Immunopeptidomics of Salmonella enterica Serovar Typhimurium-Infected Pig Macrophages Genotyped for Class II Molecules

    No full text
Salmonellosis is a zoonotic infection that has a major impact on human health; consuming contaminated pork products is the main source of such infection. Vaccination responses to classic vaccines have been unsatisfactory; that is why peptide subunit-based vaccines represent an excellent alternative. Immunopeptidomics was used in this study as a novel approach for identifying antigens coupled to major histocompatibility complex class II molecules. Three homozygous individuals having three different haplotypes (Lr-0.23, Lr-0.12, and Lr-0.21) were thus selected as donors; peripheral blood macrophages were then obtained and stimulated with Salmonella typhimurium (MOI 1:40). Although similarities were observed regarding peptide length distribution, elution patterns varied between individuals; in total, 1990 unique peptides were identified as follows: 372 for Pig 1 (Lr-0.23), 438 for Pig 2 (Lr.0.12) and 1180 for Pig 3 (Lr.0.21). Thirty-one S. typhimurium unique peptides were identified; most of the identified peptides belonged to outer membrane protein A and chaperonin GroEL. Notably, 87% of the identified bacterial peptides were predicted in silico to be elution ligands. These results encourage further in vivo studies to assess the immunogenicity of the identified peptides, as well as their usefulness as possible protective vaccine candidates.

    No full text
Antimicrobial resistance is a public health problem, since 700,000 people die each year from infections resistant to antibiotics. The World Health Organization (WHO) has warned about multi-resistant bacteria, especially Neisseria gonorrhoeae, since it registers 82 million infections in the world every year, 11 million of them in Latin America. This research was carried out by consulting articles and books published in databases such as Google Scholar, Sciencedirect, Scopus, Clinical Key, Scielo and PubMed. The situation of this microorganism is worrying, which is why it is necessary to study the resistance mechanisms of the pathogen. We found that Neisseria gonorrhoeae presents resistance mediated by plasmids that encode beta-lactamase, which mainly inhibits beta-lactam antibiotics. Additionally, chromosomally mediated resistance occurs due to mutations in genes such as penA, ponA, rpsj, gyrA, which prevent the pharmacological action of antibiotics. There are also resistance mechanisms that prevent the correct functioning of macrolides, cephalosporins, and quinolones. However, new drugs are currently in development, including zoliflodacin, geopotidacin, and solithromycin. In Colombia, Neisseria gonorrhoeae mainly affects Antioquia, Bogotá, and the Amazon according to reports from the National Health Institute in the periods between 2010 and 2022. Neisseria gonorrhoeae has a great capacity to develop complex resistance mechanisms, which represents a great challenge for the treatment of the disease in the future. Therefore, the development of new antibiotics and the repositioning of drugs currently in the market generate broad expectations. It is important to carry out greater and better epidemiological surveillance, providing updated information that guides doctors in the selection of appropriate treatments for adverse reactions to this type of medication.Incluye bibliografíaPregradoTecnólogo(a) en Regencia de Farmaci

The Influence of Playing Position on Physical, Physiological, and Technical Demands in Adult Male Soccer Matches: A Systematic Scoping Review with Evidence Gap Map

    No full text
Background: There has been an increase in studies examining the demands of soccer relative to each playing position in recent years. Understanding the physical, physiological, and technical demands on soccer players according to their positional role during competitive matches is necessary to understand match requirements and develop position-specific training practices. Thereby, there is a clear need to synthesize the information on the different profiles of each playing position. Objective: This review aimed to organize the literature investigating physical, physiological, and technical demands according to playing positions and provide a framework to identify gaps and suggestions for future studies. Methods: A systematic search was conducted in October 2023 using four electronic databases: Web of Science, SPORTDiscus, PubMed and Scopus. The review followed PRISMA 2020 guidelines and the PRISMA-ScR extension for Scoping Reviews. The studies were included if the sample comprised adult male soccer players categorized from Tier 3 to Tier 5 (i.e., highly trained/national level, elite/international level, or world class) and compared the physical, physiological, or technical parameters across playing positions. Results: A total of 178 studies met the inclusion criteria and were included in the review. The number of teams, players, and matches analyzed per study varied considerably. Although a range of classifications were reported across studies, 59% of studies classified players as central defenders, full-backs, central midfielders, wide midfielders, and forwards. The findings suggests that central and external midfielders, and external defenders cover greater total and high-speed distance than forwards or central defenders. Sprint distance was higher in external midfielders versus all other positions. Defenders and central midfielders perform more passes than external midfielders and forwards. Heart rate was the most commonly reported physiological variable across playing positions. When expressed as a percentage of maximal heart rate, midfielders presented higher mean values than all other playing positions. Conclusion: This scoping review demonstrates that there are differences in the demands on players across playing positions in soccer. Training practices in soccer should be based on the specific requirements of each positional role to ensure players can fulfill their tactical responsibilities during the game.Incluye referencias bibliográfica
