    Resistensi antibiotik merupakan isu yang genting dijadikan topik penelitian beberapa tahun belakangan ini, dimana salah satu penyebab terjadinya resistensi antibiotik adalah penggunaan antibiotik secara terus menerus dengan dosis yang tidak tepat. Salah satu alternatif yang memiliki peluang untuk melawan bakteri resisten adalah dengan menggunakan bakteriosin. Bakteriosin merupakan senyawa protein yang secara biologis aktif, memiliki berat molekul kecil dan memiliki sifat antibakteri yang umumnya bersifat bakterisidal. Penelitian ini dilakukan dengan metode kajian pustaka dari literatur yang sesuai dengan topik penelitian. Hasil penelitian menunjukkan bahwa bakteriosin banyak dihasilkan oleh bakteri asam laktat, salah satunya dari genus Lactobacillus, seperti L. paracasei, L. plantarum, L. coryniformis, dan L. crustorum, yang memiliki karakteristik menguntungkan seperti stabil terhadap pH dan suhu dan mampu bekerja pada spektrum luas dengan menghambat pertumbuhan bakteri Gram-positif dan Gram-negatif yang resistan terhadap antibiotik, dan pada umumnya memiliki mekanisme antibakteri melalui pembentukan pori-pori yang akan mempengaruhi permeabilitas membran sel dan pada akhirnya menyebabkan kematian se

    POLA PEMELIHARAAN BURUNG WALET PADA PULAU-PULAU UTAMA PENGHASIL SARANG BURUNG WALET DI INDONESIA

    Burung walet di Indonesia umumnya dibudidayakan pada rumah burung walet (RBW) secara tradisional dengan pola pemeliharaan tertentu. Pola pemeliharaan menjadi salah satu faktor pendukung bagi burung walet untuk memproduksi sarang burung walet (SBW) dengan kualitas baik secara berkelanjutan. Penelitian ini bertujuan untuk menganalisis pola pemeliharaan burung walet di RBW di pulau-pulau utama penghasil SBW di Indonesia. Suatu survei terhadap total 44 RBW di pulau Jawa, Sumatera, Sulawesi, dan Kalimantan telah dilakukan untuk mengetahui pola pemeliharaan di masing-masing pulau tersebut. Data dikumpulkan melalui wawancara secara langsung dengan menggunakan kuesioner terstruktur. Pertanyaan dalam kuesioner terdiri atas karakteristik bangunan, kebersihan, sumber makanan dan udara, dan lingkungan RBW. Hasil penelitian menunjukkan bahwa bangunan RBW umumnya bertingkat, dengan atap beton, dinding bata semen, lantai plester semen, dan sirip kayu. Rumah burung walet dibersihkan dibersihkan dengan cara digores/disapu dalam waktu kurang dari dua bulan. Kotoran burung walet umumnya digunakan untuk kebutuhan sendiri. Rumah burung walet umumnya tidak menyediakan bahan untuk menarik serangga atau pakan tambahan. Pakan burung walet umumnyaHymenoptera , dan sumber airnya adalah kolam di dalam gedung RBW. Lingkungan RBW merupakan kawasan pemukiman dan dekat dengan jalan raya. Pembinaan dan pemantauan terhadap pola pemeliharaan burung walet perlu terus dilakukan untuk mendapatkan SBW yang berkualitas baik.

    The Vermicidal Activity of Water Extract Two Types of Ethnopharmacology Against Haemonchus contortus in Kacang Goat (Capra hircus) In-vitro

    This research aims at determining the differences in the vermicidal activity between EADMAN and EADMDV in-vitro based on concentration and treatment time. It encompassed three groups of concentrations of EADMAN and EADMDV solution (2.5%, 3.5%, 4.5% from 0.2 g/mL each extract solution), a group of negative control (aqua pro injection), and a group of positive control (Oxfendazole 0.055%). The EADMAN and EADMDV was diluted in 1.5 mL aqua pro injection, then placed on petri dish with a concentration of 2.5%, 3.5%, 4.5% of 0.2 g/mL extract solution. Eight worms placed on each petri dish afterwards. The effectivity of EADMAN and EADMDV was determined based on the percentage of dead worms. The dead and live worms was counted from each immerse group for 1, 3, 5, 7 hours with four replications for each treatment. The vermicidal activity was indicated by touching the worms using a spatula; if it did not react, it was put in warm water. If the worms did not move, it was declared as the dead one, otherwise it was paralysed. Data of effective concentration from both extracts against vermicidal activity was analyzed using a two-way Anova, followed by Tukey test (P>0.05). The result showed the vermicidal activity of 2,5% EADMDV was significantly greater than 4.5% EADMDV, the negative control, and EADMAN at all concentrations. Meanwhile, its difference to 3.5% EADMDV and positive controls revealed insignificant results statistically. Therefore, 2.5% concentration of EADMDV with 7 hours of immersions gave the most effective vermicidal activity against Haemonchus contortus

    Infection of Infectious Spleen and Kidney Necrosis Virus and Viral Nervouse Necrosis in Seawater Fish Identifed Using Multiplex Polymerase Chain Reaction

    Indonesia become a maritime country with large of ocean area and beach line with marine species diversity reaches 37% of the world’s fish. This makes Indonesia have aquaculture potential that supported by an appropriate climate. However, the challenge of viral diseases caused by Infectious Spleen and Kidney Necrosis Virus (ISKNV) and Viral Nervous Necrosis (VNN) would hamper marine aquaculture and causing huge economic losses. Therefore, it is necessary to carry out early detection methods that are efficient, fast, precise and accurate to identifying these viral disease. Here we developed a multiplex-PCR (mPCR) a method that can detect simultaneously of the ISKNV and VNN. The results of this method showed high sensitivity and specificity by using MCP and CP gene target primers to detect ISKNV and VNN, respectively. From a total of 353 samples of seawater fish examined by mPCR, positive results following of single infection of ISKNV and VNN were 14 and 2, respectively. We also found that 18 grouper fish was co-infection with these viruses. From these results, it shows that the mPCR method developed has efficiency, faster and high accuracy. This founding was possible to be applied in laboratory testing or early detection system in the field for ISKNV and VNN.

    Combination of Wild ginger, Honey, and Probiotics as Growth Prometer Candidates : An in Vitro Study

    Growth Promoter Antibiotics are used to prevent disease and promote growth and production in poultry. Repeated admnistration of feed can have a microorganic resistance effect, accumulation of antibiotic residues in animal and environmental products and imbalance of normal microflora in the intestine. The antibacterial and carbohydrate content of some natural ingredients can be potential as a replacement candidate for AGP. This study aims to determine the role of a combination of wild ginger, honey, and probiotics (Bacillus subtilis and Lactobacillus acidophilus) as AGP candiate in Vitro. The antibacterial activity of the combination of wild ginger and honey against pathogens (E. coli) and their use against probiotics was tested by disk diffusion method, while the calculation of optical density values to determine the minimum inhibitory concentration and minimum bactericidal concentration was carried out on E. coli. The ability of inhibition of probiotics against pathogens is also done by the disk diffusion method. The disk diffusion test results showed the best combination was extract of 25% wild ginger aquades + 100% Lombok honey with inhibition zone diameter (8,53 ±, 03). Optical density values indicate this combination is able to inhibit and kill E. coli (DO 0,00 ±, 002) and support B. subtilis (DO 0,18 ±, 002) and L. acidophilus (DO 0.25 ±, 005) significantly better than positive control. MIC value of wild ginger aquades extract and honey combination against E. coli is wild ginger aquades extract 3.13% + Lombok honey 25%, while MBC value is wild ginger aquades extract 6,25% + Lombok honey 25%. The combination of B. subtilis and L. acidophilus showed the largest inhibitory zone diameter against E. coli (7,30 ±, 02 mm) compared to individual colonies. The combination of wild ginger and honey, in addition to inhibiting also able to kill pathogens and support the growth of probiotics, so this formula can be used as one of the replacement candidates for AGP. 

    Study of Genetic Diversity of Native Indonesian Catfish Based on Nucleotide Sequences of the ND1 Gene

    One of the native catfish to Indonesia is the baung fish (Hemibagrus). Baung fish are found in rivers in Sumatra, Kalimantan and Java. The catfish population is declining, thus conservation is needed to prevent extinction. To conduct an effective conservation efforts, molecular studies are needed to confirm the fish species from the three islands. Mitochondrial DNA is one of the markers that is often used to see the lineage and kinship of animals for conservation purposes. The purpose of this study is to determine the genetic diversity of the NADH Dehydrogenase Subunit I (ND1) gene catfish from each of these locations, can be used as a genetic marker. Samples were obtained from their natural habitat, namely Magelang (5), Palembang (3), Riau (2), Samarinda (2), 2 from Sintang, and 3 from Banjarmasin. The DNA of the fish sample was then isolated and then used as a template for amplification of DNA fragments using PCR techniques. Amplikon (PCR product) was then purified by gel extraction and then sequenced to determine the DNA sequence. The potential of DNA sequences as catfish genetic markers was proven by analyzing genetic diversity between species using the MEGA version 7.0 program (Kumar et al., 2016). The sequencing results 972 nucleotides composing the ND gene, and found differences in 268 nucleotide sites and 47 amino acid sites. Based on the nucleotide sequence of the ND1 gene, catfish from the Progo river (Magelang, Central Java), the Musi river (Palembang, Sumatra), the Kampar river (Riau), the Kapuas river (Sintang, Kalimantan), the Martapura river (Banjarmasin), the Mahakam river (Kalimantan) is included in the Hemibagrus sp.; catfish from the Elo river (Magelang, Central Java) belongs to the genus Mystus sp.; catfish from the Bengawan Solo river (Bojonegoro, East Java) belongs to the Pangasius sp

    Avian Influenza (AI) is a strategic communicable and zoonotic disease. The cause is a virus with Highly Pathogenic Avian Influenza (HPAI) subtype H5N1. The poultry market has important roles in the preservation, propagation, and spreads of the Avian Influenza (AI) virus from poultry to other species and humans. The purpose of this study was to determine the level of seroprevalence of Avian Influenza (H5N1) in free-range chickens at the Beringkit and Galiran market. A total of 120 free-range chickens were used as a sample. They have taken 60 serum samples per market. Serum removal is made from 5 merchants under the provisions of 3 samples from merchants who sell 6 to 10 free-range chickens. The sampling period is carried out for 2 months every 2 weeks 4 times. Sample testing was performed at the Denpasar Veterinary Centre with Haemagglutination (HA) and Haemagglutination Inhibition (HI) as barriers. The data titer of the antibodies obtained was analyzed by Non-Parametric Statistic Test Chi-Square (χ2) using IBM SPSS for windows. The results of the study showed that the AI subtype of the H5N1 subtype in both Beringkit Markets is 3.3% and Galiran Market is 6.7%, with seroprevalence in the two markets of 5.0% which is statistically not dissimilar (P<0.05). To prevent the transmissions of AI disease at the Beringkit Market and Galiran is recommended for vaccination and more attention to the market management and the free-range chicken maintenance system

    Anemia adalah terjadinya defisiensi eritrosit atau hemoglobin atau keduanya hingga kemampuan darah mengangkut oksigen berkurang dan sering dijumpai pada anjing. Anjing lokal  betina bernama Pogan berumur 1 tahun dengan riwayat enterektomi diperiksa dengan keluhan; lemas, kurus, nafsu makan dan minum menurun. Hasil pemeriksaan fisik;  mukosa mulut dan mata anemis, dehidrasi dan kurus. Hasil pemeriksaan hematologi rutin, total protein plasma (TPP) dan albumin menunjukkan bahwa anjing mengalami anemia mikrositik hipokromik, trombositopenia, limfositosis, terjadi penurunan nilai TPP dan hipoalbuminemia. Berdasarkan anamnesa, pemeriksaan fisik dan laboratorik anjing Pogan didiagnosis menderita anemia mikrositik hipokromik yang diakibatkan oleh defisiensi zat besi (Fe). Pengobatan dengan menggunakan preparat Fe dengan dosis 1 ml/5kgbb, subcutan selama 5 hari dan terapi suportif berupa infus serum albumin manusia dengan total volume 128 ml melalui infus intravena selama ±4 jam dan Livron (Vitamin B complex) 1tab/hari, per oral selama 5 hari memberikan hasil yang baik

    Suplemen Calcitriol Dosis Tinggi Mencegah Osteoporosis dan Memicu Urolitiasis pada Tikus Ovariektomi

    Calcitriol supplementation in ovariectomized rats for 8 weeks reduce osteoporosis risk and safe for kidney depend a proper dose. The objective of the research was to study the efectivity of high dose calcitriol for osteoporosis prevention and safe for kidney  in ovariectomized Wistar rats. Fifteen female Wistar rats at 8 weeks of age were randomly divided into three  groups of five which were sham operated rats (KN), ovariectomized rats (KOV), and ovariectomized rats+calcitriol supplementation 200ng/day (OVD200). After 8 weeks of treatment, blood samples were taken from plexus orbitalis medialis for estrogen analysis. All rats were then euthanized, left tibia and left kidney were taken for histopathological examination using hematoxylin and eosin stain. Immunohistochemistry using anti TRAP5b monoclonal antibody was also done for left tibia by streptavidin-biotin.  The results showed that estradiol level of KOV was significantly decreased compared with KN (P<0.05), meanwhile, estradiol levels of OVD200 were not significantly different compared with KOV. Histopathologic figure of kidney in KOV was not different compared with KN. Glomerulus was surrounded by Bowman‘s capsule, proximal and distal convoluted tubulus were lined with typical cuboidal epithelium. Immunohistochemistry result of proximal tibial epiphysis in KOV were shown more tartrate resistant alkaline phosphatase 5b (TRAP5b) expression in trabecular bone, which was located in bone marrow space, and trabecular speculum surface as well. Meanwhile, proximal tibia epiphysis in OVD200 had less tartrate resistant alkaline phosphatase 5b (TRAP5b) expression compared with KOV. Histopathologic figure  of proximal tibia epiphysis in group OVD200 was shown more trabecular bone speculum and less adipocyte in the bone marrow compared with KOV. Histopathologic figure of kidney in OVD200 was showed glomerular  hypertrophy and atrophy, as well as protein deposits in proximal convoluted tubules epithelium.  In conclusion, calcitriol supplementation 200ng/day in ovariectomized rats for 8 weeks prevented osteoporosis risk but trigger urolithiasis. Key words :  : Calcitriol, osteoporosis, ovariectomized rats, urolithiasi

    Brucellosis is a Brucella bacterial infectious disease that is spread from animals to humans, generally through consumption of milk, especially unpasteurized milk, or other dairy products. Veterinary public health has an important role in preventing transmission of disease to humans through both animals and foodstuffs of animal origin or other animal origin ingredients. Brucellosis is one of the problems often faced by the Animal Husbandry Department, especially at the Animal Health Unit. Handling and controlling about Brucellosis is very vital to maintain animal health and reduce the level of economic losses of farmers due to Brucellosis experienced by livestock owned. This study aims to determine the spread of Brucellosis disease by mapping in the Administrative and Service Coordination areas, to know the handling and control actions carried out by the Animal Husbandry Service throughout the implementation of the Administrative Coordination Office and Veterinary Public Health. The results of the mapping of the spread of Brucellosis in the District of Veterinary Public Health and Public Service Administration Coordination can be divided into 3 groups with the number of cases 66.
