Simultaneous determination of ezetimibe and atorvastatin calcium in spiked human plasma using SPE coupled to HPLC-MS/MS
Combination drug products containing ezetimibe (EZB) and atorvastatin calcium (ATVC) are widely used for treatment of hyperlipidemia. A rapid, simple and sensitive liquid chromatography tandem mass spectrometry (LC-MS/MS)method for determination of EZB and ATVC in spiked human plasma has been developed and validated.Plasma samples were subjected to solid phase extraction before analysis with an extraction recovery of 90.59 - 98.02%. Chromatographic separation was performed on a C18 col- umn using isocratic elutionwithin 2 min run time. The mobile phase con- sisted of acetonitrile: 1% of formic acid in water (8: 2, v/v) and was pumped at a flow rate of 0.8 mL/min. Detection of analytes was achieved by with electrospray ionization (ESI) in negative ion mode for EZB and positive ion mode for ATVC. The calibration curves were linear over the range of 1.67 83.33 ng/mL for both drugs. The precision at the lower limit of quantitation for EZB (12.97%) and ATVC (8.99%) was determined. The intra- and inter- day precisions were within 9.16%, while the accuracy ranged from 94.18 to 106.94%. The validated LC-MS/MS method was successfully employed for thedetermination of EZB and ATVC in spiked human plasma for application in therapeutic drug monitoring. 2014 Trade Science Inc. - INDI

Single-dose linezolid pharmacokinetics in critically ill patients with impaired renal function especially chronic hemodialysis patients
Renal failure patients were treated with linezolid (LZD) for proven or suspected infections by multi-resistant Gram-positive cocci. The aim of this study was to determine if dose adjustment of LZD is needed as a function of renal impairment or not, especially that a significant component of LZD is eliminated unchanged in urine. Methods: The single dose pharmacokinetics of LZD was investigated. Eighteen non-infected male subjects with various degrees of renal impairment ranged from normal to severe chronic impairment were enrolled, including end-stage renal disease (ESRD) patients maintained on hemodialysis (HD). LZD was administered as a single oral 600 mg dose, and blood samples were drawn at different times and analysed by a validated HPLC assay method. Plasma profiles were evaluated by non-compartmental and compartmental approaches. Results and Discussion: A similar rate and extent of LZD absorption and elimination and comparable body exposure was observed in both healthy subjects and acute renal failure patients. The extent of LZD exposure was significantly increased by 3-fold in ESRD patients in their off-dialysis day. Furthermore, the t1/2 and MRT values were significantly increased by ~5- and 3-fold, respec- tively. The Vd/F values of LZD did not change with renal function. A significant decrease in CL/F by ~3-fold was observed in ESRD patients in their off-dialysis day however, CL/F was significantly increased by ~4-fold during HD. Approximately half of the administered LZD dose was removed during the HD session in these selected cohorts of ESRD patients. LZD was generally well tolerated. Conclusions: The dose of LZD did not need to be adjusted for patients with acute renal dysfunction or ESRD on HD. One of the twice-daily doses should be administered after the dialysis session because almost half of the LZD dose was substan- tially removed by HD. During the first three dialysis sessions of the treatment course, to avoid potentially ineffective therapy, a supplemental dose of LZD might be given if necessary or the dose of LZD should be administered 4 h before the beginning of the HD session. Genetic algorithm derives its structure from the observation of nature. Since algorithmic art consists of generation of images on the basis of al- gorithms, algorithms can be viewed as a notation, and notation is something that music has but visual artifacts in general miss. This paper aims to discover the role of evolutionary algorithms in historical Islamic architecture. Also, we shall try to investigate the way in which the future development could occur not only through the discovery of new facts or theories, but also through the rise and dissemination of new visions having different explanation of Islamic archi- tecture that considers it as a result of serious application of formation through evolutionary genetic algorithm

Electroanalytical Determination of Gemifloxacin Mesylate in Bulk, Tablets and Human Urine Using Gold Nanoparticles Modified Carbon Paste Electrode
A simple, precise, inexpensive and sensitive voltammetric method has been developed for the determination of gemifloxacin mesylate (GEM) in the presence of tween 80 in the bulk, farmaceutical dosage forms and human urine at gold nanoparticles modified carbon paste electrode (GNCPE). The electrochemical behavior of GEM has been investigated by using cyclic voltammetry (CV) and differential pulse voltammetry (DPV) techniques. The electrochemical oxidation of GEM was an irreversible process which exhibited adsorption-diffusion controlled process behavior in Britton-Robinson (BR) buffer over the entire pH range of values from 2 to 9. The adsorptive stripping response was evaluated as a function of some variables such as pH, type of surfactant, scan rate and accumulation time. The anodic peak current varied linearly over the range from 8.0 × 10 -7 to 2.8 × 10 -5 M. The limits of detection and quantification were 7.32 × 10 M and 2.44 × 10 -7 M, respectively. The relative standard deviations and the percentage recoveries were found in the following ranges: 0.58-1.35% and 99.37-101.76%, respectively
