The VEGF decoy receptor soluble Fms-like tyrosine kinase 1 binds to macrophages.

319069.pdf (Publisher’s version ) (Open Access)BACKGROUND: Soluble Fms-like Tyrosine kinase-1 (sFLT1) is a native inhibitor of VEGF, best known for its antiangiogenic effects in preeclampsia. sFLT1 also reduces chronic inflammation and promotes tissue repair. In experimental diabetic nephropathy, we previously found that sFLT1 ameliorates kidney fibrosis and reduces the infiltration of macrophages. How sFLT1 regulates inflammation is still incompletely understood. Based on the direct association of sFLT1 with various cell types, we here studied whether sFLT1 interacts with macrophages to modulate inflammation. METHODS: Using various macrophage cell lines, sFLT1 cell surface binding was detected with flow cytometry. Enzyme studies, mass spectrometry and RNAseq were employed to identify potential sFLT1 cell surface interactors and effects of sFLT1 on macrophage signaling. RESULTS: Soluble FLT1 binds to primary macrophages, THP-1 and RAW264.7 macrophages in vitro. Alternative activation with IL-4 increases sFLT1 binding in THP-1 macrophages, whereas proinflammatory activation with IFN-γ and LPS decreases binding. Binding of sFLT1 depends on heparan sulphates, and colocalizes with the membrane heparin sulfate proteoglycan neuropilin-1. Incubation with sFLT1 reduces the gene expression of chemokine receptors. CONCLUSION: Our results show that sFLT1, while typically associated with angiogenesis, also directly interacts with macrophages. Alternative activation of macrophages by IL-4 strongly increases binding of sFLT1 to the cell surface membrane, possibly via the VEGF co-receptor neuropilin-1. Considering sFLT1's anti-inflammatory effects in animal studies, our findings indicate a novel function for sFLT1 to directly control anti-inflammatory macrophage function

321032.pdf (Publisher’s version ) (Open Access)The research presented in this thesis had the goal to examine the long-term health effects of early menopause duo to prophylactic resection of the ovaries and fallopian tubes (RRSO) in women with increased risk of familial breast- and or ovarian cancer. To summarize we did not find clues for increased risk of cardiovascular disease in women who underwent premenopausal RRSO compared to postmenopausal RRSO. Although we found that women in the premenopausal RRSO group had lower bone mineral density, we found no increased risk of osteopenia and osteoporosis compared with the postmenopausal RRSO group. Women in the premenopausal RRSO group experienced more vaginal dryness and discomfort compared with the postmenopausal RRSO group, however they reported no difference in sexual pleasure. Women in the premenopausal RRSO group reported more stress urine incontinence compared with women in the postmenopausal RRSO, however, these differences were rather small and its clinical relevance is questionable. We found no difference in urge incontinence between the pre- and postmenopausal RRSO groups. Change towards more gender equality: The role of formal sponsorship programs in a Dutch professional service firm
Item does not contain fulltextThis chapter contributes to the literature on gender equality interventions for women leaders by examining the role of formal sponsorship programs. Despite considerable scholarly attention given to the positive and negative gender implications of informal sponsorship relations, there remains a gap in knowledge regarding formal sponsorship programs and their (un)intended effects. Drawing on a longitudinal field study conducted in a professional service firm in the Netherlands, the research follows 21 women, encompassing both ethnic minorities and majorities, and their sponsors within a formal sponsorship program. Five sponsorship practices were identified: mirroring, advising, connecting and making visible, protecting and advocating. However, the majority of sponsors tended to gravitate towards the more passive practices closely aligned with mentoring. The study concludes that sponsorship relations are hard to formalize due to issues of power and trust and can even be counterproductive when dominant gendered constructions of 'leadership' and 'potential' are not addressed
From Molecules to Cells: Strategies to Target and Eliminate Autoreactive B-cells

319711.pdf (Publisher’s version ) (Closed access)In rheumatoid arthritis (RA), the immune system does not work properly and ends up attacking the person's own body. A special type of immune cell, the B cell, plays an important role in this process, in particular when these cells respond to what is known as ‘citrullinated proteins’. Normal treatments suppress the entire immune system or kill all B cells, including the good ones. In this study, we looked at how we can target only the ‘faulty’ B cells using CCP4, a substance only recognised by these cells. We created different versions of CCP4 and attached drugs or light-sensitive substances to it. The combination of CCP4 with a light-sensitive substance proved very successful: with light, we were able to precisely turn off the faulty B cells, without damaging the healthy ones. In addition, we developed special CAR T cells (a kind of smart immune cells) that, thanks to CCP4, only recognised and attacked the faulty B cells. Finally, we investigated how to turn these CAR T cells off again when needed. Costs and benefits of temporal expectations on somatosensory perception and decision-making

318892.pdf (Publisher’s version ) (Open Access)Our perception is shaped by prior expectations, including those about the timing of our sensations. These temporal expectations can be formed by recognizing patterns in the onset of sensory inputs. However, in the somatosensory domain, it remains unclear how these expectations impact the speed and accuracy of somatosensory judgments, as previous research has yielded mixed results. Here, participants used auditory tones to anticipate the onset of forces applied to their fingers and discriminated their intensity compared to a reference force. Experiment 1 showed that participants had worse discrimination sensitivity and higher thresholds for expected versus unexpected forces. Experiment 2 replicated and extended these costs to include perceptual accuracy, even when comparing expected to expectation-free forces, and further revealed reaction time benefits. Drift-diffusion modelling suggested that expectations speeded non-decisional processes while simultaneously slowing somatosensory evidence accumulation. These findings demonstrate both costs and benefits of temporal expectations in somatosensory perception and decision-making.11 p

Gezien de transdiagnostische aard van het concept emotieregulatie, wordt in dit hoofdstuk dieper ingegaan op het meten van emotieregulatie aan de hand van de verschillende niveaus zoals gedefinieerd door de Research Domain Criteria (RDoC), een theoretisch raamwerk dat de onderliggende transdiagnostische mechanismen van psychopathologie in kaart tracht te brengen. Meer specifiek wordt stilgestaan bij de fundamentele biologische en neurologische determinanten van emotieregulatie, psychofysiologische correlaten van emotieregulatie, gedragsobservaties, emotie-inductieparadigma’s en zelfrapportagematen. Ten slotte wordt besproken hoe specifieke emotieregulatievaardigheden in kaart kunnen worden gebracht
