International audienceThe scholarly culture of South India provides tools that help make precise the notion of musical rhythm, as opposed to tempo or measure. It is steeped in the Indian conception of measured time and its key concept of heterometry. There are five notions of poetic rhythm, based on seven structural levels; they are closely related to mathematical innovation, especially in combinatorics. Musical rhythm, on the other hand, requires in addition the interplay of a time unit or tempo, a structured cycle (tāla), the choice of a gait (naṭai) and of stress patterns (jati) within the cycle. After a survey of sources in the light of current musicology, we outline five concepts of rhythm in Sanskrit and Tamil poetry ; one of them gave rise to the Mathematics of combinatorics. The concert aria (kṛti) helps convey to the audience the gradual integration of rhythm and melodic line into a rhythmical harmony (Sambamoorthy). Examples taken from tonal compositions will show how these concepts also shed light on the structure of familiar works, in the spirit of recent works.
Key-enzymes involved in the biosynthesis of resveratrol-based stilbenes in Vitis spp.: a review
International audiencePolyphenols are a large class of plant specialized metabolites with interesting biological properties. Among them, the stilbene phytoalexins are particularly abundant in grapevine gaining considerable applications in the cosmetic and nutraceutical industry. The well-studied stilbene biosynthetic pathway uses phenylalanine to generate resveratrol, the primary stilbene core synthetized in grapevine. A set of enzymes further metabolize resveratrol producing a diversity of stilbenes with different properties, activities and stabilities. In grapevine, 13 enzymes have been identified with the ability to use resveratrol as a substrate, including ten peroxidases, two glycosyltransferases and one O-methyltransferase. Peroxidases and methyltransferases are the most studied enzymes in grapevine, especially under stress conditions while the role of plant laccases and hydroxylases in stilbene metabolism is still poorly described. This article provides an overview of the key enzymes involved in the synthesis of resveratrol-based derivatives in Vitis spp. Perspectives to identify new enzymes are discussed, which could be further used for stilbene bioproduction using biotechnologies
Estimating Rhizosphere Priming in European Agricultural Soils
International audienceThe rhizosphere priming effect (RPE) is a key process where the mineralization of soil organic carbon (SOC) by microorganisms is modified by the presence and activity of plant roots compared to SOC mineralization on bare soil, increasing carbon fluxes from soils to the atmosphere. However, its magnitude in agricultural systems remains uncertain. Moreover, since the RPE is not specifically accounted for in earth system models it is a source of uncertainty in global carbon loss estimates relevant to achieve climate change targets. The PrimeSCale model offers a simple framework to quantify RPE-induced SOC respiration at large spatial scales. Here we aim to estimate the RPE in specific European croplands. The PrimeSCale model estimates the RPE using root carbon input to the soil derived from the combination of MODIS gross primary production (GPP) and net primary production (NPP) data, root depth distribution, heterotrophic respiration, soil bulk density and soil organic carbon content. A central component of PrimeSCale is the RPE ratio, the relative increase in heterotrophic respiration induced by priming based on literature using living plants. Our analysis of the time period 2010-2020 covers six types of croplands (maize, wheat, oat, barley, legumes, and soy) across Europe at a 5 km resolution, down to a depth of 200 cm. The model outputs include the magnitude of the RPE ratio and RPE-induced SOC loss in these croplands and how they vary within Europe according to climate and crops. Our findings will enhance understanding of the processes behind carbon cycling in managed environments and provide insights for carbon-farming policies to better suit mitigation strategies
Landscape fragmentation caused by linear transport infrastructures (LTIs) generally disrupts functional connectivity by limiting animal movements and gene flow, processes that are essential to the persistence of populations. However, the extent of this impact varies depending on the type of infrastructure and the species involved. This study assessed the effects of a highway and a navigation canal on the actual functional connectivity of three ungulate species in the Ardennes region of France. An integrative approach was used, combining landscape genetics, GPS tracking, and camera trap monitoring. Genetic analyses revealed a barrier effect of the highway for red deer (Cervus elaphus), a moderate effect for wild boar (Sus scrofa), and no detectable impact for roe deer (Capreolus capreolus). In contrast, the canal had no significant effect on gene flow for any of the studied species. Camera monitoring of 29 non-wildlife crossing structures showed that they can facilitate movement for a wide range of small- and medium-sized terrestrial mammals, though their effectiveness varies by species. Roe deer use several structures (n=12), unlike red deer and wild boar (n=2). GPS tracking of 14 roe deers and 10 wild boars confirmed that the animals did not avoid the vicinity of the highway, but that it significantly shaped their movement patterns by limiting crossings. This research highlights the importance of using multiple empirical data sources to assess connectivity and more effectively guide conservation actions within a spatial planning policy framework.
C -Branched Iminosugars as Selective Pharmacological Chaperones of Lysosomal α-Glucosidase for the Treatment of Pompe Disease
International audienceWe report herein the design and synthesis of a series of 5-C-alkyl-deoxynojirimycins from L-sorbose, through an effi-cient and scalable method amenable to prepare a large variety of analogues. The interaction of this class of com-pounds with human acid -glucosidase (GAA), the genetically defective enzyme in patients suffering from Pompe disease, was investigated to identify pharmacological chaperones exhibiting high selectivity for this enzyme. Crystallographic analyses provided a rationale for their binding mode to GAA and chaperone activity. The effects of 5-C-phenethyl-DNJ (4c) were evaluated on GAA activity enhancement in cells from Pompe disease patients and in GAA-KO mice. The significant increase of GAA activity in the presence of 4c in various tissues, particularly in the dia-phragm, encourages further studies on this class of small molecules towards developing clinical drugs. Their chaper-one activity and excellent selectivity may offer potential benefits over the current treatments for Pompe disease

Zusammenhang, Verknüpfung and Verkettung: In Defense of Schenker’s Model of Coherence in his Analysis of Brahms’s Variations and Fugue on a Theme by Handel
Within Heinrich Schenker’s prolific theoretical output, the analysis of variations occupies a very small space. Faced with the challenge of a form whose essence is additive, Schenker responds with an analytical attitude that may seem to contradict the foundations of his theory, and which has been criticized for this reason by several theorists since the 1980s: he renounces submitting the totality of form to a single Ursatz. By examining in detail the analysis of Brahms’ Variations and Fugue on a Theme by Handel (1924), it is shown how Schenker developed an exceptional analytical strategy based on a meticulous search for connections between variations. This does not mean that Schenker neglects the specific logic of each variation: this is expressed in an “internal law”, the identification of which is one of the aims of the analysis. Connections between variations, whether they concern contiguous or non-contiguous variations, can be observed in the foreground but also, more often, in the middleground. They concern the motivic dimension of the music, the interplay of registers, or voice leading similarities at the middleground. Schenker thus replaces his usual all-encompassing vision of form controlled by the Ursatz with a model of coherence based on a dense network of mostly “secret” relationships seen as the manifestation of Brahms’s genius.
Detection of Mercury Ions at ng/L Scale by Surface Plasmon Resonance (SPR) on DNA Aptamer Biochips
International audienceMercury pollution is an important environmental problem due to its toxicity to humans, even at very low concentrations. Presently, several methods enable the detection of this element at environmental concentrations. Although sufficiently sensitive in most cases, measurements are typically made punctually. Here, we present the development of a continuous flow detection system for two mercury forms: the mercuric ion (Hg2+ ) and the methylmercury ion (CH3-Hg+). We used a compact SPR device based on gold chips functionalized with tailored aptamers. Leveraging the specific affinity between DNA aptamers and mercuric ions, this reliable technique allowed the detection of mercuric ion concentrations in the ng/L range. The high sensitivity is attributed to the expected DNA folding into a hairpin shape in the presence of mercuric ions. Circular Dichroism (CD) spectroscopy confirmed that a conformational change in the DNA occurs starting from 3 mercuric ions per aptamer. Below this threshold (1 or 2 ions), the strand does not fold, whereas from 3 ions onward, folding is observed. This study highlights the strong potential of compact SPR associated with specific DNA aptamers for the realization of a portable real-time detection device
