    An overview of reclaimed wastewater reuse in gold heap leaching

    "The intensive use of groundwater by the gold mining industry is an important sustainability concern, especially in arid and semiarid regions where groundwater is a scarce resource. Alternatives, such as water reuse, treatment and recycling, have been implemented to overcome this issue. The potential use of reclaimed wastewater in gold heap leaching, without decreasing the process efficiency, has not yet been considered. Hence, this review focuses on the physicochemical, biological, and chemical features (organic matter, microbial loads, metal ions, and anions) of reclaimed wastewater that may limit its use in gold heap leaching.

    Expresión de péptidos antigénicos y antivirales del virus sincicial respiratorio en plantas de tabaco

    "El virus sincicial respiratorio (VSR) es una de las principales causas de infecciones del tracto respiratorio bajo (IRB) en el mundo. Las infecciones por VSR causan anualmente cerca de 34 millones de episodios de IRBs agudas, 66,000 muertes y costos por más de 2.4 miles de millones de dólares. A pesar de la importancia del VSR, no existe actualmente una vacuna disponible en el mercado. En este trabajo se desarrolló una construcción que codifica epítopos antigénicos de las proteínas F, G y M del VSR optimizadas para su expresión estable en el citoplasma de células vegetales. La construcción, denominada pBI-VSR, además codifica para péptidos espaciadores ricos en prolina y una región del virus del sarampión para obtener un balance de respuesta inmune Th1/Th2 que ha sido una limitante en trabajos previos en el desarrollo de vacunas contra VSR. Con pBI-VSR se realizó la transformación genética de plantas de tabaco (N. benthamiana) vía Agrobacterium tumefaciens, de las cuales se obtuvieron siete líneas transgénicas para la generación T2. Además se diseñó mediante PCR de fusión un gen que codifica el péptido con actividad antiviral RhoA, el cual fue fusionado a dos moléculas acarreadoras, liquenasa (LicKM) y la proteína de la cápside (CP) del virus del mosaico de la alfalfa para transformar transitoriamente plantas de tabaco, usando un vector de expresión basado en el virus del mosaico del tabaco. Las proteínas de fusión RhoA-LicKM y RhoA2-FLd25CP fueron expresadas eficientemente e inhibieron la replicación del VSR in vitro en un 50% y 80% respectivamente a una menor concentración que el péptido sintético RhoA usado como control. Estos resultados indican el potencial de sistemas de producción de biofármacos en plantas y, para el caso de transformación transitoria, la producción de un péptido antiviral en un tiempo corto y la ventaja que da utilizar moléculas acarreadoras para incrementar la expresión y eficacia.""Respiratory Syncytial Virus (RSV) is one of main causes of viral lower respiratory tract illness (LRI) in infants and young children worldwide. RSV causes nearly 34 million episodes of acute LRIs annually, with an estimated 66,000 deaths and costs of more than 2.4 billion dollars per year. Despite the burden of the disease, there is no licensed vaccine against RSV. In this work we developed a construct encoding plant-optimized sequences of epitopes from the antigenic F, G and M, RSV proteins in the cytoplasm of the plant cells. This genetic construct named pBI-RSV has proline-rich linkers and a measles virus region though to balance the Th1/Th2 immune response, which has been a limiting factor on the development of vaccines against RSV in previous studies. With this construct, the genetic transformation of tobacco (Nicotiana benthamiana) via Agrobacterium tumefaciens was performed. We obtained seven transgenic lines up to the T2 generation. We also engineered a synthetic gene encoding the antiviral RhoA peptide fused to carrier polypeptides, either lichenase (LicKM) or the coat protein (CP) of alfalfa mosaic virus. These constructs were introduced into tobacco plants using transient transformation by means of a tobacco mosaic virus-based expression vector and purified the recombinant proteins. The RhoA peptide fusion proteins were efficiently expressed in N. benthamiana plants, and two of them, RhoA-LicKM and RhoA2-FLd25CP inhibited RSV in vitro replication by 50% and 80%, respectively, at lower concentrations than the synthetic peptide used as a control. These data on biological activity indicate the feasibility of expression systems in plants for biopharmaceutical production and transient transformation, the production of an antiviral RhoA peptide in a short time in plants, and the advantage of using carrier molecules to enhance the level of expression and its efficacy.

    Delayed quadratic buck converter

    "Nowadays, the microprocessors family normally require for the input source not only low voltage but also high current. There exists an increasing necessity for developing new applied techniques on topologies and control strategies, which may be able to fulfil such requirements. Although a considerable amount of applications use the conventional buck converter for step-down for DC/DC conversion, when a high step-down conversion is required is a much better choice to implement the quadratic buck converter (QBC); however, the problem for obtaining a high step-down conversion at wide duty ratio still remains. This document proposes the delayed quadratic buck converter, which offers a very high-step-down dc-dc conversion with a wide duty cycle. The QBC is modified in its topology by adding an inductor, which allows us to obtain for the duty cycle a higher increment than this normally obtained by a quadratic one. Converter behaviour and analysis are illustrated. Experimental results are also shown for a conversion from 36 to 1.5 V and an output power of 20 W.

    Analysis of genetic diversity of mezcalero agaves from central México

    "En el Altiplano Potosino, un ecosistema árido y semiárido, Agave salmiana Otto Salm Dick ssp. crassispina (Trel Gentry) es una especie utilizada para la recolección de insectos comestibles y para producción de mezcal, bebida alcohólica tradicional con denominación de origen. El aprovechamiento para producción de mezcal de las poblaciones silvestres de Agave salmiana ssp. crassispina carece de un sistema de explotación basado en el conocimiento biológico de la especie. Esto tiene efectos demográficos, y a su vez podría causar el deterioro de sus recursos fitogenéticos y colocar a la especie en situación de riesgo. Este trabajo analizó diversidad genética y estructura poblacional de tres poblaciones silvestres de Agave salmiana ssp. crassispina en San Luis Potosí, México, mediante polimorfismos de longitud de los fragmentos amplificados, AFLP. La evidencia mostró un alto nivel de diversidad genética dentro de las poblaciones analizadas y un bajo nivel de diferenciación entre ellas, probablemente debido a una fragmentación del hábitat producida por actividades antropogénicas. Postulamos que la diversidad genética en las poblaciones analizadas, a pesar de la constante explotación, se origina por el sinergismo entre la polinización cruzada y la actividad de elementos genéticos transponibles. El alto grado de diversidad genética encontrado en las poblaciones silvestres analizadas en San Luis Potosí demuestra la importancia agroecológica de Agave salmiana ssp. crassispina en el Altiplano Mexicano, una región geográfica que abarca casi la mitad del territorio mexicano. Sin embargo, uso descontrolado y manejo inapropiado de las magueyeras silvestres podría arriesgar los recursos fitogenéticos de Agave salmiana ssp. crassispina.""The Potosino Upland Plateau is an arid and semiarid ecosystem. Agave salmiana Otto Salm Dick ssp. crassispina (Trel Gentry), which grows in this environment, is a species used for gathering of edible insects and for production of mezcal. Mezcal is a Mexican alcoholic beverage with denomination of origin. Production of mezcal with wild populations of Agave salmiana ssp. crassispina lacks an exploitation system based on biological knowledge of the species. The lack of organized use has demographic effects, which leads to degradation of genetic resources and risks to this species. This study analyzed genetic diversity and population structure of three wild populations of Agave salmiana ssp. crassispina from San Luis Potosí, Mexico by AFLP's (amplified fragment length polymorphism). The evidence showed a high level of genetic diversity within populations and low level of genetic differentiation among populations, probably as a result of habitat fragmentation produced by anthropogenic activities. We postulate that the genetic diversity in the wild populations analyzed, despite their constant exploitation, is originated by the synergism between cross-pollination and the activity of transposable genetic elements. The high level of genetic diversity found in the wild populations in San Luis Potosí shows agro-ecological relevance of Agave salmiana ssp. crassispina in the Mexican upland plateau. However, uncontrolled use and inappropriate management of wild populations could risk the genetic resources of Agave salmiana ssp. crassispina.

    Effects of shade on germination traits of the endangered cycad Dioon edule (Zamiaceae)

    "The endangered cycad Dioon edule requires shade provided by filtered sunlight under the canopy of trees or maternal plants during initial growth stages. It is known that germination improves under shade, but there is no report of radiation conditions. In order to understand how photosynthetic photon flux density (PPFD) affect germination traits, we evaluated some germination indexes. A sample of three mature strobili and 200 viable seeds per strobilus were selected to evaluate seed size (length, width, and fresh weight). Two experimental treatments were established simulating shade under the oak forest canopy with PPFD 81 µmol m−2 s−1 (PPFD81), and under maternal plant canopy with PPFD 17 µmol m−2 s−1 (PPFD17), as measured previously in the study site. Means of response variables (germinability, velocity of germination, synchronization, mean germination time and relative frequency of germination) for the two treatments were compared using a t-test and a correlation analysis. No significant correlation between seed size and germination traits was detected. Germinability was higher at PPFD17 (89 %) than PPFD81 (39 %), but mean germination time was similar across treatments. The velocity of germination was greater under PPFD17 but synchronization was the opposite. The low PPFD stimulated D. edule germination, but also the spectral composition must be evaluated.""La cícada en peligro de extinción Dioon edule requiere la sombra que le proporciona el dosel o las plantas maternas al fltrar la luz del sol durante su etapa inicial de crecimiento. Se sabe que la germinación es mejor con sombra, pero no existen reportes sobre las condiciones de la radiación. Evaluamos diferentes indicadores de germinación con el propósito de entender cómo es afectada por la densidad de fujo de fotones fotosintéticos (PPFD). Una muestra de tres estróbilos maduros y 200 semillas viables por estróbilo fueron seleccionadas para evaluar el tamaño de la semilla (largo, ancho y peso fresco). Dos tratamientos experimentales se establecieron simulando la sombra bajo el dosel del encinar con densidad de fujo de fotones fotosintéticos de 81 µmol m-2 s-1 (PPFD81) y bajo las plantas maternas con densidad de fujo de fotones fotosintéticos de17 µmol m-2 s-1 (PPFD17), como se midió previamente en el sitio de estudio. La media de las variables (capacidad germinativa, velocidad de germinación, sincronización, tiempo medio de germinación y frecuencia relativa de germinación) para los dos tratamientos se compararon usando una prueba de t. Se aplicó un análisis de correlación a los datos de germinación y tamaño de semilla. Se aplicó una regresión a los predictores ambientales (temperatura, humedad relativa, densidad de fujo de fotones fotosintéticos) de la capacidad germinativa. No se detectó correlación signifcativa entre el tamaño de la semilla y las variables de germinación. La capacidad germinativa fue mayor en PPFD17 (89 %) que PPFD81 (39 %), pero el tiempo medio de germinación fue similar. La velocidad de germinación fue mayor en PPFD17 pero menor en la sincronización. El bajo PPFD estimuló la germinación de D. edule, pero también la composición espectral debe ser evaluada

    Efectos del sombreado de las espinas en aspectos de fotosíntesis para tres especies de cactáceas

    "We evaluated the effect of spine removal on three cactus species varying in spine-shading of stems. Turbinicarpus schmiedickeanus showed the greater spine shading (c. 61 %), followed by Mammillaria zephyranthoides (43 %), and Echinocactus platyacanthus (17 %). We evaluated photosynthetic activity [titratable acidity, electron transport rate, potential quantum yield of Photosystem II, effective quantum yield of Photosystem II, and non-photochemical quenching], as well as pigment content (chlorophylls a and b, total chlorophyll, chlorophyll a/b, carotenoids, and total chlorophyll/carotenoids ratio). For T. schmiedickeanus, the species having the higher spine shading, spine removal increased the CAM activity (acidity level) and the effective quantum yield of Photosystem II and diminished both electron transport rate and non-photochemical quenching at 15:00 hours. For M. zephyranthoides, the species having intermediate spine shading, spine removal also increased the acidity level but diminished effective quantum yield of Photosystem II at midday, as well as chlorophyll a, total chlorophyll, and carotenoids, although there was higher non-photochemical quenching (heat quenching) as an adaptation to avoid photosynthetic damage. Spine removal did not affect acidity level or pigment content from E. platyacanthus, the species having the lower spine shading; however, at midday this species without spines showed a decrease of effective quantum yield of Photosystem II, as well as an increase of non-photochemical quenching and electron transport rate. We suggest that a main function of cactus spines is reduction of sun exposure to avoid damage to the photosynthetic apparatus.""Se evaluó el efecto de la remoción de espinas en tres especies de cactos que varían en el sombreado de sus tallos por las espinas. Turbinicarpus schmiedickeanus mostró el mayor sombreado (c. 61 %), seguido por Mammillaria zephyranthoides (43 %) y Echinocactus platyacanthus (17 %). Se evaluó la actividad fotosintética [acidez titulable, tasa de transporte de electrones, rendimiento cuántico potencial del Fotosistema II, rendimiento cuántico efectivo del Fotosistema II y disipación no fotoquímica], así como el contenido de pigmentos (cloroflas a y b, clorofla total, clorofla a/b, carotenoides y proporción clorofla total /carotenoides). Para T. schmiedickeanus, la especie con el mayor sombreado de espinas, la remoción de espinas incrementó la actividad MAC (nivel de acidez) y el rendimiento cuántico efectivo del fotosistema y disminuyó la tasa de transporte de electrones y la disipación no fotoquímica (disipación de calor) a las 15:00 h. Para M. zephyranthoides, la especie con el sombreado de espinas intermedio, la remoción de espinas incrementó el nivel de acidez pero disminuyó el rendimiento cuántico efectivo del fotosistema al mediodía, así como también la clorofla a, la clorofla total y los carotenoides, aunque hubo mayor disipación no fotoquímica (disipación de calor) como una adaptación para evitar el daño fotosintético. La remoción de espinas no afectó el nivel de acidez ni los pigmentos de E. platyacanthus, la especie con el menor sombreado de espinas. Sin embargo, sin espinas esta especie mostró disminución del rendimiento cuántico efectivo del fotosistema al mediodía, así como también incremento en disipación no fotoquímica y tasa de transporte de electrones. Se sugiere que una función principal de las espinas es la reducción de la exposición del sol para evitar daños en el aparato fotosintético.

    Effect of graphene oxide on bacteria and peripheral blood mononuclear cells

    "Background Driven by the potential biological applications of graphene, many groups have studied the response of cells exposed to graphene oxide (GO). In particular, investigations of bacteria indicate that there are 2 crucial parameters, which so far have only been investigated separately: GO size and exposure methodology. Our study took into account both parameters. We carefully characterized the samples to catalog sizes and structural properties, and tested different exposure methodologies: exposure in saline solution and in the presence of growth media. Furthermore, we performed experiments with peripheral blood mononuclear cells exposed to our GO materials. Methods Atomic force microscopy, scanning electron microscopy, Raman spectroscopy, X-ray photoelectron spectroscopy and transmission electron microscopy were used to characterize the morphology and composition of different samples of GO: GO-H2O, GO-PBS and GO-MG. Our samples had 2D sizes of ?100 nm (GO-H2O and GO-PBS) and >2 µm (GO-MG). We tested antibacterial activity and cytotoxicity toward peripheral blood mononuclear cells of 3 different GO samples. Results A size-dependent growth inhibition of Escherichia coli (DH5 ?) in suspension was found, which proved that this effect depends strongly on the protocol followed for exposure. Hemocompatibility was confirmed by exposing peripheral blood mononuclear cells to materials for 24 hours; viability and apoptosis tests were also carried out. Conclusions Our experiments provide vital information for future applications of GO in suspension. If its antibacterial properties are to be potentiated, care should be taken to select 2D sizes in the micrometer range, and exposure should not be carried out in the presence of grow media.

    Finite-time observer-based output-feedback control for the global stabilisation of the PVTOL aircraft with bounded inputs

    "In this work, an output-feedback scheme for the global stabilisation of the planar vertical take-off and landing aircraft with bounded inputs is developed taking into account the positive nature of the thrust. The global stabilisation objective is proven to be achieved avoiding input saturation and by exclusively considering the system positions in the feedback. To cope with the lack of velocity measurements, the proposed algorithm involves a finite-time observer. The generalised versions of the involved finite-time stabilisers have not only permitted to solve the output-feedback stabilisation problem avoiding input saturation, but also provide additional flexibility in the control design that may be used in aid of performance improvements. With respect to previous approaches, the developed finite-time observer-based scheme guarantees the global stabilisation objective disregarding velocity measurements in a bounded input context. Simulation tests corroborate the analytical developments. The study includes further experimental results on an actual flying device.

    Optimization of hydrogen production by the psychrophilic strain G088

    Tesis (Maestría en Ciencias en Biología Molecular)"El hidrógeno (H2) es considerado como uno de los vectores energéticos más prometedores para el futuro, posee características únicas como un alto rendimiento energético, es renovable, limpio y su combustión es sustentable y amigable con el medio ambiente debido a que no genera subproductos tóxicos. De los métodos de producción de H2, la fermentación oscura es el método biológico más usado debido a las ventajas que presenta. Sin embargo, la mayoría de los estudios referentes a la producción fermentativa operan a temperaturas mesófilas, termófilas e incluso hipertermófilas, y poco se conoce de los procesos psicrófilos que tienen como ventaja requerir menor gasto de energía para calentar los reactores durante el proceso. En esta tesis se estudió la producción de biohidrógeno utilizando la cepa psicrófila G088 estrechamente relacionada a Polaromonas rhizosphaerae [EF127651] utilizando xilosa, glucosa, fructosa, galactosa, lactosa o sacarosa como fuente de carbono a una temperatura de 20ºC. Los resultados mostraron que la cepa G088 produjo biohidrógeno usando todos los sustratos evaluados, produciendo desde 91.7 hasta 439.8 ml para lactosa y glucosa respectivamente. Sin embargo, la glucosa fue el sustrato con el que se obtuvo la mayor velocidad de producción y rendimiento de 50.1 ml/l/h y 1.7 mol H2/mol glucosa, respectivamente. Adicionalmente, la fermentación oscura es un proceso complejo el cual puede ser influenciado por diferentes factores, por lo tanto, también se evaluó el efecto de la temperatura, el pH y la concentración de sustrato sobre la producción de biohidrógeno, la velocidad de producción y el rendimiento, empleando glucosa. Se utilizó la metodología de superficie de respuesta con un diseño experimental Box-Behnken con el fin de determinar las condiciones óptimas de producción de biohidrógeno. De acuerdo al modelo matemático, las condiciones óptimas fueron temperatura de 26.3ºC, pH de 6.2 y concentración de glucosa de 25.31 g/l para una producción de biohidrógeno predicha de 503.34 ml de H2, una velocidad de producción de 37.91 ml/l/h y un rendimiento de 1.93 mol H2/mol glucosa. Se encontró que el efecto lineal del pH y el efecto cuadrático de la temperatura, pH y concentración de glucosa fueron los términos que afectaron a la producción de biohidrógeno. Por otra parte, la velocidad de producción fue afectada únicamente por el efecto cuadrático de la temperatura y concentración de glucosa, mientras que el efecto lineal y cuadrático de los tres factores afectó a la variable de rendimiento. Las condiciones óptimas fueron probadas experimentalmente por triplicado corroborando la correspondencia con el valor predicho por el modelo matemático. Los valores experimentales de la producción de biohidrógeno, velocidad y rendimiento bajo estas condiciones fueron 513±12.48 ml, 36.5±4.10 ml/l/h y 1.81±0.04 mol H2, respectivamente. Estos resultados demuestran que la cepa G088 tiene potencial para ser usada para el desarrollo de nuevos procesos biotecnológicos para la producción de biohidrógeno.""Hydrogen (H2) is considered as one of the most promising energy carriers for the future, it has several unique characteristics, such as high energy yield, it is renewable, clean and its combustion is sustainable and environmentally friendly since it does not generate toxic byproducts. Among hydrogen production methods, dark fermentation is the biological method most widely used. However, most of the studies addressing fermentative hydrogen production operate at mesophilic, thermophilic and even hyperthermophilic temperatures and little information is available using psychrophilic microorganisms. These processes consume less energy for heating the reactors during the process. In this thesis, the biohydrogen production was studied using the psychrophilic strain G088 closely related to Polaromonas rhizosphaerae [EF127651] using xylose, glucose, fructose, galactose, lactose or sucrose as a carbon source at temperature of 20ºC. The results showed that G088 strain produced biohydrogen using all the evaluated substrates, ranging from 91.7 to 439.8 ml for lactose and glucose respectively. Nevertheless, glucose was the best substrate with maximum values of biohydrogen production rate and yield of 50.1 ml/l/h and 1.7 mol H2/ mol glucose respectively. In addition, dark fermentation is a complex process, and can be influenced by different factors; thus, the effect of temperature, pH and substrate concentration on biohydrogen production, biohydrogen production rate and yield was also studied using glucose. Response surface methodology with a Box-Behnken design was used to determine the optimal biohydrogen production conditions. According to the mathematical model, temperature 26.30ºC, pH 6.2 and glucose 25.31 g/l were the optimum conditions for a predicted biohydrogen production of 503.34 ml of H2, and a biohydrogen production rate and yield of 37.91 ml/l/h and 1.93 mol H2/ mol glucose, respectively. The linear effect of pH and the quadratic effect of temperature, pH and glucose concentration were the most significant terms affecting biohydrogen production. Otherwise, biohydrogen production rate was affected only by the quadratic effect of temperature and glucose concentration, while biohydrogen yield was affected by the lineal and quadratic effect of the three factors. The optimal production conditions were experimentally tested by triplicate corroborating the correspondence with the predicted value by the mathematical model.

    Adaptive tracking control of Euler-Lagrange systems with bounded controls

    "We solve the simultaneous closed?loop identification and tracking?control problems for fully actuated Euler-Lagrange systems under input constraints. We use a nonlinear adaptive controller reminiscent of computed?torque?type controllers in which linear correction terms are saturated in order to comply with the imposed bounds on the control inputs. Adaptation, reminiscent of gradient methods, is used also with saturation. With respect to related literature, our contribution consists in establishing uniform global asymptotic stability. Therefore, our control scheme ensures robustness with respect to bounded perturbations and uniform convergence of the estimation errors for any initial conditions.
