Seed germination of Magnolia pugana (Magnoliaceae), an endemic and endangered species from Western Mexico
"Magnolia pugana es una especie endémica y en peligro de extinción localizada en el occidente de México, que se distribuye en bosques de galería en las barrancas del sur de Zacatecas y centro-norte de Jalisco. El objetivo de este trabajo fue conocer la viabilidad de las semillas e identificar los tratamientos para romper la latencia de semillas de esta especie. Se colectaron semillas de cuatro localidades, tres en Jalisco (Centro Universitario de Ciencias Biológicas y Agropecuarias, San Lorenzo y San Nicolás) y una en Zacatecas (Palo Verde). Se aplicaron los siguientes tratamientos germinativos: (1) remoción manual del arilo, (2) remoción de arilo por lixiviación, (3) inmersión en ácido giberélico con 100 mg L-1, (4) inmersión en ácido giberélico con 200 mg L-1, (5) inmersión en ácido giberélico con 300 mg L-1 y (6) control. Se utilizaron 100 semillas por tratamiento con cinco repeticiones cada una. El tratamiento que presentó el mayor porcentaje de germinación para las cuatro localidades fue la remoción manual del arilo con 52 %, mientras que el tratamiento con menor germinación fue el control con 3.5 %. Por localidad, San Lorenzo obtuvo el mayorporcentaje de germinación con 16.5% en promedio, mientras que San Nicolás registró el menor con 5.33 %. Los tratamientos germinativos no lograron romper la latencia (porcentajes de germinación < 70 %). Las semillas de las cuatro localidades se embebieron de agua por lo que no se presenta la latencia física. Las semillas con mayor biomasa absorbieron menos agua.""Magnolia pugana is an endemic and endangered species located in western Mexico, which is distributed in gallery forests in the canyons of southern Zacatecas and north-central Jalisco. The aim of this study was to determine the seed viability and to identify the treatments to break seed dormancy in M. pugana. Seeds were collected from four locations, three of them located in Jalisco (Centro Universitario de Ciencias Biológicas y Agropecuarias, San Lorenzo and San Nicolas) and one in Zacatecas (Palo Verde). The following germinative treatments were applied: (1) manual aril removal, (2) aril removal by leaching, soaking in gibberellic acid at different concentrations: (3) 100 mg L-1, (4) 200 mg L-1, (5) 300 mg L-1, and (6) control. One hundred seeds were used per treatment with five replicates each. The treatment with the highest germination percentage for all four sites was the manual aril removal with 52 %, whereas the treatment with the lowest germination was the control with 3.5 %. By location San Lorenzo had the highest germination percentage at 16.5 %, while San Nicolas had the lowest percentage (5.33 %). Germination treatments failed to break dormancy (germination rates < 70 %). The seeds of the four locations imbibed water so no physical seed dormancy occurs. State and actuator fault estimation observer design integrated in a riderless bicycle stabilization system
"This paper deals with an observer design for Linear Parameter Varying (LPV) systems with high-order time-varying parameter dependency. The proposed design, considered as the main contribution of this paper, corresponds to an observer for the estimation of the actuator fault and the system state, considering measurement noise at the system outputs. The observer gains are computed by considering the extension of linear systems theory to polynomial LPV systems, in such a way that the observer reaches the characteristics of LPV systems. As a result, the actuator fault estimation is ready to be used in a Fault Tolerant Control scheme, where the estimated state with reduced noise should be used to generate the control law. The effectiveness of the proposed methodology has been tested using a riderless bicycle model with dependency on the translational velocity v, where the control objective corresponds to the system stabilization towards the upright position despite the variation of v along the closed-loop system trajectories.
Estudios genéticos de las mutantes de los genes BATs de Lachancea kluyveri
Tesis (Doctorado en Ciencias en Biología Molecular)"Las transaminasas de aminoácidos de cadena ramificada (BCAAT), de L-valina, Lisoleucina y L-leucina (VIL) participan en la biosíntesis y degradación de VIL. Los genes y mecanismos del metabolismo de VIL en la levadura Lachancea kluyveri no se han identificado funcionalmente. Para elucidar los elementos involucrados en ese metabolismo en L. kluyveri analizamos su genoma e identificamos el gen (LkBAT1) ortólogo al gen ScBAT1 de Saccharomyces cerevisiae. Adicionalmente, encontramos otro gen (LkBAT1bis) que es muy similar en secuencia a LkBAT1 y ScBAT1, y si codificara una proteína, ésta estaría trunca en el carboxilo teminal. Para caracterizar funcionalmente estos genes, se generaron las mutantes sencillas y la doble mutante en los genes LkBAT1 y LkBAT1bis, y se hicieron ensayos para probar el papel funcional de los genes en el metabolismo de VIL. Encontramos que el gen LkBAT1 es necesario para el metabolismo de VIL, y actividad de BCAAT en la cepa que expresa LkBAT1 pero no en la cepa mutante Lkbat1. La complementación heteróloga de genes entre S. cerevisiae y L. kluyveri confirmó que LkBAT1 y ScBAT1 son ortólogos funcionales. Se determinó que LkBat1 se localiza principalmente en mitocondria, y la regulación transcripcional de LkBAT1 no depende de la calidad de la fuente de nitrógeno. LkBAT1bis no parece participar en el metabolismo de VIL, pero ambos genes (LkBAT1 y LkBAT1bis) son necesarios para asimilar etanol, una fuente de carbono no fermentable. Estos resultados sugieren que el gen LkBAT1 codifica para una transaminasa de aminoácidos de cadena ramificada y LkBAT1 y LkBAT1bis participan en el metabolismo respiratorio.""Branched chain aminoacid transaminases (BCAAT) have a functional role on the biosynthesis and catabolism of L-valine, L-isoleucine and L-leucine (VIL). In Lachancea kluyveri, the genes and mechanisms for VIL metabolism have not been identified. To elucidate the elements involved in the biosynthesis and catabolism of VIL in L. kluyveri, we survey the genome of this yeast and we found the orthologous gene (LkBAT1) to the S. cerevisiae ScBAT1 gene. In addition, we found another ORF (LkBAT1bis) which is highly similar to LkBAT1 and this gene encodes for a hypothetical protein which is truncated at its C-terminus. To assess the function of these genes, we constructed the single mutants and the double mutant in the Lkbat1 and Lkbat1bis genes. We found that LkBAT1 is needed in the VIL metabolism and that Lkbat1 null mutant did not show transaminase activity. Heterologous complementation assays between S. cerevisiae and L. kluyveri confirmed that LkBAT1 and ScBAT1 are functional orthologous genes. Furthermore, LkBat1 is localized in the mitochondria, and the transcriptional regulation of LkBAT1 does not depend on the nitrogen source quality. LkBAT1bis has no role in VIL metabolism, however, both genes (LkBAT1 and LkBAT1bis) are needed for the assimilation of ethanol, a non-fermentable carbon source. These results suggest that LkBAT1 encodes for a BCAAT and that both genes (LkBAT1 and LkBAT1bis) have a role in the respiratory metabolism.
Instrumentation of a bicycle without driver for experimental testing of automatic control
"In this paper, the instrumentation of a riderless bicycle prototype for control purposes is addressed. The proposed instrumentation is based on the mathematical model of a bicycle, in which the dynamical variables correspond to the translational velocity, the angular position of the handlebar and the velocity of the roll angle of its general framework. Throughout the paper, a convenient and simple way to measure these variables is detailed explained, in order to provide an instrumented prototype ready to be used for testing automatic control laws. This corresponds to the main contribution of the present paper. The dynamical model from which the proposed instrumentation has been designed, corresponds to a Linear Parameter Varying system, considering the translational velocity as time-varying parameter. The results of the instrumentation and its effectiveness are shown from experimental tests.
Flower, fruit phenology and flower traits in Cordia boissieri (Boraginaceae) from northeastern Mexico
"We characterized variations in Cordia boissieri flowers and established if these variations occur between plants or between flowering events. Flowering and fruiting was measured for 256 plants. A GLM test was used to determine the relationship between flowering and fruit set processes and rainfall. We performed measurements of floral traits to detect variations within the population and between flowering events. The position of the anthers with respect to the ovary was determined in 1,500 flowers. Three out of four flowering events of > 80% C. boissieri plants occurred after rainfall events. Only one flowering event occurred in a drought. Most plants flowered at least twice a year. The overlapping of flowering and fruiting only occurred after rainfall. Anthesis lasted three-to-five days, and there were two flower morphs. Half of the plants had longistylus and half had brevistylus flowers. Anacahuita flower in our study had 1-4 styles; 2-9 stamens; 6.5-41.5 mm long corolla; sepals from 4.5-29.5 mm in length; a total length from 15.5-59 mm; a corolla diameter from 10.5-77 mm. The nectar guide had a diameter from 5-30.5 mm; 4-9 lobes; and 5 distinguishable nectar guide colors. The highest variation of phenotypic expression was observed between plants.
McCabe-Thiele reactivo: Método corto incluyendo eficiencia vapor líquido reactivo
"A graphic McCabe-Thiele method is developed for application to fully reactive column configuration. The extended method takes in to account several aspects as classic McCabe-Thiele method under transformed reaction-invariant composition variables as: minimum number of reactive theoretical stages, number of reactive theoretical stages, minimum reflux ratio, feed tray location, to and / or bottom flow. Reaction equilibrium is assumed on each stage, vapor-liquid equilibrium is assumed on all stages and is also considered a reactive Murphree vapor tray efficiency. The methodology is illustrated in two ideal reactive systems. The results of the reactive McCabe-Thiele method agree with those obtained using a rigorous method in commercial simulator AspenONE Aspen Plus (Radfrac module).""Un método gráfico basado en el método McCabe-Thiele es desarrollado para la aplicación del diseño de columnas reactivas. El método toma en cuenta varios aspectos del método clásico McCabe-Thiele bajo variables reactivas transformadas: número mínimo de etapas reactivas teóricas, número de etapas reactivas, reflujo mínimo, ubicación del plato de alimentación flujos de destilado y fondos. Se asume: equilibrio reactivo para cada etapa, equilibrio vapor líquido y es considerado una eficiencia reactiva Murphee para la fase de vapor. La metodología es ilustrada en dos sistemas reativos ideales. Los resultados del método McCabe-Thiele reactivo concuerdan con los obtenidos usando un método riguroso en un simulador comercial AspenONE Aspen Plus (Radfrac module).
Efecto de la composición de mezclas de metabolitos procedentes de la fermentación oscura para su uso en la producción de biohidrógeno en celdas de electrólisis microbiana
Tesis (Maestría en Ciencias Ambientales)"La celda de electrólisis microbiana (CEM) es un dispositivo que surge como una alternativa a los sistemas actuales de producción de biohidrógeno (Bio-H2). A la fecha diversos estudios han evaluado algunos parámetros de este sistema (voltaje aplicado, inóculo, configuración de la celda, etc.) con el fin de optimizarlos. El sustrato por excelencia para las CEMs suele ser acetato, sin embargo, hay estudios que reportan el empleo de mezclas de ácidos grasos volátiles (AGVs), lo cual da indicios de que los efluentes de fermentación oscura, ricos en AGVs, pueden emplearse como sustrato en CEMs. No obstante, el efecto de otros componentes de tales efluentes, tal como el etanol y el lactato, no han sido explorados. El objetivo general de este trabajo fue evaluar el efecto de la variación de la fracción de etanol y lactato en una mezcla definida de acetato, propionato y butirato, para la producción de Bio-H2 en CEMs. Para cumplir el objetivo se recurrió a celdas de tipo H, separadas por una membrana de intercambio catiónico, se empleó como inóculo un efluente de un fermentador productor de Bio-H2 y lixiviados de composta, como voltaje se probaron 0.6 y 1 V. Se evaluó el efecto en el sistema al variar la mezcla de sustratos, que estuvo conformada por acetato, propionato y butirato, los cuales se mantuvieron en una proporción fija, mientras que las proporciones de lactato y etanol variaron. Al evaluar el inóculo procedente de la fermentación oscura aplicando 0.6 V y diferentes concentraciones de AGVs fue evidente que la cantidad de especies electroactivas presentes en el inóculo no fue la suficiente, puesto que la generación de corriente fue muy inferior a lo reportado en la literatura (~38 mA/m2), aunado a esto no se observó formación de Bio-H2. Por lo tanto se decidió utilizar lixiviados de composta de los cuales se sabe tienen una comunidad electroactiva importante, para este caso se aplicaron los mismos voltajes: 0.6 y 1.0 V, siendo con éste último el ensayo con el cual se obtuvo una mejor respuesta en la producción de hidrógeno (0.0325 m3 H2/m3 anolito-d). Una vez definido el inóculo, voltaje y concentración de la mezcla base (lixiviados de composta, 1.0 V y 2.5 g DQO/L) se evaluaron cuatro mezclas de lactato y etanol, en las cuales la DQO se mantuvo fija (1 g/L) pero las proporciones de éstos sustratos variaron de manera tal que de las mezclas uno a la cuatro la concentración de lactato disminuyó, mientras que la de etanol aumentó.""Microbial electrolysis cell (MEC) is a device that is an alternative to current biohydrogen production systems (Bio-H2). To date few studies have evaluated system parameters (applied voltage, inoculum, cell configuration, etc.) in order to optimize them. The substrate of choice for the MECs is usually acetate, however, some studies have reported the use of mixtures of volatile fatty acids (VFA), which are produced during dark fermentation. Nonetheless the effects of other components present in the mixture, such as ethanol and lactate, have not yet been explored. The overall objective of this study was to evaluate the effect of varying the fraction of ethanol and lactate in a defined mixture of acetate, propionate and butyrate, for the production of Bio-H2 in MECs. To achieve the objective cells type H were used, separated by a cation exchange membrane, it were used as inoculum both effluent from a fermenter producing Bio-H2 and leachate compost. As voltage were tested 0.6 and 1 V, and it was assessed their effect on the system by varying the mixture of substrates, which consisted of acetate, propionate and butyrate, which remained in a fixed ratio while the proportions of lactate and ethanol varied. Assessing the inoculum from dark fermentation using 0.6 V and under different concentrations of VFA it was evident its low electroactive capacity since the current generation was much lower than that reported in the literature (~ 38 mA/m2), additionally Bio-H2 was not observed. Therefore leachate compost was used as inoculum, which is known to have a significant electroactive community; in this case two voltages were applied: 0.6 and 1.0 V. From this trial 1.0 V was used because it was the potential to which it was obtained a better response in the hydrogen production (0.0325 m3/m3 anolyte-d). Having defined the inoculum, voltage and concentration of the base mixture (compost leachate, 1.0 V and 2.5 g COD/L), four assays of different lactate and ethanol ratios were evaluated, in which COD (1 g/L) remained fixed but the proportions of these substrates varied so that lactate concentration decreased while that of ethanol increased in the mixtures.
Chaotic attractors based on unstable dissipative systems via third-order differential equation
"In this paper, we present an approach how to yield 1D, 2D and 3D-grid multi-scroll chaotic systems in R3 based on unstable dissipative systems via third-order differential equation. This class of systems is constructed by a switching control law(SCL) changing the equilibrium point of an unstable dissipative system. The switching control law that governs the position of the equilibrium point varies according to the number of scrolls displayed in the attractor.
Caracterización de una cepa de preadipocitos humanos como modelo experimental para estudios de adipogénesis in vitro
Tesis (Maestría en Ciencias en Biología Molecular)"El estudio del tejido adiposo aprovecha modelos experimentales in vitro constituidos por cultivos primarios de preadipocitos o líneas celulares adiposas. Los primeros representan con alta fidelidad el metabolismo del linaje adiposo in vivo, pero tienen un tiempo de vida limitado y una decreciente capacidad adipogénica al subcultivarse. Las segundas muestran capacidad proliferativa ilimitada pero no necesariamente poseen todos los atributos funcionales del linaje adiposo. Las líneas celulares adipogénicas más empleadas son de origen múrido pero tienen diferencias genéticas y metabólicas con las células humanas. Aunque se han descrito líneas celulares adipogénicas humanas, hasta ahora su uso es limitado y no hay un modelo in vitro de adipogénesis humana de uso generalizado. Aquí caracterizamos una cepa de preadipocitos subcutáneos humanos de naturaleza clonal, PASH-4, para explorar su utilidad como modelo in vitro de la adipogénesis humana. Las células PASH-4 proliferaron durante 30 subcultivos consecutivos mostrando tiempos de duplicación de 47 h y 95 h en los subcultivos 17 y 26, y capacidad clonogénica del 7% y 2% en los subcultivos 15 y 25, respectivamente. Las células PASH-4 mantuvieron su capacidad adipogénica aun en el subcultivo 26, expresando los marcadores adipogénicos tempranos C/EBPβ y C/EBPδ aun en preadipocitos. Las células PASH-4 fueron diploides (46XY) al menos hasta el subcultivo 20. Estas características hacen de la cepa PASH-4 un modelo útil para estudios de adipogénesis humana in vitro y una población de preadipocitos humanos normales adecuada para generar una línea establecida de preadipocitos humanos.""The study of adipose tissue takes advantage of in vitro experimental models constituted by cultures of primary preadipocytes or adipose cell lines. The first ones represent with high fidelity the metabolism of the cell adipose lineage but have a limited life span and decreasing capacity when subcultivated. The second ones show an ilimited proliferative capacity but they do not necessarily have all the functional attributes of the adipose cell lineage. The most commonly used adipogenic cell lines have a murine origin and present genetic and metabolic differences withrespect to the human cells. Although human adipogenic cell lines have been described, its use is limited, and currently there is not an vitro model system widely used for the study of human adipogenesis. Here we characterize a clonal cell strain of human subcutaneous preadipocytes, PASH-4, to explore its utility as an in vitro model system for human adipogenesis. The PASH-4 cells proliferated for 30 serial subcultures, showing population doubling times of 47 h and 95 h in the 17 and 26 subculture, and a clonogenic capacity of 7% and 2% in the 15 and 25 subculture, respectively. The PASH-4 cells maintained their adipogenic capacity until the 26 subculture, expressing the early adipose molecular markers C / EBPβ and C/EBPδ even as preadipocytes. The PASH-4 cells were diploid (46XY) at least until 20 subculture. All these characteristics make the PASH-4 strain a useful model for in vitro human adipogenesis studies and a population suitable of normal human preadipocytes to generate an established cell line of human preadipocytes
