Clinical and Correlated Responses among Steroid Hormones and Oxidant/Antioxidant Biomarkers in Pregnant, Non-Pregnant and Lactating CIDR-Pre-Synchronized Dromedaries (Camelus dromedarius)
ArticleOverproduction of free radicals is controlled by antioxidant defense mechanisms. These defense mechanisms are achieved by antioxidant enzymes such as catalase (CAT). The current study aimed to assess the changes in steroid hormones, oxidant/antioxidants biomarkers, lipid profiles/liver functions indices, renal function biomarkers and minerals metabolism in non-pregnant, lactating or pregnant one-humped she-camels (Camelus dromedarius) pre-synchronized with controlled internal drug releasing. The study also focused on the correlational relationships between steroid hormones and the oxidant/antioxidant biomarkers, lipid profiles and liver functions indices, renal functions and mineral metabolism in these she-camels. The study was conducted on apparently healthy dromedary she-camels (n = 60) during breeding season. A sexually active camel-bull was introduced to she-camels pre-synchronized with CIDR. Fifty to sixty days after natural mating, she-camels were examined for pregnancy. She-camels were divided into three main groups according to both pregnancy and lactation as following: pregnant (PREG, n = 38) which was kept as control one, non-pregnant and lactating (LACT, n = 8), and non-pregnant and non-lactating she-camels (NPREG, n= 14). Steroid hormones, i.e., progesterone (P4), estradiol (E2) and cortisol, oxidant indictors, i.e., malondialdehyde (MDA), antioxidant biomarkers, i.e., superoxide dismutase (SOD), total antioxidant capacity (TAC), CAT and reduced glutathione (GSH), lipid profiles indices, renal functions and related minerals were assessed. The present study confirmed the efficacy of using CIDR for synchronization in she-camels. Significant elevations in serum steroids hormones in PREG compare with LACT and NPREG. The highest concentrations of MDA as lipid peroxidation and oxidative stress indictors and lowest levels of antioxidant biomarkers except for SOD, i.e., TAC, CAT and GSH, were reported in PREG compared with LACT and NPREG. PREG showed the highest liver enzymes activities and lowest total protein values. Remarkable increases in serum concentrations of renal function parameters and phosphorous (P) were observed in PREG when compared with the other two groups. The investigated she-camels revealed significant correlation between steroid hormones and the oxidant biomarkers, antioxidant biomarkers, liver functions, renal functions and minerals metabolism parameters. P4 showed positive correlations with antioxidant biomarkers, i.e., TAC, CAT and GSH, and serum aspartate aminotransferase (AST) activities, whereas negative correlations were reported between P4 and renal functions biomarkers, i.e., blood urea nitrogen (BUN), creatinine (Cr) and creatinine kinase (CK), and minerals metabolism parameters, i.e., P and magnesium (Mg), in CIDR pre-synchronized she-camels. In contrast, E2 and cortisol showed negative correlations with antioxidant biomarkers, i.e., TAC, CAT and GSH, lipid profiles/liver functions indices, i.e., AST, alkaline phosphatase (ALP) and γ-glutamyl transferase (GGT), CK and Mg, however, positive correlations were demonstrated between E2 and cortisol, and MDA, Cr and P in investigated she-camels. In conclusion, the present study confirmed the efficacy of using CIDR for synchronization in she-camels. The highest MDA levels as indictors for oxidative stress and the lowest antioxidant levels, i.e., TAC, CAT and GSH, except for SOD in pregnant she-camels, were attributable to physiological oxidative stress as excellent compensatory responses observed in the PREG group to face such a physiologic stage. Moreover, lower P levels in non-pregnant she-camels would be contributed to failure of conception or early embryonic death. The investigated she-camels revealed significant correlations between steroid hormones and the oxidant indicators, antioxidant biomarkers, lipid profile indices and renal functions biomarkers that provided better understanding for physiological stress during pregnancy in camels.journal articl
The unique physiological features of the broiler pectoralis major muscle as suggested by the three-dimensional ultrastructural study of mitochondria in type IIb muscle fibers
ArticleTypical skeletal muscles are composed of mixed muscle fiber types, which are classified as slow-twitch (type I) and fast-twitch (type II) fibers, whereas pectoralis major muscles (PMs) in broiler chickens are 100% composed of type IIb fast-twitch fibers. Since metabolic properties differ among muscle fiber types, the combination of muscle fiber types is involved in physiological functions and pathological conditions in skeletal muscles. In this study, using serial block-face scanning electron microscopy, we compared three-dimensional (3D) mitochondrial properties in type IIb fibers in broiler PMs and those in type I fibers of broiler gastrocnemius muscles (GMs) heterogeneously composed of slow- and fast-twitch muscle fibers. In type I fibers in the GMs, elongated mitochondria with numerous interconnections to form a substantial network among myofibrils were observed. Along with lipid droplets sandwiched by mitochondria, these features are an adaptation to effective oxidative respiration and constant oxidative damage in slow-twitch muscle fibers. In contrast, type IIb fibers in the PMs showed small and ellipsoid-shaped mitochondria with few interconnections and no lipid droplets, forming a sparse network. The mitochondrial spatial network comprises of active mitochondrial dynamics to reduce mitochondrial damage; therefore, type IIb fibers possess physiologically low capacity to maintain mitochondrial wellness due to static mitochondrial dynamics. Based on 3D mitochondrial properties, we discussed the contrasting physiological functions between type I and IIb fibers and proposed a high contractile power and low stress resistance as unique physiological properties of broiler PMs.journal articl
Comparison of RGB and Multispectral Unmanned Aerial Vehicle for Monitoring Vegetation Coverage Changes on a Landslide Area
ArticleThe development of UAV technologies offers practical methods to create landcover maps for monitoring and management of areas affected by natural disasters such as landslides. The present study aims at comparing the capability of two different types of UAV to deliver precise information, in order to characterize vegetation at landslide areas over a period of months. For the comparison, an RGB UAV and a Multispectral UAV were used to identify three different classes: vegetation, bare soil, and dead matter, from April to July 2021. The results showed high overall accuracy values (>95%) for the Multispectral UAV, as compared to the RGB UAV, which had lower overall accuracies. Although having lower overall accuracies, the vegetation class of the RGB UAV presented high producer’s and user’s accuracy over time, comparable to the Multispectral UAV results. Image quality played an important role in this study, where higher accuracy values were found on cloudy days. Both RGB and Multispectral UAVs presented similar patterns of vegetation, bare soil, and dead matter classes, where the increase in vegetation class was consistent with the decrease in bare soil and dead matter class. The present study suggests that the Multispectral UAV is more suitable in characterizing vegetation, bare soil, and dead matter classes on landslide areas while the RGB UAV can deliver reliable information for vegetation monitoring.journal articl

ArticleThe purpose of this study was to compare effects of different amounts of carbohydrate (CHO) in beverage on plasma glucose (GLU) concentration during exercise after feeding breakfast. Seven healthy Japanese male subjects performed 30-min cycling at an intensity on the 75% of maximal load at 3-h after feeding breakfast under 3 types of CHO (0 g, 30 g and 150 g) beverage conditions. The subjects ingested 500 mL of beverage dissolved each amount of glucose at 30-min before the exercise. The GLU concentrations were measured at 35- and 10-min before the exercise and every 5-min during the exercise. The minimum GLU concentrations under the 30 g condition for all subjects were lower than a criterion for hypoglycemic (72 mg / dL). The mean value also fell below the criterion at from 10- to 20-min during exercise. The minimum GLU concentration under the 150 g condition of only one subject was lower than the criterion, but the mean value did not fall below the criterion. Significant negative correlations were found between the subjects’ maximum oxygen uptake (VO_2max) and the decreases in GLU concentration from maximum to minimum under both the 30 g (r = -0.947, p < 0.01) and 150 g conditions (r = -0.884, p < 0.01). This study clarified that feeding breakfast at 3-h before the exercise followed by the 30 g CHO beverage ingestion at 30 min before the exercise induced exercise-induced hypoglycemia, and that the subjects with higher VO_2max had the greater decreases in GLU concentration.journal articl
ブドウ灰色カビ病に対する微生物農薬としてのヴィンヤード野生酵母の探索
Article自然発酵ワイン沈殿物,本学キャンパスおよび市販の植物材料から野生酵母225株を分離した.この中から選抜した59株を用いブドウ灰色カビ病の病原菌Botrytis cinereaに対する抗カビ作用をin vitroで評価したところ,強い抗カビ作用を示す20株が得られた.この内,最も多くの16株が得られたSaccharomyces cerevisiae について,遺伝子多型解析による株鑑別を行い,抗カビ作用を比較したところ,株型ごとに抗カビ作用の強さが異なることがわかった.このとき,S. cerevisiaeの多型解析には自動rRNA遺伝子間多型解析(ARISA)が従来の手法に比べ極めて有用性が高いことがわかった.The antifungal activity against Botrytis cinerea, a pathogen of grape gray mold disease, was validated in vitro for 59 of 225 wild yeast strains isolated from naturally fermented wine samples (69 strains), our university campus plants (129 strains), and commercially available fruits (27 strains). Twenty strains showing strong antifungal activity were identified. The majority of the strains were Saccharomyces cerevisiae (16 of the 20 strains). The S. cerevisiae strains were differentiated by genetic polymorphism analyses with comparisons of their antifungal activities. As a result, it was clarified that each strain type showed different antifungal activity, and that automatic rRNA intergenic spacer analysis was extremely useful for the differentiation of S. cerevisiae strains when compared to other conventional polymerase chain reaction-based methods.journal articl
Radiotherapy-induced tumor lysis syndrome in a dog with thymoma
ArticleA 13-year-old, female, mixed-breed dog with a huge cranial mediastinal mass underwent radiotherapy (RT). On the following day, the dog presented with lethargy and anorexia. Hematological examination revealed elevated levels of blood urea nitrogen, creatinine, inorganic phosphorus, potassium, lactate dehydrogenase, creatine phosphokinase and aspartate aminotransferase, decreased calcium level, and metabolic acidosis. Urine output markedly decreased. The patient recovered with fluid therapy and diuretic therapy; however, died suddenly from an unknown cause 11 days after RT completion. Histopathological examination after necropsy showed thymoma in the cranial mediastinum and extensive tubular necrosis of both kidneys which may be due to RT-induced tumor lysis syndrome (TLS). This report suggests that the risk of TLS should be evaluated in dogs with thymoma who undergo RT.journal articl
National parks, protected areas and biodiversity conservation in North Korea: opportunities for international collaboration
ArticleNational parks and protected areas represent one of the best ways to safeguard nature and preserve biodiversity, especially endemic and endangered species. Currently, more than 110 countries have some form of protected areas within their borders. North Korea’s comprehensive network of national parks and protected areas are rich in biodiversity and ecological value; however, contemporary research on the integrity of North Korea’s protected lands is incomplete. In this paper, we utilize a remote sensing approach to explore North Korea’s protected area system and investigate the current state of biodiversity conservation. We find that several key biodiversity areas have experienced severe tree loss and habitat fragmentation since 2000, indicating that the effectiveness of national park governance and management toward guarding against biodiversity loss is inadequate. To address the issue of biodiversity conservation, we recommend pathways for international cooperation aimed at enhancing conservation efforts and comment on the steps North Korea can take to achieve the United Nations Convention on Biological Diversity’s Aichi Targets and Sustainable Development Goals.journal articl
Effects of temporary cessation of milking following intramammary cefazolin sodium infusion on residual antibiotic in lactating dairy cows
ArticleThe aims of studies were to estimate the withdrawal period of antibiotic from milk after the intramammary infusion of cefazolin sodium (CEZ) in cows with difficulties in frequent milk discharge due to disease such as teat injury. The period was compared among cows milked twice a day after 150 or 450 mg of CEZ were administered to all quarters (Study 1, 2) and the cows in which milking of front-right quarter was ceased for five days after administration of these infusions to only that quarter (Study 3). In Studies 1 and 2, the median of 17.66 µg/ml and 83.18 µg/ml of CEZ were detected in the samples of first milking after intramammary administration, respectively; however, there was no residual antibiotic by 72 hr in all cows. In Study 3, the median of 1.96 µg/ml of CEZ was detected in the sample after the resumption of milking at 120 hr, and the residual was eliminated by 174 hr. The withdrawal period may be prolonged by the cessation of milking after administration, and the period is the total time from cessation to 72 hr after the resumption of milking.journal articl
USP2-Related Cellular Signaling and Consequent Pathophysiological Outcomes
ArticleUbiquitin specific protease (USP) 2 is a multifunctional deubiquitinating enzyme. USP2 modulates cell cycle progression, and therefore carcinogenesis, via the deubiquitination of cyclins and Aurora-A. Other tumorigenic molecules, including epidermal growth factor and fatty acid synthase, are also targets for USP2. USP2 additionally prevents p53 signaling. On the other hand, USP2 functions as a key component of the CLOCK/BMAL1 complex and participates in rhythmic gene expression in the suprachiasmatic nucleus and liver. USP2 variants influence energy metabolism by controlling hepatic gluconeogenesis, hepatic cholesterol uptake, adipose tissue inflammation, and subsequent systemic insulin sensitivity. USP2 also has the potential to promote surface expression of ion channels in renal and intestinal epithelial cells. In addition to modifying the production of cytokines in immune cells, USP2 also modulates the signaling molecules that are involved in cytokine signaling in the target cells. Usp2 knockout mice exhibit changes in locomotion and male fertility, which suggest roles for USP2 in the central nervous system and male genital tract, respectively. In this review, we summarize the cellular events with USP2 contributions and list the signaling molecules that are upstream or downstream of USP2. Additionally, we describe phenotypic differences found in the in vitro and in vivo experimental models.journal articl
Vertical distribution of tree fine roots in the tephra profile with two buried humic soil layers
ArticleSurface humic soils, where fine roots are mainly distributed, can be accidentally buried due to coverage by deposits such as volcanic ash. This buried humic soil may influence the vertical distribution of fine roots because soil organic matter strongly affects soil functions. However, fine root distributions in buried humic soils are little understood. In order to elucidate the effects of buried humic soils on fine root distribution, we investigated fine root biomass and soil characteristics in a soil profile down to 3.3 m with two buried humic soils formed by tephra in Tomakomai, Hokkaido, Japan. In this profile, fine root biomass decreased with soil depth, but increased in buried humic soils that had higher soil total carbon (C) content and higher fine soil ratio than buried nonhumic soils. These results lead us to surmise a preferential development of active fine roots in buried humic soils rich in organic C rather than nonhumic soils.journal articl
