Characterization of Carbofuran Hydrolase Enzyme Bacillus Cereus Strain HS06 and Bacillus Paramycoides Strain BSAS15
Bacillus cereus strain HS06 and Bacillus paramycoides strain BSAS15 are two bacteria known to have the ability to degrade carbofuran, an active compound used in pesticides which are very dangerous to the environment. This study aims to determine the characteristic of the carbofuran hydrolase extracelllular and intracellular from B. cereus strain HS06 and B. paramycoides strain BSAS15. Extracelllular and intracellular enzyme of carbofuran hydrolase were isolated from B. cereus strain HS06 and B. paramycoides strain BSAS15 cultures, enzyme activity was measured and tested for the effect of temperature (30-50oC), pH (5-9) and metals (Na, Ca, Fe, Mg, Mn). One unit of carbofuran hydrolase enzyme activity is defined as the amount of enzyme that can produce 1 mol of carbofuran-7-phenol per minute per ml of enzyme with carbofuran as the substrate. The results indicate that the carbofuran hydrolase intracellular enzyme produced by B. cereus and B. paramycoides has a higher activity than that from extracellular enzymes. The intracellular enzyme activiy of B. cereus has an optimum condition at 40°C, pH 9, and an increase in enzyme activity with the addition of Fe metal ions. Meanwhile, B. paramycoides at 40°C, pH 7, and with the addition of Fe metal. The carbofuran hydrolase enzyme from B. cereus and B. paramycoides have different optimal effectiveness characteristics with higher activity in its intracellular than the extracellular enzymes.50 PagesSkripsi Sarjan

The Relationship Between The Level of Parental Knowledge About Cleft Lip and Palate and Delays in Referral to Hospitals in Medan
Cleft lip and palate (CLP) is a common congenital orofacial disorder that requires early correction to minimize its impact and optimize treatment success. However, delays in patient referral to a hospital or doctor are often caused by parents' lack of awareness and knowledge about the impact of CLP and the importance of early management, including the use of feeding plates as the initial treatment. This delay in referral can hinder the achievement of optimal functional and aesthetic benefits for patients. To determine the factors that influence the delay in CLP management, as well as the relationship between the level of parental knowledge (general about CLP and specifically about feeding plates) and the delay in referral. A cross-sectional analytical survey was conducted on 103 parents of CLP patients in two hospitals in Medan City. Data were collected using a questionnaire, with the independent variable being the level of parental knowledge and the dependent variable being the delay in referral. Most (70.8%) parents had a moderate level of knowledge. There was a significant relationship (P=0.019) between the level of knowledge and referral delay. Patients with poor knowledge experienced a higher referral delay (77.8%) compared to those with good knowledge (38.1%). The level of parental knowledge is a factor that greatly influences the delay in referral/management of CLP diagnosis. There was a significant relationship between the level of parental knowledge, both regarding CLP and feeding plates, and referral delay (P<0.05).92 PagesSkripsi Sarjan
The Effect Of Customer Experience And Glad2glow Cushion Product Variety On Decisions Repurchase In Students University Of North Sumatera
This study aims to analyze the influence of Customer Experience and Product
Variety on Glad2Glow Cushion Repurchase Decisions in University of North
Sumatra students. The increasingly competitive growth of the local cosmetics
industry requires companies to be able to provide a satisfactory product use
experience while providing a variety of products that are relevant to consumer
needs. Although Glad2Glow is one of the well-known local cushion brands, the
level of consumer repurchase decisions still depends on how the user experience
and product diversity are able to meet user expectations. This study uses an
associative quantitative approach with a survey method. The research sample
amounted to 104 respondents who were selected using the purposive sampling
technique, namely University of North Sumatra students who had used Cushion
Glad2Glow more than once. Primary data was collected through the distribution of
questionnaires, while secondary data was obtained from journals, books, and other
supporting references. Data analysis was carried out using SPSS which included
validity, reliability, classical assumption test, descriptive analysis, multiple linear
regression, determination coefficient, t-test, and F test. Likewise, Product Variety
has been proven to have a positive and significant effect partially. Simultaneously,
the two independent variables were also proven to have a positive and significant
effect on the Repurchase Decision, which was shown by the value of Fcal 71.054 >
Ftable 2.70 and the significance value of 0.000 < 0.05. This confirms that product
user experience and a variety of product choices play an important role in increasing
consumers' tendency to repurchase Glad2Glow Cushion.119 PagesSkripsi Sarjan
Isolation and Selection of L-Asparaginase–Producing Microorganisms from Tempeh
Tempeh is a soybean-based fermented product rich in amino acids and
beneficial microorganisms, including L-asparaginase–producing species. The aim
of this study was to obtain L-asparaginase-producing bacteria and fungi from
tempeh. Tempeh in two different types of packaging was obtained from tempeh
industry in Medan Tuntungan. Bacteria and fungi were isolated from the tempeh
and subsequently characterized macroscopically and microscopically. The ability
of the isolates to produce L-asparaginase was assessed qualitatively on M9 medium
by observing the color change to pink, and quantitatively by determining enzyme
activity and specific activity values. The most promising bacterial isolate with the
highest L-asparaginase activity was identified molecularly using 16S rRNA
primers, while fungal isolates were identified morphologically using standard
identification book. A total of 24 microbial isolates were obtained from the tempeh
samples, consisting of 7 bacterial isolates, 10 lactic acid bacterial (LAB), and 7
fungal. The bacterial and fungal isolates displayed variation in both colony
morphology and microscopic characteristics. Qualitative screening yielded 6
bacterial isolates and 4 fungal isolates with potential L-asparaginase activity,
whereas none of the LAB isolates showed enzyme production. The highest Lasparaginase
activity among bacterial isolates was recorded for isolate TDN 04,
with a specific activity of 338,448U/mg in value, and the highest fungal activity was
recorded for isolate JTP3, with a specific activity of 445,75 U/mg in value .
Molecular identification showed that isolate TDN 04 shared 99.53% similarity with
Klebsiella aerogenes, while fungal isolate JTP3 exhibited morphological
characteristics consistent with the genus Aspergillus. Klebsiella aerogenes isolate
TDN 04 and Aspergillus sp. isolate JTP3 are therefore the bacterial and fungal
isolates from tempeh with the highest L-asparaginase activity, demonstrating
potential in converting asparagine into aspartic acid and ammonia.67 PagesTesis Magiste
Problematics of Copyright Protection Against Cinematographic Piracy Crimes on the Tiktok Social Media Platform
The development of information technology has transformed patterns of media
consumption in society, with TikTok emerging as a short-video-based social media
platform that plays a significant role in global promotion and entertainment. However,
behind its creative potential, TikTok has also become a space in which violations of
Intellectual Property Rights are widespread, particularly film piracy as a form of
cinematographic work. Normatively, criminal copyright law functions as a repressive
instrument to address such violations, as stipulated in Article 113 of Law Number 28
of 2014 on Copyright. This research examines the role and effectiveness of criminal
copyright provisions in combating film piracy on the TikTok platform, while also
identifying gaps in law enforcement arising from the massive, anonymous, and crossjurisdictional
nature of cybercrime.
This study employs a normative legal research method with a descriptiveanalytical
character, using secondary data sources consisting of primary, secondary,
and tertiary legal materials, which are qualitatively analyzed through statutory and
case approaches.
The findings show that although Law Number 28 of 2014 on Copyright provides
criminal protection for the economic rights of cinematographic works as an ultimum
remedium, its effectiveness in combating digital piracy on platforms such as TikTok
remains limited. This ineffectiveness arises from weaknesses in legal substance, law
enforcement capacity, and legal culture, which collectively undermine legal certainty
and justice for copyright holders. Therefore, strengthening future enforcement requires
legal reconstruction through sanction reform and the adoption of a hybrid offense
regime that distinguishes between non-commercial users, subject to complaint-based
offenses and restorative justice, and commercial offenders, who should be treated as
ordinary offenses to ensure responsive and proportional copyright protection in the
digital era.161 PagesTesis Magiste
Analysis of Social Resilience and Double Burden of Sandwich Generation (Social Practice Study on Generation Z Students at the University of North Sumatra)
This study analyzes social practices, differences, and relationships between dual roles and dual burdens on social resilience among Generation Z students at the University of North Sumatra who belong to the sandwich generation and non sandwich generation particularly, open sandwich generaton category. This phenomenon emerged amid demographic transition and increasing academic demands. Using a descriptive and correlational quantitative approach, the study involved 100 respondents (50 from the sandwich generation and 50 from the non sandwich generation). Data were analyzed using the Independent Sample T-Test and Spearman's Correlation through SPSS software. The results showed significant differences in all variables studied. The sandwich generation group had a much higher mean dual role and dual burden, but a lower level of social resilience than the non-sandwich generation group. Correlation analysis revealed that dual roles were significantly related to social resilience, while dual burdens as a whole did not show a significant correlation. A key finding of this study is the existence of “paradoxical resilience,” in which high dual burdens actually encourage positive adaptive effects on intergenerational communication and time
management. This is reinforced by high-quality family relationships and the use of digital platforms as social capital that neutralizes the negative effects of dual burdens. In conclusion, sandwich generation students at USU exhibit unique adaptation patterns; despite being burdened by roles and finances, they possess strong adaptive capacities thanks to cultural capital. Academic institutions are
advised to provide flexible policy support to reduce the structural burdens on students in this group.197 PagesSkripsi Sarjan
Manufacturing Of Activated Carbon From Cassava Peels (Manihot Utilissima) For Reduction Of Iron (Fe) Metal Levels In Drilled Well Water
Borehole water in some areas often contains iron metal (Fe) that exceeds the threshold set by Permenkes No. 2 Year 2023, which is 0.2 mg/L. Excess iron content can reduce water quality and cause health problems. This study aims to reduce Fe metal levels using activated carbon from cassava peel (Manihot esculenta) and determine the characteristics of activated carbon crystallinity through X-Ray Diffraction (XRD) tests. Cassava peels were dehydrated through drying and oven, carbonized at 400°C and chemically activated using 30% NaOH solution. The adsorption process was carried out for 90 minutes with adsorbent mass variations of 1; 2; and 3 g in 100 mL of solution. The initial Fe level was 8.78 mg/L. After adsorption, the Fe concentration decreased to 6.99 mg/L (20.39% efficiency) at 1 g; 5.5 mg/L (37.36%) at 2 g; and 7.51 mg/L (14.36%) at 3 g. The XRD test results showed the amorphous structure of the adsorbent. XRD test results showed an amorphous structure with a dominant peak of carbon 2θ = 23.94˚ in the [002] plane and a weak peak 2θ = 44.6˚ in the [100] plane. Activated carbon from cassava peel has the potential as an alternative adsorbent, but Fe levels have not met the drinking water quality standards set.79 PagesSkripsi Sarjan

Comparative Experimental Study of High Strength Concrete (HSC) with the Addition of Steel Fiber and Palm Shell Ash (PSA) and Normal Concrete
This study aims to analyse the effect of palm shell ash (PSA) as a partial cement replacement material combined with steel fibres on the mechanical properties of concrete, particularly High Strength Concrete (HSC) and the flexural behaviour of composite concrete beams. Palm shell ash was used at 15% by weight of cement, while steel fibres were added at 2% by volume of concrete. The experimental program included both fresh and hardened concrete tests, namely slump flow, T50 time, V-funnel, L-box, compressive strength, splitting tensile strength, and flexural strength tests. The results of fresh concrete tests indicated that all parameters met EFNARC standards, with slump flow values ranging from 610–765 mm, T50 times between 5.83–6.03 seconds, V-funnel flow times of 11.29–18.15 seconds, and L-box ratios between 0.907–0.95. These results confirm that the mix achieved adequate flowability and stability to be categorised as Self-Compacting Concrete (SCC). The compressive strength results demonstrated a significant improvement, increasing from an average of 30.89 MPa (normal concrete) to 90.25 MPa (HSC), corresponding to a 65.7% enhancement. Similarly, the splitting tensile strength rose from 2.95 MPa to 5.63 MPa, while flexural strength increased from 6.27 MPa to 16.69 MPa with the addition of HSC layers ranging from 1 cm to 5 cm thickness. The improvement in strength is attributed to the pozzolanic reaction of palm shell ash, which contributes to the formation of calcium silicate hydrate (C–S–H), and the reinforcing effect of steel fibres that enhance ductility and crack resistance. Flexural beam displacement analysis revealed that increasing the HSC layer thickness improved flexural stiffness and reduced maximum deflection. Therefore, the combination of 15% palm shell ash and 2% steel fibres effectively enhances the mechanical and structural performance of concrete while promoting the use of environmentally friendly materials in modern concrete technology.182 PagesTesis Magiste
