    Behavioral Investigation of the Effects of Gabapentin on the Serotonin Pathway of the Descending Pain Inhibitory System

    新潟大学Niigata University博士(医学)近年,神経障害性疼痛治療薬としてガバペンチノイドを使用することが多い.ガバペンチノイドは電位依存性Ca2+チャネルのα2δ サブユニットと⾼い親和性を持ち,当初は抗けいれん薬として開発されたが,メカニズムの研究が進み,下行性疼痛抑制系を活性化して脊髄のノルアドレナリンの放出を増加させることが知られるようになった.下行性抑制系にはノルアドレナリン系の他にセロトニン系が存在するが,ガバペンチノイドの作用にセロトニンが関与しているという報告はほとんどない.そこで本研究では,左L5脊髄神経を結紮した神経障害性疼痛モデル(SNL)ラットを用いて,ガバペンチン(GBP)が下行性抑制系のセロトニン経路に及ぼす影響を,行動実験により検討した.SNL ラットにGBPを腹腔内投与すると約60分後から足底への機械的刺激に対する逃避閾値の改善が認められ,その効果はおよそ240分後まで持続した.GBPを投与したSNL ラットの脊髄くも膜下腔にセロトニン受容体拮抗薬を投与すると逃避閾値の改善が消失したことから,GBPの鎮痛作用にセロトニン系が関与している可能性が示唆された.しかし,パラクロロフェニルアラニンでセロトニンを枯渇させたSNL ラットにGBPを投与すると,GBPによる鎮痛効果は認められたものの,セロトニン受容体拮抗薬を投与しても鎮痛効果は拮抗されなかった.一方,ノルアドレナリンを枯渇させたSNLラットでは,GBPの鎮痛効果そのものが認められなかった.これらの結果から,GBPの鎮痛作用はノルアドレナリン系を介した下行性疼痛抑制系促進の影響が大きく,脊髄での鎮痛作用においてセロトニンが関与している可能性はあるものの,他の系よりも影響は少ないと考えられた."新潟医学会雑誌. 2023, 137(11), 399-409.新大院博(医)第1148

    Effect of Mandibular Bilateral Distal Extension Denture Design on Masticatory Performance

    新潟大学Niigata University博士(歯学)Purpose: Removable partial dentures have a wide range of applications as missing tooth prostheses, and the denture design can be quite diverse. However, the factors affecting masticatory function remain unclear. We investigated the effect of denture design, especially the use of indirect retainers and major connectors, on masticatory function in patients with mandibular bilateral distal extension dentures. Methods: Fifty-three patients with mandibular Kennedy Class I and Eichner classification B occlusal support were included in this study. Masticatory performance (MP) was assessed using gummy jelly. For the dentures, the type of major connector (bar or apron), presence or absence of indirect retainers, number of rests, and number of artificial teeth were evaluated. The number of functional teeth and occlusal support, Eichner classification, maxillary denture usage, mandibular residual ridge height, and duration of denture use were also evaluated. The factors influencing MP were evaluated using the Mann-Whitney U test, Spearman's correlation coefficient, and logistic regression analysis. Results: MP was significantly higher when the major connector was bar-type or when there was an indirect retainer than with an apron-type or no indirect retainer. MP was positively correlated with the number of functional teeth, occlusal support, and number of rests. Logistic regression analysis revealed that the number of functional teeth and the presence or absence of an indirect retainer were factors that significantly influenced the MP. Conclusion: An indirect retainer setting is useful for achieving good masticatory performance in mandibular bilateral distal extension dentures.Journal of Prosthodontic Research. 2023.新大院博(歯)第538

    新潟大学Niigata University博士(歯学)Cholesterol plays an important role in cancer progression, as it is utilized in membrane biogenesis and cell signaling. Cholesterol-lowering drugs have exhibited tumor-suppressive effects in oral squamous cell carcinoma (OSCC), suggesting that cholesterol is also essential in OSCC pathogenesis. However, the direct effects of cholesterol on OSCC cells remain unclear. Here, we investigated the role of cholesterol in OSCC with respect to caveolin-1 (CAV1), a cholesterol-binding protein involved in intracellular cholesterol transport. Cholesterol levels in OSCC cell lines were depleted using methyl-β-cyclodextrin and increased using the methyl-β-cyclodextrin-cholesterol complex. Functional analysis was performed using timelapse imaging, and CAV1 expression in cholesterol-manipulated cells was investigated using immunofluorescence and immunoblotting assays. CAV1 immunohistochemistry was performed on surgical OSCC samples. We observed that cholesterol addition induced polarized cell morphology, along with CAV1 localization at the trailing edge, and promoted cell migration. Moreover, CAV1 was upregulated in the lipid rafts and formed aggregates in the plasma membrane in cholesterol-added cells. High membranous CAV1 expression in tissue specimens was associated with OSCC recurrence. Therefore, cholesterol promotes the migration of OSCC cells by regulating cell polarity and CAV1 localization to the lipid raft. This study aimed to explore the effect s of excess glutamate on astrocytes both in vitro and in vivo. Methods We used astrocyte-enriched culture s (AECs), in which microglia were removed from mixed glial cultures, to investigate the effects of extracellu lar glutamate on these cells by microarray, quantitative PCR, ELISA, and immunostaining. We also examined the production of lipocalin 2 (Lcn2) by immunohistochemistry in the brains of mice after status epilepticus induced by pilocarpine and by ELISA in the cerebrospinal fluid (CSF) of patients characterised by status epilepticus Results Microarray analysis identified Lcn2 as a factor upregulated in AECs by excess glutamate; glutamate addition increased Lcn2 in the cytoplasm of astrocytes and AECs released Lcn2 in a concentration dependent manner. Lcn2 production was reduced by chemical inhibition of metabotropic glutamate receptor or siRNA knockdown of metabotropic glutamate receptor 3 .     新潟大学Niigata University博士(医学)Abstract NRF2 is a transcription factor responsible for antioxidant stress responses that is usually regulated in a redox-dependent manner. p62 bodies formed by liquid-liquid phase separation contain Ser349-phosphorylated p62, which participates in the redox independent activation of NRF2. However, the regulatory mechanism and physiological significance of phosphorylation remain unclear. Herein, we identify ULK1 as a kinase responsible for phosphorylation of p62. ULK1 co-localizes with p62 bodies, and directly interacts with p62. This phosphorylation allows KEAP1 to be retained within p62 bodies, activating NRF2. p62S351E/+ mice are phosphomimetic knock-in mice in which Ser351 corresponding to human Ser349 is replaced by Glu. These mice, but not phosphodefective p62S351A/S351A mice, exhibit NRF2 hyperactivation and growth retardation, the latter caused by malnutrition and dehydration due to obstruction of the esophagus and forestomach secondary to hyperkeratosis. p62S351E/+ mice are a phenocopy of systemic Keap1-knockout mice. Our results expand our understanding of the physiological importance of the redox-independent NRF2 activation pathway and provide new insight into the role of phase separation in this process.The EMBO Journal. 2023, 42, e113349.新大院博(医)第1146
