    First epidemiological survey of Toxoplasma gondii in Galapagos sea lions (Zalophus wollebaeki)

    International audienceToxoplasma gondii is the protozoan parasite responsible for toxoplasmosis, a zoonosis that represents a health risk for mammals, including marine species. Felines are the only definitive hosts of this parasite, playing a critical role in the introduction and maintenance of the pathogen in a new environment. Recent data demonstrate the contamination by T. gondii of the terrestrial and seawater environment of the Galapagos archipelago, in the Pacific Ocean. Little is known about the exposure of Galapagos’ threatened species to T. gondii, although introduced domestic cats in the archipelago are known to be seropositive for T. gondii. We documented for the first time exposure to T. gondii of Galapagos sea lions (Zalophus wollebaeki), an endemic and emblematic species of the archipelago. The modified agglutination test revealed the presence of antibodies against T. gondii in 61 of 77 plasma samples collected in 2016–2017 from 2- to 4-year-old wild sea lions live-handled in their breeding sites on the inhabited island of San Cristóbal. Antibodies were also detected in 4 of 19 serum samples (21%) from sea lions whose corpses were found in 2021 on the same island. In addition, T. gondii DNA was detected in a lung sample from one necropsied pup and a tissue cyst-like structure was found in another, suggesting infection. These results, together with the high prevalence of antibodies in 2 to 4-year-olds, indicate that Galapagos sea lions are frequently exposed to T. gondii and raise concerns that toxoplasmosis may pose a threat to this endemic species

    19F NMR study of the interaction between an amino acid derived ligand and the transmembrane protein TSPO

    International audienceTranslocator protein (TSPO, 18 kDa), previously known as peripheral-type benzodiazepine receptor, is an evolutionarily conserved transmembrane protein involved in various physiological processes and patho-physiological conditions. The endogeneous TSPO ligand is a polypeptide of 9 kDa, but dipeptides with biological activity have been previously synthesized and characterized. Herein, we synthesized a phenyl alanine derived ligand with a 19F labelling which opens prospective for 19F-MRI and potential 18F-PET applications. We characterized the coexistence of two conformers and performed interaction studies with the recombinant mouse TSPO (mTSPO) in different membrane-mimicking environments using 19F NMR hence enabling structure/function characterizations. A change in the mTSPO environment from pure detergent to lipid/detergent mixture reveals different exchange rates between bound and free ligand forms. Competition experiments with the high-affinity drug ligand (R)-PK 11195 suggest that phenyl alanine derived ligand binds in the same protein cavity.</div

    Comparaison d'une séquence IRM de flux 4D et de la simulation numérique pour l'étude hémodynamique des anévrismes intracrâniens

    International audienceLa séquence IRM de flux 4D permet l’évaluation de la vitesse du sang. Initialement développée pour les grands vaisseaux, son application dans les petits vaisseaux cérébraux pose des défis. L’adaptation de la séquence IRM de flux 4D aux artères cérébrales nécessite une augmentation de la résolution spatiale, entraînant une augmentation du temps d’acquisition, la rendant ainsi cliniquement inapplicable. Nous avons alors développé une méthodologie d’optimisation pour réduire ce temps tout en maintenant la précision des vitesses. Une séquence de référence et plusieurs séquences accélérées ont été acquises, puis comparées voxel à voxel via une régression linéaire permettant de choisir un compromis optimal entre accélération et précision, réduisant le temps d’acquisition d’un facteur 24,6. Une fois la séquence optimisée, nous avons extrait les données de vitesse et la géométrie vasculaire à l’aide du logiciel interne MRProject et les avons utilisées pour alimenter des modélisations numériques de l'écoulement sanguin dans une géométrie réaliste, adaptées aux images IRM. Nous résolvons les équations de Stokes stationnaires pour des écoulements laminaires à l'aide de la méthode des éléments finis, considérant que cette approximation des équations de Navier-Stokes est pertinente pour les petites artères cérébrales.Nous souhaitons évaluer la pertinence de marqueurs hémodynamiques de l'évolution des anévrismes, dilatations locales de la paroi artérielle pouvant entrainer un accident vasculaire cérébral (AVC) et améliorer le suivi des anévrismes par IRM. Par exemple, nous calculons la contrainte de cisaillement pariétale (Wall Shear Stress, WSS) sur les images IRM ainsi que sur les résultats de simulation numérique
