Das Kälteschockprotein YB-1 vermittelt pro- und anti-inflammatorische Effekte in einem Lipopolysaccharid(LPS)-induzierten Entzündungsmodell der Maus
Das Kälteschockprotein Y-box binding protein-1 (YB-1) ist ein zentraler Mediator in der
Regulation von Entzündungsprozessen. Diese Arbeit untersucht die Rolle von YB-1 im
systemischen Modell einer Lipopolysaccharid(LPS)-vermittelten Entzündung.
Mäuse mit einem Knock-out des Ybx1-Gens weisen nach Verabreichung von LPS eine höhere
Überlebensrate auf. Die Abwesenheit von YB-1 führt zu einer geringeren Zytokinfreisetzung.
Zudem kann bei Ybx1-Knock-out-Tieren eine verringerte Gerinnungsaktivierung und eine
verminderte Organschädigung nachgewiesen werden. Erstmalig wurde in dieser Arbeit ein
Mausstamm mit einer Sekretionsdefizienz für YB-1 generiert. Sekretionsdefiziente Tiere
zeigen nach LPS-Gabe eine verstärkte inflammatorische Reaktion, die mit einer erhöhten
Letalitätsrate einhergeht.
Diese Arbeit liefert neue Erkenntnisse über die duale Rolle von YB-1 im systemischen
Entzündungsgeschehen. Es konnte gezeigt werden, dass
intrazelluläres YB-1 pro-
inflammatorische Prozesse
fördert, während extrazelluläres YB-1 als zentraler anti-
inflammatorischer Regulator agiert

Widespread decline of Ground Beetles in Germany
Aim
Many insect species are facing existential crises, primarily due to diverse human activities. Most insect assessments, however, are based on relatively short time series or some iconic species. Here, we assess how the occupancy of ground beetles has changed in Germany over the last 36 years.
Location
Germany.
Methods
In close collaboration with taxonomic experts from natural history societies, we compiled the best available occurrence data for ground beetles in Germany, estimated the changes in species occupancy over time, and related these changes to species traits and characteristics.
Results
We obtained trends for 383 species and found that 52% of species significantly declined, and 22% significantly increased in site occupancy over the last 36 years. The remainder of the species (26%) all showed a mean negative trend, albeit nonsignificant. Species classified as non-threatened in the German red list declined at a similar rate as threatened species, with 64% of the Near Threatened species experiencing significant declines (highest among all red list categories). Across all traits, we found that large (compared to medium-sized) and omnivore (compared to predator) species declined less.
Conclusions
Since ground beetles are key predators in many natural and agricultural ecosystems that play an important role in pest control and the food chain, their decline should raise concerns. Thus, we urgently plead for more harmonised and systematic monitoring of this insect group

Perceived biodiversity : is what we measure also what we see and hear?
Biodiversity is crucial for human health and well-being. Perceived biodiversity—people's subjective experience of biodiversity—seems to be particularly relevant for mental well-being.
Using photographs and audio recordings of forests that varied in levels of biodiversity, we conducted two sorting studies to assess how people perceive visual and acoustic diversity and whether their perceptions align with species richness and proxies for forest structural diversity (‘actual diversity’). Per study, 48 participants were asked to sort the stimuli according to any similarity-based sorting criteria they liked (‘open sorts’) and perceived diversity (‘closed sorts’).
The main perceived visual forest characteristics identified by participants in the open visual sorts were vegetation density, light conditions, forest structural attributes and colours. The main perceived acoustic forest characteristics identified in the open acoustic sorts comprised bird song characteristics, physical properties such as volume, references to the time of day or seasonality and evoked emotions.
Perceived visual and acoustic diversity were significantly correlated with actual visual and acoustic diversity, respectively.
We further computed several objective visual and acoustic diversity indices from the photos and audio recordings, for example, a Greenness Index or the Acoustic Complexity Index, and assessed their relevance for perceived and actual diversity. While all acoustic diversity indices were significantly associated with perceived and actual acoustic diversity, for the visual sense, the Greenness Index successfully captured both perceived and actual visual diversity.
Our results suggest that people can perceive variations in biodiversity levels. Our identified visual and acoustic forest characteristics may help to better understand perceived diversity and how it differs from how diversity is measured in biological studies. We present one visual and several acoustic diversity indices that quantify aspects of perceived and actual diversity. These indices may serve as cost-efficient tools to manage and plan greenspaces to promote biodiversity and mental well-being
Simulative Optimierungsmethodik eines Erdgasmotors mittels einer aktiv gespülten Vorkammerzündkerze
Vor dem Hintergrund der Erreichung der Klimaziele und der dringenden Notwendigkeit,
Treibhausgasemissionen zu reduzieren, werden im Großmotorenbereich zunehmend
kohlenstoffarme bzw. kohlenstofffreie Kraftstoffe eingesetzt. Dazu gehören Erdgas und
Ammoniak, die eine deutlich geringere Zündwilligkeit als konventionelle Marinekraftstoffe
wie Diesel oder Schweröl aufweisen. Zur Erzielung der besseren Ergebnisse hinsichtlich eines hohen Wirkungsgrades und niedriger NOx-Rohemissionen ist ein Magerbrennverfahren von Vorteil. Dies hat zur Folge, dass die Zündwilligkeit der Kraftstoff-Luft-Gemische bei den ohnehin zündunwilligen Kraftstoffen weiter abnimmt und eine konventionelle Hakenzündkerze nicht in der Lage ist, eine effiziente und vollständige Verbrennung solcher Gemische sicherzustellen. Zudem steigt das Risiko einer unvollständigen Verbrennung des Kraftstoffs, der im Falle von Erdgas je nach Betrachtungszeitraum ein 30- bis 87-fach höheres Treibhausgaspotenzial als Kohlendioxid aufweist. Ein zuverlässiges und leistungsfähiges Zündverfahren gewinnt daher zunehmend an Bedeutung. Eine aktiv gespülte Vorkammerzündkerze ist eines der leistungsfähigsten Zündsysteme, bei dem die Gemischanreicherung und Zündung in einer Vorkammer erfolgen und anschließend turbulente Fackelstrahlen schnell in den Hauptbrennraum eindringen und dort eine beschleunigte Verbrennung auslösen.
Ziel dieser Dissertation ist die Entwicklung einer zeit- und ressourceneffizienten Methodik für die Auslegung und Optimierung einer aktiv gespülten Vorkammerzündkerze unter Nutzung der Synergie von numerischen und empirischen Methoden. Diese Optimierungsmethodik besteht aus aufeinanderfolgenden Auslegungs- und Validierungsschritten, wobei einerseits die Simulationskomplexität schrittweise erhöht wird und andererseits geeignete zeit- und ressourceneffiziente Validierungsmethoden eingesetzt werden. Zunächst wird eine eindimensionale Auslegungsmethode zur Geometrieoptimierung und Randbedingungsermittlung entwickelt. Darauf aufbauend erfolgt eine industrietaugliche Reynolds-averaged Navier–Stokes (RANS) Modellierung der Einzelprozesse wie Spülung und Gemischbildung in der Vorkammer mit anschließender Validierung und Optimierung mittels hochgenauer direkter numerischer Simulation. Dabei werden die Genauigkeit und der Rechenaufwand des entwickelten Modells optimiert. Abschließend wird auf Basis der gewonnenen Erkenntnisse ein RANS-Einzylindermodell mit aktiv gespülter Vorkammerzündkerze aufgebaut und anhand von Motorversuchen validiert. Ein zentrales Ziel dieser Dissertation ist es, zeiteffiziente, genaue und stabile numerische Methodik für Optimierungsaufgaben der Vorkammerzündkerze zu finden und zu untersuchen. Dabei wird untersucht, wie sich die Simulationsgenauigkeit bei Variation der Motorbetriebsparameter verändert und wo noch Verbesserungspotential besteht.In response to climate change and the urgent need to reduce greenhouse gas emissions, the use of low- and zero-carbon fuels is increasing in the large engine sector. These include natural gas and ammonia, which are significantly less flammable than conventional marine fuels such as diesel or heavy fuel oil. In order to achieve better results in terms of high efficiency and low raw NOx raw emissions, a lean combustion process is advantageous. As a result, the flammability of the fuel-air mixtures is further reduced for fuels that are already difficult to ignite, and a conventional spark plug cannot ensure efficient and complete combustion of such mixtures. There is also an increased risk of incomplete fuel combustion, which in the case of natural gas has a greenhouse gas potential 30 to 87 times higher than carbon dioxide, depending on the period under consideration. Reliable and effective ignition, therefore, becomes increasingly important. An active scavenged pre-chamber spark plug is one of the most powerful ignition systems, where mixture enrichment and ignition take place in a pre-chamber and turbulent flame jets then rapidly penetrate into the main combustion chamber, triggering accelerated combustion. This thesis aims to develop a time and resource-efficient optimization methodology for the development of actively scavenged pre-chamber spark plugs using the synergy of numerical and empirical methods. This optimization methodology consists of successive modeling and validation steps, where, on the one hand, the simulation complexity is progressively increased. On the other hand, appropriate time and resource-efficient validation methods are used. The first step is to develop a one-dimensional design method for geometry optimization and boundary condition determination. Based on this, an industrially suitable Reynolds-averaged
Navier–Stokes (RANS) modeling of the individual processes such as scavenging and mixture formation in the pre-chamber is carried out with subsequent validation and optimization utilizing high-accuracy direct numerical simulation. The accuracy and computational complexity of the developed model are then optimized. Finally, a single-cylinder RANS model with an active scavenged pre-chamber spark plug is developed and validated by engine testing. A central objective of this thesis is to find and investigate time-efficient, accurate, and stable numerical methods for optimization of a pre-chamber spark plug. Auswirkungen eines Ausdauertrainings auf die Tortuositas der Arteria cerebri posterior bei gesunden jungen Erwachsenen mit geringer Alltagsaktivität
In dieser Studie wurde bei 40 gesunden jungen Probanden und Probandinnen untersucht,
welchen Einfluss Ausdauertraining auf die Gefäßtortuositas der Arteria cerebri posterior (PCA)
und damit die vaskulären, zerebralen Strukturen nimmt. Dabei wurden die Probanden und
Probandinnen
in eine Kontroll- und eine Trainingsgruppe, welche ein 4-monatiges
Laufbandtraining absolvierte, randomisiert. Vor Beginn, nach 4 Wochen und nach Abschluss
der Studie wurden Belastungstests zur Erhebung des Fitnessstandes und angiographische 7
Tesla MRT-Aufnahmen durchgeführt. Diese wurden zur Auswertung der Gefäßtortuositas
genutzt, indem an verschiedenen Segmenten der PCA die euklidische, d.h. die lineare Distanz
zwischen Anfangs- und Endpunkt, sowie die geodätische Länge, d.h. die tatsächliche
Gefäßlänge, vermessen wurden. Nach 4 Monaten zeigte sich bei Erhöhung der Fitness eine
signifikante Abnahme der Gefäßtortuositas bei dem Segment P3 der PCA und der Arteria
temporalis posterior (PT), während sich in den weiteren untersuchten Gefäßabschnitten keine
signifikanten Änderungen zeigten (Arteria calcarina (CA) und Arteria parietooccipitalis (POA)).
Insgesamt zeigt diese Studie, dass kardiovaskuläres Training die Tortuositas des zerebralen
Gefäßsystems bei jungen Erwachsenen moduliert. Die Ergebnisse dieser Studie geben
Anlass, aeroben Ausdauersport als Intervention weiter zu untersuchen, da dieser als
Therapiemöglichkeit bei verschiedenen Erkrankungen, wie zum Beispiel der Demenz bei
Alzheimer-Krankheit, erfolgsversprechend sein kann

Pathologist-like explainable AI for interpretable Gleason grading in prostate cancer
The aggressiveness of prostate cancer is primarily assessed from histopathological
data using the Gleason scoring system. Conventional artificial intelligence
(AI) approaches can predict Gleason scores, but often lack
explainability, which may limit clinical acceptance. Here, we present an alternative,
inherently explainable AI that circumvents the need for post-hoc
explainability methods. Themodel was trained on 1,015 tissuemicroarray core
images, annotated with detailed pattern descriptions by 54 international
pathologists following standardized guidelines. It uses pathologist-defined
terminology and was trained using soft labels to capture data uncertainty. This
approach enables robust Gleason pattern segmentation despite high interobserver
variability. Themodel achieved comparable or superior performance
to direct Gleason pattern segmentation (Dice score: 0:713±0:003 vs.
0:691±0:010) while providing interpretable outputs. We release this dataset to
encourage further research on segmentation in medical tasks with high subjectivity
and to deepen insights into pathologists’ reasoning
