What are the current trends in G protein-coupled receptor targeted drug discovery?
G protein-coupled receptors (GPCRs) constitute a superfamily of membrane proteins which have in common the presence of 7 transmembrane α-helices linked by several intra- and extracellular loops. This particular structure is important for extracellular stimulus recognition through the amino acids forming the extracellular loops (class B, C and F of GPCRs) or by the transmembrane α-helices (class A and B of GPCRs). Their structure is also essential for signal transduction via intracellular proteins (G proteins, arrestins and kinases – GRKs-) [Citation1]. These proteins are attached to the intracellular loops and/or to the intracellular regions of the α-helices of the GPCRs, even when they are not activated by extracellular ligands or drugs

A study protocol for an experimental study for a pharmaceutical care programme to improve chronic complex disease management: PCAF programme
The prevalence of elderly patients with chronic illnesses, associated polypharmacy and a high risk of functional and cognitive impairment has increased in recentyears. The care of Complex Chronic Patients (CCP) therefore represents a challenge for health systems in developed countries around the world. This paper presentsa study protocol that will explore the effects of an intervention within a pharmaceutical care programme (PCAF) on optimising CCP care and on the suitability ofthe organisational methods proposed for the PCAF programme. This is a multicentre experimental study. The setting will be primary health care and community pharmacies in the so-called ‘autonomous community’ of Cataloniain Spain. The participants will be CPP attended to Primary Healthcare Centres with the CCP code in their electronic medical record. The included CCP will bedistributed in two strata depending on whether they will be considered likely to receive the arranged medicinal treatment as Monitored Dosage System accordingto benefits based on criteria or not. They will then be assigned by random stratified sampling at a ratio of 1:1 to the control or intervention groups. A sample size of860 patients is estimated. Patients will undergo 12 months follow-up. The primary outcome of this study will be the medication adherence measured by the MoriskyGreen Test and the billing/prescription ratio
Self-assessment scale of basic mental health action competencies: development and evaluation of the psychometric properties
Background: The assessment of adolescents' mental health competencies requires validated instruments. Thus, we sought to develop and evaluate the psychometric properties of a scale in which adolescents self-assess their competencies to act in mental health. Methods: A psychometric study was conducted. The data was collected in Portugal between May 2022 and September 2023. Firstly, a focus group was held with 9 experts to generate the scale items. The data was examined using content analysis. Secondly, a modified e-Delphi technique was used to obtain the opinion of 97 healthcare workers on the scale statements and the classification method. An analysis of response frequencies was carried out. Thirdly, cognitive interviews about the scale were conducted with 8 adolescents. Content analysis was performed. Finally, the scale was applied to 514 adolescents. Item sensitivity assessment and factor analysis were performed. The scale's convergent and divergent validity, internal consistency and test-retest reliability were also examined.
Results: A 20-item scale emerged, featuring four factors: perception of knowledge about mental health, mental health self-care, interpersonal relationships and mental health first aid. Cronbach's alpha and McDonald's omega varied from 0.79 to 0.92, the composite reliability from 0.70 to 0.89, and the intraclass correlation coefficient from 0.77 to 0.80. Conclusions: The scale was created and demonstrated good validity and reliability. It facilitates the assessment of adolescents' mental health needs and the results of educational interventions
Preservice Secondary School Teachers’ Knowledge and Competencies When Reflecting on the Incorporation of Gamification in the Teaching of Mathematics
This study analyses how preservice mathematics teachers reflect on the incorporation of gamification in their teaching practices after participating in a training cycle focused on this active methodology. The cycle, applied to 31 students in the Secondary Mathematics Teacher Education Master’s programme in Catalonia, integrated game elements—such as narratives, quests, and badges—alongside the Didactical Suitability Criteria as a tool for developing competency in the analysis and evaluation of didactical suitability. The reasons preservice teachers provided for using gamification in mathematics teaching were analysed at two points: upon completing the training cycle and after implementing teaching proposals incorporating gamification with secondary education students in Catalonia. The results show that preservice teachers show an improvement in their reflective competency regarding gamification, as well as increased use of this active methodology. Additionally, the knowledge and competencies considered relevant by preservice teachers for working with gamification in mathematics teaching were identified. It is concluded that training in the use of gamification in mathematics teaching is necessary to apply this methodology appropriately, and that tools such as the Didactical Suitability Criteria are fundamental for reflection on, and improvement in, the use of active methodologie
Cortical macro- and microstructural changes in isolated rapid eye movement sleep behavior disorder
Cortical mean diffusivity (cMD), a marker of cortical microstructural changes in neurodegenerative disorders, remains unexplored in isolated REM sleep behavior disorder (iRBD), a prodromal stage of Lewy body (LB) diseases. Its relationship with cortical thickness (CTh), clinical features, and neuropsychological performance is also unknown. We assessed cMD and CTh in thirty-six patients with iRBD at high risk of conversion and 29 healthy controls (HC), examining associations with clinical and cognitive measures. Effect sizes were calculated using Cohen's d. Patients with iRBD showed increased cMD in rostral and caudal cortical regions compared to HC (d > 0.5). Cortical thinning was restricted to caudal areas. Higher cMD correlated with longer iRBD duration, later onset, MDS-UPDRS III, apathy, and poorer performance on Grooved Pegboard and Symbol-Digit Modality Tests. These findings suggest that cMD may be more sensitive than CTh and serve as a valuable imaging biomarker for detecting early cortical changes in prodromal LB diseases.
Satisfaction in the Intensive Care Unit (ICU). Patient opinion as a cornerstone
Objective: To study the agreement between the level of satisfaction of patients and their families referred to the care and attention received during admission to the ICU. Design: A prospective, 5-month observational and descriptive study was carried out. Setting: ICU of Marqués de Valdecilla University Hospital, Santander (Spain). Subjects: Adult patients with an ICU stay longer than 24 h, who were discharged to the ward during the period of the study, and their relatives. Intervention: Instrument: FS-ICU 34 for assessing family satisfaction, and an adaptation of the FS-ICU 34 for patients. The Cohen kappa index was calculated to assess agreement between answers. Results: An analysis was made of the questionnaires from one same family unit, obtaining 148 pairs of surveys (296 questionnaires). The kappa index ranged between 0.278 and 0.558, which is indicative of mild to moderate agreement. Conclusions: The families of patients admitted to the ICU cannot be regarded as good proxies, at least for competent patients. In such cases, we must refer to these patients in order to obtain first hand information on their feelings, perceptions and experiences during admission to the ICU. Only when patients are unable to actively participate in the care process should their relatives be consulted
