    The impact of once daily dosing on protein binding of clozapine and norclozapine

    Purpose/background: Clozapine and norclozapine are highly protein bound. Currently, clozapine is increasingly prescribed once daily (QD). Higher (once daily) doses may theoretically lead to saturation of protein binding of (nor)clozapine, resulting in increased unbound fractions. This study investigated whether protein binding of clozapine and norclozapine becomes saturated at higher concentrations. Secondly, the correlation between unbound (nor)clozapine fractions and alpha-1 acid glycoprotein (AGP) concentrations was studied.Methods/procedures: From 44 patients taking clozapine QD or twice daily a total of 319 blood samples were collected at different time points within a dose interval. AGP concentrations were measured in samples drawn just before clozapine intake. A validated liquid chromatography-tandem mass spectrometry method was used for quantification of the (nor)clozapine concentrations. Ultrafiltration was used to separate the bound and unbound molecules. The relation between total concentrations and fractions, and between unbound fractions and AGP concentrations were investigated using linear mixed model analysis.Findings/results: There was no significant correlation between total clozapine ( P = 0.270) and norclozapine ( P = 0.678) concentrations and its unbound fractions with total clozapine concentrations up to 1500 μg/L. A statistically significant (negative) correlation between AGP concentrations and clozapine ( P = 0.000) and norclozapine ( P = 0.028) unbound fractions was found.Implications/conclusions: In general, total concentrations remain suitable for therapeutic drug monitoring if clozapine is used once daily. Future research is needed to define if higher total concentrations are warranted in case of lower unbound fractions due to increased AGP concentrations

    This contribution discusses the concept of ‘mnemohistory’ through an analysis of the video game Remember Me. The game’s protagonist Nilin has the ability to manipulate other people’s memories by subtly changing details, which changes how they see themselves and act. The author analyses how the game connects memories to identity and consciousness based on Locke’s theory. It also explores themes of sin, temptation, paradise lost, and climate change. The contribution argues that the game shows how the past shapes the present through remembered history rather than objective facts. It raises questions about memory manipulation and who owns memories

    On relative income and life datisfaction: The moderating role of GDP per capita, culture and income inequality

    In his seminal book ‘Social limits to growth’ Hirsch argues that the importance of positional goods (those things of which the value depends on how much other people own) increases with economic welfare. This would suggest that relative income affects subjective well-being more in rich countries than in poor countries. This paper tests this hypothesis by analysing the effect of relative income on life satisfaction in 101 countries using a comprehensive dataset from the Integrated World Values Survey and the World Bank. The results contradict the hypothesis of Hirsch: life satisfaction in rich countries is less affected by relative income and more affected by absolute income than in poor countries. The results are robust when splitting the sample into four sub samples for high, middle-high, middle-low-, and low-income countries, using alternative measures of relative income, tests for over-control bias, and bounds analysis. Delving into factors that explain these results, we find that national culture, measured by individualism and generalized trust, negatively and income inequality positively moderate the relationship between relative income and life satisfaction

    Faint-hearted?:Bespiegelingen over de rol en verantwoordelijkheden van collectieve-belangenbehartigers onder de WAMCA

    Uit de Engelse en de Nederlandse rechtspraak blijkt dat het niet makkelijk is voor (de bestuurders van) collectieve-belangenbehartigers om de toetsing van hun expertise en handelwijze door de rechter met vlag en wimpel te doorstaan. Hoewel beide systemen van collectieve-belangenbehartiging van elkaar verschillen, hebben zij meer gemeen dan op het eerste gezicht lijkt en valt er wat te leren uit de recente ervaringen in het Verenigd Koninkrijk. Dit artikel bevat een overzicht van de Nederlandse wet- en regelgeving die op stichtingen die als collectieve-belangenbehartigers optreden van toepassing is. Ook wordt aandacht besteed aan de knelpunten die zij in de praktijk onder de WAMCA ervaren. Zo kan het lastig zijn om geschikte bestuurskandidaten te vinden, en kunnen de (administratieve) verplichtingen die uit de wet en/of de Claimcode voortvloeien een (te) grote (financiële) last zijn. Steeds wordt aandacht besteed aan de relatie met de procesfinancier, omdat deze commerciële partijen een belangrijke rol spelen in collectieve acties en dat gevolgen heeft voor de governance van deze stichtingen. De auteurs doen aanbevelingen aan (bestuurders van) deze collectieve-belangenbehartigers op basis van de Nederlandse en de Engelse rechtspraak en op basis van hun eigen (praktijk)ervaringen met collectieve acties. De conclusie is dat de Nederlandse wet- en regelgeving bestuurders in staat stelt de redelijke belangen van procesfinanciers in acht te nemen. Daarnaast zijn de bestuurders gebaat bij een besluitvormingsproces, waarbij zij op cruciale momenten desgewenst onafhankelijke experts kunnen raadplegen en verschil van inzichten met de procesfinancier via adequate escalatiemechanismen kunnen laten toetsen en beslechten. Ook een herziening van de Claimcode ligt na de introductie van de WAMCA en de recente ervaringen voor de hand, alsmede een (her)overweging van een one-tier-board. Hoewel er nog veel onzekerheid is over de procedure onder de WAMCA, is één ding zeker: collectieve-belangenbehartiging is niet voor de faint-hearted. Ze is voor ervaren professionals en gaat gepaard met verantwoordelijkheden en kwaliteitseisen. Een verdere professionalisering van de collectieve-belangenbehartiging is daarom een stap in de goede richting

    Do teams alleviate or exacerbate overreaction in beliefs?

    We investigate whether teams exhibit increased or reduced overreaction in expectation formation relative to individuals. Using preregistered randomized experiments that directly elicit expectations about future returns, we find that teams display lower belief overreaction to recent investment performance. A quantitative decomposition shows that this team effect stems primarily from a ''self-selection'' mechanism, whereby the most biased team member chooses to influence the team decision less. An LLM-based analysis of team interactions reinforces this result. A complementary analysis of US equity mutual fund managers operating both individually and as part of a team yields consistent evidence of lower overreaction in teams
