In this dataset, you will find RNA sequencing data derived from human intestinal tissue and corresponding intestinal organoid monolayers from the jejunum and ileum region of the intestine for adults and ileum region for pediatric donors (0-18y). The samples have been sequenced in two batches. Intestinal samples were collected from adult and pediatric patients (0-18 years olds). Intestinal stem cells were used to culture intestinal organoid monolayers. Intestinal tissue and enteroid monolayers were RNA sequenced to identify similarilies/differences focussed on drug transporters and intestinal integrity markers

324217.pdf (Publisher’s version ) (Open Access)Recent public corruption scandals in South Africa have created a social landscape where trust in the motives of businesspeople and politicians has sharply declined. I argue that a by-product of this is constraints on local brokers' potential to act as change agents because they are perceived to be associated with the state and the market. This ethnographic case study explores the efforts of two community leaders-cum-brokers, from a small rural township, to transform a natural sand mine into a community enterprise. Their project received lacklustre support from local residents, who suspected it could be a cover for fraudulent dealings, a suspicion informed by fears of becoming entangled in corruption, whether willingly or unwittingly. I also discuss how the metaphor of "state capture" has been scaled down to the local level and used by residents to make sense of both the town's lack of development and the limits to enacting change.14 p

322450.pdf (Publisher’s version ) (Open Access)BACKGROUND: The aim of prehabilitation is to optimize patient specific modifiable risk factors before major surgery, in order to enhance the individual resilience. Due to the lack of universal guidelines, prehabilitation has been conducted in various ways, making it difficult to estimate its effect. In the Netherlands, proposed uniform prehabilitation protocols were developed. The aim of this study is to analyse clinical outcomes of prehabilitation when implemented as standard of care according to the proposed uniform protocols. METHODS: Uniform prehabilitation protocols were created based on the multimodal program of the PREHAB randomized controlled trial. All hospitals in the Netherlands that implemented prehabilitation according to the proposed protocols, for patients undergoing elective colorectal surgery, will be asked to participate. This study will recruit 535 patients who underwent prehabilitation and 535 one-to-one nearest neighbour propensity score matched patients who did not undergo prehabilitation (historical cohort). Clinical outcomes will be compared between the prehabilitation group and the historical cohort group using regression analyses. The primary outcome of interest is 90-day presence of postoperative complications. In addition, length of hospital stay and readmissions will be analysed. DISCUSSION: Prehabilitation has been proven to reduce the risk of complications and hospital length of stay. Prehabilitation has however been defined in various ways, since there is no standardized program. This multicentre cohort study will estimate the clinical effect of prehabilitation implemented as standard of care according to proposed uniform protocols. Progress and pitfalls when evaluating the unintended effects of public policy: The case of German international development cooperation

321741.pdf (Publisher’s version ) (Open Access)The study of unintended effects of policies is a key debate among evaluation scholars. Through complexity theory, we argue that unintended effects of (international) public actions are inevitable and question the reliability of evaluations in providing a correct and complete picture of public policy. We use a machine-learning-assisted text-mining case study approach, examining 254 programme evaluations of German international development cooperation as a 'least likely case'. While German evaluations focus more on unintended effects than Dutch, Norwegian and American evaluations, their treatment is not always correct or complete. There is an overidentification of unintended effects and a bias towards positive ones, with certain types of unintended effects overlooked. We explore explanations for the observed weaknesses, including an overreliance on linear thinking and insufficient guidance for the evaluators on identifying unintended effects. We conclude with concrete suggestions to improve implementation of the Organization for Economic Co-operation and Development guidelines that are essential to make public administration more effective and trusted.21 p
