    Optimization of HIB Value Combination of Tween 60 and Span 80 on Cream Formulation of Ethanol Extract of Green Tea Leaves (Camellia Sinensis L.)

    Green tea (Camellia sinensis L.) is known to have ability to protect skin against free radicals. This is supported by polyphenol compound catechin. This research aims to determine the optimum Hydrophilic-Lipophilic Balance (HLB) value of Tween 60 and Span 80 compositions on the optimum cream formula of ethanol extract of green tea leaves. Tea leaves are extracted by macerating using 70% ethanol. Catechin in extract is known from Thin Layer Chromatography (TLC) test with silica gel 60 F254 as stationary phase and ethyl acetate:aquadest:formic acid (18:1:1 v/v) as mobile phase. Antioxidant activity is determined by 2,2-Diphenyl-1-picryhydrazyl (DPPH) method and value of Inhibition Concentration 50% (IC50) is then calculated. Formula optimization using Design Expert® version 7.1.5 (DX 7) software, Simplex Lattice Design (SLD) method with two components Tween 60 and Span 80. Cream is characterized according to physical properties organoleptic, homogeneity, viscosity, pH, spreadability, adhesiveness, and cream type. The optimum formula obtained is then tested for physical stability for 4 weeks at room temperature (28±2°C) and data are statistically analyzed using one-way ANOVA. The extract contains catechin proved with Retention factor (Rf) value 0.8 and has antioxidant activity with IC50 value 56.35 ppm. 6.4% Tween 60 and 3.6% Span 80 result an optimum HLB value 11.1. It has viscosity 2897.50±35.94 mPa.s, spreadability 18.44±0.06 cm2, adhesiveness 0.85±0.05 seconds, and pH 4.530±0.002. Statistical test shows that the cream is significantly altered at pH, but does not significantly change in viscosity, spreadability, and adhesiveness after being stored for 4 weeks

    The mangosteen peels (Garcinia mangostana L.) has high free radical scavengers activity. This study aims to determine the physical and chemical conditions that are formulated in SNEDDS and nanoemulsion preparations. SNEDDS was made with various concentrations of Tween 80, PEG 400, and Virgin Coconut Oil (VCO) with a ratio of 4,98:1,02:1. Nanoemulsions were made with the addition of water to SNEDDS. Stability observation results were analyzed by statistics. The results of physical stability test show that all test samples in the research, in nanoemulsion, SNEDDS fraction, base SNEDDS, SNEDDS vitamin E did not undergo separation, precipitation, cracking, and creaming. On the chemical stability test of nanoemulsion and SNEDDS fraction there is no difference between IC50 before and after storage. While on basic SNEDDS and vitamin E nanoemulsion there is a difference between IC50 before and after storage

    Macroscopy and Microscopy Study of Scopraia dulcis L.

    A morphological diversity analysis of Scoparia dulcis by macroscopy and microscopy has been conducted. The aim of this research is to determine the morphological and anatomical character of S. dulcis which grow in Opak watershed, Special Region of Yogyakarta. Samples were taken from Sleman (Cangkringan, Ngemplak Kalasan, and Prambanan) and Bantul (Imogiri and Pundong) region. Selection of sampling location is based on altitude difference. The samples are the third leaf order from the tip of the stem. They were measured in length and width. Microscopy analysis was performed by observing the number of glandular trichomes, palisade cells in the epidermis and stomata in the upper and lower epidermis. The data obtained were analyzed using principal component analysis (PCA). The results show that based on morphological and anatomical assessment, there were found 3 clusters of S. dulcis, i.e. Cangkringan-Ngemplak-Prambanan, Kalasan, and Imogiri-Pundong cluster

    Research of radical scavenging activity of sambiloto (Andrographis paniculata Nees) and patikan kebo (Euphorbia hirta L) ethanolic extracts and their combination toward 2,2-Diphenyl-1-Picrylhidrazyl (DPPH) has conducted. Subjects of this experiment comprise sambiloto ethanolic extract, patikan kebo ethanolic extract, dechlorophyllated extract of sambiloto and combinations among those three. Dechlorophyllation by electrocoagulation method is done to reduce chlorophyll content thereby increasing the level of active compound in the sambiloto extract. The results of dechlorophyllation showed that only total flavonoid content in the extract of sambiloto increased, meanwhile the radical scavenging activity results showed that patikan kebo ethanolic extract has the lowest IC50 value (22,36 µg/mL) compared to sambiloto ethanolic extract (IC50 value 499,03 µg/mL). Extract combination may increase DPPH radical scavenging activity of the sambiloto extract (IC50 value 76,06 µg/mL) but not higher than that of patikan kebo single extract

    Heme Polymerization Inhibition by Tithonia diversifolia (Hemsley) A.Gray Leaves Fractions as Antiplasmodial Agent and Its Cytotoxicity on Vero Cells

    Previous research revealed that the extracts and fractions of Tithonia diversifolia (Hemsley) A.Gray leaves had antiplasmodial activity in vitro. For further development as an antiplasmodial agent, the mechanisms of action and safety of compounds are important to disclose. Heme polymerization inhibition is one of the main targets of antiplasmodial action. The aim of the study was to investigate the activity of T. diversifolia fractions in inhibiting heme polymerization and its cytotoxic effect on Vero cells. Heme polymerization inhibition assay from Bassilico and cytotoxic test on Vero cell using MTT method were conducted for three fractions (F5, F6, and F7) of T. diversifolia leaves. The inhibitory activity of heme polymerization expressed as IC50 and cytotoxicity effect expressed as CC50 were determined by probit analysis. The best heme polymerization inhibition activity was F5 with IC50 = 162.20 ± 57.81 μg/mL followed by F6 and F7 with IC50 216.30 ± 26.56 and 231.54 ± 44.26 μg/mL respectively. All the fractions had a low cytotoxic effect with CC50 for F5, F6, and F7 were over than 100, 34.81 ± 9.94 and 56.26 ± 6.73 μg/mL, respectively and the toxicity index fraction is below 10 or categorized as low selectivity. Conclusion: The fraction of T. diversifolia inhibited heme polymerization in vitro and had low cytotoxic effect on Vero cells but no selective toxicity. Further research using pure compounds may improve its selectivity

    Syzygium aromaticum as an immunomodulator contains main active compound eugenol which is able to stimulate lymphocyte proliferation and the production of macrophages. Lymphocytes have a very important role to provide protection in the body against infection. This study aims to prove the effects of extract Syzygium aromaticum leaf against increased proliferation of lymphocytes, lymphoblast and macrophages of mice Balb/c of induced Salmonella typhimurium. The method used in this study was experimental with post test only control group. Mice Balb/c were divided into 4 groups as a control group and treatment induced of Salmonella typhimurium. The first treatment group were administrated extracts of 15mg /kgbw, the second treatment 75mg/kgbw, the third treatment of 150mg/kgbw for 12days. ANOVA test showed a significant difference in lymphocyte proliferation but not lymphoblast and macrophages

    Oxidative stress has been known to contribute to Alzheimer’s disease. Acetylcholinesterase (AChE) enzyme may lead to Alzheimer’s disease as a neurotransmitter. Antioxidants may have protective activities against oxidative damage and Alzheimer’s disease. Acetylcholinesterase inhibitors also can be used in the treatment of various neurological disorders for management of Alzheimer’s disease. This study aimed to determine antioxidant and anticholinesterase effects of Garcinia picrorrhiza Miq. fruit extract (GpKar) and its compounds, xanthone. Antioxidant activity was measured by H2O2 scavenging inhibitory activity, while anticholinesterase activity was measured using modified Ellman method. GpKar has higher H2O2 scavenging inhibitory activity (IC50= 967.28 µg/ml) compared to xanthone (IC50= 1198.95 µg/ml). In the anticholinesterase inhibitory activity, GpKar has lower activity (IC50= 70.25 µg/ml) compared to xanthone (11.80 µg/ml). In summary, GpKar has higher antioxidant activity but lower anticholinesterase activity compared to its compounds, xanthone. However, GpKar has potency as antioxidant agent to prevent Alzheimer’s disease

    Determination of FICI of Ethanolic Extract of Aloe Vera Skin Leaves (Aloe vera (L.) Burm.f.) and Gentamicin Sulphate againts Staphylococcus aureus

    The main therapy of wounds infection using antibiotic such as gentamicin sulfate can be applied topically. Increasing incidence of antibiotic resistance forces new strategy to combine plant extract and antibiotic. These two combinations are expected to reduce the incidence of microbial resistance. Ethanolic extract of Aloe vera skin leaves contain anthraquinone that has antimicrobial activity. The aim of this study was to determine the effect and Fractional Inhibitory Concentration Index (FICI) from combination of ethanolic extract of Aloe vera skin leaves and gentamicin sulphate which can inhibit the growth of Staphylococcus aureus. Minimum Inhibitory Concentration (MIC) determination was used Kirby-Bauer disc diffusion method. The concentration of extract solution used were 1,25; 2,5; 5; 10 mg/mL while the solution concentration of gentamicin sulfate used were 2,5; 5; 15; 25 µg/mL. Solution DMSO was used as negative control. Combination solution was made with volume ratio 1:1 from MIC of extract and gentamicin shulphate. The result showed that the MIC of the of ethanolic extract of Aloe vera skin leaves was about 2.5 mg/mL and MIC gentamicin sulphate was about 5 µg/mL. Combination of ethanolic extract of Aloe vera skin leaves and gentamicin sulfate can inhibit the growth of Staphylococcus aureus with zone of inhibition 7,63 mm. FICI of combination was 2 and its antibacterial activity was indifferent compared to single extract and single gentamicin sulphate

    The Effectiveness Test of Oil Phase Ointment Containing Snakehead Fish (Channa striata) Extract on Open Stage II Acute Wounded Wistar Strain Male Rats

    The snakehead (Channa striata) contained fatty acids omega-3 and omega-6 are useful as nutrients in accelerating the wound healing process. This study aims to determine the effectiveness of the oil phase extract ointment snakehead in the healing of acute wounds open stage II. Testing the effectiveness of wound healing using 6 groups: normal, gel bioplacenton (positive control), ointment base (negative control), ointments snakehead extract oil phase concentration of 10; 20 and 40% of the 4 rats with acute wounds open stage II and observation until day 16 injury. The area of the wound was measured with the image J program Macbiophotonic, calculated% healing power, and AUC values. A statistical test to the total AUC values per rat with SPSS for Windows 22.0 program using One Way ANOVA and Post-hoc LSD test. The results showed a significant difference in the negative group with ointment concentration of 20% (p <0.05). The treatment group phase ointment fish oil extract is effective for wound healing is best to have a concentration of 20% for percentage of wound healing power of 97.157% and the average AUC value of 865.683% of the day

    Dechlorophyllation of Cosmos caudatus Kunth., Morinda citrifolia, and Mangifera indica L. Leaves Methanolic Extract by Electrocoagulation Technique

    The present of chlorophyll is not expected in the isolation process of plant active constituent. Electrocoagulation is a potential dechlorophyllation method. This research aims to know the effectivity of electrocoagulation in the dechlorophyllation process of Cosmos caudatus Kunth., Morinda citrifolia, and Mangifera indica L. leaves methanolic extract as well as the effect to the total phenolic content. Electrocoagulation are performed using copper, silver, aluminum and iron plates as the electrode. Dechlorophyllation by extraction using n-hexane is performed as reference. The % absorbance of chlorophyll and % of total phenolic content of dechlorophyllated samples are measured spectrophotometrically. Electrocoagulation process reduces % absorbance of chlorophyll in Cosmos caudatus Kunth., Morinda citrifolia, and Mangifera indica L. leaves methanolic extract. The more the duration of electrocoagulaton process the lower the % absorbance of chlorophyll obtained. % absorbance of chlorophyll of the electrocoagulated extract according to paired t-test (P=0.95) are significantly different with the previous. One way ANOVA continued with LSD (P=0.95) shows that the % absorbance of chlorophyll from the electrocoagulated extract are significantly different with those extracted with n-hexane. Electrocoagulation process reduces total phenolic content along with duration of electrocoagulation
