    Glycerol Reduces Cross Hybridization on Nitrocellulose Membrane

    Lateral flow assay (LFD) based nucleic acid lateral flow (NALF)  method has been developed recently. The method met point of care testing (POCT) as simple and rapid procedures, less equipment, and can be performance by less skilled technician. NALF based on nucleic acid hybridizationis  more economical then immunochromatography assay which use antibody-antigen recognition. Cross hybridization has issued while used to differentiate organism with high GC content and high homology as high similarity genome. Some techniques has applied to give high stringency condition avoid cross hybridization reaction but need more procedure to apply. We found glycerol applied to buffer assay could reduce cross hybridization on nitrocellulose membrane. The study used 2 kinds of high stringency buffer as PBS and SSC bases and high concentration of ssDNA amplicon as sample. Without glycerol ingredient gave cross hybridization signal on test line. But used glycerol could reduce those even omitted with PBS based buffer assay. Beside those, glycerol could significantly increased hybridization signal in SSC based buffer assay (p<0.05)

    Chocolate poisoning has long been recognized as a common cause mostly in dogs, although many species are susceptible. Contributing factors include indiscriminate eating habits and readily available sources of chocolate. In general, the poisoning resulted from a lack of public knowledge of the health hazard to dogs that may be imposed by these products.Chocolate is derived from the seeds of the plant Theobroma cacao, and the main toxic components are the methylxanthine alkaloids theobromine and caffeine, causing gastrointestinal, cardiovascular and central nervous signs. Diagnosis is based on history of exposure, along with clinical signs. Amphetamine or cocaine toxicosis, and ingestion of antihistamines, antidepressants, or other CNS stimulants should be considered in the differential diagnosis. Stabilization of symptomatic animals is a priority in treating chocolate toxicosis. Although there is no specific antidote, supportive management includes induction of vomiting and administration of activated charcoal, oxygen, and intravenous fluids. Preventing exposure is the key to reducing the incidence of these poisoning episodes. Therefore, it is important to increase the knowledge of dogs owners with regard to foodstuffs that must not be fed to dogs and should be stored outside their reach

    Banda aceh which part of the area is a coastal area and lowland, is an endemic area of dengue hemorrhagic fever (DHF). The case of DHF in Banda Aceh is always fluctuating from year to year so it needs to be analyzed the relationship of the environment to the distribution of DHF. The purpose of this study was to analyze the relationship between settlement density and settlement area to the distribution of DHF in Banda Aceh City. This research uses visual interpretation and overlay methods using quantitative descriptive research design. Descriptive research was conducted using a survey method based on a cross-sectional study. The survey was carried out on the density of settlements and was associated with the incidence of DHF spatially. The type of data used is primary data obtained through direct observation using the Global Positioning System (GPS) to see the distribution of DHF and interpretation data of remote sensing images to see patterns of settlement density. While secondary data from the Banda Aceh City Public Works and Spatial Planning Office and the Banda Aceh City Health Office. The number of samples was all DHF sufferers in 2017 totaling 236 people spread in the city of Banda Aceh. Data were analyzed using ArcGIS and processed statistically using Chi-Square. Sparse residential density areas of 46.7% have 3 points of high category DHF cases, areas of medium settlement density of 34.4%have 7 points of high category DBD cases and areas of densely populated densities of 18.9% have 5 points of high category DBD cases. Statistical test results showed a value of P> 0.05, meaning that there was no relationship between settlement density and the incidence of dengue cases in Banda Aceh City. There is no relationship between the density of settlements with the area of settlements to the distribution of dengue hemorrhagic fever (DHF) in the city of Banda Aceh

    Antibiotics are substances that capable of inhibiting the growth of or killing microorganisms. The presence of residues in the tissues are associated with continuous use of antibiotics for long periods of time. The antibiotic residue that is often detected in freshwater fishery products is tetracycline. One method of analysis of tetracycline residues in fish meat is widely developed using high performance liquid chromatography (HPLC). This research was aimed to validate the method of analysis of tetracycline content in tilapia meat by using HPLC Shimadzu 6.1. The mobile phase consisting of methanol: acetonitrile: oxalic acid (5:15:80) with 1 ml / min flow rate, detector UV Vis with wavelength 355 nm, and C18Shim-pack column size 150 L x 4,6 mm at temperature of 30oC. The result of this research showed values corresponding to validation criteria based on parameters of specificity, precision, accuracy, linearity, limit of detection, and limit of quantification. Tetracycline analysis using HPLC tool has good and acurate validity as the first step in detecting the tetracycline level in tilapia meat

    Melamine, Asam Suanurat dan Melamin-Sianurat: Kaitan dengan Penyakit Saluran Perkencingan Hewan

    Numerous cases of acute renal failure in dogs and cats from 2004-2007 were associated with the ingestion of dog and cat pet food products and the suspected contaminant was identified as melamine and its derivatives such as cyanuric acid and melamine cyanurate. Melamine is a chemical used primarily for the manufacture of plastics, coatings, commercial filters, adhesives and compounds in the plate-making industry and kitchen appliance.But then these compounds have been abused to increase the content of nitrogen compounds in human food products and animal feed products, respectively. Increasing the content of nitrogen compounds from melamine aims to increase the value of false protein food and feed. Melamine basically has a wide security limit so it is relatively non-toxic if consumed by humans and animals. However, many studies and case reports show that melamine can cause damage to the kidneys especially if present together with cyanuric acid in the body.This paper aims to provide a simple overview and discussion in cases of damage to the urinary tract in dogs and cats that are likely caused by toxicity of melamine and its derivatives are likely to have occurred in Indonesia.

    This study was aimed to determine the effect of insulin supplementation in maturation and/or culture medium on nuclear maturation rate and the early bovine embryo development in vitro. Oocytes were collected and matured in maturation media without (IVM I-) or with (IVM I+) 10 ug/µL insulin at incubator 5% CO2 and the temperature of 39 °C, for early embryonic development evaluation, oocytes were divided into 4 groups, without the supplementation of insulin to the maturation medium and culture (IVM I-/IVC I-), insulin supplementation only the maturation medium (IVM I+/IVC I-), insulin supplementation only in the culture medium (IVM I-/IVC I+), and the combination of insulin supplementation to the maturation medium and culture (IVM I+/IVC I+). The result showed that supplementation of insulin to the maturation medium increased number nuclear maturation was higher 87.7% (P<0.05) compared to treatment without supplementation of insulin (70.1%). Cleavage rate in treatment IVM was higher IVM I-/IVC I- (55.8%), IVM I+/IVC I- (64.1%), IVM I-/IVC I+ (59.9%) (P<0.05). Result of the other were showed that early bovine embryo on day-4 cultured (IVC) reached 16 cell on treatment IVM I-/IVC I+(31,9%) and IVM I+/IVC I+ (27,1%) were higher compared to treatment IVM I-/IVC I-(2.9%) and IVM I+/IVC I- (2.5%) (P<0.05). In conclusion, supplementation of insulin to maturation medium and culture medium can increase nuclear maturation rate and improved early embryo cleavage rate

    A. galli infection caused high economic losses and decreased the amount and quality of egg production. The existence of anthelmintic resistance lead to the development of research on alternative treatments for A. galli. This aim of the research was to know LC50 of Areca catechu on infestation of A. galli. In this research A. galli was taken from a chicken slaughterhouse in Terban, Yogyakarta, and was divided into 8 groups for in vitro research. Group I was treated with an 10% infusion of A. catechu; group II was treated with 12.5% infusion of A. catechu; group III was treated with a 15% infusion of A. catechu; group IV was treated with a 17.5% infusion of A. catechu; group V was treated with 20% infusion of A. catechu; group VI was treated with a 22.5% infusion of A. catechu; group VII was treated with 25% A. catechu infusion and group VIII as negative control (0.9% NaCl). A. galli mortality is recorded every one hour until worm mortality is 100%. The results were then analyzed using the Reed and Muench method. Based on the in vitro test the infusion of Areca catechu against Ascaridia galli can cause death in worms with an LC 50 calculation of 21. 18%

    Distribusi neuron serotonergik pada hipokampus Lasiwen (Myotis sp) sebagai satwa liar yang berpotensi menjadi reservoir virus rabies

    Myotis sp is an Indonesian microchiropteran, insectivore bat that potential to be a reservoir for rabies virus. Rabies is fatal viral encephalitis and  bat could  act as wildlife potential reservoirs for the rabies virus and can transmit the disease to humans as the zoonotic disease. Hippocampus  is one of diagnostic tissue for rabies disease and serotonergic neuron could involved in pathogenesis of rabies disease.  The aim of the study is to investigate the distribution of serotonergic neurons in  Myotis sp hippocampus. Five Myotis sp. were captured from wild population in Central Java, and were humanly anesthetized using ketamine and xylazin. The animals were perfused intracardially using NaCl 0.9% as the pre-rinse followed by 10% formaldehyde to fix it. The cerebrums were  collected and processing the  for paraffin embedding. Cerebrums were sectioning in saggital sections,  12 µm thickness serially with 120 µm intervals.    The tissues  were staining immunohistochemistry using antibody to serotonin (1/300; Bioss, Cat. No: bs-1126R) 2 night incubation in 4 oC temperature. The solution for blocking background, secondary antibody, avidin-biotin-peroxidae complex and chromogen using  kit Starr Trek Universal HRP Detection System (Biocare Medical, Cat No: STUHRP700) and were analyzed descriptivelly. The results show that serotonergic neuros were distributed in the all area of  the of Myotis sp hippocampus. In  dentate gyrus neuron serotonergic (Sert-IR)s are round in shape and  mostly distributed in the middle layer, few in the  superficially also deeper layers. In the hippocampus , the Sert-IR neurons are pyramidal in shape and distributed in the CA1, Ca2 and CA3 areas. In subiculum, the Sert-IR neurons  are pyramidal in shape, more wider distributed than in the CA1 with no differences between outer layer and deeper layer.  The conclutions of this research are the serotonergic neurons are distributed in the all area of hippocampu

    Pengaruh Pemberian Serum Albumin Manusia Terhadap Peningkatan Kadar Albumin dalam Darah pada Anjing Penderita Hipoalbuminemia

    Albumin terdiri dari 50% total protein plasma dan merupakan protein utama yang sangat penting di dalam tubuh hewan. Hipoalbuminemia dapat terjadi akibat berbagai kondisi, seperti kehilangan protein nefropati, kehilangan protein enteropati, diare kronis dan akut atau perdarahan berat, serta kondisi malnutrisi dan malabsorbsi. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian serum albumin manusia 20% intravena terhadap kadar albumin dalam darah anjing penderita hipoalbuminemia. Hewan yang digunakan dalam penelitian ini adalah 5 ekor anjing sakit yang hasil pemeriksaan kadar albuminnya menunjukkan di bawah batas normal atau hipoalbuminemia. Anjing sakit yang menderita hipoalbuminemia akan diberi serum albumin manusia 20% melalui infus intravena selama ± 4jam.  Sehari setelah diterapi, dilakukan pengambilan darah untuk diperiksa kadar albumin. Hasil yang diperoleh berupa data pemeriksaan kadar albumin dalam darah sebelum dan sesudah diterapi akan dianalisis dengan T-tes berpasangan. Hasil analisis menunjukkan bahwa terjadi peningkatan kadar albumin secara signifikan dari rata-rata 1,98 ± 0,356 menjadi 2,28 ± 0,257 (P< 0,05) setelah diterapi serum albumin manusia. Penelitian ini menunjukkan bahwa pemberian serum albumin manusia melalui infus intravena mampu meningkatkan kadar albumin pada anjing sakit penderita hipoalbuminemia

    Uji Ekstrak n-Hexana Rumput Kebar (Biophytum petersianum Klotzsch) pada Tikus Wistar Hiperkolesterolemia

    This study aims to determine the secure level of the n-hexane extract of kebar grass on hypercholesterolemia rats based the acute oral toxicity test. Based the literature, no report on the toxic dosage before, initial dose was started at 300 mg/kg body weight (BW) and to be increased 2000 to 5000 mg/kg BW. Toxicity data were set for 24 hours and continued until 14 days after treatment for several parameters, i.e : toxicity symptoms, death of animals, changes in body weight and the manifestation of toxicity effects. At the end of the test, the rats were sacrificed and organs were taken for abnormalities (macroscopy). The results of this study showed that the administration of the n-hexane extract of grass kebar at all doses did not show the toxicity symptoms, mortality and body weight change. The absolute and relative weights and gross pathology observation of the internal organ were normal. Conclusions: the n-hexane extract of grass kebar is safe and the LD50 was determined on category 5, unclassified or minimum practically non toxi
