    Towards understanding the long-term fate of biochar in Terra Preta

    Terra Preta soils, or anthropogenic dark earths (ADE), were formed beneath pre-Columbian settlements in Central Amazonia. Unlike surrounding weathered soils poor in soil organic matter, ADE soils are rich in nutrients and C, largely because of the biochar that has remained in the soil for over 2000 years. The reasons underlying this stability of particulate biochar remain unclear. This study aimed to elucidate stabilization processes by (i) characterizing the chemistry of biochar particles in ADE and (ii) comparing surface and interior chemical composition. Analyses included bulk chemical composition, benzene polycarboxylic acids as biochar markers, microstructure and surface elemental composition, functional groups, and the distribution of organic and inorganic compounds and their interactions within the particles. Bulk chemical analysis revealed relatively low total organic carbon and black carbon contents, suggesting mineral-organic interactions. Surfaces showed high oxygen content, indicating oxidation, and microscopy revealed mineral-organic coatings attached to particle surfaces. These coatings likely protect the aromatic biochar core from further degradation. Elemental depth profiles further indicated increasing C, K, and Ca contents with particle depth, suggesting mineral–biochar interactions extend into the interior. Thus, both surface coatings and internal interactions contribute to the long-term stability of ADE biochar

    Loss of Schwann cell's normal rhythmic core clock gene expression and gain of rhythmic expression of oncogenic driver genes in malignant NF1-associated peripheral nerve sheath tumor

    Neurofibromatosis type 1 (NF1) is an autosomal dominant tumor syndrome caused by pathogenic variants in the NF1 gene. Beside tumor formation, patients often have sleep disturbances, suggesting circadian involvement. Previous NF1 studies have implicated MAPK pathway, cAMP-PKA, calcium signaling, and ALK in circadian regulation, and shown disrupted rhythms in murine astrocytes lacking NF1. However, whether human Schwann cells show rhythmic gene expression remains unknown, although impaired rhythms may contribute to tumorigenesis. In this study, we analyzed normal human Schwann cells and NF1-derived malignant peripheral nerve sheath tumors (MPNST) for rhythmic gene expression. Cultured cells were synchronized via serum shock, and mRNA levels of core clock and associated genes (e.g. ARNTL, JUN, TGFA, CLOCK, VEGFA, MYC) were quantified at defined intervals. We observed rhythmic core clock gene expression in normal Schwann cells, demonstrating intrinsic circadian oscillations in peripheral glia. In contrast, MPNST lacked rhythmicity in core clock genes, instead showing de novo rhythmic expression of oncogenes and growth factors like MYC and VEGFA. Thus, loss of clock gene rhythmicity (desynchronization) and emergence of rhythmic oncogene expression (synchronization) in NF1-associated MPNST further our understanding of peripheral glial physiology and tumorigenesis. These insights suggest that chronotherapeutic strategies may be beneficial for NF1-associated MPNST

    Selective oral decontamination of the esophagus to reduce microbial burden in patients undergoing esophagectomy for esophageal cancer (SODA) : first results from a proof-of-principle study

    Background/Objectives: Postoperative pneumonia and other infectious complications after robotic-assisted minimally invasive esophagectomy still contribute to morbidity and mortality. Selective oral decontamination of the esophagus prior to surgery might reduce the rate of infectious complications. However, its impact on the esophageal microbiota is unknown. Therefore, this study aimed to analyze whether selective oral decontamination of the esophagus prior to surgery reduces postoperative pneumonia rates and alters the esophageal microbiome. Methods: We conducted a proof-of-principle study including 22 patients who underwent robotic-assisted minimally invasive esophagectomy. Thirteen patients were treated with 50 mg amphotericin B, 8 mg tobramycin, and 10 mg colistin orally 7 days prior to surgery, intraoperatively, and 5 days postoperatively. The remaining nine patients received standard-of-care treatment (no oral decontamination). The esophageal microbiome was assessed using 16S rRNA gene amplicon libraries which were annotated using the Ribosomal Data Project. The incidence of postoperative (at discharge from hospital or 30 days, whichever was later) infectious complications was assessed. Results: Selective oral decontamination was associated with reduced overall rates of infectious complications (7.7% vs. 55.5%, p = 0.008) and postoperative pneumonia (0% vs. 33.3%, p = 0.007). Alterations in the esophageal microbiome depending on selective oral decontamination were detectable. The microbiomes of patients with infectious complications showed higher abundances of Neisseria and lower abundances of Streptococcus than samples without infectious complications. Conclusions: Selective oral decontamination reduced the rate of postoperative complications, postoperative pneumonia in particular, after robot-assisted esophagectomy. Alterations in the microbiome were also evident following decontamination. Further studies with larger sample sizes are necessary to confirm these data

    Einfluss des Massepfades auf vestibulär evozierte myogene Potentiale bei elektrischer Stimulation über ein Cochlea-Implantat

    Die Studie untersuchte den Einfluss des Massepfades auf elektrische zervikale und okuläre vestibulär evozierte myogene Potenziale (e-VEMP) bei Cochlea-Implantat-Trägern. E-VEMP wurden mittels unterschiedlicher Stimulationsmodi (monopolar basal, monopolar apikal, bipolar transmodiolar) ausgelöst. Bei 43% der untersuchten Probandinnen und Probanden konnten e-VEMP nachgewiesen werden, teils ohne Antwort bei vorheriger Knochenleitungsstimulation. Die höchste Antwortrate (40%) zeigte sich bei monopolarer Stimulation über die basale Elektrode E3. Die Amplituden der e-VEMP waren vergleichbar mit denen bei Knochenleitungsstimulation, jedoch mit kürzeren Latenzen. Teilweise lag die e-VEMP-Schwelle unterhalb der elektrischen Hörschwelle. Fazit: Die Auslösung von e-VEMP hängt vom Massepfad und Stimulationsniveau ab. Eine vestibuläre Kostimulation durch das CI ist insbesondere bei hoher Stimulationsintensität und basaler monopolarer Stimulation wahrscheinlich

    Successes and limitations of pretrained YOLO detectors applied to unseen time-lapse images for automated pollinator monitoring

    Pollinating insects provide essential ecosystem services, and using time-lapse photography to automate their observation could improve monitoring efficiency. Computer vision models, trained on clear citizen science photos, can detect insects in similar images with high accuracy, but their performance in images taken using time-lapse photography is unknown. We evaluated the generalisation of three lightweight YOLO detectors (YOLOv5-nano, YOLOv5-small, YOLOv7-tiny), previously trained on citizen science images, for detecting ~ 1,300 flower-visiting arthropod individuals in nearly 24,000 time-lapse images captured with a fixed smartphone setup. These field images featured unseen backgrounds and smaller arthropods than the training data. YOLOv5-small, the model with the highest number of trainable parameters, performed best, localising 91.21% of Hymenoptera and 80.69% of Diptera individuals. However, classification recall was lower (80.45% and 66.90%, respectively), partly due to Syrphidae mimicking Hymenoptera and the challenge of detecting smaller, blurrier flower visitors. This study reveals both the potential and limitations of such models for real-world automated monitoring, suggesting they work well for larger and sharply visible pollinators but need improvement for smaller, less sharp cases

    Drawing on previously unknown sources, this article documents the physicist Johannes Stark's unsuccessful attempt to publish a declaration by German Nobel laureates in support of the August 1934 referendum. Following the death of Reich President Paul von Hindenburg, this referendum aimed to legitimize the transfer of the two highest state offices—Reich President and Reich Chancellor—to one single person, thereby making Adolf Hitler the head of state and Führer of the German people. At the suggestion of the Propaganda Ministry, Stark sent a telegram to eleven “Aryan German Nobel laureates” in chemistry and physics, and urged them to sign a public appeal to participate in the referendum and support “the great national commitment of the entire German people” to Adolf Hitler as “Führer of the German people.” Responses to Stark's telegram ranged from wholehearted support to rejection. The majority of the laureates declined to lend their signature, and the initiative ultimately failed. The episode reveals that Stark—although at the height of his influence as president of the Physikalisch-Technische Reichsanstalt (Imperial Institute of Physics and Technology) and the Deutsche Forschungsgemeinschaft (German Research Foundation)—was largely isolated within the scientific community of the so-called Third Reich

    Für die ökologische Wende der gegenwärtigen Gesellschaft bildet die Transformation zu einer nachhaltigen chemischen Industrie eine der zentralen Voraussetzungen. Denn die materiellen Grundlagen dieser Gesellschaft sind zum größten Teil von der chemischen Industrie hervorgebracht. Diese Situation stellt eine große Herausforderung dar, könnte aber auch eine Chance bieten, die gesellschaftliche Stellung der Chemie neu zu definieren, indem sie sich glaubwürdig mit den gesellschaftlichen Veränderungsprozessen verknüpft, die unter der Überschrift Just transition zusammengefasst werden. Wertewandel in Industrie und Gesellschaft scheinen notwendig zu sein, um mit gängigen Rationalitäten, Ansprüchen und Verhaltensmustern in der materiellen Kultur der Gegenwart zu brechen und neue dauerhaft zu begründen. Eine konstruktiv kritische sozial- und kulturwissenschaftliche Begleitung kann dazu beitragen, diese „weichen“ Aspekte der Transformation zu erfassen und zu adressieren. Die zu begleitenden Transformationsprozesse umfassen dabei sowohl die in den Produktionsnetzwerken der chemischen Industrie vorherrschenden Handlungsmuster, Organisationsformen und Zielvorgaben als auch die gesellschaftlichen Umgänge mit „der Chemie“ und ihren Produkten.Das Just Transition Center ist ein Projekt der Martin-Luther-Universität Halle-Wittenberg und beschäftigt sich mit Übermorgen-Fragen für einen gerechten, nachhaltigen und klimaneutralen Übergang in der Strukturwandelregion Mitteldeutsches Revier. Dafür arbeiten 17 interdisziplinäre Forschungsteams und vier Revierscouts eng mit den Akteuren in der Fokusregion südliches Sachsen-Anhalt zusammen und entwickeln Lösungen und Strategien für die Transformationsgesellschaft

    Eine retrospektive Betrachtung des Geburtsmodus bei vorangegangener Sectio caesarea

    Einleitung: Die Zielsetzung, ist die Auswertung der retrospektiv erhobenen Daten in Hinblick auf den Entbindungsmodus von Frauen mit Zustand nach einmaliger Sectio und der darauffol- genden Geburt. Sämtliche Faktoren, die im Rahmen dieser Ausarbeitung zu einer erneuten Schnittentbindung oder einer vaginalen Geburt geführt haben, werden aufgeführt und anschlie- ßend analysiert sowie auch das fetale Outcome bei den Geburtsmodi betrachtet und verglichen. Methodik: Die Daten aus dem MEDICO-KIS Programm beziehen sich auf die Entbindungen des Universitätsklinikums Magdeburg (2014-2018). In diesem Zeitraum wurden n=560 Frauen mit einem vorangegangenen Kaiserschnitt detektiert. Einschlusskriterium war der Z.n. einma- liger Sectio. Ausgeschlossen wurden Patientinnen mit einer weiteren Entbindung nach Sectio und Mehrlingsschwangerschaften. Zur Auswertung der Daten wurde das Programm SPSS ver- wendet. Ergebnisse: 49 % der Frauen strebten den TOLAC an. Davon erlebten fast 54 % eine VBAC und 46 % eine sekundäre Re-Sectio. 14 % der VBAC erhielten eine Geburtseinleitung. Prädiktivmarker für die VBAC waren das Alter der Mutter < 35 Jahre und eine vaginale Entbindung vor der Sectio. Erkrankungen, wie Diabetes (p=0.04) oder (Prä-) Eklampsie (p=0.011) führten vermehrt zu einer Sectio. 51 % der Kohorte wurden durch eine primäre Re-Sectio entbunden. Die Anzahl der Wunsch-Re-Sectiones betrug 0,5 %, der Plazenta praevia 1,4 % und der abnormal invasiven Plazenta 1,1 %. Schlussfolgerung: Die Schwangere sollte in der Geburtsplanung individuell betrachtet werden, um die für sie vernünftige und sichere Entbindungsform zu eruieren, indem Faktoren berücksichtigt werden, die ein Risiko und ggf. Misserfolg des TOLAC bedeuten könnten. Bei Z.n. Sectio haben Frauen mit vorheriger vag. Entbindung, maternalem Alter unter 35 Jahren sowie Ausbleiben von Risikofaktoren wie bspw. Diabetes oder HELLP-Syndrom/(Prä-) Eklampsie besonders gute Voraussetzungen für eine VBAC. Ziel sollte sein, die TOLAC-Rate anzuheben und die Re-Sectio-Rate zu senken
