccygeal vein of adult healthy M. reticulatus. Blood sample put into tube with EDTA then analized at Internal Department Laboratory, Faculty of Veterinary Medicine UGM. Based on blood samples analize results, adult healthy Java M. reticulatus shows the higher value on parameters of erytrocyte, leucocyte, PCV, MCH, heterophyl, limphocyte and monocyte, while haemoglobin, TPP, MCV, MCHC, azurofil and monosit were lower than Kalimantan M. reticulatus. There were no significant differences (P>0,05) between blood profile of Java and Kalimantan M. reticulatus. It could be concluded that blood profile of Java and Kalimantan M. reticulatus is not different
Kombinasi One-step Reverse Transcriptase Polymerase Chain Reaction (RT-PCR) dan Nucleic Acid Lateral Flow (NALF) sebagai Metode Deteksi Gen env-tm Virus Jembrana Strain Tabanan 1987
Jembrana disease is an acute viral disease in Bali cattle that have short incubation period, high mortality and morbidity rate. Early diagnostic methods are needed to prevent the further spreading of this disease. In this study, we combined One-step RT-PCR and NALF methods to detect env-tm gene of Jembrana virus Tabanan 1987 strain. Viral RNA isolated from a spleen of the infected cattle was used as the template. One-step RT-PCR procedure was performed using oligoprobes labeled digoxigenin and right primer sequence that were designed using the primer3plus program based on the conserves region of env-tm gene from NCBI database. The product of one-step RT-PCR was tested using NALF method instead of electrophoresis. Positive result was shown by the appearance of dark lines on the test line of digoxigenin in NALF dipstick device
