    BulletinPancetta (raw bacon) was prepared using a commercially available dehydrant sheet (Okamoto Co., Ltd.), and the occurrence of spoilage and food poisoning bacteria was investigated. Pancetta is pork belly that has been salted over an extended period and is generally not heat-treated. It is prepared through long-term aging and dehydration, has a strong umami flavor, and is prized in Europe as a luxury ingredient. Recently, it has attracted attention due to its easy preparation using dehydrant sheets even at home in Japan. However, its microflora remains poorly understood. In this study, raw bacon was prepared using a commercially available dehydrant sheet, referring to a publicly available recipe for pancetta, and the prevalence of putrefactive and food poisoning bacteria was investigated. Additionally, we investigated whether the microflora changes depending on the length of the treatment period with the dehydrant sheet. Food hygiene microorganisms were detected following conventional methods. For the viable bacteria, Staphylococcus aureus, cold putrefactive bacteria, coliform bacteria, Escherichia coli, and Clostridia, the raw materials were appropriately diluted, and the number of bacteria was quantified. The presence or absence of Listeria and Salmonella was determined after the enrichment culture. Meat weight (%) decreased from 90% to 84%, 83%, and 76% as the dehydrant sheet treatment period (1, 2, 3, and 4 weeks, respectively) increased. The number of viable, cold putrefactive, and coliform bacteria decreased with weight reduction. When fresh meat was inoculated with S. aureus and E. coli at a rate of 105 /g and treated with a dehydrated sheet for 2 weeks, both bacteria did not decrease. However, in a similar study that inoculated meat at a rate of 102/g, S. aureus and E. coli were undetected after 2 weeks of treatment.departmental bulletin pape

    Reduced pigmentation and thyroid hormone disruption in zebrafish embryos caused by industrial sludge near Bohai Bay, China

    journal articleIn recent years, pollution caused by the discharge of industrial wastewater into Bohai Bay has posed a potential threat to the health of surrounding residents. Sludge was collected from the outlet of a factory that discharges effluent into Bohai Bay, and alcohol extracts of sludge (SE) were prepared. We confirmed by UPLC-MS/MS analysis that the SE contained PAHs, including fluorene, pyrene, and phenanthrene. Zebrafish embryos as animal models were exposed to 0.1, 0.3, 0.5, 1 and 5 ​mg/mL SE from 2 to 4 ​h post-fertilization (hpf) until 120 hpf. The results showed that SE caused a concentration-dependent increase in mortality and a decrease in hatchability. We found that SE significantly reduced eye pigmentation and decreased the movement of embryos and larvae. In addition, SE decreased triiodothyronine (T3) content and down-regulated the mRNA expression of some thyroid hormone-related genes including TPO and Thrβ, and caused the up-regulation of Dio2 and Dio3 at 120 hpf. Exposure to three individual PAHs found in SE, namely fluorene, pyrene, and phenanthrene, caused morphological and transcriptional changes that were similar to those caused by SE exposure. These findings indicate that PAHs in SE can reduce the pigmentation of zebrafish, which may be related to the genetic changes associated with thyroid hormones, and that zebrafish eye pigmentation can be used as an indicator of PAHs exposure.journal articl
