    Genetic Diversity of Dicranopteris and Sticherus from Rokan Hulu, Riau Based on ISSR Marker

    Genetic diversity of eleven taxa consisted of Dicranoperis speciosa, D. curanii, and D. linearis with seven varieties and Sticherus truncatus with two varieties from Rokan Hulu, Riau was analyzed using ISSR markers generated from 10 primers. Nine out of ten ISSR primers produced a high level of polymorphism, with six of them showed 100% polymorphism. The genetic similarity was calculated using Jaccard's similarity coefficient, and cluster analysis using the Unweighted Pair-Group Method with Arithmetic Mean. The result showed that the genetic similarity of the eleven taxa under study ranged from 0.377 to 0.627 indicated a moderate level of genetic diversity and the clusters did not separate Dicranopteris from Sticherus.

    Morphological Characterization and Seed Germination Study of Wild Banana Musa acuminata var. flava (Ridl.) Nasution

    Wild bananas provide important genetic materials for further banana improvement, therefore they need to be conserved and studied. This study aimed to describe morphological characteristics of plant and seed and also to study the seed germination of wild banana M. acuminata var. flava (Ridl.) Nasution. The morphological characteristics were observed descriptively by referring to the descriptor for banana. The internal and external morphology of the seeds were observed using a digital microscope. The germination testing was carried out by a completely randomized design, using fresh seeds extracted from a bunch of fruits with two ripeness levels i.e. fully-ripe (yellow peel) and under-ripe (green-yellow peel). The data resulted was then analyzed using an independent t-test. The results showed that M. acuminata var. flava is characterized as a perennial herb; pseudostem height ≥3 m; male bud like a top with prominent green-yellow bracts; fruit curved and tasted mild-sweet when ripe. The seed is angular with wrinkled surface, and dark brown-black color when ripe. The longitudinal section showed parts of the seeds comprising the seed coat, outer and inner integument, embryo, endosperm, chalazal mass, micropyle cap and channel. The seeds are classified as orthodox, with hypogeal type and gradual germination pattern. The seeds extracted from fully-ripe fruit germinated faster with higher germination percentage and growth variables (root number and plant height). Thus, it is suggested to use physiologically mature seeds (seeds from fully-ripe fruits) which should be separated from the seeds of under-ripe fruits to lower the heterogeneity.

    Biostimulant Activity of Sargassum sp. Extracts on Early Growth of Zea mays L. and the Phytohormones Content Analysis

    Seaweed has been gaining global interest in agriculture for the development of marine-based plant biostimulants. This research aimed to study the effect of three different liquid extracts of Sargassum sp., acidic, alkaline, and water extract, on the germination and early growth of maize and to evaluate the phytohormones content responsible for the growth. Phytohormones content including Indole-3-acetic acid (IAA), gibberellins (GA), kinetin and zeatin were analyzed using high-performance liquid chromatography (HPLC) and bioassay was performed twice on maize. Parameters observed on the bioassay were germination percentage, number of roots, shoot length, shoot weight and root weight under 4 different concentrations with 0.5; 1.5; 3.5; and 5% in the first bioassay and 3.5% concentration in the second bioassay. Both bioassays following randomized complete design and the data were analyzed using one-way ANOVA using post hoc test of Duncan Multiple Range Test (DMRT) at error probability of 5% in Genestat software. Phytohormones content in the seaweed extract indicated that alkaline extract was rich in IAA, gibberellin, and zeatin content, while water extract showed the highest kinetin content. The first bioassay indicated that lower concentration of the seaweed extracts gave better growth in all extracts, therefore a 3.5% concentration was chosen for the second bioassay with higher replication for each treatment. The second bioassay confirmed alkaline extract resulted in the highest germination while the highest seedling height, number of roots, shoot and root weight were resulted from acidic extract treatment. In conclusion, Sargassum sp. extracts obtained from acidic, alkaline, and water-based extraction methods, were able to improve the shoot and root growth of maize plants. The acidic extract showed the highest growth promotion among other extracts with the lowest phytohormones content.

    Utilization of Coffee Pulp Waste Composted with Cellulolytic Actinomycetes to Enhance Chili Plant Growth

    The abundant volume of coffee bean pulp as a  by-product of the post-harvest processing is an important source of soil organic matter if it is properly handled. The alternative way to use coffee bean pulp waste to reduce the impact of environmental pollution is composting. This study aims to determine the ability of actinomycetes to degrade coffee pulp, to identify the physical and chemical characteristics of coffee pulp compost, and to evaluate the effect of coffee pulp compost on chili plant growth. The results showed that 7 isolates of actinomycetes were able to hydrolyze coffee pulp in vitro with a hydrolytic index of 1.7-3.81. The treatment of coffee pulp compost with the addition of a starter of cellulolytic actinomycetes (P2) at the end of the three-week incubation period showed the highest organic N (25 mg/kg), P (7.05 mg/kg), and K (33 mg/kg), t compared to other treatments. The effect of giving coffee pulp compost towards the growth of chili plants shows that  the coffee pulp composted with zeolite 5% (w/w) increased the height of the chili plants by 37.6%, while in coffee pulp composted by cellulolytic actinomycetes 5% (v/w) increased the number of leaves by 96% and plant biomass by 25%.  Based on the results of this research, coffee pulp compost has the potential to be used as biological fertilizer to increase plant growth, both composted by zeolite and cellulolytic actinomycete

    Carbon Stock Potential of Gara Gola Natural Vegetation in East Hararghe Zone, Eastern Ethiopia

    This study was conducted at Gara Gola, in the Oromia regional state of Ethiopia, to examine the carbon sequestration potentials under three altitudinal gradients [i.e., Lower altitude (LA: 1500–1800 m.a.s.l.); Middle altitude (MA: 1801–2000 m.a.s.l.) and upper altitude (UA: 2001–2300 m.a.s.l.)]. A total of 60 quadrats of 20m x 20m, 5m x 5m, and 1m x 1m with six horizontal transect lines were employed to gather data on the tree, shrub, herbaceous, and soil, respectively. To estimate organic carbon percentage, soil parameters were collected from three soil profiles (i.e., 0 to 10 cm, 10 to 20 cm, and 20 to 30 cm). The mean total carbon stock of the study area was 641.18 Mg ha-1. MA had relatively higher TC than the other gradients. But the LA had the lowest TC stock, due to a high amount of human and animal interference. The results showed that the UA had significantly higher above-ground (AGC) and below-ground carbon (BGC) stocks with 147.3±39.4 Mg ha-1 and 18.37±7.8 Mg ha-1, respectively, compared to other gradients. However, LA had the lowest AGC (66.8±8.7 Mg ha-1) and BGC (12.06±2.6 Mg ha-1). Lower altitude exhibited a significantly higher SOC value than the other two altitudinal gradients followed by MA. The UA had the lowest SOC value. SOC across the three soil profiles follows a reduction trend from topsoil depth to lower soil depth with significant variation. In conclusion, LA should embrace better ecological, policy, and socioeconomic considerations than the other gradients

    Single-dose Acute Oral Toxicity Study of Chloroform Extract of Snake Plant (Sansevieria trifasciata Prain.) Leaf in Wistar Rats (Rattus norvegicus Berkenhout, 1769)

    Sansevieria trifasciata is one of popular ornamental plants which also believed possessing therapeutic effects due to their phytochemical constituents. Secondary metabolites of plants can be toxic to other organisms; therefore, toxicity studies must be carried out to investigate adverse effects prior to further exploration as potent candidates of medicinal plants. This research aimed to evaluate toxicity and safety of consuming chloroform extract of S. trifasciata leaf (CESTL) in acute phase using female Wistar rats as model animal. Procedure referred to OECD Guidelines for the Testing of Chemicals, Test No. 420: Acute Oral Toxicity - Fixed Dose Procedure with single-dose administration of 2000 mg/kg bw. Results demonstrated that during 14 days of the experiment, neither mortality and sublethal effects as signs of toxicity were detected. There were no significant differences during the experiment between treatment groups and control in body weight, core temperature, individual and social behavior, food and water intake, as well as hematological profile, clinical biochemistry parameters, and relative organ weight (visceral organs indices). Almost all values were maintained within normal range (baseline) with fluctuation as normal physiological dynamics appeared relatively similar in all groups. Therefore, it can be concluded that no-observed-adverse-effect-level (NOAEL) for single-dose oral administration of CESTL with the dose 2000 mg/kg bw and can be classified in the hazard of Category 5 based on Globally Harmonized System of Classification and Labelling of Chemicals (GHS). Based on this finding, we will continue to conduct further study to assess the repeated-dose acute oral toxicity.

    Effect of Carbon Source Variations on Growth, Physiological Stress, and Saponin Levels of Talinum paniculatum Gaertn. Adventitious Roots

    Monosaccharide and disaccharide as carbon sources can affect the production of secondary metabolites. The study aims to determine the effect of variations in carbon sources on growth, physiological stress, and saponin levels of the adventitious roots of Talinum paniculatum Gaerthn. Adventitious roots are subculture in liquid MS medium treated with various sugars: 3% sucrose, 3% glucose, 3% fructose, 3% lactose, 3% maltose, 3% dextrose, sucrose + fructose (1.5% + 1.5%), sucrose + glucose (1.5% + 1.5%), glucose + fructose (1.5% + 1.5%), sucrose + dextrose (1.5% + 1.5%) for 6 weeks. The results of this study show that the 3% fructose treatment produces the highest fresh and dry biomass, which are 1.30 g and 0.23 g compared to the control. The morphology of adventitious roots in the treatment of carbon source variation is not different from the control treatment. The highest MDA (malondialdehyde) levels are found in the sucrose + fructose treatment (1.5% + 1.5%). Meanwhile, the highest proline levels are found in the 3% maltose treatment. Saponin levels analyzed using thin layer chromatography show the data in the form of color intensity and stain area based on ImageJ software analysis. The 3% fructose treatment shows the highest color intensity and stain area compared to the control. Variations in carbon sources affect physiological stress, biomass, and saponin levels of adventitious roots of T. paniculatum, but do not effect on root morphology

    Leaf Morphometric and Chlorophyll Content Study of Bisbul (Diospyros discolor Willd.) at the Bogor Botanical Garden

    Bisbul (Diospyros discolor Willd.) is one of the collections from the Bogor Botanical (BBG). The wide distribution of this plant in Southeast Asia's tropical forests creates many morphometric variations of this species. The diversity of leaf morphometric variations of a plant species can be the basis for grouping various species. This study aimed to determine the morphometric diversity of Bisbul leaves from various locations of origin. The study took samples of bisbul leaves from Bogor Botanical Gardens from three accessions, namely the Philippines, West Java, and Papua. All the trees are over 30 years old. The leaves are taken based on a horizontal position, vertical crown, and leafage. The results showed that the origin of the location gave different multiplier values to calculate the area, namely the Philippines y = 0.733x + 0.034; West Java y = 0.765x-2,949; and Papua y = 0.758x-1.389. The length to leaf width ratio also has differences, namely, the Philippines, which is 2.64, West Java 2.65, and Papua 2.81. The chlorophyll content in young leaves increases in old leaves. The samples from Papua also have the highest chlorophyll content compared to the Philippines and West Java. This difference indicates morphometric variations between the three, even within one species. Some environmental conditions may affect shade areas and tree age. In addition, DNA research from accession D. discolor is also needed to determine the cause of the morphometric variation.

    Molecular Bird Sexing of Small Yellow-crested Cockatoo (Cacatua sulphurea, Gmelin 1788) Using Polymerase Chain Reaction Method

    The yellow-crested cockatoo (Cacatua sulphurea, Gmelin 1788) is an endemic bird in eastern part of Indonesia with monomorphic characteristics and included in the list of endangered birds.  A method of sex determine in monomorphic birds is by molecular sexing which is based on the PCR amplification of the chromodomain helicase DNA-binding 1 (CHD-1) gene of the bird sex chromosome. This study was aimed to sex determine of the C. sulphurea by amplifying the CHD-1 gene on the Z and W chromosomes and comparing the PCR amplification results from peripheral blood and plucked feathers samples. The samples used were four birds of C. sulphurea from the Wildlife Rescue Centre (WRC), Yogyakarta. The feathers obtained from the ventral wings of each bird were plucked. Through the cutting of the birds' nails, the peripheral blood samples were collected into microhematocrit tubes which contained Heparin. The amplification of the CHD-1 gene used the PCR method with specific primers, such as NP, P2, and MP. Moreover, the PCR results were visualized on 1.5% agarose gel using UV-Transilluminator, at a wavelength of 280 nm. The PCR products (amplicons) were in 297 bp and 392 bp DNA bands, depending on the sex of the bird being tested. It was also observed that the male C. sulphurea produced single 392 bp DNA fragment of the Z chromosome. However, the female birds produced two DNA fragments of the Z and W chromosomes, with a length of 297 bp and 392 bp. The results showed that samples obtained from peripheral blood produced clearer DNA bands compared to plucked feather. It concludes that the extracted DNA from peripheral blood samples have a better quality compared to samples from plucked feathers. Amplification of the CHD-1 gene in male C. sulphurea generated only a single DNA fragment in size of 392 bp, so the four C. sulphurea were male birds

    Aboveground Forest Carbon Stock in Protected Area: A Case Study of Bukit Tigapuluh National Park, Indonesia

    The role of protected areas has been expanded into climate change mitigation, specifically on Reducing Emissions from Deforestation and Forest Degradation (REDD+). A reliable and practical method for measuring, reporting and verifying carbon stock is an essential component for REDD+. This study aims to recognize the characteristic and estimate aboveground forest carbon (AGC) stock in the tropical protected tropical area using a combination of terrestrial forest inventory and spatial data. A 168 cluster plots totaling 33.6 hectares were taken proportionally based on the percentage of forest cover types (dryland primary natural forest/DPF and dryland secondary natural forest/DSF) using a traditional forest inventory method (more than 5 cm dbh). Results showed that Bukit Tigapuluh National Park secured a significant AGC stock which has been estimated to be 269.2 [247.07; 291.43] tC/ha or 35,823,639 [32,872,312; 38,774,966] tC in total, being stored in approximately 133,051 hectares of the tropical rain forest. This result was higher than other studies in non-protected areas but slightly lower than other studies within protected areas. This finding supported the argument that protected areas possess a higher figure of AGC stock than other forest management units. The high amount of forest carbon biomass in the protected areas shall be very important assets for conducting the role of conservation for REDD+.
